| 2 | Analysis of DNA methylation in different maize tissues显示文摘DNA methylation plays an important role in gene expression regulation during biological development and tissue differentiation in plants. This study adopted methylation-sensitive Amplified fragment length polymorphism (AFLP) to compare the levels of DNA cyto- sine methylation at CCGG sites in tassel, bracteal leaf, and ear leaf from maize inbred lines, 18 White and 18 Red, respectively, and also examined specific methylation patterns of the three tissues. Significant differences in cytosine methylation level among the three tissues and the same changing tendency in two inbred lines were detected. Both MSAP (methylation sensitive amplification polymorphism) ratio and full methylation level were the highest in bracteal leaf, and the lowest in tassel. Meanwhile, different methylation levels were ob- served in the same tissue from the inbred lines, 18 White and 18 Red. Full methylation of internal cytosine was the dominant type in the maize genome. The differential methylation patterns in the three tissues were observed. In addition, sequencing of nine differentially me- thylated fragments and the subsequent blast search revealed that the cytosine methylated 5′-CCGG-3′sequences were distributed in re- peating sequences, in the coding and noncoding regions. Southern hybridization was used to verify the methylation polymorphism. These results clearly demonstrated the power of the MSAP technique for large-scale DNA methylation detection in the maize genome, and the complexity of DNA methylation change during plant growth and development. | 3 | Three-dimensional morphometric analysis for hepatectomy of centrally located hepatocellular carcinoma:A pilot study显示文摘AIM: To describe a three-dimensional model(3DM) to accurately reconstruct anatomic relationships of centrally located hepatocellular carcinomas(HCCs).METHODS: From March 2013 to July 2014, reconstructions and visual simulations of centrally located HCCs were performed in 39 patients using a 3D subject-based computed tomography(CT) model with customdeveloped software. CT images were used for the 3D reconstruction of Couinaud's pedicles and hepatic veins, and the calculation of corresponding tumor territories and hepatic segments was performed using Yorktal DMIT software. The respective volume, surgical margin, and simulated virtual resection of tumors were also estimated by this model preoperatively. All patients were treated surgically and the results were retrospectively assessed. Clinical characteristics, imaging data, procedure variables, pathologic features, and postoperative data were recorded and compared to determine the reliability of the model.RESULTS: 3D reconstruction allowed stereoscopic identification of the spatial relationships between physiologic and pathologic structures, and offered quantifiable liver resection proposals based on individualized liver anatomy. The predicted values were consistent with the actual values for tumor mass volume(82.4 ± 109.1 m L vs 84.1 ± 108.9 m L, P = 0.910), surgical margin(10.1 ± 6.2 mm vs 9.1 ± 5.9 mm, P = 0.488), and maximum tumor diameter(4.61 ± 2.16 cm vs 4.53 ± 2.14 cm, P = 0.871). In addition,the number and extent of portal venous ramifications, as well as their relation to hepatic veins, were visualized. Preoperative planning based on simulated resection facilitated complete resection of large tumors located in the confluence of major vessels. The results indicated that the content of endonuclease sensitive sites (ESS) was reduced by He-Ne laser irradiation, which formed in cells irradiated by enhanced ultraviolet-B. | 7 | Effectiveness of fluidized pellet bed for removing soluble contaminants显示文摘Fluidized pellet bed(FPB) has been successfully applied in water and wastewater treatment. However,the removal mechanism of contaminants especially the soluble ones,is still unclear. This study aimed to evaluate the effectiveness of FPB reactor for removing soluble contaminants from synthetic wastewater. By only coagulation through jar test operation with addition of polyaluminium chloride(PACl) as primary coagulant and polyacryamide(PAM) as coagulant-aid,the removals of soluble chemical oxygen demand(COD) ,total phosphorus(TP) ,and NH4+-N were found to be only 2.2%-7.5%,5.7%-25.5%,and 9.9%-18.5%,respectively. However,by FPB operation under the same dosage of coagulants,these values increased to 82.7%,37.2%,and 50%,indicating that the formation of pellets in the FPB effectively enhanced the removal of soluble contaminants. By careful comparison of the settleablility and filterability of the pollutants after coagulation,the originally soluble contaminants could be divided into three groups,namely:(1) coagulated-and-settleable,(2) coagulated-but-nonsettleable,and(3) uncoagulable. It was found that not only the first two groups but also a large part of the third group could be effectively removed by FPB operation. | 9 | Adjuvant radiotherapy in central hepatocellular carcinoma after narrow-margin hepatectomy:A 10-year real-world evidence显示文摘Objective:A prospective randomized control study investigated the feasibility and efficacy of adjuvant radiotherapy on patients with central hepatocellular carcinoma(HCC)after narrow-margin hepatectomy(<1 cm).This study presents an updated 10-year real-world evidence to further characterize the role of adjuvant radiotherapy.Methods:Patients with central HCC after narrow-margin hepatectomy(<1 cm)were prospectively assigned to adjuvant radiotherapy group and control group.Patients'outcome,adverse events,long-term recurrence and survival rates were investigated.Results:The 1-,5-,and 10-year recurrence-free survival(RFS)rates were 81.0%,43.9%,and 38.7%,respectively in adjuvant radiotherapy group and 71.7%,35.8%,and 24.2%,respectively in control group(log-rank test,P=0.09).The 1-,5-,and 10-year overall survival(OS)rates were 96.6%,54.7%,and 42.8%,respectively in adjuvant radiotherapy group and 90.2%,55.1%,and 30.0%,respectively in control group(log-rank test,P=0.20).The 1-,5-,and 10-year RFS rates for patients with small HCC(≤5 cm)were 91.1%,51.6%,and 48.4%,respectively in adjuvant radiotherapy group and 80.0%,36.6%,and 26.6%,respectively in control group(log-rank test,P=0.03).Landmark analysis demonstrated that patients with small HCC in adjuvant radiotherapy group had a significantly improved OS in second five years after treatment in comparison to patients in control group(log-rank test,P=0.05).Conclusions:Our updated results showed a sustained clinical benefit on reducing recurrence,improving longterm survival for small central HCC by adjuvant radiotherapy after narrow-margin hepatectomy.Long-term survival data also indicated that hepatectomy is an optimal treatment for selected patients with central HCC. | Weiqi Rong Weibo Yu Liming Wang Fan Wu Kai Zhang Bo Chen Chengli Miao Liguo Liu Songlin An Changcheng Tao Weihu Wang Jianxiong Wu | 2020 | Chinese Journal of Cancer Research2020,32,5: | 6 |
| 10 | Water softening by induced crystallization in fluidized bed显示文摘Fluidized bed and induced crystallization technology were combined to design a new type of induced crystallization fluidized bed reactor. The added particulate matter served as crystal nucleus to induce crystallization so that the insoluble material, which was in a saturated state, could precipitate on its surface. In this study, by filling the fluidized bed with quartz sand and by adjusting water p H, precipitation of calcium carbonate was induced on the surface of quartz sand, and the removal of water hardness was achieved.With a reactor influent flow of 60 L/hr, a fixed-bed height of 0.5 m, pH value of 9.5, quartz sand nuclear diameter of 0.2–0.4 mm, and a reflux ratio of 60%, the effluent concentration of calcium hardness was reduced to 60 mg/L and 86.6% removal efficiency was achieved. The resulting effluent reached the quality standard set for circulating cooling water. Majority of the material on the surface of quartz sand was calculated to be calcium carbonate based on energy spectrum analysis and moisture content was around 15.994%. The composition and structure of the prepared compounds were determined by the chemical analysis, elemental analysis, IR spectroscopy, UV, FS spectroscopy, X ray powder diffraction, 1H NMR and solubility. | 16 | Fabrication and characterization of novel meso-porous carbon/n-octadecane as form-stable phase change materials for enhancement of phase-change behavior显示文摘In this study, series of novel composite phase change materials(PCMs) were prepared through vacuum impregnation by using meso-porous carbon as a supporting matrix and n-octadcane as PCMs.The meso-porous carbon material was prepared through one-pot co-assembly method, using resorcinol and formaldehyde as carbon precursor, tetraethoxysilane as silica sources and triblock copolymer F127 as a template. And the phase behaviors of n-octadcane confined in the nano-porous structure of the meso-porous carbon were further investigated. Fourier transform-infrared spectroscopy spectra show that n-octadecane was effectively encapsulated in the porous structure of mesoporous carbon and the composite PCMs were successfully prepared. Differential scanning calorimetry results confirm that the composite PCMs possess a good phase change behavior, fast thermal-response rate and excellent thermal cycling stability. In addition, the composite PCMs possess expected heat storage and heat release properties. | 17 | The D253N Mutation in the Polymerase Basic 2 Gene in Avian Influenza(H9N2) Virus Contributes to the Pathogenesis of the Virus in Mammalian Hosts显示文摘Mutations in the polymerase basic 2(PB2) gene of avian influenza viruses are important signatures for their adaptation to mammalian hosts. Various adaptive mutations have been identified around the 627 and nuclear localization sequence(NLS) domains of PB2 protein, and these mutations contribute to the replicative ability of avian influenza viruses.However, few studies have focused on adaptive mutations in other regions of PB2. In this study, we investigated the functional roles of the D253N mutation in PB2 in an H9N2 virus. This mutation was found to affect an amino acid residue in the middle domain of the PB2 protein. The virus with the D253N mutation showed higher polymerase activity and transiently increased viral replication in human cells. However, the mutant did not show significant differences in viral replication in the respiratory tract of mice upon infection. | 18 | Differentially Expressed Genes in Heads and Tails of Angelica Sinensis diels: Focusing on Ferulic Acid Metabolism显示文摘Objective: To explore the scientific connotation of the discrepant pharmaceutical activities between the head and tail of Angelica sinensis diels(AS), an important herb extensively utilized in Chinese medicine, by the approach of transcriptome sequencing. Methods: Ten samples of AS were randomly collected in Min County, Gansu Province of China. Transcriptome sequencing of AS was accomplished in a commercial ILLumina Hi Seq-2000 platform. The transcriptome of each head and tail of AS were fixed in a gene chip, and detected under the procedure of Illumina Hi Seq-2000. Differentially expressed unigenes between the heads and tails of AS were selected by Shanghai Biotechnology Corporation(SBC) online analysis system, based on Gene Ontology, Kyoto Encyclopedia of Genes and Genomes and relevant bioinformatic database. Results: Totally 63,585 unigenes were obtained from AS by high-throughput sequencing platform. Among which 3359 unigenes were identified as differentially expressed unigenes between the heads and tails of AS by SBC analysis system scanning. Of which 15 differentially expressed unigenes participate in the metabolic regulation of phenylpropanoid biosynthesis(PB) pathway and ferulic acid metabolites, in response to the distinguished pharmaceutical actions of the heads and tails of AS. | 20 | Identification of proteinaceous binding media for the polychrome terracotta army of Emperor Qin Shihuang by MALDI-TOF-MS显示文摘A proteinaceous binding media for the polychrome terracotta army of Emperor Qin Shihuang has been identified using matrix-assisted laser desorption/ionization time-of-flight mass spectrometry(MALDI-TOF-MS).Methods for the preparation of the model samples and the elimination of interferences have been evaluated,as well as ageing processes.A method involving the complexation of EDTA in combination with dialysis was used to eliminate any interference in the polychrome layers taken from the historical samples.The model samples were aged by being buried in loess to replicate the ageing process of the historical samples as closely as possible.The optimum conditions for the extraction and subsequent enzymatic hydrolysis of the extracted protein were investigated.Under the optimized conditions,the‘‘peptide mass fingerprints’’of the proteinaceous binding media from the historical and model samples were determined by MALDITOF-MS.The results revealed that animal glue was used as the binding media for the polychrome layers of Emperor Qin Shihuang’s terracotta army.It also shows that animal glue used as binding media has a long history in China. | Hongtao Yan Jingjing An Tie Zhou Yin Xia Bo Rong | 2014 | Chinese Science Bulletin2014,59,21: | 3 |
