| 3 | Wld^(S),Nmnats and axon degeneration-progress in the past two decades显示文摘A chimeric protein called Wallerian degeneration slow(Wld^(S))was first discovered in a spontaneous mutant strain of mice that exhibited delayed Wallerian degeneration.This provides a useful tool in elucidating the mechanisms of axon degeneration.Over-expression of WldS attenuates the axon degeneration that is associated with several neurodegenerative disease models,suggesting a new logic for developing a potential protective strategy.At molecular level,although Wld^(S)is a fusion protein,the nicotinamide mononucleotide adenylyl transferase 1(Nmnat1)is required and sufficient for the protective effects of Wld^(S),indicating a critical role of NAD biosynthesis and perhaps energy metabolism in axon degeneration.These findings challenge the proposed model in which axon degeneration is operated by an active programmed process and thus may have important implication in understanding the mechanisms of neurodegeneration.In this review,we will summarize these recent findings and discuss their relevance to the mechanisms of axon degeneration. | Yan Feng Tingting Yan Zhigang He Qiwei Zhai | 2010 | Protein & Cell2010,1,3: | 2 |
| 4 | Expression of phenylalanine ammonia lyase as an intracellularly free and extracellularly cell surface-immobilized enzyme on a gut microbe as a live biotherapeutic for phenylketonuria显示文摘Phenylketonuria(PKU),a disease resulting in the disability to degrade phenylalanine(Phe)is an inborn error with a 1 in 10,000 morbidity rate on average around the world which leads to neurotoxicity.As an potential alternative to a protein-restricted diet,oral intake of engineered probiotics degrading Phe inside the body is a promising treatment,currently at clinical stage II(Isabella,et al.,2018).However,limited transmembrane transport of Phe is a bottleneck to further improvement of the probiotic’s activity.Here,we achieved simultaneous degradation of Phe both intracellularly and extracellularly by expressing genes encoding the Phe-metabolizing enzyme phenylalanine ammonia lyase(PAL)as an intracellularly free and a cell surface-immobilized enzyme in Escherichia coli Nissle 1917(EcN)which overcomes the transportation problem.The metabolic engineering strategy was also combined with strengthening of Phe transportation,transportation of PAL-catalyzed trans-cinnamic acid and fixation of released ammonia.Administration of our final synthetic strain TYS8500 with PAL both displayed on the cell surface and expressed inside the cell to the Pah^(F263S)PKU mouse model reduced blood Phe concentration by 44.4%compared to the control Ec N,independent of dietary protein intake.TYS8500 shows great potential in future applications for PKU therapy. | Yu Jiang Bingbing Sun Fenghui Qian Feng Dong Chongmao Xu Wuling Zhong Rui Huang Qiwei Zhai Yu Jiang Sheng Yang | 2023 | Science China(Life Sciences)2023,66,1: | 1 |

| 6 | Investigating the function of Akt by tet-off inducible expression system显示文摘A tet-off inducible cell line named BBT derived from BA/F3β cell line was constructed and the effect of this inducible expression system was significant when detected by tet-off responded luciferase reporter gene assay. Then tet-off responded Akt expression plasmid was transfected into BBT cells, and the stable cell lines were screened. The result of Northern blot showed that the expression of akt was significantly inducible. The clone with the best inducible effect was selected and named BBA for investigating the function of Akt. We found that Akt could significantly inhibit zinc-induced decrease of cell viability when assayed by MTT method. | 12 | Interleukin-2 mediates the peripheral analgesia of interferon alpha显示文摘Interferon-α (IFN-α) and interleukin-2 (IL-2) are crucial cytokines in immune system. They also play an important role in nerve system. It has been reported that IL-2 can induce the central analgesia. It is demonstrated that IFN-α also can induce the peripheral analgesia, which can be blocked by naloxone as IL-2. Furthermore, the analgesic effect of IFN-α is reversible by monoclonal anti-IL-2 antibody. In vitro experiments show that IFN-α significantly increases the production of IL-2 in a dose dependent manner.
