| 1 | Establishment of cell clones with different metastatic potential from the metastatic hepatocellular carcinoma cell line MHCC97显示文摘ALM To establish clone cells with different metastatic potential for the study of metastasis-related mechanisms. METHODS Cloning procedure was performed on parental hepatocellular carcinoma (HCC) cell line MHCC97. andbiological characteristics of the target clones selected by in vivo screening were studied.``RESULTS Two clones with high MHCC97-H and IowMHCC9--L1 metastatic potential were isolated from theparent cell line. Compared with MHCC97-L. MHCC97-H hadsmaller cell size average cell diameter 43 um vs 50 μmand faster in vitro and in vivo growth rate tumor celldoubling time was 34.2 h vs 60.0 h. The main ranges ofchromosomes were 5.5 58 in MHCC97-H and 57 62 inMHCC97-L. | 2 | New proofs of the recent climate warming over the Tibetan Plateau as a result of the increasing greenhouse gases emissions显示文摘A striking climate warming over the Ti- betan Plateau during the last decades has been re- vealed by many studies, but evidence linking it to human activity is insufficient. By using historical ob- servations, here we show that the in situ climate warming is accompanied by a distinct decreasing trend of the diurnal range of surface air temperature. The ERA40 reanalysis further indicates that there seems to be a coherent warming trend near the tro- popause but a cooling trend in the lower stratosphere. Moreover, all these features can be reproduced in two coupled climate models forced by observed CO2 concentration of the 20th century but cannot be pro- duced by the fixed external conditions before the industrial revolution. | 4 | Effects of the tyrosine protein kinase inhibitor genistein on the proliferation,activation of cultured rat hepatic stellate cells显示文摘AIM: Hepatic stellate cell (HSC) plays a pivotal role inliver fibrosis and is considered as the therapeutic targetfor the treatment of hepatic fibrosis. Tyrosine proteinkinase plays an important role in the proliferation,activation of HSC. The purpose of the study is toinvestigate the effects of the tyrosine protein kinaseinhibitor genistein onthe proliferation and activationof cultured rat HscMETHODS: Rat HSC were isolated from Wistar rats by insitu perfusion of collagenase and pronase and single-stepdensity Nycodenz gradient. Culture-activated HSC wereserum-starved and incubated with 10-3 to 10-5 mol/Lconcentration of genistein for 24, 48 or 72 h. In PDGF-induced HSC proliferation, HSC were stimulatel with L0μg@L-1PDGF-BB for 15 min, and then treaeed with genisteinfor the same time. Cell proliferation was measured byMTr assay and based on flow cytometric analysis of cellcycle. The a-smooth muscle actin (α-SMA) expression inHSC was studied with confocal laser microscopy and flowcytometry. c-fos c-jun and cyclin D1 expression in HSCwas also detected by flow cytometry.RESULTS: Genistein inhibited basal and PDGF-inducedproliferation of HSC at the concentration of 10-8 to 10-5mol/L, and treatment with 10-7 mol/L concentration ofgenistein for 48 h inhibited the HSC proliferationsignificantly (the inhibition rate was 70.3 %, P<0.05).Immunofluorescence detected by confocal lasermicroscopy and flow cytometry showed that treatmentwith 10-7 mol/L genistein for 48 h suppressed theexpression of α-SMA significantly in HSC (the specificfluorescence intensity were 60.2±21.5 vs 35.3±11.6and 12.8± 10.4 vs9.54±6.39, respectively, both P<0.05).The intensity of c-fos, c-jun and cydin D1 expression ofHSCs treated with 10-7 mol/L genistein for 48 h wasalso significantly decreased compared with the controls.CONCLUSION: Genistein influences proliferation of HSC,suppresses the expression of α-SMA in HSC and tinhibits the intensity of c-fos, c-jun and cyclin D1expression of HSCs. | 5 | Metastatic human hepatocellular carcinoma models in nude mice and cell line with metastatic potential显示文摘Metastatic human HCC model is needed for the studies on mechanism and intervention of metastatic recurrence. By using orthotopic implantation of histologically intact tissues of 30 surgical specimens, a patient like metastatic model of human HCC in nude mice (LCI-D20)and a Iow metastatic model of human HCC in nude mice LCI-D35 ) have been established. All mice with transplanted LCI-D20 tumors exhibited extremely high metastatic ability including spontaneous metastasis to liver, lungs, lymph nodes and peritoneal seeding.Remarkable difference was also found in expression of some of the invasiveness related genes and growth factors between the LCI-D20 and LCI-D35 tumors. PAI-Iincreased gradually following tumor progression in LCID20 model, and correlated with tumor size and AFP level,Phasic expression of tissue intercellular adhesion molecule-I in this model was also observed. Using corneal micropocket model, it was demonstrated that the vascular response induced by LCI-D20 tumor was stronger than that induced by LCI-D35 tumor. Similar report on metastatic human HCC model in nude mice and human HCC cell line with metastatic potential was rarely found in the literature. | 9 | Rapeseed research and production in China显示文摘Rapeseed(Brassica napus L.) is the largest oilseed crop in China and accounts for about 20% of world production.For the last 10 years,the production,planting area,and yield of rapeseed have been stable,with improvement of seed quality and especially seed oil content.China is among the leading countries in rapeseed genomic research internationally,having jointly with other countries accomplished the whole genome sequencing of rapeseed and its two parental species,Brassica oleracea and Brassica rapa.Progress on functional genomics including the identification of QTL governing important agronomic traits such as yield,seed oil content,fertility regulation,disease and insect resistance,abiotic stress,nutrition use efficiency,and pod shattering resistance has been achieved.As a consequence,molecular markers have been developed and used in breeding programs.During 2005–2014,215 rapeseed varieties were registered nationally,including 210 winter-and 5 spring-type varieties.Mechanization across the whole process of rapeseed production was investigated and operating instructions for all relevant techniques were published.Modern techniques for rapeseed field management such as high-density planting,controlled-release fertilizer,and biocontrol of disease and pests combined with precision tools such as drones have been developed and are being adopted in China.With the application of advanced breeding and production technologies,in the near future,the oil yield and quality of rapeseed varieties will be greatly increased,and more varieties with desirable traits,especially early maturation,high yield,high resistance to biotic and abiotic stress,and suitability for mechanized harvesting will be developed.Application of modern technologies on the mechanized management of rapeseed will greatly increase grower profit. | Qiong Hu Wei Hua Yan Yin Xuekun Zhang Lijiang Liu Jiaqin Shi Yongguo Zhao Lu Qin Chang Chen Hanzhong Wang | 2017 | The Crop Journal2017,5,2: | 25 |
| 10 | Chaihu-Shugan-San exerts an antidepressive effect by downregulating miR-124 and releasing inhibition of the MAPK14 and Gria3 signaling pathways显示文摘Dysregulation of mi R-124 has been reported to be involved in the pathophysiology of depression. Chaihu-Shugan-San, a traditional Chinese medicine, has antidepressive activity; however, the underlying mechanisms remain unclear. In this study, to generate a rodent model of depression, rats were subjected to a combination of solitary confinement and chronic unpredictable mild stress for 28 days. Rats were intragastrically administered Chaihu-Shugan-San(2.835 m L/kg/d) for 4 weeks, once a day. Real-time reverse-transcription quantitative polymerase chain reaction, mi RNA microarray, western blot assay and transmission electron microscopy demonstrated that ChaihuShugan-San downregulated mi R-124 expression and upregulated the m RNA and protein levels of mitogen-activated protein kinase 14(MAPK14) and glutamate receptor subunit 3(Gria3). Chaihu-Shugan-San also promoted synapse formation in the hippocampus. The open field test, sucrose consumption test and forced swimming test were used to assess depression-like behavior. After intragastric administration of Chaihu-Shugan-San, sucrose consumption increased, while the depressive behaviors were substantially reduced. | 12 | Incidence and mortality of lung cancer in China, 2008-2012显示文摘Objective: To analyze the incidence and mortality rates of lung cancer in China from 2008 to 2012.Methods: Incident and death cases of lung cancer were retrieved from the National Central Cancer Registry(NCCR) database collecting from 135 cancer registries in China during 2008-2012.The crude incidence and mortality rates of lung cancer were calculated by area(urban/rural),region(eastern,middle,western),gender and age group(0,1-4,5-9,…,85+).China census in 2000 and Segi's world population were applied for agestandardized rates.JoinPoint(Version 4.5.0.1) model was used for time trend analysis.Results: The crude incidence rate of lung cancer was 54.66/100,000 which ranked the first in overall cancers.The age-standardized incidence rates by China population(ASIRC) and by World population(ASIRW) were35.13/100,000 and 34.86/100,000,respectively.The crude mortality of lung cancer in China was 45.60/100,000 and it was the first cause of cancer-related death in overall cancers.The age-standardized mortality rates by Chinese standard population(ASMRC) and by world standard population(ASMRW) were 28.57/100,000 and 28.22/100,000,respectively.Incidence and mortality rates of lung cancer were higher in males than in females and higher in urban areas than in rural areas.Eastern areas had the highest incidence and mortality rates followed by middle and western areas.Incidence and mortality rates of lung cancer retained low level in age groups before 40 years old but increased greatly after and peaked in age group of 80-84.During 2003-2012,the temporal trend of the incidence rate of lung cancer in both sexes in China was general stable(P<0.05).The lung cancer incidence rate increased by 0.71% per year in females(P<0.05) and 2.26% per year in rural areas(P<0.05).The mortality rate of lung cancer decreased slightly annually during 2003-2012 in China(P>0.05).In urban areas,it declined by 0.76%per year(P<0.05),but rose by 2.09% per year(P<0.05) in rural areas.Conclusions: Appropriate targeted prevention,early detection and treatment programs should be carried out to control the local burden of lung cancer. | Shuzheng Liu Qiong Chen Lanwei Guo Xiaoqin Cao Xibin Sun Wanqing Chen Jie He | 2018 | Chinese Journal of Cancer Research2018,30,6: | 19 |

| 15 | Evaluation of novel decellularizing corneal stroma for cornea tissue engineering applications显示文摘·AIM:To develop a new decellularization method depended upon the natural corneal structure and to harvest an ideal scaffold with good biocompatibilities for corneal reconstruction.METHODS:The acellular cornea matrix (ACM) were prepared from de-epithelium fresh porcine corneas (DFPCs) by incubation with 100% fresh human sera and additional electrophoresis at 4℃. Human corneal epithelial cells (HCEs) were used for the cytotoxicity tests of ACM. ACM were implanted into the Enhanced Green Fluorecence Protein (eGFP) transgenic mouse anterior chamber for evaluation of histocompatibility.RESULTS:HE and GSIB4 results showed fresh porcine cornea matrix with 100% human sera and electrophoresis could entirely decellularize stromal cell without reducing its transparency. ACM has no cytotoxic effect ex vivo. | 17 | Chemical and optical properties of aerosols and their interrelationship in winter in the megacity Shanghai of China显示文摘A field campaign on air quality was carried out in Shanghai in winter of 2012. The concentrations of NO, NO2, NOx, SO2, CO, and PM2.5increased during haze formation. The average masses of SO42-, NO3-and NH4+were 10.3, 11.7 and 6.7 μg/m3 during the haze episodes, which exceeded the average(9.2, 7.9, and 3.4 μg/m3) of these components in the non-haze days. The mean values for the aerosol scattering coefficient(b sp), aerosol absorption coefficient(b ap) and single scattering albedo(SSA) were 288.7, 27.7 and0.91 Mm-1, respectively. A bi-peak distribution was observed for the mass concentrations of CO, NO, NO2, and NOx. More sulfate was produced during daytime than that in the evening due to photochemical reactions. The mass concentration of NH4+achieved a small peak at noontime. NO3-showed lower concentrations in the afternoon and higher concentrations in the early morning. There were obvious bi-peak diurnal patterns for b sp and b ap as well as SSA. b sp and b ap showed a positive correlation with PM2.5mass concentration.(NH4)2SO4, NH4NO3, organic mass, elemental carbon and coarse mass accounted for 21.7%, 19.3%, 31.0%, 9.3% and 12.3% of the total extinction coefficient during non-haze days, and 25.6%, 24.3%, 30.1%, 8.1% and 8.2% during hazy days. Organic matter was the largest contributor to light extinction. | 20 | Investigations on Nitrate Pollution of Soil, Groundwater and Vegetable from Three Typical Farmlands in Beijing Region, China显示文摘The aim of this study was to determine the nitrate pollution status of soil, groundwater, and vegetable from three typical farmlands (croplands, vegetable fields, and orchards) in Beijing region. During the investigation, hundreds of the soil, groundwater, and vegetable samples from three typical farmlands were collected and analyzed. In addition, attributes of all samples were recorded for data analysis. The results showed that nitrate was substantially accumulated in soil profiles, while the soil nitrate concentrations of vegetable fields and orchards were higher than those of croplands. Nitrate concentration in 0-30 cm soil of vegetable field and orchard were 3.8 and 1.2 times of that of cropland, respectively. Nitrate content of groundwater in vegetable field was 13.8 mg L-1 (with the over-standard ratio 44.8%), which was 2.8 folds of that in cropland. Nitrate concentration of groundwater under orchard was 9.3 mg L-1 (with the over-standard ratio 23.5%), which was 1.9 folds of that in cropland. High concentrations of the nitrate in vegetables were detected, particularly green leafy vegetables ranked first with 2 685.5 mg kg-1, followed by rhizome vegetables, cabbages, and fruit vegetables. The nitrate over-standard ratios of rhizome vegetables, green leafy vegetables, fruit vegetables, and cabbages were 80.9, 37.9, 29.7, and 2.2%, respectively.
