| 1 | Gambogic acid induces mitochondria-dependent apoptosis by modulation of Bcl-2 and Bax in mantle cell lymphoma JeKo-1 cells显示文摘Aim: To study the mechanisms in gambogic acid (GA) -induced JeKo-1 human Mantle Cell Lymphoma cell apoptosis in vitro. Methods: The proliferation of GA-treated JeKo-1 cells was measured by CCK-8 assay and Ki-67 immunocytochemical detection. Apoptosis, cell cycle and mitochondrial membrane potential were measured by flow cytometric analysis. Caspase-3,-8 and -9 were detected by colorimetric assay. Bcl-2 and Bax were analyzed by Western blotting. Results: GA inhibited cell growth in a time-and dose-dependent manner. GA induces apoptosis in JeKo-1 cells but not in normal bone marrow cells, which was involved in reducing the membrane potential of mitochondria, activating caspases-3, -8 and -9 and decreasing the ratio of Bcl-2 and Bax without cell cycle arresting. | 4 | Control mechanism for the upper airway collapse in patients with obstructive sleep apnea syndrome: a finite element study显示文摘Obstructive sleep apnea syndrome (OSAS) is characterized by recurrent collapses of the upper airway, which lead to repetitive transient hypoxia, arousals and finally sleep fragmentation. Both anatomical and neuromuscular factors may play key roles in the pathophysiology of OSAS. The purpose of this paper was to study the control mechanism of OSAS from the mechanical point of view. A three-dimensional finite element model was developed, which not only reconstructed the realistic anatomical structure of the human upper airway, but also included surrounding structures such as the skull, neck, hyoid, cartilage and soft tissues. The respiration process during the normal and apnea states was simulated with the fluid-structure interaction method (FSI) and the computational fluid dynamics method (CFD). The airflow and deformation of the upper airway obtained from the FSI and the CFD method were compared and the results obtained under large negative pressure during an apnea episode were analyzed. The simulation results show that the FSI method is more feasible and effective than the CFD method. | 5 | Resource and Environmental Quality Changes and Adjustment Principles for Sustainable Development in Rapidly Developing Coastal Region of Southeastern China显示文摘Problems in water-land resources and environmental quality increase with the fast economic development in the coastal region of southeastern China. This region has the highest density in population, industry and towns in China, and the large export-oriented economy makes itself a strategic importance while China enters into international markets. The problems facing to this region include arable land reduction, land degradation aggravation, high percentage of built-up land with high rate of expansion, expanded non-point pollution, deteriorated water quality, decreasing biodiversity, destroyed ecosystems, severe air pollution, frequently occurred acid rain, and multi-pollutants. The important research fields in the near future should include a) influence of high-intensity exploitation of resources on changes of environment quality and its feedback; b) interface processes, key mechanisms and adjustment principles for degradation of water-, land-, and air-resources and environmental quality; c) evolvement processes and nurturing theory of biodiversity and ecological resources; and d) spatio-temporal variation and human-induced effects on regional resources and environment quality. | 6 | Evaluation of the Customer Satisfaction Index for Wellhead Blowout Preventers of China’s Petroleum Industry——A new method based on PLS显示文摘This paper establishes an evaluation model of the customer satisfaction index for the wellhead blowout preventers of China’s petroleum industry based on evaluation models of the customer satisfaction index at home and aboard, and by considering the consuming situation in China and the features of the China’s petroleum industry. For the existence of: (1) multiple correlations among the factors in the model; (2) the variables need to be explained, but that are hard to observe; (3) the customer satisfaction degree of observation variables appears the shape of skewness or two or three peaks, the correlations between the satisfaction index and its factors cannot be described by common multiple regression. This paper uses a partial least squares (PLS) method based on principal components and typical correlative analysis to solve the problem. When PLS is used in the model of the customer satisfaction index of the wellhead blowout preventers, the latent variables and the explanation degree coefficient of the manifest variable to the corresponding latent variables are estimated by PLS path analysis, and the influencing coefficient among the latent variables in the model is estimated by PLS regression analysis. | 7 | Nitrogen Mineralization of Prunings of Six N_2-Fixing Hedgerow Species in a Dry Valley of the Jinsha River显示文摘A litterbag experiment of 12 weeks was conducted to study nitrogen mineralization process of prunings of six nitrogen-fixing hedgerow species in a dry valley of the Jinsha River. Prunings were incorporated into soil or used as mulch. The results indicated that pruning N of the six hedgerow species was mineralized fast in the first week and then decreased slowly in the rest of the study period. When prunings were incorporated into soil, the amount of nitrogen mineralized by the end of the first week accounted for 69.9%, 58.2%, 54.5%, 43.0%, 29.6% and 20.6% of the total N in prunings of Desmodium rensonii, Tephrosia Candida, Leucaena leucocephala, Albizia yunnanensis, Acacia dealbata, and Acacia mearnsii, respectively. When prunings of L. leucocephala were used as mulch materials, the amount of nitrogen mineralized in the first week was 16.2% less than that of prunings incorporated into soil. The mineralization pattern of pruning N could be simulated by an exponent model Nt% = N01% (1 ?exp(梜1t))+ No2% (1 ?exp(梜2t) where Nt% is cumulative mineralized N in time t, N01 % and No2 % are readily and less readily mineralizable N in prunings, respectively, and k1 and k2 are rate constants. A half-life period of pruning nitrogen mineralization could also be determined by this model. The nitrogen content in the pruning residues decreased quickly in the first week but fluctuated thereafter. CCHFV is widely distributed in countries around the world.Outbreaks of CCHFV infection in humans have occurred in prior years in Xinjiang Province, China.Epidemiological surveys have detected CCHFV RNA in ticks and animals; however, few isolates were identified. In this study, we identified and isolated a new CCHFV strain from Hyalomma asiaticum asiaticum ticks collected from north of Tarim Basin in Xinjiang, China. A preliminary investigation of infection and antigens expression of CCHFV was performed in newborn mice. The target tissues for CCHFV replication in newborn mice were identified. The analysis of the phylogenetic relationships with other Chinese strains suggested that diverse genotypes of CCHFV have circulated in Xinjiang for years. | 11 | Leaching of Nutrient Elements in a Red Soil Derived from Quaternary Red Clay显示文摘A red soil derived from Quaternary red clay was employed to study nutrient leaching with soil columns repacked in laboratory. The objective was to identify the effects of fertilization practices on leaching patterns and magnitudes of Ca2+, Mg2+, K+, NH+, and NO-. The treatments were CK (as a control), CaCO3,CaSO4, MgCO3, Ca(H2PO4)2, urea, KCI, and multiple (a mixture of the above-mentioned fertilizers). The fertilizers were added to the bare surface of the soil columns, and then the columns were leached with 120 mL deionized water daily through peristaltic pumps over a period of 92 days. | 12 | An efficient algorithm to compute transient pressure responses of slanted wells with arbitrary inclination in reservoirs显示文摘Compared with vertical and horizontal wells, the solution and computation of transient pressure responses of slanted wells are more complex. Vertical and horizontal wells are both simplified cases of slanted wells at particular inclination, so the model for slanted wells is more general and more complex than other models for vertical and horizontal wells. Many authors have studied unsteady-state flow of fluids in slanted wells and various solutions have been proposed. However, until now, few of the published results pertain to the computational efficiency. Whether in the time domain or in the Laplace domain, the computation of integration of complex functions is necessary in obtaining pressure responses of slanted wells, while the computation of the integration is complex and time-consuming. To obtain a perfect type curve the computation time is unacceptable even with an aid of high-speed computers. The purpose of this paper is to present an efficient algorithm to compute transient pressure distributions caused by slanted wells in reservoirs. Based on rigorous derivation, the transient pressure solution for slanted wells of any inclination angle is presented. Assuming an infinite-conductivity wellbore, the location of the equivalent-pressure point is determined. More importantly, according to the characteristics of the integrand in a transient pressure solution for slanted wells, the whole integral interval is partitioned into several small integral intervals, and then the method of variable substitution and the variable step-size piecewise numerical integration are employed. The amount of computation is significantly reduced and the computational efficiency is greatly improved. | 13 | Spatial Distribution and Changes of Heavy Metals of Agricultural Lands in Typical Pregrading Coast in Dongtai City,Jiangsu Province,China显示文摘According to the historical changes of coastal lines, seven soil sampling districts, from land to sea, were arranged in Dongtai City, Jiangsu Province to sample soils from surface and profile. Concentrations of seven major heavy metals (HMs), granularity, pH, organic matters and C/N of the soil samples were analyzed. Results show that concentrations of heavy metals in agricultural land present a certain spatial variance, decreasing from land to sea. Pollution assessment indicates that the agricultural soils were not polluted by HMs, but the potential pollution of Cu and Hg needs to be alerted. Different HMs accumulate in the surface and sub-surface of the soil profiles, and concentrations of Hg and Pb decrease significantly with the increment of soil depth. Concentrations of HMs exhibit a significantly negative correlation to pH, but have no significant relation with organic matters in soil. Principle component analysis show that the concentrations of HMs relate to the land use history. Concentrations of Hg, Ni and Cr in soil are closely related with land use history, and concentrations of Pb, Cu and Cr are affected by land use history as well as other factors. Chinese Antarctic astronomy has begun to flourish in recent years, and it is an urgent need in basic astronomical work to measure and analyze the optical turbulence spatiotemporal distribution in the Antarctic region. We analyzed turbulence data measured by a mobile atmospheric parameter measurement system from 30 December 2013 to 10 February 2014 at Antarctic Taishan Station. Because there is a discrepancy between the refractive index structure constant C_n^2 measured by an ultrasonic anemometer with a single-point temperature structure function method and by micro-thermometer, a new method to measure C_n^2 with a temperature spectrum method is proposed herein. Through comparing long-term continuous C_n^2 data derived from ultrasonic anemometer with those via the new method and micro-thermometer, trend, magnitude and measured weak turbulence of ~2×10^(-16)m^(-2/3) are generally satisfactory. We present the initial results of C2 n measurement at Antarctic Taishan Station using micro-thermal sensors and a three-dimensional sonic anemometer at height ~2.0 m above the snow surface. The site testing experiments were carried out during the 30 th Chinese National Antarctic Research Expedition(CHINARE). We collected about 1 000 h of data between 30 December 2013 and 10 February 2014. The C2 n curve exhibits clear daily structures, with two peaks around midnight and midday and two troughs around 7:30 and 17:00 local time(UTC+5). The mean C2 n is 2.7×10-15 m-2/3 and the 25 th and 75 th percentiles of the C215 n cumulative distribution are 9.6×10-16 m-2/3 and 6.2×10- m-2/3, respectively. | 18 | The Effect of Online Channel Addition on Store Performance:Empirical Evidence from Chinese Chain Retailers显示文摘With the rapid development of e-Commerce and takeaway platforms, retailers have gradually developed multi-channel operations. However, limited empirical studies explored the effects of an online channel offered by takeaway platforms on the store performance. Does an online channel addition have a synergy effect or a cannibalization effect on store performance? We empirically investigate these effects by analyzing a large dataset including diverse samples collected from multiple retailers across various regions. The dataset includes 2115 stores across 25 retailers for 10 months that includes two types of retail formats and covers 16 provinces and 21 cities of China. We study the impacts of the newly introduced online channel on the incumbent offline channel and the overall store performance.The empirical results reveal that the online channel addition mainly has a synergy effect. Specifically,for the overall store, it has a positive effect on the sales and product variety, whereas it has a negative effect on the basket size. Surprisingly, an online channel addition also has a positive effect on the offline sales. Our study adds novel values to multi-channel retailing literature by empirically researching the cannibalization and synergy effect of a new type of online channel, offered by takeaway platforms. However, ARC1 only causes partial breakdown of the self-incompatibility response, and other SRK downstream factors may also participate in the self-incompatibility signaling pathway. In the present study, to search for SRK downstream targets, a plant U-box protein 3 (BoPUB3) was identified from the stigma of Brassica oleracea L. BoPUB3 was highly expressed in the stigma, and its expression was increased with the stigma development and reached to the highest level in the mature-stage stigma. BoPUB3, a 76.8-kDa protein with 697 amino acids, is a member of the PUB-ARM family and contains three domain characteristics of BoARC1, including a U-box N-terminal domain, a U-box motif, and a C-terminal arm repeat domain. The phylogenic tree showed that BoPUB3 was close to BoARC1. The synteny analysis revealed that B. oleracea chromosomal region containing BoPUB3 had high synteny with the Arabidopsis thaliana chromosomal region containing AtPUB3 (At3G54790). In addition, the subcellular localization analysis showed that BoPUB3 primarily localized in the plasma membrane and also in the cytoplasm. The combination of the yeast two-hybrid and in vitro binding assay showed that both BoPUB3 and BoARC1 could interact with SRK kinase domain, and SRK showed much higher level of β-galactosidase activity in its interaction with BoPUB3 than with BoARC1.
