3Circumvent PEGylation dilemma by implementing matrix metalloproteinase-responsive chemistry for promoted tumor gene therapy显示文摘Surface modification by poly(ethylene glycol)(PEGylation)has been acknowledged as a powerful strategy in minimizing non-specific reactions for biomedical devices.Once applied into manufacture of drug/gene delivery systems,PEGylation has demonstrated to significantly improve their biocompatibility and stealthiness in physiological environment.Nonetheless,reluctant cell membrane affinities thus cellular uptake efficiencies owing to PEGylation brought up further issues that are imperative to be resolved.Pertain to this PEGylation dilemma,we attempted to introduce peptide(GPLGVRG)linkage between block copolymer of PEG-poly{N’-[N-(2-aminoethyl)-2-aminoethyl]aspartamide}PAsp(DET),wherein the cationic PAsp(DET)could self-assemble with pDNA into nanoscaled complex core.Noteworthy was the peptide linkage whose amino acids sequence could be specifically recognized and degraded by matrix metalloproteinases(MMPs)(overexpressed in extracellular milieu of tumors).Therefore,our subsequent studies validated facile detachment of PEGylation from the aforementioned polyplex micelles upon treatment of MMPs,which elicited improved cytomembrane affinities and cellular uptake efficiencies.In addition,promoted escape from endosome entrapment was also confirmed through direct endosome membrane destabilization by PAsp(DET),which was further elucidated to be attributable to dePEGylation as well as elevated charged density of PAsp(DET)in acidic endosomes.These benefits from dePEGylation eventually contributed to promoted gene expression at the affected cells and potent tumor growth suppression based on anti-angiogenic approach.Therefore,our developed strategy has provided a facile approach in overcoming the dilemma of PEGylation,which could be informative in design of drug/gene delivery systems.Jingyun Wang Hao Wang Hongyan Cui Peng Sun Xi Yang Qixian Chen 2020Chinese Chemical Letters2020,31,12:0
4Regional source contributions to fine particulate matter of less studied cities in Beijing-Tianjin-Hebei region in 2017显示文摘Located in the central area of the North China Plain, the Beijing-Tianjin-Hebei region experiences severe air pollution, especially for Hebei province with five cities among the top 10 cities with the worst air quality nationwide. However, less studies have investigated local and regional contributions to cities in Hebei in comparison to Beijing and Tianjin. In this study, a source-oriented version of the Community Multiscale Air Quality (CMAQ) model is used to quantify inter- and intra-province transport of PM_(2.5) in 11 cities within Hebei in 2017. PM_(2.5) and its components vary seasonally with high levels in winter and low levels in summer. Local contributions to total PM_(2.5) within cities are 23%–61%, while intra-province transport contributes 10%–30%, and inter-province transport contributes 23%–46%, with top contributors of Shandong (4%–22%), Henan (3%–15%) and Shanxi (2%–11%). The primary components in most cities are mainly from local sources (30%–80%), and the secondary components are mainly from inter-province contributions (30%–66%). Local sources have the highest contributions to total PM_(2.5) in Shijiazhuang (∼49%), followed by Shanxi (∼12%) and Xingtai (∼7%). Secondary components from local and intra-province emissions are the major cause of heavy pollution events. 5Promising roles of non-exosomal and exosomal non-coding RNAs in the regulatory mechanism and as diagnostic biomarkers in myocardial infarction显示文摘Non-exosomal non-coding RNAs(non-exo-ncRNAs)and exosomal ncRNAs(exo-ncRNAs)have been associated with the pathological development of myocardial infarction(MI).Accordingly,this analytical review provides an overview of current MI studies on the role of plasma non-exo/exo-ncRNAs.We summarize the features and crucial roles of ncRNAs and reveal their novel biological correlations via bioinformatics analysis.The following contributions are made:(1)we comprehensively describe the expression profile,competing endogenous RNA(ceRNA)network,and“pre-necrotic”biomarkers of non-exo/exo-ncRNAs for MI;(2)functional enrichment analysis indicates that the target genes of ncRNAs are enriched in the regulation of apoptotic signaling pathway and cellular response to chemical stress,etc.;(3)we propose an updated and comprehensive view on the mechanisms,pathophysiology,and biomarker roles of non-exo/exo-ncRNAs in MI,thereby providing a theoretical basis for the clinical management of MI.Jingru LI Haocheng MA Xinyu WU Guihu SUN Ping YANG Yunzhu PENG Qixian WANG Luqiao WANG 2023Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2023,24,4:0
6Activation of pyroptosis by specific organelle-targeting photodynamic therapy to amplify immunogenic cell death for anti-tumor immunotherapy显示文摘Pyroptosis,a unique lytic programmed cell death,inspired tempting implications as potent anti-tumor strategy in pertinent to its potentials in stimulating anti-tumor immunity for eradication of primary tumors and metastasis.Nonetheless,rare therapeutics have been reported to successfully stimulate pyroptosis.In view of the intimate participation of reactive oxygen species(ROS)in stimulating pyroptosis,we attempted to devise a spectrum of well-defined subcellular organelle(including mitochondria,lysosomes and endoplasmic reticulum)-targeting photosensitizers with the aim of precisely localizing ROS(produced from photosensitizers)at the subcellular compartments and explore their potentials in urging pyroptosis and immunogenic cell death(ICD).The subsequent investigations revealed varied degrees of pyroptosis upon photodynamic therapy(PDT)towards cancerous cells,as supported by not only observation of the distinctive morphological and mechanistic characteristics of pyroptosis,but for the first-time explicit validation from comprehensive RNA-Seq analysis.Furthermore,in vivo anti-tumor PDT could exert eradication of the primary tumors,more importantly suppressed the distant tumor and metastatic tumor growth through an abscopal effect,approving the acquirement of specific anti-tumor immunity as a consequence of pyroptosis.Hence,pyroptosis was concluded unprecedently by our proposed organelles-targeting PDT strategy and explicitly delineated with molecular insights into its occurrence and the consequent ICD.Shuang Zeng Chen Chen Liuwei Zhang Xiaosheng Liu Ming Qian Hongyan Cui Jingyun Wang Qixian Chen Xiaojun Peng 2023Bioactive Materials2023,,7:0
