| 1 | ShRNA-mediated gene silencing of β-catenin inhibits growth of human colon cancer cells显示文摘AIM: To observe the gene silencing mediated by the specific shRNA targeted against β-catenin and its effect on cell proliferation and cycle distribution in the human colon cancer cell line Colo205. METHODS: Two shRNA plasmid vectors against β-catenin were constructed and transfected into Colo205 cells with LipofectamineTM2000. The down-regulations of β-catenin, c-myc and cyclinD1 expressions were detected by RT-PCR and western blot analysis. The cell proliferation inhibitions were determined by MTT assay and soft agar colony formation assay. The effect of these two β-catenin shRNAs on cell cycle distribution and apoptosis was examined by flow cytometry. RESULTS: These two shRNA vectors targeted against β-catenin efficiently suppressed the expression of β-catenin and its down stream genes, c-myc and cyclinD1. The expression inhibition rates were around 40%-50% either at the mRNA or at the protein level. The shRNA-mediated gene silencing of β-catenin resulted in significant inhibition of cell growth both on the culture plates and in the soft agar. Moreover, the cancer cells showed significant G0/G1 arrest and increased apoptosis at 72 h post transfection due to gene silencing. CONCLUSION: These specific shRNAs targeted against β-catenin could have a gene silencing effect and block the WNT signaling pathway. They could inhibit cell growth, increase apoptosis, and induce cell cycle arrest in Colo205 cells. | 2 | Management of granulomatous lobular mastitis: an international multidisciplinary consensus(2021 edition)显示文摘Granulomatous lobular mastitis(GLM) is a rare and chronic benign inflammatory disease of the breast. Difficulties exist in the management of GLM for many front-line surgeons and medical specialists who care for patients with inflammatory disorders of the breast. This consensus is summarized to establish evidence-based recommendations for the management of GLM. Literature was reviewed using PubMed from January 1, 1971 to July 31, 2020. Sixty-six international experienced multidisciplinary experts from 11 countries or regions were invited to review the evidence.Levels of evidence were determined using the American College of Physicians grading system, and recommendations were discussed until consensus. Experts discussed and concluded 30 recommendations on historical definitions,etiology and predisposing factors, diagnosis criteria, treatment, clinical stages, relapse and recurrence of GLM. GLM was recommended as a widely accepted definition. In addition, this consensus introduced a new clinical stages and management algorithm for GLM to provide individual treatment strategies. In conclusion, diagnosis of GLM depends on a combination of history, clinical manifestations, imaging examinations, laboratory examinations and pathology.The approach to treatment of GLM should be applied according to the different clinical stage of GLM. | 3 | Determination of Lignin in Marine Sediment Using Alkaline Cupric Oxide Oxidation-Solid Phase Extraction-on-Column Derivatization-Gas Chromatography显示文摘Lignin serves as one of the most important molecular fossils for tracing Terrestrial Organic Matters (TOMs) in marine environment. Extraction and derivatization of lignin oxidation products (LOPs) are crucial for accurate quantification of lignin in marine sediment. Here we report a modification of the conventional alkaline cupric oxide (CuO) oxidation method, the modification consisting in a solid phase extraction (SPE) and a novel on-column derivatization being employed for better efficiency and reproducibility. In spiking blanks, recoveries with SPE for the LOPs are between 77.84% and 99.57% with relative standard deviations (RSDs) ranging from 0.57% to 8.04% (n=3), while those with traditional liquid-liquid extraction (LLE) are from 44.52% to 86.16% with RSDs being from 0.53% to 13.14% (n=3). Moreover, the reproducibility is greatly improved with SPE, with less solvent consumption and shorter processing time. The average efficiency of on-column derivatization for LOPs is 100.8% ± 0.68%, which is significantly higher than those of in-vial or in-syringe derivatization, thus resulting in still less consumption of derivatizing reagents. Lignin in the surface sediments sampled from the south of Yangtze River estuary, China, was determined with the established method. Recoveries of 72.66% to 85.99% with standard deviation less than 0.01mg/10g dry weight are obtained except for p-hydroxyben-zaldehyde. The lignin content Σ8 (produced from 10 g dry sediment) in the research area is between 0.231 and 0.587 mg. | 5 | Association of Overlapped and Un-overlapped Comorbidities with COVID-19 Severity and Treatment Outcomes: A Retrospective Cohort Study from Nine Provinces in China显示文摘Objective Several COVID-19 patients have overlapping comorbidities. The independent role of each component contributing to the risk of COVID-19 is unknown, and how some non-cardiometabolic comorbidities affect the risk of COVID-19 remains unclear.Methods A retrospective follow-up design was adopted. A total of 1,160 laboratory-confirmed patients were enrolled from nine provinces in China. Data on comorbidities were obtained from the patients’ medical records. Multivariable logistic regression models were used to estimate the odds ratio(OR) and 95% confidence interval(95% CI) of the associations between comorbidities(cardiometabolic or non-cardiometabolic diseases), clinical severity, and treatment outcomes of COVID-19.Results Overall, 158(13.6%) patients were diagnosed with severe illness and 32(2.7%) had unfavorable outcomes. Hypertension(2.87, 1.30–6.32), type 2 diabetes(T2 DM)(3.57, 2.32–5.49),cardiovascular disease(CVD)(3.78, 1.81–7.89), fatty liver disease(7.53, 1.96–28.96), hyperlipidemia(2.15, 1.26–3.67), other lung diseases(6.00, 3.01–11.96), and electrolyte imbalance(10.40, 3.00–26.10)were independently linked to increased odds of being severely ill. T2 DM(6.07, 2.89–12.75), CVD(8.47,6.03–11.89), and electrolyte imbalance(19.44, 11.47–32.96) were also strong predictors of unfavorable outcomes. Women with comorbidities were more likely to have severe disease on admission(5.46,3.25–9.19), while men with comorbidities were more likely to have unfavorable treatment outcomes(6.58, 1.46–29.64) within two weeks.Conclusion Besides hypertension, diabetes, and CVD, fatty liver disease, hyperlipidemia, other lung diseases, and electrolyte imbalance were independent risk factors for COVID-19 severity and poor treatment outcome. | 8 | Sources of nitrous and nitric oxides in paddy soils: Nitrification and denitrification显示文摘Rice-paddies are regarded as one of the main agricultural sources of N 2O and NO emissions. To date, however, specific N2O and NO production pathways are poorly understood in paddy soils. 15N-tracing experiments were carried out to investigate the processes responsible for N2O and NO production in two paddy soils with substantially different soil properties. Laboratory incubation experiments were carried out under aerobic conditions at moisture contents corresponding to 60% of water holding capacity. The relative importance of nitrification and denitrification to the flux of N2O was quantified by periodically measuring and comparing the enrichments of the N2O, NH+4-N and NO. 3-N pools. The results showed that both N2O and NO emission rates in an alkaline paddy soil with clayey texture were substantially higher than those in a neutral paddy soil with silty loamy texture. In accordance with most published results, the ammonium N pool was the main source of N2O emission across the soil profiles of the two paddy soils, being responsible for 59.7% to 97.7% of total N2O emissions. The NO. 3-N pool of N2O emission was relatively less important under the given aerobic conditions. The rates of N2O emission from nitrification(N2On) among different soil layers were significantly different, which could be attributed to both the differences in gross N nitrification rates and to the ratios of nitrified N emitted as N2O among soil layers. Furthermore, NO fluxes were positively correlated with the changes in gross nitrification rates and the ratios of NO/N2O in the two paddy soils were always greater than one(from 1.26 to 6.47). | 9 | Hydrolyzed seawater pearl tablet modulates the immunity via attenuating Th1/Th2 imbalance in an immunosuppressed mouse model显示文摘OBJECTIVE: To investigate whether Hydrolyzed Seawater Pearl tablet(HSPT) could modulate the Th1/Th2 imbalance in an immunosuppressed mouse model with Th1 to Th2 shift induced by Cyclosporine A(Cs A) which can be used in the clinical treatment of Th2 to Th1 shift diseases, and explore the possible mechanism for the adjuvant therapeutic efficacy of HSPT on recurrent respiratory infections(RRI) and acquired immune deficiency syndrome(AIDS).METHODS: The mice were randomly divided into six groups of five animals each, namely normal group, model group, lentinan polysaccharide tablet(LPT) group and three HPST treated groups. HPST treated groups were administered with HPST(0.51,1.02, 2.04 g/kg) via intragastric gavage(i.g) for 30 consecutive days. LPT used as reference drug for positive control, LPT group was administered with LPT(8.2 mg/kg) for 30 consecutive days. Normal group and model group were received distilled water. The animals in model group, LPT group and HPST treated groups were injected intraperitoneally with Cs A(50 mg/kg) to establish the immunosuppressed mice model with Th1 to Th2 shift on the20 th, 22 nd and 24 th day, one hour after the administration of the respective treatment. Animals were sacrificed one hour after the last administration to collect blood and splenic tissue. The proportion of T cells including CD8+ and CD4+ T cells, Th1 and Th2 in peripheral blood of experimental mice were measured by flow cytometric. The protein level in serum and m RNA level in splenic tissue of experimental mice for interleukin(IL)-2, IL-12, interferon-γ(IFN-γ), IL-4, IL-6, IL-10 and IL-13 were measured by enzyme linked immunosorbent assay and fluorescence quantitative polymerase chain reaction respectively.RESULTS: HSPT elevated the proportion of T cells including both CD8+ and CD4+ T cells, in which the proportion of Th1 and Th2 cells increased, while the ratio of Th1/Th2 cells decreased in peripheral blood of the immunosuppressed mouse model with Th1 to Th2 shift induced by Cs A. | 14 | AlN Precipitates and Microstructure in Non-oriented Electrical Steels Produced by Twin-roll Casting Process显示文摘Aluminum nitride(AlN) precipitates and microstructure of 4 wt.%(Si+Al) non-oriented electrical steel were investigated.The 2.0 mm thick cast strips with three different silicon/aluminum(Si/Al) ratios were produced by twin-roll casting process,then the strips were reheated,warm rolled,cold rolled and annealed.The microstructure and AlN precipitates were characterized using optical microscopy,scanning electron microscopy and transmission electron microscopy.The results showed that with the increase of Si/Al ratio,on the one hand,the casting microstructure changed from columnar grains to equiaxed grains,and the uniformity of annealed microstructure was improved;On the other hand,the number of AlN precipitates in cast strips reduced meanwhile the distribution became dispersed.By the reheat treatment,the size and distribution of the AlN precipitates can be changed.Moreover,the grain size of the annealed strips is in the range of 20-50 μm,at the same time,many AlN precipitates were located at grain boundaries.Therefore,controlling the Si/Al ratio is a simple method to obtain desired microstructure.Then AlN precipitates in non-oriented electrical steel prepared by twin-roll casting process hinder markedly the recrystallized grains growth.A compatible reheat treatment can be an approach worth exploring to control the behavior of AlN precipitates. | Ting ZHANG Xiaoming ZHANG Zhiyuan GUO Yuqian WANG Chenggang LI Liangyun LAN | 2013 | Acta Metallurgica Sinica(English Letters)2013,26,4: | 5 |
| 15 | CDER167,a dual inhibitor of URAT1 and GLUT9,is a novel and potent uricosuric candidate for the treatment of hyperuricemia显示文摘Urate transporter 1(URAT1)and glucose transporter 9(GLUT9)are important targets for the development of uric acid-lowering drugs.We previously showed that the flexible linkers of URAT1 inhibitors could enhance their potency.In this study we designed and synthesized CDER167,a novel RDEA3710 analogue,by introducing a linker(methylene)between the naphthalene and pyridine rings to increase flexibility,and characterized its pharmacological and pharmacokinetics properties in vitro and in vivo.We showed that CDER167 exerted dual-target inhibitory effects on both URAT1 and GLUT9:CDER167 concentration-dependently inhibited the uptake of[^(14)C]-uric acid in URAT1-expressing HEK293 cells with an IC_(50) value of 2.08±0.31μM,which was similar to that of RDEA3170(its IC_(50) value was 1.47±0.23μM).Using site-directed mutagenesis,we demonstrated that CDER167 might interact with URAT1 at S35 and F365.In GLUT9-expressing HEK293T cells,CDER167 concentration-dependently inhibited GLUT9 with an IC_(50) value of 91.55±15.28μM,whereas RDEA3170 at 100μM had no effect on GLUT9.In potassium oxonate-induced hyperuricemic mice,oral administration of CDER167(10 mg·kg^(−1)·d^(−1))for 7 days was more effective in lowering uric acid in blood and significantly promoted uric acid excretion in urine as compared with RDEA3170(20 mg·kg^(−1)·d^(−1))administered.The animal experiment proved the safety of CDER167.In addition,CDER167 displayed better bioavailability than RDEA3170,better metabolic stability and no hERG toxicity at 100μM.These results suggest that CDER167 deserves further investigation as a candidate antihyperuricemic drug targeting URAT1 and GLUT9. | Ze-an Zhao Yu Jiang Yan-yu Chen Ting Wu Qun-sheng Lan Yong-mei Li Lu Li Yang Yang Cui-ting Lin Ying Cao Ping-zheng Zhou Jia-yin Guo Yuan-xin Tian Jian-xin Pang | 2022 | Acta Pharmacologica Sinica2022,43,1: | 4 |
| 16 | Highly Selective Biomimetic Flexible Tactile Sensor for Neuroprosthetics显示文摘Biomimetic flexible tactile sensors endow prosthetics with the ability to manipulate objects,similar to human hands.However,it is still a great challenge to selectively respond to static and sliding friction forces,which is crucial tactile information relevant to the perception of weight and slippage during grasps.Here,inspired by the structure of fingerprints and the selective response of Ruffini endings to friction forces,we developed a biomimetic flexible capacitive sensor to selectively detect static and sliding friction forces.The sensor is designed as a novel plane-parallel capacitor,in which silver nanowire-3D polydimethylsiloxane(PDMS)electrodes are placed in a spiral configuration and set perpendicular to the substrate.Silver nanowires are uniformly distributed on the surfaces of 3D polydimethylsiloxane microcolumns,and silicon rubber(Ecoflex^(■))acts as the dielectric material.The capacitance of the sensor remains nearly constant under different applied normal forces but increases with the static friction force and decreases when sliding occurs.Furthermore,aiming at the slippage perception of neuroprosthetics,a custom-designed signal encoding circuit was designed to transform the capacitance signal into a bionic pulsed signal modulated by the applied sliding friction force.Test results demonstrate the great potential of the novel biomimetic flexible sensors with directional and dynamic sensitivity of haptic force for smart neuroprosthetics. | Yue Li Zhiguang Cao Tie Li Fuqin Sun Yuanyuan Bai Qifeng Lu Shuqi Wang Xianqing Yang Manzhao Hao Ning Lan Ting Zhang | 2020 | Research2020,,1: | 4 |

| 18 | Efficacy of water fraction from Dioscorea cirrhosa on oxidative stress and apoptosis in H9c2 cardiomyocytes induced by H2O2显示文摘OBJECTIVE:To investigate the efficacy of water fraction from Dioscorea cirrhosa(WF)on oxidative damage and apoptosis of cardiomyocytes induced by H2O2,and to study its mechanism.METHODS:Cell viability was measured by the MST assay kit.The content of malondialdehyde(MDA),release of lactate dehydrogenase(LDH)and activity of catalase(CAT)and superoxide dismutase(SOD)were detected by biochemical kit.The content of reactive oxygen species(ROS)was assessed by nonfluorescent probe 2′,7′-dichlorofluorescin diacetate(DCFH-DA).JC-1 was used to analyze the mitochondrial membrane potential(mtΔΨ)and Annexin-V-FITC/PI staining was applied to assess apoptosis of H9c2 by flow cytometry.Moreover,the expression of B-cell lymphoma-2(Bcl-2),Bcl-2-associated X(Bax),caspase-3,caspase-9,cleaved-caspase-3 and cleaved-caspase-9 proteins was determined by western blot analysis.RESULTS:WF increased cell viability and decreased LDH leakage in H9c2 cells exposed to H2O2.WF treatment decreased ROS and MDA level,enhanced SOD and CAT activities,improved mtΔΨand inhibited apoptosis.Western blot analysis demonstrated that the ratio of Bcl-2/Bax was increased and the expression cleaved-caspase-3,caspase-3,cleaved-caspase-9 and caspase-9 were decreased in group treated with WF.CONCLUSION:WF protects H9c2 myocardial cells on H2O2-induced oxidative stress and apoptosis by scavenging ROS,improving antioxidant capacity,protecting mitochondrial and regulating the proteins expression related to apoptosis. | LIU Chunhua PAN Jie WANG Aimin LAN Yanyu LI Yongjun LU Yuan SUN Jia WANG Yonglin LIU Ting LU Dingyan YOU Jingrui | 2021 | Journal of Traditional Chinese Medicine2021,41,1: | 4 |
| 19 | Si substituted hydroxyapatite nanorods on Ti for percutaneous implants显示文摘An ideal intraosseous transcutaneous implant should form a tight seal with soft tissue,besides a requirement of osseointegration at the bone-fixed position.Si substituted hydroxyapatite(Si-HA)nanorods releasing Si ion and simulating nanotopography of natural tissue were designed on Ti to enhance fibroblast response in vitro and biosealing with soft tissue in vivo.Si-HA nanorods were fabricated by alkali-heat treatment followed with hydrothermal treatment.The hydrothermal formation mechanism of Si-HA nanorods was explored.The surface characteristic of Si-HA nanorods was compared with pure HA nanorods.Fibroblast behaviors in vitro and skin response in vivo on different surfaces were also evaluated.The obtained results show that the substitution of Si did not significantly alter the phase component,morphology,roughness and wettability of HA,but additional Si and more Ca were released from Si-HA into medium.Comparing to pure HA nanrods and Ti substrate,Si-HA nanrods enhanced cell behaviors including proliferation,fibrotic phenotype and collagen secretion in vitro,and reduced epithelial down growth in vivo.The enhanced cell response and biosealing should be due to the releasing of Ca,Si and nanotopography of Si-HA nanorods.Si-HA nanorods can be a potential coating to accelerate skin integration for percutaneous implants in clinic. | Kai Li Yang Xue Ting Yan Lan Zhang Yong Han | 2020 | Bioactive Materials2020,5,1: | 4 |
| 20 | A 6.2–9.5 GHz receiver for Wimedia MB-OFDM and China UWB standard显示文摘An integrated 6.2–9.5 GHz UWB receiver for both WiMedia MB-OFDM and China UWB standard is proposed. The receiver can cover Group#3, Group#4 for WiMedia and Group#2–5 for China UWB standard respectively. The fully differential receiver consists of a wideband LNA and a down conversion mixer with high gain and low gain mode. A low-pass filter with controllable gain and cut-o? frequency along with a programmable gain amplifier are also included. The chip was fabricated in TSMC 0.13-μm RF CMOS process. Measurement results show that the receiver achieves voltage gain from 21 to 69 dB with 2 dB/step and an NF of 3.2–4.6 dB over 6.2–9.5 GHz and 1-dB compression point of ?21 dBm. The intermediate frequency of the receiver can be switched between 264 and 132 MHz, which enables the receiver applicable for both WiMedia and China UWB standard.
