1Herbal decoctosome is a novel form of medicine显示文摘Traditionally, herbal medicine is consumed by drinking decoctions produced by boiling herbs with water. The functional components of the decoction are heat stable. Small RNAs(sRNAs) were reported as a new class of functional components in decoctions. However, the mechanisms by which sRNAs survive heat treatment of the decoction and enter cells are unclear.Previous studies showed that plant-derived exosome-like nanoparticles(ELNs), which we call botanosomes, could deliver therapeutic reagents in vivo. Here, we report that heat-stable decoctosomes(ELNs) from decoctions have more therapeutic effects than the decoctions in vitro and demonstrate therapeutic efficacy in vivo. Furthermore, sRNAs, such as HJT-sRNA-m7 and PGY-sRNA-6, in the decoctosome exhibit potent anti-fibrosis and anti-inflammatory effects, respectively. Decoctosome is comprised of lipids, chemical compounds, proteins, and s RNAs. A medical decoctosome mimic is called bencaosome. A single lipid sphinganine(d22:0) identified in the decoctosome was mixed and heated with the synthesized sRNAs to form the simplest bencaosome. This simple bencaosome structure was identified by critical micelle concentration(cmc) assay that sRNAs coassembled with sphinganine(d22:0) to form the lipid layers of vesicles. The heating process facilitates co-assembly of sRNAs and sphinganine(d22:0) until a steady state is reached. The artificially produced sphinganine-HJT-sRNA-m7 and sphinganinePGY-sRNA-6 bencaosomes could ameliorate bleomycin-induced lung fibrosis and poly(I:C)-induced lung inflammation, respectively, following oral administration in mice. 3Changing Proportions of HIV-1 Subtypes and Transmitted Drug Resistance Among Newly Diagnosed HIV/AIDS Individuals—China,2015 and 2018显示文摘Introduction:With the expansion of human immunodeficiency virus(HIV)antiretroviral therapy(ART),HIV drug resistance is becoming more and more serious.This study describes the changing prevalence of HIV-1 subtypes and transmitted drug resistance(TDR)among newly diagnosed individuals in China,2015 and 2018.Methods:A total of 8,980 individuals in 2015 and 2018 from 31 provincial-level administrative divisions(PLADs)were enrolled in this study.Viral RNAs were amplified and sequenced using an in-house polymerase chain reaction(PCR)protocol.The Stanford HIV Drug Resistance Database(HIVdb)was used to predict susceptibility to 12 antiretroviral drugs.Results:The prevalence of TDR was not significantly increased over time.The prevalence of TDR was 3.8%and 4.4%in 2015 and 2018,respectively(P=0.13).The prevalence of CRF55_01B increased from 2.3%in 2015 to 3.9%in 2018(P<0.001).The drug resistance prevalence of nonnucleoside reverse transcriptase inhibitors(NNRTI)increased from 2.4%in 2015 to 3.3%in 2018(P<0.01).The prevalence of E138(P<0.001),H221(P=0.03),and V179(P<0.001)mutations increased from 0.30%,0.09%,and 0.70%in 2015 to 1.10%,0.30%,and 1.70%in 2018,respectively.Conclusions:HIV drug resistance affects the effect of antiretroviral treatment,so the monitoring of HIV TDR should be strengthened to control the transmission of HIV drug resistance.Jingjing Hao Shan Zheng Mengze Gan Aobo Dong Ruihua Kang Miaomiao Li Shuai Zhao Jing Hu Chang Song Lingjie Liao Yi Feng Yiming Shao Yuhua Ruan Hui Xing 2021China CDC weekly2021,3,53:13
4Distributions of chlorophyll a and carbon fixed strength of phyto-plankton in autumn of the southern Huanghai Sea waters显示文摘Chlorophyll a (Chl a) concentration and primary productivity (PP), namely, carbon fixed strength of phytoplankton along four transects in the southern Huanghai Sea (SHS) were studied for their distribution features and controlling mechanisms based on the investigations from 17 October to 3 November 2005. The Chl a concentration in the study waters dynamically changed spatially. Surface Chl a concentrations ranged from 0.11 to 2.38 mg/m 3 with higher and lower values observed in the nutrient-laden inshore waters and central part of the SHS occupied by oligotrophic current, respectively. The vertical distribution of Chl a concentration showed a predominant pattern of subsurface concentration maximum profile. It followed the previous result of the deep dissolved oxygen concentration maximum profile, which was significantly correlated with phytoplankton and regional water mass. The primary productivity of carbon in autumn of the SHS, ranging from 95 to 1 634 mg/(m 2 ·d) mainly varied with nutrient condition, especially phosphate concentration in seawater and hydrological condition. Furthermore, associating the present study results together with previous studies, the annual value of carbon fixed production of phytoplankton in the entire marginal seas of East China (including the Bohai Sea, the Huanghai Sea and the East China Sea) was estimated to be 222 Mt, which accounted for 2% of that in the global margins. Besides, it was as 16.2 times as the annual value of apparent carbon sink strength (13.96 Mt) in the marginal seas of East China. 5Genome Warehouse: A Public Repository Housing Genome-scale Data显示文摘The Genome Warehouse(GWH)is a public repository housing genome assembly data for a wide range of species and delivering a series of web services for genome data submission,storage,release,and sharing.As one of the core resources in the National Genomics Data Center(NGDC),part of the China National Center for Bioinformation(CNCB;http://gffzz77e3413bc06540edsv6n6wncw5bup6cv9.ffgz.tsg.suse.edu.cn),GWH accepts both full and partial(chloroplast,mitochondrion,and plasmid)genome sequences with different assembly levels,as well as an update of existing genome assemblies.For each assembly,GWH collects detailed genome-related metadata of biological project,biological sample,and genome assembly,in addition to genome sequence and annotation.To archive high-quality genome sequences and annotations,GWH is equipped with a uniform and standardized procedure for quality control.Besides basic browse and search functionalities,all released genome sequences and annotations can be visualized with JBrowse.By May 21,2021,GWH has received 19,124 direct submissions covering a diversity of 1108 species and has released 8772 of them.Collectively,GWH serves as an important resource for genomescale data management and provides free and publicly accessible data to support research activities throughout the world.GWH is publicly accessible at http://gffzz77e3413bc06540edsv6n6wncw5bup6cv9.ffgz.tsg.suse.edu.cn/gwh.Meili Chen Yingke Ma Song Wu Xinchang Zheng Hongen Kang Jian Sang Xingjian Xu Lili Hao Zhaohua Li Zheng Gong Jingfa Xiao Zhang Zhang Wenming Zhao Yiming Bao 2021Genomics, Proteomics & Bioinformatics2021,19,4:12
6Prognostic implications of epidermal growth factor receptor variant Ⅲ expression and nuclear translocation in Chinese human gliomas显示文摘Objective: To determine the prognostic implications and clinical significance of epidermal growth factor receptor variant Ⅲ(EGFRvⅢ) expression and EGFRvⅢ nuclear translocation in Chinese human gliomas.Methods: We retrospectively examined EGFRvⅢ expression and EGFRvⅢ nuclear translocation using immunohistochemistry in specimens of 240 Chinese patients with glioma, including 84 World Health Organization(WHO) II gliomas, 84 WHO Ⅲ gliomas and 72 glioblastomas(WHO IV). Factors that correlated with EGFRvⅢ and EGFRvⅢ nuclear translocation expression were analyzed by the Chi-square test. Kaplan-Meier methodology and Cox regression were used for the survival analysis.Results: Log-rank tests showed that patient age, Karnofsky performance scale(KPS) score, tumor grade,EGFRvⅢ expression, EGFRvⅢ nuclear translocation, 1 p/19 q codeletion, isocitrate dehydrogenase(IDH)mutation, Ki-67 labeling index and O6-methylguanine-DNA methyltransferase(MGMT) status(P<0.05) were significantly correlated with overall survival(OS) time. Multivariate Cox regression analysis revealed that patient age, tumor grade, EGFRvⅢ nuclear translocation, 1 p/19 q codeletion, and IDH mutation(P<0.05) were significantly correlated with OS. Patients with a high level of EGFRvⅢ nuclear translocation(≥7%) had both significantly shorter OS [hazard ratio(HR): 1.920, 95% confidence interval(95% CI): 1.228-3.003, P=0.004] and progression-free survival(PFS) times(HR: 1.661, 95% CI: 1.116-2.471, P=0.012) than those with a low level of EGFRvⅢ nuclear translocation(<7%).Conclusions: A high level of EGFRvⅢ nuclear translocation in glioma is an independent factor indicating a poor prognosis, but EGFRvⅢ expression is not an independent clinical prognostic factor. 9Cartilage-targeting peptide-modified dual-drug delivery nanoplatform with NIR laser response for osteoarthritis therapy显示文摘Cartilage-targeting delivery of therapeutic agents is still an effective strategy for osteoarthritis(OA)therapy.Recently,scavenging for reactive oxygen species(ROS)and activating autophagy have been increasingly reported to treat OA effectively.In this study,we designed,for the first time,a dual-drug delivery system based on metal organic framework(MOF)-decorated mesoporous polydopamine(MPDA)which composed of rapamycin(Rap)loaded into the mesopores and bilirubin(Br)loaded onto the shell of MOF.The collagen II-targeting peptide(WYRGRL)was then conjugated on the surface of above nanocarrier to develop a cartilage-targeting dual-drug delivery nanoplatform(RB@MPMW).Our results indicated the sequential release of two agents from RB@MPMW could be achieved via near-infrared(NIR)laser irritation.Briefly,the rapid release of Br from the MOF shell exhibited excellent ROS scavenging ability and anti-apoptosis effects,however responsively reduced autophagy activity,to a certain extent.Meanwhile,following the NIR irradiation,Rap was rapidly released from MPDA core and further enhanced autophagy activation and chondrocyte protection.RB@MPMW continuously phosphorylated AMPK and further rescued mitochondrial energy metabolism of chondrocytes following IL-1βstimulation via activating SIRT1-PGC-1αsignaling pathway.Additionally,the cartilage-targeting property of peptide-modified nanocarrier could be monitored via Magnetic Resonance(MR)and IVIS imaging.More significantly,RB@MPMW effectively delayed cartilage degeneration in ACLT rat model.Overall,our findings indicated that the as-prepared dual-drug delivery nanoplatform exerted potent anti-inflammation and anti-apoptotic effects,rescued energy metabolism of chondrocytes in vitro and prevented cartilage degeneration in vivo,which thereby showed positive performance for OA therapy.Song Xue Xiaojun Zhou Weilin Sang Cong Wang Haiming Lu Yiming Xu Yiming Zhong Libo Zhu Chuanglong He Jinzhong Ma 2021Bioactive Materials2021,6,8:5
10A comparison between acute large vessel occlusion in the posterior circulation and anterior circulation after endovascular treatment:the ANGEL-ACT registry experience显示文摘Background and purpose Endovascular treatment(EVT)has shown an overwhelming benefit for acute anterior circulation artery occlusion(ACO).Whether it can achieve the same outcomes in posterior circulation artery occlusion(PCO)has not been well explained.We aimed to evaluate the characteristics and prognosis of ACO and PCO after EVT in a nationwide registry.Method The present analysis was based on the prospective ANGEL-ACT Registry in China between November 2017 and March 2019.Demographic data,periprocedural times,recanalisation rate,intracranial haemorrhage(ICH)and 90-day functional outcomes were compared between the ACO and PCO groups.Results A total of 1793 patients were analysed including 397(22.1%)consecutive patients with PCO and 1396(77.9%)patients with ACO treated with EVT.A larger proportion of patients with PCO had intracranial atherosclerotic disease and received extra angioplasty during EVT.Successful recanalisation and 90-day favourable functional outcomes did not differ significantly between the two groups.Patients with PCO showed lower 24-hour ICH and symptomatic ICH rates.There was a trend towards higher mortality rate in the PCO group(22.09%vs 14.44%;adjusted OR 1.286(95%CI 0.820 to 2.017),p=0.2731),especially when the onset to puncture time was over 6 hours(30.77%vs 11.13%;adjusted OR 2.673(95%CI 1.280 to 5.583),p=0.0089,interactive p=0.0002).Conclusions In this large prospective multicentre registry,there was a significant difference in the characteristics and periprocedural features between patients with PCO and ACO.However,successful recanalisation and 90-day favourable functional outcomes in PCO were equivalent to those in ACO.Xuan Sun Jingyu Zhang Xu Tong Baixue Jia Dapeng Mo Ning Ma Feng Gao Lian Liu Xiaochuan Huo Ligang Song Yiming Deng Zhongrong Miao 2022Stroke & Vascular Neurology2022,7,4:4

12A hybrid genipin-crosslinked dual-sensitive hydrogel/nanostructured lipid carrier ocular drug delivery platform显示文摘The objective of this study was to develop a novel hybrid genipin-crosslinked dual-sensitive hydrogel/nanostructured lipid carrier(NLC) drug delivery platform. An ophthalmic antiinflammatory drug, baicalin(BN) was chosen as the model drug. BN –NLC was prepared using melt-emulsification combined with ultra-sonication technique. Additionally, a dual pH-and thermo-sensitive hydrogel composed of carboxymethyl chitosan(CMCS) and poloxamer 407(F127) was fabricated by a cross-linking reaction with a nontoxic crosslinker genipin(GP). GP-CMCS/F127 hydrogel was characterized by FTIR, NMR, XRD and SEM. The swelling studies showed GP-CMCS/F127 hydrogel was both pH-and thermo-sensitive. The results of in vitro release suggested BN –NLC gel can prolong the release of baicalin comparing with BN eye drops and BN –NLC. Ex vivo cornea permeation study was evaluated using Franz diffusion cells. The apparent permeability coefficient(Papp) of BN –NLC gel was much higher(4.46-fold) than that of BN eye drops. Through the determination of corneal hydration levels, BN –NLC gel was confirmed that had no significant irritation to cornea. Ex vivo precorneal retention experiments were carried out by a flow-through approach. The results indicated that the NLC-based hydrogel can prolong precorneal residence time. 13Circ-Smad5 retards the G1/S transition of cell cycle via inhibiting the activity of wnt/lef/cyclind1 signaling in JB6 cells显示文摘Circular RNAs are a large class of noncoding RNAs.Smad5 functions in cell differentiation,cell proliferation and metastasis.It has been reported that lnc-Smad5 can inhibit the proliferation of diffuse large B cell lymphoma.However,the function of circ-Smad5 has not yet been reported.Lentivirus vectors were constructed to establish circ-Smad5 upregulated and circ-Smad5 downregulated cell models.A CCK-8 assay was used to detect the proliferation of JB6 cells.FACS was used to analyze the cell cycle in the cell models.Western blot,immunofluorescence staining and TOP/FOP flash dual luciferase activity assays were used to determine the activity of the Wnt signaling pathway.The results revealed that the expression level of circ-Smad5 in JB6 cells was significantly lower than the expression level of linearizedSmad5.Compared with the control group,the percentage of S phase cells and the expression level of cyclin D1 protein were significantly higher in the sh-circ-Smad5 group.In the sh-circSmad5 group,b-catenin and LEF-1 were significantly increased,p-b-catenin was significantly decreased,and the relative activity of the TOP/FOP reporter gene was higher compared to the control group levels.These phenomena could be reversed by treating with Wnt signaling inhibitor PNU-74654.We conclude that the circ-Smad5 retards the proliferation and the cell cycle progression of JB6 cells.Thus,circ-Smad5 may function by inhibiting the activation of Wnt/b-catenin/Lef 1 signaling,which inhibits the expression of cyclin D1.To the best of our knowledge,we are the first to report the function of circ-Smad5.Xiaogen Ma Fei Xiang Zhuo Pei Jiafeng Miao Pan Wu Xiaofeng Song Yuhong Li Yiming Zhang 2021Genes & Diseases2021,8,3:3
14Epidermal self-powered sweat sensors for glucose and lactate monitoring显示文摘Sweat could be a carrier of informative biomarkers for health status identification;therefore,wearable sweat sensors have attracted significant attention for research.An external power source is an important component of wearable sensors,however,the current power supplies,i.e.,batteries,limit further shrinking down the size of these devices and thus limit their application areas and scenarios.Herein,we report a stretchable self-powered biosensor with epidermal electronic format that enables the in situ detec-tion of lactate and glucose concentration in sweat.Enzymatic biofuel cells serve as self-powered sensing modules allowing the sweat sensor to exhibit a determination coefficient(R2)of 0.98 with a sensitivity of 2.48 mV/mM for lactate detection,and R2 of 0.96 with a sensitivity of 0.11 mV/μM for glucose detection.The microfluidic channels developed in an ultra-thin soft flexible polydimethylsiloxane layer not only enable the effective collection of sweat,but also provide excellent mechanical properties with stable performance output even under 30%stretching.The presented soft sweat sensors can be integrated at nearly any location of the body for the continuous monitoring of lactate and glucose changes during normal daily activities such as exercise.Our results provide a promising approach to develop next-generation sweat sensors for real-time and in situ sweat analysis.Xingcan Huang Jiyu Li Yiming Liu Tszhung Wong Jingyou Su Kuanming Yao Jingkun Zhou Ya Huang Hu Li Dengfeng Li Mengge Wu Enming Song Shijiao Han Xinge Yu 2022Bio-Design and Manufacturing2022,5,1:3

16Microstructures and mechanical properties of titanium-reinforced magnesium matrix composites: Review and perspective显示文摘Currently, many gratifying signs of progress have been made in magnesium(Mg) matrix composites(MMCs) by virtue of their high mechanical properties both at room and elevated temperatures. Although the commonly used reinforcements in MMCs are ceramic particles,they often provide improved yield and ultimate stresses by a significant loss in ductility. Therefore, hard metallic phases were introduced as alternative candidates for the manufacturing of MMCs, especially titanium(Ti). It has a high melting point, high Young’s modulus, high plasticity, low level of mutual solubility with Mg matrix, and closer thermal expansion coefficient to that of Mg metal than that of ceramic particles. It is highly preferable to provide both high ultimate stress and ductility in Mg matrix. However, many critical challenges for the fabrication of Ti-reinforced MMCs remain, such as Ti’s homogeneity, low recovery rate, and the optimization of interfacial bonding strength between Mg and Ti, etc. Meanwhile, different fabrication methods have various effects on the microstructures, mechanical properties, and the interfacial strength of Ti-reinforced MMCs. Hence, this review placed emphasis on the microstructural characteristics and mechanical properties of Ti-reinforced MMCs fabricated by different techniques. The influencing factors that govern the strengthening mechanisms were systematically compared and discussed. 17Applying DNA Barcoding to Identify Actinidia in the Northeast of China显示文摘Identification of kiwifruit germplasm materials is the basis of protecting and utilizing these resources. However,identifying Actinidia arguta varieties based on morphology is difficult,especially for non-specialists. In this study,we collected 180 specimens comprising 60 varieties of three species( A. arguta,A. kolomikta,and A. polygama) from the Northeast of China. The emphasis of our study was on the feasibility of identifying A. arguta varieties. Here,we used common analysis methods( genetic distance,phylogenetic analysis and the DNA barcoding gap) and SNPs analysis to evaluate the discriminatory power of different DNA barcoding markers. The results revealed that common methods were insufficient to identify A. arguta varieties but SNPs analysis based on DNA barcoden was the potential method for identifying A. arguta varieties.
