3LHCGR and ALMS1 defects likely cooperate in the development of polycystic ovary syndrome indicated by double-mutant mice显示文摘Polycystic ovary syndrome(PCOS)is a heterogeneous disorder with evidence of polygenetic components,and obesity may be a risk factor for hyperandrogen ism.Previous studies have shown that LHCGR is en riched in the ovary and LHCGR deficiency causes infertility without typical PCOS phenotypes.ALMS1 is implicated in obesity and hyperandrogenism,the common phenotypes among PCOS patients.Through whole-exome sequencing of 22 PCOS families and targeted candidate gene sequencing of additional 65 sporadic PCOS patients,we identified potential causative mutations in LHCGR and ALMS1 in a sibling-pair PCOS family and three sporadic PCOS patients.The expression of LHCGRL638 P in granulosa-like tumor cell line(KGN)cells promoted cyclic adenosine monophosphate production and granulosa cell proliferation,indicating that LHCGRL638 P is an activating mutation.Lhcgr~(L642 P/L642 P)mice showed an irregular estrous cycle,reduced follicles with dynamic folliculogenesis,and increased testosterone(T),estradiol(E2),and dehydroepiandrosterone.Lhcgr~(+/L642 P)AIms1~(+/PB)mice displayed increased T and E2 but decreased late secondary and preovulatory follicles.We showed that activating mutation of LHCGR likely plays important roles in the pathophysiology of PCOS involving abnormal reproductive physiology,whereas ALMS1 deficiency may promote anovulatory infertility via elevated androgens,suggesting that the disturbed LHCGR and ALMS1 cooperatively induce PCOS phenotypes,characterized as anovulation and hyperandrogenemia frequently observed in PCOS patients with obesity.Li Yu Lina Wang Wufan Tao Wenxiang Zhang Shuanghao Yang Jian Wang Jia Fei Rui Peng Yiming Wu Xiumei Zhen Hong Shao Weiyue Gu Rong Li Bai-Lin Wu Hongyan Wang 2021Journal of Genetics and Genomics2021,48,5:3

5YB1 regulates miR-205/200b-ZEB1 axis by inhibiting microRNA maturation in hepatocellular carcinoma显示文摘Background:Y-box binding protein 1(YB1 or YBX1)plays a critical role in tumorigenesis and cancer progression.However,whether YB1 affects malignant transformation by modulating non-codingRNAs remains largely unknown.This study aimed to investigate the relationship between YB1 and microRNAs and reveal the underlying mechanism by which YB1 impacts on tumor malignancy via miRNAs-mediated regulatory network.Methods:The biological functions of YB1 in hepatocellular carcinoma(HCC)cells were investigated by cell proliferation,wound healing,and transwell invasion assays.The miRNAs dysregulated by YB1 were screened by microarray analysis in HCC cell lines.The regulation of YB1 on miR-205 and miR-200b was determined by quantitative real-time PCR,dual-luciferase reporter assay,RNA immunoprecipitation,and pull-down assay.The relationships of YB1,DGCR8,Dicer,TUT4,and TUT1 were identified by pull-down and coimmunoprecipitation experiments.The cellular co-localization of YB1,DGCR8,and Dicer were detected by immunofluorescent staining.The in vivo effect of YB1 on tumor metastasis was determined by injecting MHCC97H cells transduced with YB1 shRNA or shControl via the tail vein in nude BALB/c mice.The expression levels of epithelial tomesenchymal transition markerswere detected by immunoblotting and immunohistochemistry assays.Results:YB1 promoted HCC cell migration and tumor metastasis by regulating miR-205/200b‒ZEB1 axis partially in a Snail-independent manner.YB1 suppressedmiR-205 and miR-200b maturation by interacting with the microprocessors DGCR8 and Dicer as well as TUT4 and TUT1 via the conserved cold shock domain.Subsequently,the downregulation of miR-205 and miR-200b enhanced ZEB1 expression,thus leading to increased cell migration and invasion.Furthermore,statistical analyses on gene expression data from HCC and normal liver tissues showed that YB1 expression was positively associated with ZEB1 expression and remarkably correlated with clinical prognosis.Conclusion:This study reveals a previously undescribed mechanism by which YB1 promotes cancer progression by regulating the miR-205/200b‒ZEB1 axis in HCC cells.Furthermore,these results highlight that YB1 may play biological functions via miRNAs-mediated gene regulation,and it can serve as a potential therapeutic target in human cancers.Xiumei Liu Di Chen Huan Chen Wen Wang Yu Liu Yawei Wang Chao Duan Zhen Ning Xin Guo Wuxiyar Otkur Jing Liu Huan Qi Xiaolong Liu Aifu Lin Tian Xia Hong-xu Liu Hai-long Piao 2021Cancer Communications2021,41,7:1

7The effect of different cancer therapies on clinical outcomes of assisted reproductive technology in breast cancer patients显示文摘Objective To evaluate the clinical outcomes of assisted reproductive technology(ART)on fertility preservation and infertility treatment in breast cancer patients who had undergone different cancer therapies.Methods 20 infertile females who had undergone breast cancer treatments during 2011–2018 were studied retrospectively.The patients were divided into two groups based on their cancer treatment methods and their needs of fertility preservation:the combined treatment group,who had both breast cancer surgery combining with any of the three treatments(adjuvant endocrine therapy,radiotherapy or chemotherapy),and the surgery only group.A group of infertile females without breast cancer history were used as a control group.An aromatase inhibitor Letrozole-based ovarian micro-stimulation protocol was used in females from the three groups for in vitro fertilization and embryo transfer.The ART clinical outcomes were evaluated by using the parameters of antral follicle count(AFC),the ratio of FSH/LH,oocyte retrieval number,2 pronucleus(2 PN)fertilization rate,high-quality embryo rate,clinical pregnancy rate,and delivery outcome.Results The surgery only group had significantly lower ratio of FSH/LH than the combined treatment group and the control group.No significant difference on the ART clinical outcomes,evaluated by the aforementioned criteria,were found between the three groups.Conclusions Breast cancer surgery with adjuvant therapies,but not surgery alone,may damage ovarian function.The best time-limited window to preserve fertility for breast cancer patients is after surgery but before the initiation of adjuvant therapies.Importantly,the clinical outcomes of aromatase inhibitor-based ovary micro-stimulation in breast cancer patients are similar to that in non-breast cancer controls.Mengmeng Zhang Xueling Song Caihong Ma Xiumei Zhen Hongbin Chi Xiaowei Zhang Rong Li Jie Yan Jie Qiao 2021Gynecology and Obstetrics Clinical Medicine2021,1,1:0
8Effects of Dachengqi Decoctions Made from Raw Rhubarb and Vinegar-processed Rhubarb on Levels of Endotoxin,NO and TNF-αin Serum显示文摘[Objectives]The effects of Dachengqi decoctions made from raw rhubarb and vinegar-processed rhubarb on serum endotoxin,NO and TNF-αlevels were investigated.[Methods]Total 105 mice were randomly divided into Dachengqi decoction made from raw rhubarb groups(6,10 g/kg),Dachengqi decoction made from vinegar-processed rhubarb groups(6,10 g/kg),positive control group,blank control group and model group.After administration at a dose of 20 mL/kg,the levels of endotoxin,NO and TNF-αin serum were determined.The effects of Dachengqi decoctions made from raw rhubarb and vinegar-processed rhubarb on serum endotoxin,NO and TNF-αlevels in mice were compared.[Results]Dachengqi decoctions made from raw rhubarb and vinegar-processed rhubarb both increased the NO level and reduced the endotoxin and TNF-αlevels in serum of ABP mice.[Conclusions]Dachengqi decoctions made from raw rhubarb and vinegar-processed rhubarb have different effects on endotoxin,NO and TNF-αcontents.These changes correspond to the effects of Dachengqi Decoctions made from raw rhubarb and vinegar-processed rhubarb.The change in the content of rhubarb anthraquinones has a certain effect on the efficacy of Dachengqi decoction.Wen ZHONG Zujie QIN Jiangcun WEI Zhen XIE Xiumei MA Xianyi SHI Fengxian ZHAO Qian HAN 2020Medicinal Plant2020,11,3:0
