1The Predictive Value of Baseline HBs Ag Level and Early Response for HBs Ag Loss in Patients with HBe Ag-positive Chronic Hepatitis B during Pegylated Interferon Alpha-2a Treatment显示文摘Objective To explore the predictive value of baseline HBsAg level and early response for HBsAg loss in patients with HBeAg-positive chronic hepatitis B during pegylated interferon alpha-2a treatment. Methods A total of 121 patients with HBeAg-positive chronic hepatitis B who achieved HBsAg loss were enrolled; all patients were treated with PEG-IFNα-2a 180 μg/week. Serum HBV DNA and serological indicators(HBsAg, anti-HBs, HBeAg, and anti-HBe) were determined before and every 3 months during treatment. Results The median treatment time for HBsAg loss was 84 weeks(7-273 weeks), and 74.38%(90 cases) of the patients needed extended treatment(> 48 weeks). The correlation between baseline HBsAg levels and the treatment time of HBsAg loss was significant(B = 14.465, t = 2.342, P = 0.021). Baseline HBsAg levels together with the decline range of HBsAg at 24 weeks significantly correlated with the treatment time of HBsAg loss(B = 29.862, t = 4.890, P = 0.000 and B = 27.993, t = 27.993, P = 0.005). Conclusion Baseline HBsAg levels and extended therapy are critical steps toward HBsAg loss. 3BLT1 in dendritic cells promotes Th1/Th17 differentiation and its deficiency ameliorates TNBS-induced colitis显示文摘Leukotriene B4(LTB4)synthesis is enhanced in the colonic mucosa in patients with inflammatory bowel disease(IBD).BLT1,a highaffinity receptor for LTB4,exhibits no effect on the progression of dextran sodium sulfate(DSS)-induced colitis,which mostly relies on innate immunity.Here,we reported that BLT1 regulates trinitrobenzene sulfonic acid(TNBS)-induced colitis,which reflects CD4+T-cell-dependent adaptive immune mechanisms of IBD.We found that BLT1 signaling enhanced the progression of colitis through controlling the production of proinflammatory cytokines by dendritic cells(DCs)and modulating the differentiation of Th1 and Th17.BLT1−/−mice displayed an alleviated severity of TNBS-induced colitis with reduced body weight loss and infiltrating cells in the lamina propria.BLT1 deficiency in DCs led to reduced production of proinflammatory cytokines,including IL-6,TNF-α,and IL-12,and these results were further confirmed via treatment with a BLT1 antagonist.The impaired cytokine production by BLT1−/−DCs subsequently led to reduced Th1 and Th17 differentiation both in vitro and in vivo.We further performed a conditional DC reconstitution experiment to assess whether BLT1 in DCs plays a major role in regulating the pathogenesis of TNBS-induced colitis,and the results indicate that BLT1 deficiency in DCs also significantly reduces disease severity.The mechanistic study demonstrated that BLT1-regulated proinflammatory cytokine production through the Gαiβγsubunit-phospholipase Cβ(PLCβ)-PKC pathway.Notably,we found that treatment with the BLT1 antagonist also reduced the production of proinflammatory cytokines by human peripheral blood DCs.Our findings reveal the critical role of BLT1 in regulating adaptive immunity and TNBS-induced colitis,which further supports BLT1 as a potential drug target for adaptive immunity-mediated IBD.Jinfeng Zhou Weiming Lai Wanjie Yang Juping Pan Hu Shen Yingying Cai Cuixia Yang Ningjia Ma Yue Zhang Ru Zhang Xin Xie Zhongjun Dong Yuan Gao Changsheng Du 2018Cellular & Molecular Immunology2018,15,12:9

7Research on Hepatitis B virus Genotypes and Subgenotypes among Bai Nationality in Dali, Yunnan Province显示文摘To investigate the distribution of hepatitis B virus (HBV) genotypes and subgenotypes among the Bai nationality in Dali, a total of 100 serum samples from patients with chronic HBV-infection were collected for the detection of HBV genotypes and subgenotypes by genotype-specific primers and restriction fragment length polymorphism (RLFP), respectively. Among the 100 samples, the proportions of genotype B, C and mixed genotype (B+C) were 41%, 25% and 34%, respectively. All the genotype B strains belonged to subgenotype Ba. In genotype C, 84% were Subgenotype Cs and 12% were subgenotype Ce. The distribution of genotypes B, C and B+C showed no significant difference between male and female patients (P=0.182) and among the age groups of patients (P=0.812). The rates of HBeAg/HBeAg positivity were no significantly different among genotypes B, genotype C and mixed genotype (B+C) (P=0.077/P=0.663). In Dali, genotypes B, B+C and C existed among Bai nationality with chronic HBV-infection, and genotype B was the major genotype. Subgenotypes Ba and Cs were the predominant strains in patients with HBV genotype B/C infection. 8Homoharringtonine is a safe and effective substitute for anthracyclines in children younger than 2 years old with acute myeloid leukemia显示文摘Homoharringtonine (HHT),a plant alkaloid from Cephalotaxus harringtonia,exhibits a unique anticancer mechanism and has been widely used in China to treat patients with acute myeloid leukemia (AML) since the 1970s.Trial SCMC-AML-2009 presented herein was a randomized clinical study designed based on our previous findings that pediatric AML patients younger than two years old may benefit from HHT-containing chemotherapy regimens.Patients randomized to arm A were treated with a standard chemotherapy regimen comprising mainly of anthracyclines and cytarabine (Ara-C),whereas patients in arm B were treated with HHTcontaining regimens in which anthracyclines in all but the initial induction therapy were replaced by HHT.From February 2009 to November 2015,59 patients less than 2 years old with de novo AML (other than acute promyelocytic leukemia) were recruited.A total of 42 patients achieved a morphologic complete remission (CR) after the first course,with similar rates in both arms (70.6% vs.72.0%).At the end of the follow-up period,40 patients remained in CR and 5 patients underwent hematopoietic stem cell transplantation in CR,which could not be considered as events but censors.The 5-year event-free survival (EFS) was 60.2%± 9.6% for arm A and 88.0%± 6.5% for arm B (P=0.024).Patients in arm B experienced shorter durations of leukopenia,neutropenia,and thrombocytopenia and had a lower risk of infection during consolidation chemotherapy with high-dosage Ara-C.Consequently,the homoharringtonine-based regimen achieved excellent EFS and alleviated hematologic toxicity for children aged younger than 2 years with de novo AML compared with the anthracycline-based regimen.Xiaoxiao Chen Yanjing Tang Jing Chen Ru Chen Longjun Gu Huiliang Xue Ci Pan Jingyan Tang Shuhong Shen 2019Frontiers of Medicine2019,13,3:3

10MoS2/C/C nanofiber with double-layer carbon coating for high cycling stability and rate capability in lithium-ion batteries显示文摘MoS2 has attracted a lot of interest in the field of lithium-ion storage as ananode material owing to its high capacity and two-dimensional (2D)-layerstructure. However, its electrochemical properties, such as rate capability andcycling stability, are usually limited by its low conductivity, volume variation, andpolysulfide dissolution during lithiation/delithiation cycling. Here, a designedtwo-layer carbon-coated MoS2/carbon nanofiber (MoS2/C/C fiber) hybrid electrodewith a double-layer carbon coating was achieved by a facile hydrothermal andsubsequent electrospinning method. The double carbon layer (inner amorphouscarbon and outer carbon fiber) shells could efficiently increase the electronconductivity, prevent the aggregation of MoS2 flakes, and limit the volumechange and polysulfide loss during long-term cycling. The as-prepared MoS2/C/Cfiber electrode exhibited a high capacity of up to 1,275 mAh/g at a currentdensity of 0.2 A/g, 85.0% first cycle Coulombic efficiency, and significantlyincreased rate capability and cycling stability. 11Two-dimensional correlation analysis of continuous online in situ ATR-FTIR on the adsorption of butyl xanthate at the surface of α-PbO显示文摘The adsorption behavior of butyl xanthate on the surface of lead oxide was investigated using continuous online in situ attenuated total reflectance Fourier transform infrared(ATR-FTIR) spectroscopy technique and two dimensional(2D) correlation analysis.The adsorbed layer studied was prepared by coating α-PbO particles onto the surfaces of the ZnSe crystal.The appearance of spectral peaks at 1203 cm^(-1),1033 cm^(-1) and their red shift indicated the formation and aggregation of xanthate at the surface of α-PbO.According to 1R intensity changes after rinsing with deionized water and a NaOH solution,the adsorption was proved to be a chemisorption type.The competition between xanthate and OH for the surfaces leads to desorption of xanthate at higher pH.The technique of 2D correlation ATR-FTIR spectroscopy was used to evaluate the changing order of spectral intensities in the adsorption process,and the results indicated that xanthate micelles were formed at the surfaces.The adsorption kinetics of butyl xanthate was found to be a pseudo-second-order reaction model and the adsorption capacity of butyl xanthate at α-PbO was as high as 281 mg g^(-1) after 150 min.Qi Shen Ying-Ju Fan Wei-Min Zhang Bo-Li Zhu Ru Wang Zhong-Xi Sun 2015Chinese Chemical Letters2015,26,2:2
12High proportion strontium-doped micro-arc oxidation coatings enhance early osseointegration of titanium in osteoporosis by anti-oxidative stress pathway显示文摘The excessive accumulation of reactive oxygen species(ROS)under osteoporosis precipitates a microenvironment with high levels of oxidative stress(OS).This could significantly interfere with the bioactivity of conventional titanium implants,impeding their early osseointegration with bone.We have prepared a series of strontium(Sr)-doped titanium implants via micro-arc oxidation(MAO)to verify their efficacy and differences in osteoinduction capabilities under normal and osteoporotic(high OS levels)conditions.Apart from the chemical composition,all groups exhibited similar physicochemical properties(morphology,roughness,crystal structure,and wettability).Among the groups,the low Sr group(Sr25%)was more conducive to osteogenesis under normal conditions.In contrast,by increasing the catalase(CAT)/superoxide dismutase(SOD)activity and decreasing ROS levels,the high Sr-doped samples(Sr75% and Sr100%)were superior to Sr25% in inducing osteogenic differentiation of MC3T3-E1 cells and the M2 phenotype polarization of RAW264.7 cells,thus enhancing early osseointegration.Furthermore,the results of both in vitro cell co-culture and in vivo studies also showed that the high Sr-doped samples(especially Sr100%)had positive effects on osteoimmunomodulation under the OS microenvironment.Ultimately,the collated findings indicated that the high proportion Sr-doped MAO coatings were more favorable for osteoporosis patients in implant restorations.Xinkun Shen Kai Fang Kendrick Hii Ru Yie Zixin Zhou Yiding Shen Shuyi Wu Yue Zhu Zhennan Deng Pingping Ma Jianfeng Ma Jinsong Liu 2022Bioactive Materials2022,7,4:2

16Phase Analysis of Titanium in Steel and Some Aspects on Their Application显示文摘Phase analysis of titanium in steel has been carried out on thirty samples with different Ti-content from 0.036%- 0.204%.The various heat treatments for the samples are to anneal at 1050-1200 C for a while.followed by cooling at a rate of 1,10.20 ℃/s.respectively.It is achieved to separate and determine fine particle TiC.coarse particle TiC,Ti_2CS and TiN extracted from steel by controlling the oxidation potential and acidity of solutions.The results of quantitative analysis of the individual phase show that 0.09%— 0.13%Ti-content is enough to eliminate free nitrogen and transform MnS into Ti_2CS in steel effectively.In the case of0.06%-0.10%Ti-content combined with annealing at 1050 C and quick cooling at 20 ℃/s,a proper amount of fine particle TiC can be precipitated and the rate of TiC(fine)/TiC(total)is high,which are beneficial to increase strength and toughness of steel.More Ti-content or cooling slowly will precipitate more coarse particle TiC which decreaces toughness severely.The proposed methods for quantitative phase analysis of titanium in steel are given in detail.CHEN Ming Hao and SHEN Ru Mei 1994Journal of Iron and Steel Research(International)1994,1,1:1
17Microarray gene expression analysis of chemosensitivity for docetaxel,cisplatin and 5-fluorouracil(TPF)combined chemotherapeutic regimen in hypopharyngeal squamous cell carcinoma显示文摘Objective: To screen out a set of candidate genes which could help to determine whether patients with hypopharyngeal squamous cell carcinoma(HSCC) could benefit from docetaxel, cisplatin and 5-fluorouracil(TPF)induction chemotherapy.Methods: Gene-expression profiles in 12 TPF-sensitive patients were compared to 9 resistant controls by microarray analysis. Subsequently, expression levels of potential biomarkers in chemosensitive cell line Fa Du after TPF treatment were observed by quantitative real-time polymerase chain reaction(q RT-PCR).Results: Through microarray analysis, 1,579 differentially expressed genes were identified, of which 815 were up-regulated in TPF chemotherapy-responsive tissues whereas 764 were down-regulated. Gene ontology(GO)analysis suggested these genes participating in physiological processes including transcription and its regulation,cellular signal transduction and metabolic process. Additionally, Kyoto Encyclopedia of Genes and Genomes(KEGG) database revealed that MAPK and Jat/STAT signaling pathways occupied important roles in TPF chemotherapeutic sensitivity. Moreover, in vitro cell culture experiments revealed the expression alternations of IL-6, MAPK14, JUN, CDK5 and CAMK2 A exposed to TPF treatment by q RT-PCR, whilst providing an insight into the mechanism underlying TPF chemotherapeutic response in HSCC.Conclusions: These results provided a battery of genes related to TPF chemotherapeutic sensitivity and might act as molecular targets in HSCC treatment.
