1Progress in non-invasive detection of liver fibrosis显示文摘Liver fibrosis is an important pathological precondition for hepatocellular carcinoma.The degree of hepatic fibrosis is positively correlated with liver cancer.Liver fibrosis is a series of pathological and physiological process related to liver cell necrosis and degeneration after chronic liver injury,which finally leads to extracellular matrix and collagen deposition.The early detection and precise staging of fibrosis and cirrhosis are very important for early diagnosis and timely initiation of appropriate therapeutic regimens.The risk of severe liver fibrosis finally progressing to liver carcinoma is>50%.It is known that biopsy is the gold standard for the diagnosis and staging of liver fibrosis.However,this method has some limitations,such as the potential for pain,sampling variability,and low patient acceptance.Furthermore,the necessity of obtaining a tissue diagnosis of liver fibrosis still remains controversial.An increasing number of reliable non-invasive approaches are now available that are widely applied in clinical practice,mostly in cases of viral hepatitis,resulting in a significantly decreased need for liver biopsy.In fact,the noninvasive detection and evaluation of liver cirrhosis now has good accuracy due to current serum markers,ultrasound imaging,and magnetic resonance imaging quantification techniques.A prominent advantage of the non-invasive detection and assessment of liver fibrosis is that liver fibrosis can be monitored repeatedly and easily in the same patient.Serum biomarkers have the advantages of high applicability(>95%)and good reproducibility.However,their results can be influenced by different patient conditions because none of these markers are liver-specific.The most promising techniques appear to be transient elastography and magnetic resonance elastography because they provide reliable results for the detection of fibrosis in the advanced stages,and future developments promise to increase the reliability and accuracy of the staging of hepatic fibrosis.This article aims to describe the recent progress in the development of non-invasive assessment methods for the staging of liver fibrosis,with a special emphasize on computer-aided quantitative and deep learning methods.Chengxi Li Rentao Li Wei Zhang 2018Cancer Biology & Medicine2018,15,2:39

3Rational design, synthesis, and biological evaluation of novel C6-modified geldanamycin derivatives as potent Hsp90 inhibitors and anti-tumor agents显示文摘Heat shock protein 90(Hsp90) is an appealing anticancer drug target that provoked a tremendous wave of investigations. Geldanamycin(GA) is the first identified Hsp90 inhibitor that exhibited potent anticancer activity, but the off-target toxicity associated with the benzoquinone moiety hampered its clinical application. Until now, structure optimization of GA is still in need to fully exploit the therapeutic value of Hsp90. Due to the structural complexity and synthetic challenge of this compound family, conventional optimization is bound to be costly but high efficiency is expected to be reachable by combining the art of rational design and total synthesis. Described in this paper is our first attempt at this approach aiming at rational modification of the C6-position of GA. The binding affinities towards Hsp90 of compound 1(C6-ethyl) and 2(C6-methyl) were designed and predicted by using Discovery Studio. These compounds were synthesized and further subjected to a thorough in vitro biological evaluation. We found that compounds1 and 2 bind to Hsp90 protein with the IC_(50) of 34.26 nmol/L and 163.7 nmol/L, respectively. Both compounds showed broad-spectrum antitumor effects. Replacing by ethyl, compound 1 exhibited more potent bioactivity than positive control GA, such as in G2/M cell cycle arrest, cell apoptosis and client proteins degradations. 5Dual-layer efficiency enhancement for future passive optical network显示文摘Ever-increasing demands on bandwidth compel us to improve the bandwidth utilization of access networks. It is more important to enhance the efficiency of passive optical networks(PON) than other access technologies, as they are the primary solutions to various access scenarios. To satisfy the demands of improved bandwidth efficiency, we propose a dual-layer(data link layer and network layer) efficiency enhancement scheme for future PON system. Under this architecture, a data-link-layer flexible coding scheme is introduced to extend the downstream link throughput, and a network-layer centralized active queue management(AQM) scheme is introduced to increase bandwidth efficiency. Both of these technologies are designed to suit the future centralized cooperative control mode. The results of an experimental demonstration of the network coding PON scheme reveal the efficiency of the proposed architecture. The results of simulations show that the centralized AQM scheme has a positive influence on queuing delay and jitter. 6Artificial intelligence-driven autonomous optical networks: 3S architecture and key technologies显示文摘In the optical networks,the dynamicity,the complexity and the heterogeneity have dramatically increased owing to the deployment of advanced coherent techniques,and the optical cross-connect technologies and diverse network infrastructures pose great challenges in the optical network management and maintenance for the network operators.In this review,we propose a'3 S'architecture for AI-driven autonomous optical network,which can aid the optical networks operated in'self-aware'of network status,'self-adaptive'of network control,and'self-managed'of network operations.To support these functions,a number of artificial intelligence(AI)-driven techniques have been investigated to improve the flexibility and the reliability from the device aspect to network aspect.Adaptative erbium-doped fiber amplifier(EDFA)controlling is an example for the device aspect,which provides a power self-adaptive capability according to the network condition.From the link aspect,adaptive fiber nonlinearity compensation,optical monitoring performance and quality of transmission estimation are developed to monitor and alleviate the link-dependent signal impairments in an automatic way.From the network aspect,traffic prediction and network state analysis methods provide the self-awareness,while automatic resource allocation and network fault management powered by AI enhance the self-adaptiveness and self-management capabilities.Benefit from the sufficient network management data,powerful data-mining capability and matured computation units,these AI techniques have great potentials to provide autonomous features for optical networks,including the network resource scheduling and the network customization.Yuefeng JI Rentao GU Zeyuan YANG Jin LI Hui LI Min ZHANG 2020Science China(Information Sciences)2020,63,6:0
7Conversion of cropland into agroforestry land versus naturally-restored grassland alters soil macro-faunal diversity and trophic structure in the semi-arid agro-pasture zone of northern China显示文摘Restoration of cropland(termed 'Farm') after abandonment including shrubs(termed 'Shrub'),trees(termed 'Tree') and natural grassland(termed 'Grass') has become a routine process aimed to improve land productivity and control desertification. During this restoration process, soil macro-faunal diversity, and trophic structure were investigated at four types of sites(Farm, Shrub, Tree, and Grass)during growing season in the semi-arid agro-pasture zone of northern China. Results indicated that the Staphylinidae family was found to dominate at the Grass, Shrub, and Tree sites, whiles larval Pyralidae individuals were found at the Grass site only. The density of the omnivores(i.e., Formicidae family) was significantly(P<0.05) greater at the Grass site than at the Tree and Farm sites. The total density and richness of predator and phytophages were found to be markedly(P<0.05) greater at the Grass site than at the Farm site. Meanwhile, we found the taxon richness of predators was significantly(P<0.05) higher at the Shrub site than at the Farm and Tree sites. Compared with the Farm and afforested Shrub/Tree sites,the Grass site had greater density, taxon richness, and Shannon index(P<0.05). In conclusion, natural restoration of abandoned croplands toward grassland was an effective strategy relative to artificial afforestation for improvement of soil biological diversity.
