3Electroacupuncture regulates the stress-injury-repair chain of events after cerebral ischemia/reperfusion injury显示文摘Inflammation after stroke is the main cause of cerebral ischemia/reperfusion injury. Cascading events after injury can lead to cell death. Heat shock protein 70 and other endogenous injury-signaling molecules are released by damaged cells, which can lead to systemic stress reactions. Protecting the brain through repair begins with the stress-injury-repair signaling chain. This study aimed to verify whether acupuncture acts through this chain to facilitate effective treatment of ischemic stroke. Rat models of cerebral ischemia/reperfusion injury were established by Zea Longa's method, and injury sites were identified by assessing neurological function, 2,3,5-triphenyltetrazolium chloride staining, and hematoxylin-eosin staining. Electroacupuncture at acupoints Baihui(DU20) and Zusanli(ST36) was performed in the model rats with dilatational waves, delivered for 20 minutes a day at 2–100 Hz and an amplitude of 2 m A. We analyzed the blood serum from the rats and found that inflammatory cytokines affected the levels of adrenotrophin and heat shock protein 70, each of which followed a similar bimodal curve. Specifically, electroacupuncture lowered the peak levels of adrenocorticotrophic hormone and heat shock protein 70. 4CRISPR/Cas9-mediated PINK1 deletion leads to neurodegeneration in rhesus monkeys显示文摘Dear Editor,PINK1 mutations cause autosomal recessive and early-onset Parkinson’s disease(PD)with selective neurodegeneration.Unfortunately,current PINK1 knockout(KO)mouse1,2 and pig models3,4 are unable to recapitulate the selective and overt neurodegeneration seen in PD patients.Furthermore,endogenous Pink1 in the mouse brain is expressed at very low levels and can only be detected via immunoprecipitation,5 meaning that PINK1’s function in the mammalian brain needs to be assessed using larger animals that are closer to humans.We previously used CRISPR/Cas9 to target the monkey gene in one-cell stage embryos.6 Using the same approach,we designed two gRNAs to target exon 2(T1)and exon 4(T2),which encode a kinase domain in the PINK1 gene of rhesus monkeys(Fig.1a).CRISPR/Cas9 and PINK1 gRNAs were injected into one-cell stage rhesus monkey embryos.A T7E1 assay and sequencing of PCR products from the injected embryos showed high efficiency(61.5%)in targeting PINK1(Fig.1b and Supplementary information,Fig.S1).Transfer of 87 embryos to 28 surrogate rhesus monkeys resulted in 11 pregnancies(39.2%)(Fig.1b).Eleven fetuses developed to term and were born naturally.Of these live monkeys,eight carried PINK1 mutations(M),and three were wild type(WT).However,three mutant monkeys(M1,M3 and M4)were newborn triplets that struggled to survive and died 3-4 days after birth.One WT newborn monkey also died after a difficult labor.Another mutant monkey(M2)died 7 days after birth without noticeable warning signs or symptoms.The other three mutant monkeys(M6,M7 and M8)have lived for three years;M5,however,reduced its food intake and showed weakness at the age of 1.5 years,and died 30 days after anesthesia for MRI examination.Weili Yang Yunbo Liu Zhuchi Tu Chong Xiao Sen Yan Xishan Ma Xiangyu Guo Xiusheng Chen Peng Yin Zhengyi Yang Su Yang Tianzi Jiang Shihua Li Chuan Qin Xiao-Jiang Li 2019Cell Research2019,29,4:12
5First mass measurement of short-lived nuclides at HIRFL-CSR显示文摘Recent commissioning of the HIRFL-CSR has demonstrated its ability to perform direct mass measurement for short-lived nuclides. Projectile fragments produced by the 78Kr ions at 481.88 MeV/u were separated with the new radioactive beam line in Lanzhou (RIBLL2), and injected into and stored in the experimental storage ring (CSRe). By operating the CSRe as an isochronous mass spectrometry, a typical mass resolution around 2.0×10?5 has been achieved. The masses for 63Ge, 65As and 67Se were measured for the first time. The measured masses are compared with theoretical predictions and the location of the proton drip-line for As isotopes is discussed. 6A Fault Diagnosis Method of Power Systems Based on an Improved Adaptive Fuzzy Spiking Neural P Systems and PSO Algorithms显示文摘A new fault diagnosis method based on improved Adaptive fuzzy spiking neural P systems(in short,AFSN P systems) and Particle swarm optimization(PSO)algorithm is presented to improve the efficiency and accuracy of diagnosis for power systems in this paper. AFSN P systems are a novel kind of computing models with parallel computing and learning ability. Based on our previous works, this paper focuses on AFSN P systems inference algorithms and learning algorithms and builds the fault diagnosis model using improved AFSN P systems for diagnosing effectively. The process of diagnosis based on AFSN P systems is expressed by matrix successfully to improve the rate of diagnosis eminently. Furthermore, particle swarm optimization algorithm is introduced into the learning algorithm of AFSN P systems, thus the convergence speed of diagnosis has a big progress. An example of 4-node system is given to verify the effectiveness of this method. 71~H NMR-based serum metabolic profiling in compensatedand decompensated cirrhosis显示文摘AIM: To study the metabolic profiling of serum samples from compensated and decompensated cirrhosis patients. METHODS: A pilot metabolic profiling study was conducted using three groups: compensated cirrhosis patients (n = 30), decompensated cirrhosis patients (n = 30) and healthy controls (n = 30). A 1H nuclear magnetic resonance (NMR)-based metabonomics approach was used to obtain the serum metabolic profiles of the samples. The acquired data were processed by multivariate principal component analysis and orthogonal partial least-squares discriminant analysis (OPLS-DA). RESULTS: The OPLS-DA model was capable of distinguishing between decompensated and compensated cirrhosis patients, with an R2Y of 0.784 and a Q2Y of 0.598. Twelve metabolites, such as pyruvate, phenylala-nine and succinate, were identified as the most influential factors for the difference between the two groups. The validation of the diagnosis prediction showed that the accuracy of the OPLS-DA model was 85% (17/20). 9Efficient microwave-assisted photocatalytic degradation of endocrine disruptor dimethyl phthalate over composite catalyst ZrO_x /ZnO显示文摘A highly active ZrO x /ZnO catalyst for microwave-assisted photocatalytic (MW/PC) degradation of endocrine disruptor dimethyl phthalate (DMP) has been prepared via cetyltrimethylammonium assisted hydrothermal method. The ZrO x /ZnO was characterized by XPS, XRD, UV-Vis, BET and SEM techniques. The XPS result showed that Zr oxides with different valences (+2, +3, +4) co-existed in ZrO x /ZnO. By using the ZrO x /ZnO (0.1 g), the TOC removal efficiency of DMP (100 mL of 50 mg/L) was 88% after 30 min reaction, which was about 15% higher than P25 TiO 2 . It was found that the removal process of DMP by MW/PC followed pseudo first-order kinetics in all cases, and ZrO x /ZnO significantly accelerated the degradation of DMP. The degradation half-life time of DMP was shortened 45% compared with P25 TiO 2 . A possible catalytic mechanism was proposed based on microwave response and interfacial charge transfer. ZrO x /ZnO could be reused for six times without obvious decrease in catalytic activity. 10Effects of cyclooxygenase-2 on human esophageal squamous cell carcinoma显示文摘AIM:To study the relationship between the cyclooxy-genase(COX)-2 gene and the proliferation and apopto-sis of esophageal squamous carcinoma EC109 cells.METHODS:The techniques of RNA interference(RNAi)and cell transfection,as well as the levels of oncogenic-ity in nude mice,were used to study the role of COX-2 in the esophageal squamous carcinoma cell(ESCC)line EC109.Following RNAi and transfection,Western blot-ting analysis was used to determine the expression of the COX-2 protein.The 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-tetrazolium bromide(MTT)reduction assay was used to evaluate cell growth,and flow cytometry was used to detect cell apoptosis.RESULTS:Western blotting analysis demonstrated that COX-2 expression was significantly reduced in EC109 cells treated with COX-2-specif ic short interfering RNA(siRNA)but was increased in EC109 cells transfected with COX-2.Furthermore,COX-2 siRNA treatment in-hibited cell proliferation(P < 0.01)and induced apop-tosis in EC109 cells,as determined by an MTT assay and by flow cytometry,respectively.In contrast,trans-fected COX-2 led to increased cell proliferation(P < 0.05)and decreased apoptosis in EC109 cells.In addition,combination treatment of cells with COX-2 siRNA and aspirin had a synergistic effect(P < 0.01).For experi-ments measuring tumorigenicity,xenograft tumors of a greater volume and weight were found in the COX-2 group compared with other groups(P < 0.05).A large dose of aspirin inhibited tumor growth in nude mice ef-fectively(P < 0.05),and the rate of tumor suppression was 51.8% in the high-dose aspirin group.CONCLUSION:COX-2 plays a very critical role in ESCC carcinogenesis,and COX-2 siRNA combined with aspirin has the potential to be an anticancer therapy for the treatment of ESCC.Li Zhang Yong-Dong Wu Peng Li Jun Tu Ying-Lin Niu Cai-Min Xu Shu-Tian Zhang 2011World Journal of Gastroenterology2011,17,41:9
11Endothelium-Mimicking Multifunctional Coating Modified Cardiovascular Stents via a Stepwise Metal-Catechol-(Amine)Surface Engineering Strategy显示文摘Stenting is currently the major therapeutic treatment for cardiovascular diseases.However,the nonbiogenic metal stents are inclined to trigger a cascade of cellular and molecular events including inflammatory response,thrombogenic reactions,smooth muscle cell hyperproliferation accompanied by the delayed arterial healing,and poor reendothelialization,thus leading to restenosis along with late stent thrombosis.To address prevalence critical problems,we present an endothelium-mimicking coating capable of rapid regeneration of a competently functioning new endothelial layer on stents through a stepwise metal(copper)-catechol-(amine)(MCA)surface chemistry strategy,leading to combinatorial endothelium-like functions with glutathione peroxidase-like catalytic activity and surface heparinization.Apart from the stable nitric oxide(NO)generating rate at the physiological level(2:2×10^(-10) mol/cm^(2)/min lasting for 60 days),this proposed strategy could also generate abundant amine groups for allowing a high heparin conjugation efficacy up to∼1μg/cm^(2),which is considerably higher than most of the conventional heparinized surfaces.The resultant coating could create an ideal microenvironment for bringing in enhanced antithrombogenicity,anti-inflammation,anti-proliferation of smooth muscle cells,re-endothelialization by regulating relevant gene expressions,hence preventing restenosis in vivo.We envision that the stepwise MCA coating strategy would facilitate the surface endothelium-mimicking engineering of vascular stents and be therefore helpful in the clinic to reduce complications associated with stenosis.Ying Yang Peng Gao Juan Wang Qiufen Tu Long Bai Kaiqin Xiong Hua Qiu Xin Zhao Manfred FMaitz Huaiyu Wang Xiangyang Li Qiang Zhao Yin Xiao Nan Huang Zhilu Yang 2020Research2020,,1:8
12OsSPL18 controls grain weight and grain number in rice显示文摘Grain weight and grain number are two important traits directly determining grain yield in rice. To date,a lot of genes related to grain weight and grain number have been identified; however, the regulatory mechanism underlying these genes remains largely unknown. In this study, we studied the biological function of OsSPL18 during grain and panicle development in rice. Knockout (KO) mutants of OsSPL18exhibited reduced grain width and thickness, panicle length and grain number, but increased tiller number. Cytological analysis showed that OsSPL18 regulates the development of spikelet hulls by affecting cell proliferation. qRT-PCR and GUS staining analyses showed that OsSPL18 was highly expressed in developing young panicles and young spikelet hulls, in agreement with its function in regulating grain and panicle development. Transcriptional activation experiments indicated that OsSPL18is a functional transcription factor with activation domains in both the N-terminus and C-terminus, and both activation domains are indispensable for its biological functions. Quantitative expression analysis showed that DEP1, a major grain number regulator, was significantly down-regulated in OsSPL18 KO lines.Both yeast one-hybrid and dual-luciferase (LUC) assays showed that OsSPL18 could bind to the DEP1promoter, suggesting that OsSPL18 regulates panicle development by positively regulating the expression of DEP1. Sequence analysis showed that OsSPL18 contains the OsmiR156k complementary sequence in the third exon; 5?RLM-RACE experiments indicated that OsSPL18 could be cleaved by OsmiR156k. 13Removal of cyanide adsorbed on pyrite by H_2O_2 oxidation under alkaline conditions显示文摘Large amounts of cyanide tailings are produced during the cyanidation process in gold extraction,which are hazardous solid wastes due to the toxic cyanide.Pyrite is one of the main minerals in cyanide tailings.The removal of cyanide adsorbed on pyrite by H_2O_2 oxidation under alkaline conditions was investigated in this study.It was found that the removal efficiency was positively correlated with pH from 5 to 12,but remained almost constant when pH was higher than 12.The highest cyanide removal efficiency of 91.10% was achieved by adding no less than 0.6 wt.% of H_2O_2.Cyanide removal was positively correlated with the CN^-adsorption amount between 1.06 and 8.5 mg/g,and temperature between 25 and 85°C.The removal of cyanide adsorbed on pyrite by H_2O_2 oxidation under alkaline conditions was due to the oxidation of pyrite.Hexacyanoferrate,thiocyanate and sulfate were generated with mole ratios of about 2.03:1.12:3.17 during the cyanide removal.?2018 The Research Center for Eco-Environmental Sciences,Chinese Academy of Sciences.Yubo Tu Peiwei Han Lianqi Wei Xiaomeng Zhang Bo Yu Peng Qian Shufeng Ye 2019Journal of Environmental Sciences2019,31,4:8

16An analytic study on the deflection of subway tunnel due to adjacent excavation of foundation pit显示文摘Predicting and estimating the response of subway tunnel to adjacent excavation of foundation pit is a research focus in the field of underground engineering.Based on the principle of two-stage method and incremental method, an analytic approach is suggested in this paper to solve this problem in an accurate and rapid way,and the upheavals of tunnel due to adjacent excavation are solved by analytic method. Besides, the presented method is used in the practical engineering case of Shenzhen Metro Line 11 and verified by numerical simulation and in situ measurement. Finally, a parametric analysis is performed to investigate the influence of different factors on tunnel's deflection. Some useful conclusions have been drawn from the research as below: The deflection results of tunnel obtained from analytic method are nearly consistent with the results getting from numerical analysis and measured data, which verified the accuracy and rationality of presented method. The excavation size has a significant impact on both the displacement values and influenced range of tunnel. However, the relative distance only impacts the displacement values of tunnel, but not the influenced range of tunnel. 17Atomic Co/Ni dual sites with N/P-coordination as bifunctional oxygen electrocatalyst for rechargeable zinc-air batteries显示文摘Metal-nitrogen-carbon(M-N-C)single-atom catalysts exhibit desirable electrochemical catalytic properties.However,the replacement of N atoms by heteroatoms(B,P,S,etc.)has been regarded as a useful method for regulating the coordination environment.The structure engineered M-N-C sites via doping heteroatoms play an important role to the adsorption and activation of the oxygen intermediate.Herein,we develop an efficient strategy to construct dual atomic site catalysts via the formation of a Co_(1)-PN and Ni1-PN planar configuration.The developed Co_(1)-PNC/Ni1-PNC catalyst exhibits excellent bifunctional electrocatalytic performance in alkaline solution.Both experimental and theoretical results demonstrated that the N/P coordinated Co/Ni sites moderately reduced the binding interaction of oxygen intermediates.The Co_(1)-PNC/Ni1-PNC endows a rechargeable Zn-air battery with excellent power density and cycling stability as an air-cathode,which is superior to that of the benchmark Pt/C+IrO_(2).This work paves an avenue for design of dual single-atomic sites and regulation of the atomic configuration on carbon-based materials to achieve high-performance electrocatalysts.Botao Hu Aijian Huang Xuejiang Zhang Zheng Chen Renyong Tu Wei Zhu Zhongbin Zhuang Chen Chen Qing Peng Yadong Li 2021Nano Research2021,14,10:6
18Patterns of change in the population and spatial distribution of oriental white storks (Ciconia boyciana) wintering in Poyang Lake显示文摘Using total counts in simultaneous annual surveys,we monitored the population size and spatial distribution of oriental white storks(Ciconia boyciana) wintering in Poyang Lake between 1998 and 2011.Results showed that Poyang Lake wetland is an important wintering ground for oriental white storks,with an annual average population number of 2 305±326.The population sizes in 2004,2005,2010,and 2011 were higher than the highest-ever estimate of its global population.In 2005,we recorded 3 789 individuals,which was the maximum population number within the period of 1998–2011.The storks inhabited 52 lakes,with the greatest distance between these lakes being 180.3 km.The storks presented a clustered distribution pattern in the Poyang Lake wetland,irrespective of the number of individuals or occurrence frequencies.Shahu,Dahuchi,Banghu,and Hanchihu were most frequently used lakes and had the largest annual average numbers of storks.There was a significant positive correlation between occurrence frequency and annual average number of storks in the lakes.Most of the lakes important for storks were covered by existing nature reserves,though some lakes outside the reserves were also frequently used.About 64.9%± 5.5% of the storks were found in nature reserves.In addition,the storks more frequently used and clumped in significantly larger flocks in lakes within nature reserves than lakes outside.Zhen-Hua WEI Yan-Kuo LI Peng XU Fa-Wen QIAN Ji-Hong SHAN Xiao-Bin TU 2016Zoological Research2016,37,6:6
