1Endothelial cells-targeted soluble human Delta-like 4 suppresses both physiological and pathological ocular angiogenesis显示文摘Due to its essential roles in angiogenesis, Notch pathway has emerged as an attractive target for the treatment of pathologic angiogenesis. Although both activation and blockage of Notch signal can impede angiogenesis, activation of Notch signal may be more promising because it was shown that long-term Notch signal blockage resulted in vessel neoplasm. However, an in vivo deliverable Notch ligand with highly efficient Notch-activating capacity has not been developed. Among all the Notch ligands, Delta-like4(Dll4) is specifically involved in angiogenesis. In this study, we generated a novel soluble Notch ligand h D4 R, which consists of the Delta-Serrate-Lag-2 fragment of human Dll4 and an arginine-glycine-aspartate(RGD) motif targeting endothelial cells(ECs). We demonstrated that h D4 R could bind to ECs through its RGD motif and effectively triggered Notch signaling in ECs. Further, we confirmed that h D4 R could suppress angiogenesis in vitro as manifested by network formation assay and sprouting assay. More importantly, h D4 R efficiently repressed neonatal retinal angiogenesis and laser-induced choroidal neovascularization(CNV) as well in vivo. 2A hybrid genipin-crosslinked dual-sensitive hydrogel/nanostructured lipid carrier ocular drug delivery platform显示文摘The objective of this study was to develop a novel hybrid genipin-crosslinked dual-sensitive hydrogel/nanostructured lipid carrier(NLC) drug delivery platform. An ophthalmic antiinflammatory drug, baicalin(BN) was chosen as the model drug. BN –NLC was prepared using melt-emulsification combined with ultra-sonication technique. Additionally, a dual pH-and thermo-sensitive hydrogel composed of carboxymethyl chitosan(CMCS) and poloxamer 407(F127) was fabricated by a cross-linking reaction with a nontoxic crosslinker genipin(GP). GP-CMCS/F127 hydrogel was characterized by FTIR, NMR, XRD and SEM. The swelling studies showed GP-CMCS/F127 hydrogel was both pH-and thermo-sensitive. The results of in vitro release suggested BN –NLC gel can prolong the release of baicalin comparing with BN eye drops and BN –NLC. Ex vivo cornea permeation study was evaluated using Franz diffusion cells. The apparent permeability coefficient(Papp) of BN –NLC gel was much higher(4.46-fold) than that of BN eye drops. Through the determination of corneal hydration levels, BN –NLC gel was confirmed that had no significant irritation to cornea. Ex vivo precorneal retention experiments were carried out by a flow-through approach. The results indicated that the NLC-based hydrogel can prolong precorneal residence time. 3Influence of hydroxypropyl methylcellulose,methylcellulose, gelatin, poloxamer 407 and poloxamer 188 on the formation and stability of soybean oil-in-water emulsions显示文摘Macromolecules of polysaccharides, proteins and poloxamers have a hydrophobic portion and a hydrophilic one that can be used as emulsifiers. Parts of these emulsifiers are safe pharmaceutical excipients, which can replace the irritant low molecular weight surfactants to formulate emulsions for the pharmaceutical field. This project focused on preparing O/W emulsions stabilized with polymers for pharmaceuticals such as polysaccharides, proteins and poloxamers, including hydroxypropyl methylcellulose (HPMC), methylcellulose (MC),gelatin, poloxamer 407 (F127) and poloxamer 188 (F68). Emulsion physical stability was assessed by centrifugation, autoclaving sterilization and droplet size measurements. The stabilization mechanisms of emulsions were determined by interfacial tension and rheological measurements. 4Current sustained delivery strategies for the design of local neurotrophic factors in treatment of neurological disorders显示文摘Although therapeutic potential of neurotrophic factors(NTFs)has been well recognized for over two decades,attempts to translate that potential to the clinic have been disappointing,largely due to significant obstacles in delivery,including inadequate protein dose/kinetics released at target sites.Considerable efforts have been made to improve the therapeutic performance of NTFs.This articles reviews recent developments in localized delivery systems of NTFs for the neurological disorders treatments with a main focus on sustained delivery strategies.Different non-covalent binding approaches have been employed to immobilize proteins in hydrogels,microspheres,electrospun nanofibers,and their combined systems,which serve as depots for sustained local release of NTFs.The challenges associated with current NTFs delivery systems and how these systems can be applied to neurological diseases and disorders have been discussed in the review.In conclusion,optimal delivery systems for NTFs will be needed for reliable and meaningful clinical benefits;ideally,delivering a time and dose-controlled release of bioactive multiNTFs at different individual optimal kinetics to achieve multi-functions in target tissues is significant preferred.Hongzhuo Liu Yanyan Zhou Shichao Chen Meng Bu Jiayu Xin Sanming Li 2013Asian Journal of Pharmaceutical Sciences2013,8,5:2

9Chiral mesoporous silica nano-screws as an efficient biomimetic oral drug delivery platform through multiple topological mechanisms显示文摘In the microscale, bacteria with helical body shapes have been reported to yield advantages in many bio-processes. In the human society, there are also wisdoms in knowing how to recognize and make use of helical shapes with multi-functionality. Herein, we designed atypical chiral mesoporous silica nano-screws(CMSWs) with ideal topological structures(e.g., small section area, relative rough surface,screw-like body with three-dimension chirality) and demonstrated that CMSWs displayed enhanced bioadhesion, mucus-penetration and cellular uptake(contributed by the macropinocytosis and caveolaemediated endocytosis pathways) abilities compared to the chiral mesoporous silica nanospheres(CMSSs)and chiral mesoporous silica nanorods(CMSRs), achieving extended retention duration in the gastrointestinal(GI) tract and superior adsorption in the blood circulation(up to 2.61-and 5.65-times in AUC).After doxorubicin(DOX) loading into CMSs, DOX@CMSWs exhibited controlled drug release manners with pH responsiveness in vitro. Orally administered DOX@CMSWs could efficiently overcome the intestinal epithelium barrier(IEB), and resulted in satisfactory oral bioavailability of DOX(up to 348%).CMSWs were also proved to exhibit good biocompatibility and unique biodegradability. 13Genetic Types, Spatiotemporal Distribution of Ore Deposits and Sources of Ore-forming Materials in the Xuancheng Area, Anhui Province显示文摘In recent years,several large and medium-sized ore deposits have been discovered in the shallow cover of Xuancheng,Anhui Province,indicating that this area has a productive metallogenic geological background and may be a potential prospecting region.Based on systematic investigation,the geological and mineralization characteristics of porphyry Cu-Au deposits and skarn Cu-Mo-W deposits in this region have been summarized.Zircon U-Pb dating(LA-ICPMS)of the Chating quartz-diorite porphyry and the Kunshan biotite pyroxene diorite yield concordia ages of 145.5±2.1 Ma and 131.8±2.1 Ma,respectively.Meanwhile,the Re-Os dating analyses for molybdenite from the Shizishan and Magushan skarn Cu-Mo deposits yielded 133.81±0.86 Ma and 143.8±1.4 Ma ages,respectively.When viewed in conjunction with previous studies,it is suggested that twostage(the early stage of 145-135 Ma and the late stage of 134-125 Ma)magmatism may have occurred during the Mesozoic in Xuancheng region.Early stage intrusive rocks are distributed along both sides of the Jiangnan deep fault(JDF).The intrusive rocks to the north of the JDF are mainly quartzdiorite porphyry and granodiorite(porphyry)rocks,related to porphyry Cu-Au deposits and skarn-type Cu-Mo-W deposits.These deposits belong to the first stage of the porphyry-skarn copper gold metallogenic belt of the Middle-Lower Yangtze Metallogenic Belt(MLYB),associated with the high potassium calc-alkaline intermediate-acid intrusions.The magmatic and ore-forming materials are mainly derived from the enriched lithospheric mantle.South of the JDF,the Magushan granodiorite is a representative intrusive rock of the first stage I-type granite,which hosts the Magushan Cu-Mo skarn deposit,similar to the W-Mo-Cu skarn deposits in the Eastern Segment of the Jiangnan Uplift Metallogenic Belt(ESJUB).The magmatic and metallogenic materials mainly came from the Neoproterozoic basement,with the possible participation of a small amount of mantle components.The late stage magmatism was dominated by volcanic rocks with a small amount of intrusive rocks,which were consistent with the limited volcanic-intrusive activities in the second stage of the MLYB.The H-O stable isotopes of ore deposits in the region indicate that the ore-forming hydrothermal fluids of the porphyry and skarn deposits were mostly of magmatic water for the ore-forming stage,the percentage of meteoric water obviously increasing during the late ore-forming stage.The ore-forming materials of the deposits are mainly from the deep magma with a few sedimentary wall rocks,according to the stable carbon isotopes of the carbonates in the ore deposits.Additionally,according to previous research,the molybdenite from the MLYB has a higher Re content than that of the ESJUB.The higher content of Re in the molybdenite from the Shizishan deposit is identical to that of MLYB rather than ESJUB,whereas Re characteristics in molybdenite of Magushan deposit are similar to that of ESJUB.The differences in Re characteristics indicate the different deep processes and ore-forming material sources(mainly mantle composition for the former and crustal materials for the latter)of these ore deposits on opposite sides of the JDF.LU Sanming ZHANG Zanzan ZHAO Lili LI Jianshe ZHAO Zhuang QI Huasheng 2020Acta Geologica Sinica(English Edition)2020,94,6:1
14A 3D Investigation of Geological Structure and Its Relationship to Mineralization in the Nanling-Xuancheng Ore District,Middle-Lower Yangtze River Metallogenic Belt,China显示文摘The Nanling-Xuancheng ore region of Anhui Province is located in the Middle-Lower Yangtze River metallogenic belt.Insufficient exploration and research have been carried out in this newly defined ore district,although the Chating large porphyry Cu-Au deposit and a few middle-sized skarntype Cu polymetallic deposits have been discovered.In this study,we carried out high-resolution seismic reflection,magnetotelluric,gravity,and magnetic investigations,and constructed the 3 D geological structure of the uppermost crust in a depth range of 0-5 km using a comprehensive inversion of the new data constrained by previous deep-drilling data.We hence proposed some new insights to understand the mineralization processes of this district.A system of alternating ridges and valleys is suggested as the major structure pattern,composed of“two-layer structure”of the basins and“three-layer structure”of anticlines.Moreover,a conjugated fault system and its distribution features are revealed in our models,including the Jiangnan fault,Zhouwang fault,and Kunshan thrust nappe.The Jiangnan and Kunshan faults are suggested to have controlled the diagenesis and metallogenesis.Two deep concealed plutons located in Chating and Magushan are found,forming the Mesozoic diorite-felsic intrusions.These intrusions are believed to be the causes of hydrothermal deposits such as the Chating deposit and the Magushan deposit.Sanming Lu Xueyi Lan Lili Zhao Zanzan Zhang Xiaoyong Yang Zhuang Zhao Dong Guo Xiaochun Xu Yongsheng Wang Jianshe Li Huasheng Qi 2022Journal of Earth Science2022,33,3:0
15Research on cutting characteristics of fiber bundle with high-speed photography显示文摘Cutting is an essential and complicated process in many fields.Efficient and low-consumption cutting operations are of great significance for environmental protection and energy conservation.The development of high performance cutting parts relies on a deep understanding of the cutting process and cutting mechanism.In this research,a new type of cutting test bench with high-speed photography was developed,and the cutting tests were conducted on the jute fiber bundle from quasi-static cutting at 10 mm/s to dynamic cutting in the speed range of 0.6-2.4 m/s.The cutting process was captured by a high-speed camera.Analysis shows that compression exists before quasi-static cutting,and the compression force curve with respect to the compression ratio follows an exponential function.The cutting speed has a significant effect on cutting energy.The cutting energy consumption is not a monotonous function of cutting speed owing to the combined effect of elastic deformation and friction of fibers.The cutting energy increases with increasing cutting speed in the range of 0.6-1.2 m/s due to the increase of the friction within fibers and the friction between the blade and fibers.The cutting energy decreases with increasing cutting speed in the range of 1.2-1.8 m/s,and tends to be a fixed value when the cutting speed exceeds 1.8 m/s due to the stabilized elastic deformation and friction coefficient.From the perspective of energy saving,it is meaningless to increase the blade speed excessively when cutting fiber bundles.Zhitao He Huiling Ding Sanming Du Zhen Li Jiangtao Ji Jian Li Yongzhen Zhang 2020International Journal of Agricultural and Biological Engineering2020,13,3:0
16Association between the physical stability of flurbiprofen suspension and the interaction of HPMC/SDS显示文摘The anionic surfactant sodium dodecylsulfate(SDS) has improved the physical stability of flurbiprofen(FBP) suspension, which was suspended by 0.2%(w/v) hydroxypropylmethyl cellulose(HPMC, K4 M). Therefore, the physical stability of FBP suspensions and the interaction of HPMC/SDS were studied, and a certain association between them was revealed. The antisolvent precipitation method was used to prepare suspensions. The apparent drug concentration from different sites was evaluated to get the dispersion of drug actually. The process of flocculation and deflocculation with the addition of SDS was caught by analyzing the morphology of the suspended particles. The physical stability of the FBP suspensions was characterized mainly by measuring the re-dispersion time, the zeta potential and particle size. Meanwhile, conductivity measurements were carried out to obtain the characteristic concentrations of SDS in HPMC/SDS system. The viscosities, the abilities for improving the solubility and wettability of FBP in the separate and mixed HPMC/SDS solutions were also contrasted respectively. The suspensions prepared with HPMC/SDS possessed better physical stability. The suspensions were uniform when the concentration of SDS was between the critical adsorption concentration(CAC) and the polymer saturation point(PSP). After PSP, the uniformity became worse and worse until the SDS was enough to form a deflocculation state. Besides, the re-dispersion time of FBP suspensions was longest when the concentration of SDS around CAC and shorter by shorter after the critical micelle concentration(CMC).
