| 1 | Sperm DNA damage and its clinical relevance in assessingreproductive outcome显示文摘The routine examination of semen, which assesses sperm concentration, percentage motility and morphology,does not identify subtle defects in sperm chromatin architecture. The focus on the genomic integrity of the male gamete has intensified recently due to the growing concern that genetic diseases may be transmitted via assisted reproductive techniques (ART). Accordingly, the intent of this review is to describe the details of the informationpertaining to mitochondfial/nuclear sperm DNA damage with an emphasis on its clinical significance and its relationship with male infertility. Assessment of sperm DNA damage appears to be a potential tool for evaluating semen samples prior to their use in ART. Testing DNA integrity may help select spermatozoa with intact DNA or with the least amount of DNA damage for use in assisted conception. | 5 | Viral Regulation of RNA Granules in Infected Cells显示文摘RNA granules are cytoplasmic, microscopically visible, non-membrane ribo-nucleoprotein structures and are important posttranscriptional regulators in gene expression by controlling RNA translation and stability. TIA/G3BP/PABP-specific stress granules(SG) and GW182/DCP-specific RNA processing bodies(PB) are two major distinguishable RNA granules in somatic cells and contain various ribosomal subunits, translation factors, scaffold proteins, RNA-binding proteins, RNA decay enzymes and helicases to exclude m RNAs from the cellular active translational pool. Although SG formation is inducible due to cellular stress, PB exist physiologically in every cell. Both RNA granules are important components of the host antiviral defense. Virus infection imposes stress on host cells and thus induces SG formation. However, both RNA and DNA viruses must confront the hostile environment of host innate immunity and apply various strategies to block the formation of SG and PB for their effective infection and multiplication. | 6 | Effect of Alstonia scholaris bark extract on testicular function of Wistar rats显示文摘Aim:To evaluate the antifertility effect of Alstonia scholaris bark extract in male rats.Methods:In male Wistar rats Alstonia scholaris bark extract was given by oral route at a dose of 200mg/day for60days,The fertility and testicular function were assessed by mating tests,sperm motility,sperm concentration,biochemical indices and testicular cell population dynamics.Results:Oral feeding with the extract at a dose of 200mg/day for the period of 60days did not cause body weight loss,while the weights of testes,epididymides,seminal vesicle and ventral prostate were significantly reduced.The production of step-19 spermatids was reduced by 79.6%in treated rats.The popu-lation of preleptotene and pachytene spermatocytes were decrased by61.9%and60.1%,respectively.Spematogo-nia and Sertoli cell population were also affected.The seminiferous tubule and Leydig cell nuclear area were reduced significantly(P<0.01)when compared to the controls..Reduced sperm count and motility yesulted in a total supprs-sion of fertility.A significant fall in the protein and sialic acid content of the testes,epididymides,seminal vesicle and ventral prostate as well as glycogen content of testes were also noticed.The fructose content in the seminal vesicle was lowered whereas the testicular cholesterol was elevated as compared with the controls.The following com-pounds were obtained from the extract with chromatographic separation over Si-gel column;α-amyrin,β-amyrin,lupiol acetate,venenative,rhazine and yohimbine,Conclusion:At the dose level employed,Alstonia scholaris bark extract has a significant antifertility effect in male rast;the primary site of action may be post meiotic germ cells(Step19spermatids). | R. To understand its etiopathogenesis we developed ananimal model by repeated portal endotoxemia inducedthrough the gastrosplenic vein.METHODS: Twenty-nine rabbits (1.5-2.0 kg) were divid-ed into control (group n = 13) and experimental ( groupn = 16) groups. Heat killed E. coli were injected throughan indwelling cannula into the gastrosplenic vein in pre-sensitized animals. The animals were sacriflced at 1, 3 and6 months.RESULTS: The mean portal pressure in group animalswas significantly (P < 0. 05) higher than in group at 1(17.5 ±3.4 vs 10.4±2.2 mmHg), 3 (17.8±1.3 vs7.2 +3.6mmHg), and 6 (19.8±3.1 vs 10.3±4.8 mmHg) months.Similarly, the mean splenic weight in group was signifi-cantly greater than in group (P <0.05). Histopathologi-cally, the spleen showed medullary congestion, hemosid-rin-laden macrophages and mild fibrosis. Histologically,the liver had normal parenchyma with mild portal lympho-cytic infiltrates and kupffer cell hyperplasia. No significantanomalies were detected by liver function tests.CONCLUSIONS: The rabbit model showed significantsplenomegaly with a persistent increase in portal pressureand mild fibrosis without hepatic parenchymal injury, quiteakin to non-cirrhotic portal fibrosis as seen in humans.
