1Integrative lipidomic and transcriptomic study unravels the therapeutic effects of saikosaponins A and D on non-alcoholic fatty liver disease显示文摘Nonalcoholic fatty liver disease(NAFLD)has become one of the most prominent causes of chronic liver diseases and malignancies.However,few therapy has been approved.Radix Bupleuri(RB)is the most frequently used herbal medicine for the treatment of liver diseases.In the current study,we aim to systemically evaluate the therapeutic effects of saikosaponin A(SSa)and saikosaponin D(SSd),the major bioactive monomers in RB,against NAFLD and to investigate the underlying mechanisms.Our results demonstrated that both SSa and SSd improved diet-induced NAFLD.Integrative lipidomic and transcriptomic analysis revealed that SSa and SSd modulated glycerolipid metabolism by regulating related genes,like Lipe and Lipg.SSd profoundly suppressed the fatty acid biosynthesis by downregulating Fasn and Acaca expression and promoted fatty acid degradation by inducing Acox1 and Cpt1 a expression.Bioinformatic analysis further predicted the implication of master transcription factors,including peroxisome proliferator-activated receptor alpha(PPARα),in the protective effects of SSa and SSd.These results were further confirmed in vitro in mouse primary hepatocytes.In summary,our study uncoded the complicated mechanisms underlying the promising anti-steatosis activities of saikosaponins(SSs),and provided critical evidence inspiring the discovery of innovative therapies based on SSa and SSd for the treatment of NAFLD and related complications.Xiaojiaoyang Li Junde Ge Yajing Li Yajie Cai Qi Zheng Nana Huang Yiqing Gu Qi Han Yunqian Li Rong Sun Runping Liu 2021Acta Pharmaceutica Sinica B2021,11,11:15
2Exploration of spatial and temporal characteristics of PM2.5 concentration in Guangzhou, China using wavelet analysis and modified land use regression model显示文摘This article attempts to detail time series characteristics of PM2.5 concentration in Guangzhou(China)from 1 June 2012 to 31 May 2013 based on wavelet analysis tools,and discuss its spatial distribution using geographic information system software and a modified land use regression model.In this modified model,an important variable(land use data)is substituted for impervious surface area,which can be obtained conveniently from remote sensing imagery through the linear spectral mixture analysis method.Impervious surface has higher precision than land use data because of its sub-pixel level.Seasonal concentration pattern and day-by-day change feature of PM2.5 in Guangzhou with a micro-perspective are discussed and understood.Results include:(1)the highest concentration of PM2.5 occurs in October and the lowest in July,respectively;(2)average concentration of PM2.5 in winter is higher than in other seasons;and(3)there are two high concentration zones in winter and one zone in spring.Fenglei Fan Runping Liu 2018Geo-Spatial Information Science2018,21,4:2
3A novel pulmonary fibrosis murine model with immune-related liver injury显示文摘Idiopathic pulmonary fibrosis(IPF),characterized by aggravated alveolar destruc-tion and fibrotic matrix deposition,tendentiously experiences the stage called acute exacerbation IPF(AE-IPF)and progresses to multiple organ damage,especially liver injury.Recent studies have found a variety of immune microenvironment disorders associated with elevated IPF risk and secondary organ injury,whereas current animal models induced with bleomycin(BLM)could not completely reflect the pathologi-cal manifestations of AE-IPF patients in clinic,and the exact underlying mechanisms are not yet fully explored.In the current study,we established an AE-IPF model by tracheal administration of a single dose of BLM and then repeated administrations of lipopolysaccharide in mice.This mouse model successfully recapitulated the clinical features of AE-IPF,including excessive intrapulmonary inflammation and fibrosis and extrapulmonary manifestations,as indicated by significant upregulation of Il6,Tnfa,Il1b,Tgfb,fibronectin,and Col1a1 in both lungs and liver and elevated serum aspartate transaminase and alanine transaminase levels.These effects might be attributed to the regulation of Th17 cells.By sharing this novel murine model,we expect to pro-vide an appropriate experimental platform to investigate the pathogenesis of AE-IPF coupled with liver injury and contribute to the discovery and development of targeted interventions.Kexin Jia Jianzhi Wu Yijie Li Jia Liu Runping Liu Yajie Cai Yinhao Zhang Xiaojiaoyang Li 2023Animal Models and Experimental Medicine2023,6,3:1

9Edge-enriched N, S co-doped hierarchical porous carbon for oxygen reduction reaction显示文摘The development of carbon materials with high electrochemical performance for next-generation energy device is emerging, especially N, S co-doped carbon materials have sparked intensive attention. However,the exploration of N, S co-doped carbon with well-defined active sites and hierarchical porous structures are still limited. In this study, we prepared a series of edge-enriched N, S co-doped carbon materials through pyrolysis of thiourea(TU) encapsulated in zeolitic imidazolate frameworks(TU@ZIF) composites,which delivered very good oxygen reduction reaction(ORR) performance in alkaline medium with onset potential of 0.94 V vs. reversible hydrogen electrode(RHE), good stability and methanol tolerance. Density functional theory(DFT) calculations suggested that carbon atoms adjacent to N and S are probable active sites for ORR intermediates in edge-enriched N, S co-doped carbon materials because higher electron density can enhance O_(2)adsorption, lower formation barriers of intermediates, improving the ORR performance comparing to intact N, S co-doped carbon materials. 10Optimizing diabetic kidney disease animal models:Insights from a meta-analytic approach显示文摘Diabetic kidney disease(DKD)is a prevalent complication of diabetes,often leading to end-stage renal disease.Animal models have been widely used to study the pathogenesis of DKD and evaluate potential therapies.However,current animal models often fail to fully capture the pathological characteristics of renal injury observed in clinical patients with DKD.Additionally,modeling DKD is often a time-consuming,costly,and labor-intensive process.The current review aims to summarize modeling strategies in the establishment of DKD animal models by utilizing meta-analysis related methods and to aid in the optimization of these models for future research.A total of 1215 articles were retrieved with the keywords of“diabetic kidney disease”and“animal experiment”in the past 10 years.Following screening,84 articles were selected for inclusion in the meta-analysis.Review manager 5.4.1 was employed to analyze the changes in blood glucose,glycosylated hemoglobin,total cholesterol,triglyceride,serum creatinine,blood urea nitrogen,and urinary albumin excretion rate in each model.Renal lesions shown in different models that were not suitable to be included in the metaanalysis were also extensively discussed.The above analysis suggested that combining various stimuli or introducing additional renal injuries to current models would be a promising avenue to overcome existing challenges and limitations.In conclusion,our review article provides an in-depth analysis of the limitations in current DKD animal models and proposes strategies for improving the accuracy and reliability of these models that will inspire future research efforts in the DKD research field.Fanghong Li Zhi Ma Yajie Cai Jingwei Zhou Runping Liu 2023Animal Models and Experimental Medicine2023,6,5:0
11Modulation of type I interferon signaling by natural products in the treatment of immune-related diseases显示文摘Type I interferon(IFN)is considered as a bridge between innate and adaptive immunity.Proper activation or inhibition of type I IFN signaling is essential for host defense against pathogen invasion,tumor cell proliferation,and overactive immune responses.Due to intricate and diverse chemical structures,natural products and their derivatives have become an invaluable source inspiring innovative drug discovery.In addition,some natural products have been applied in clinical practice for infection,cancer,and autoimmunity over thousands of years and their promising curative effects and safety have been well-accepted.However,whether these natural products are primarily targeting type I IFN signaling and specific molecular targets involved are not fully elucidated.In the current review,we thoroughly summarize recent advances in the pharmacology researches of natural products for their type I IFN activity,including both agonism/activation and antagonism/inhibition,and their potential application as therapies.Furthermore,the source and chemical nature of natural products with type I IFN activity are highlighted and their specific molecular targets in the type I IFN pathway and mode of action are classified.In conclusion,natural products possessing type I IFN activity represent promising therapeutic strategies and have a bright prospect in the treatment of infection,cancer,and autoimmune diseases.LI Shuo FAN Guifang LI Xiaojiaoyang CAI Yajie LIU Runping 2023Chinese Journal of Natural Medicines2023,21,1:0
12Design of mesoporous ZnCoSiOx hollow nanoreactors with specific spatial distribution of metal species for selective CO_(2)hydrogenation显示文摘In heterogeneous catalysis,the precise placement of active components to perform unique functions in cooperation with each other is a tremendous challenge.The migration of matter on micro/nano-scale caused by diffusion is a promising pathway for design of catalytic nanoreactors with precise active sites location and controllable microenvironment through compartmentalization and confinement effects.Herein,we report two categories of mesoporous ZnCoSiOx hollow nanoreactors with different metal distributions and microenvironment engineered by the diffusion behavior of metal species in confined nanospace.Double-shelled hollow structures with well-distributed metal species were obtained by adopting core@shell structured ZnCo-zeolitic imidazolate framework(ZIF)@SiO2 as a template and employing three stages of hydrothermal treatment including the decomposition of ZIF,diffusion of metal species into the silica shell,and Ostwald ripening.Additionally,the formation of yolk@shell structure with a collective(Zn-Co)metal oxide as the yolk was achieved by direct pyrolysis of ZnCo-ZIF@SiO2.In CO_(2)hydrogenation,ZnCoSiOx with double-shelled hollow structures and yolk@shell structures respectively afford CO and CH_(4)as main product,which is related with different dispersion and location of active sites in the two catalysts.This study provides an efficient method for the synthesis of catalytic nanoreactors on the basis of insights of the atomic diffusion in confined space at the mesoscale.Xinyao Wang Runping Ye Melis S.Duyar Cameron Alexander Hurd Price Hao Tian Yanping Chen Na Ta Hao Liu Jian Liu 2023Nano Research2023,16,4:0
13Epigenetic modification in liver fibrosis:Promising therapeutic direction with significant challenges ahead显示文摘Liver fibrosis,characterized by scar tissue formation,can ultimately result in liver failure.It’s a major cause of morbidity and mortality globally,often associated with chronic liver diseases like hepatitis or alcoholic and non-alcoholic fatty liver diseases.However,current treatment options are limited,highlighting the urgent need for the development of new therapies.As a reversible regulatory mechanism,epigenetic modification is implicated in many biological processes,including liver fibrosis.Exploring the epigenetic mechanisms involved in liver fibrosis could provide valuable insights into developing new treatments for chronic liver diseases,although the current evidence is still controversial.This review provides a comprehensive summary of the regulatory mechanisms and critical targets of epigenetic modifications,including DNA methylation,histone modification,and RNA modification,in liver fibrotic diseases.The potential cooperation of different epigenetic modifications in promoting fibrogenesis was also highlighted.Finally,available agonists or inhibitors regulating these epigenetic mechanisms and their potential application in preventing liver fibrosis were discussed.In summary,elucidating specific druggable epigenetic targets and developing more selective and specific candidate medicines may represent a promising approach with bright prospects for the treatment of chronic liver diseases.Runping Liu Yajing Li Qi Zheng Mingning Ding Huiping Zhou Xiaojiaoyang Li 2024Acta Pharmaceutica Sinica B2024,14,3:0

16Waist circumference cutoff points for Chinese adults with metabolic syndrome defined by the International Diabetes Federation显示文摘Objective To investigate the appropriate waist circumference (WC) cutoff points for central obesity in the middle-aged and elderly Beijing residents by the metabolic syndrome definition of the International Diabetes Federation (IDF). Methods A total of 2,344 Beijing residents aged ≥40 years were investigated. They answered questionnaires, received physical examinations, and underwent plasma glucose and lipid profile measurement. Those non-diabetic subjects underwent a 75g oral glucose tolerance test. All data were analyzed to calculate the appropriate WC cutoff points for central obesity reaching the diagonsis of MS. Results 1) Both in males and females, the triglyceride (TG), systolic blood pressure, diastolic blood pressure and fasting plasma glucose (FPG) increased linearly with WC, and the high density lipoprotein cholesterol (HDL-C) decreased linearly with WC (P<0.05). 2)The prevalence of elevated TG, reduced HDL-C, elevated blood pressure, elevated FBG, or ≥ 2 of these factors increased with WC (P<0.05). 3) Based on the receiver operating characteristic (ROC) curve analysis and Youden index, the WC values for central obesity and for detecting BMI ≥ 25 kg/m2 were about 90 cm for men and 80 cm for women. 4) The odds ratio for the presence of two or more metabolic risk factors increased abruptly in men with WC ≥ 90 cm and in women with WC ≥ 80 cm.
