1Oncogenic AURKA-enhanced N6-methyladenosine modification increases DROSHA mRNA stability to transactivate STC1 in breast cancer stem-like cells显示文摘RNase III DROSHA is upregulated in multiple cancers and contributes to tumor progression by hitherto unclear mechanisms.Here,we demonstrate that DROSHA interacts withβ-Catenin to transactivate STC1 in an RNA cleavage-independent manner,contributing to breast cancer stem-like cell(BCSC)properties.DROSHA mRNA stability is enhanced by N6-methyladenosine(m^(6)A)modification which is activated by AURKA in BCSCs.AURKA stabilizes METTL14 by inhibiting its ubiquitylation and degradation to promote DROSHA mRNA methylation.Moreover,binding of AURKA to DROSHA transcript further strengthens the binding of the m^(6)A reader IGF2BP2 to stabilize m^(6)A-modified DROSHA.In addition,wild-type DROSHA,but not an m^(6)A methylation-deficient mutant,enhances BCSC stemness maintenance,while inhibition of DROSHA m^(6)A modification attenuates BCSC traits.Our study unveils the AURKA-induced oncogenic m^(6)A modification as a key regulator of DROSHA in breast cancer and identifies a novel DROSHA transcriptional function in promoting the BCSC phenotype.Fei Peng Jie Xu Bai Cui Qilan Liang Sai Zeng Bin He Hong Zou Manman Li Huan Zhao Yuting Meng Jin Chen Bing Liu Shasha Lv Peng Chu Fan An Zifeng Wang Junxiu Huang Yajing Zhan Yuwei Liao Jinxin Lu Lingzhi Xu Jin Zhang Zhaolin Su Zhiguang Li Fangjun Wang Eric W-FLam Quentin Liu 2021Cell Research2021,31,3:9

3Interfacial characteristics and mechanical properties of additive manufacturing martensite stainless steel on the Cu-Cr alloy substrate by directed energy deposition显示文摘Copper/steel is a typical bimetal functional material,combining the excellent electrical and thermal conductivity of copper alloy and the high strength and hardness of stainless steel.There has been recent interest in manufacturing copper/steel bimetal by directed energy deposition(DED)due to its layer-bylayer method.However,cracks tend to form on the copper/steel interface because of the great difference in thermal expansion coefficient and crystal structure between copper and steel.In this work,interfacial characteristics and mechanical properties of the copper/steel bimetal were studied from one layer to multilayers.The laser power has a great influence on the Cu element distribution of the molten pool,affecting the crack formation dramatically on the solidification stage.Cracks tend to form along columnar grain boundaries because of the Cu-rich liquid films and spherical particles in the cracks.Crack-free and good metallurgical bonding copper/steel interface is formed at a scanning velocity of 800 mm/min and the laser power of 3000 W.The ultimate tensile strength(UTS)and the break elongation(EL)of the vertically combined crack-free copper/steel bimetal are 238.2±4.4 MPa and 20.6±0.7%,respectively.The fracture occurs on the copper side instead of the copper/steel interface,indicating that the bonding strength is higher than that of the Cu-Cr alloy.The UTS of the horizontally combined crack-free copper/steel bimetal is 746.7±22.6 MPa,which is 200%higher than that of the Cu-Cr alloy substrate.The microhardness is 398.6±5.4 HV at the steel side and is 235.3±64.1 HV at the interface,which is400%higher than that of the Cu-Cr alloy substrate.This paper advances the understanding of the interfacial characteristics of heterogeneous materials and provides guidance and reference for the fabrication of multi-material components by DED.Wenqi Zhang Hailong Liao Zhiheng Hu Shasha Zhang Baijin Chen Huanqing Yang Yun Wang Haihong Zhu 2021Journal of Materials Science & Technology2021,,31:2

7Numerical Simulation of Asphaltene Precipitation and Deposition during Natural Gas and CO_(2) Injection显示文摘Asphaltene deposition is a significant problem during gas injection processes,as it can block the porous medium,the wellbore,and the involved facilities,significantly impacting reservoir productivity and ultimate oil recovery.Only a few studies have investigated the numerical modeling of this potential effect in porous media.This study focuses on asphaltene deposition due to natural gas and CO_(2) injection.Predictions of the effect of gas injection on asphaltene deposition behavior have been made using a 3D numerical simulation model.The results indicate that the injection of natural gas exacerbates asphaltene deposition,leading to a significant reduction in permeability near the injection well and throughout the reservoir.This reduction in permeability strongly affects the ability of gas toflow through the reservoir,resulting in an improvement of the displacement front.The displacement effi-ciency of the injection gas process increases by up to 1.40%when gas is injected at 5500 psi,compared to the scenario where the asphaltene model is not considered.CO_(2) injection leads to a miscible process with crude oil,extracting light and intermediate components,which intensifies asphaltene precipitation and increases the viscosity of the remaining crude oil,ultimately reducing the recovery rate.Shasha Feng Yi Liao Weixin Liu Jianwen Dai Mingying Xie Li Li 2024Fluid Dynamics & Materials Processing2024,20,2:0

9Temperature determines the shift of thermal neutral zone and influences thermogenic capacity in striped hamsters显示文摘The thermoneutral zone(TNZ)reflects the adaptation of mammals to their natural habitat.However,it remains unclear how TNZ shifts in response to variations in ambient temperature.To test the hypothesis that ambient temperature plays a key role in determining TNZ variations between seasons,we measured metabolic rate,body temperature,and cytochrome c oxidase(COX)activity of several visceral organs in striped hamsters(Cricetulus barabensis)either acclimated to semi-natural conditions over a year,or subjected to a gradual decrease in mean temperature from 30±1°C to-15±1°C.The TNZ range in striped hamsters differed seasonally,with a wider TNZ and a lower lower-critical temperature in winter compared to summer.The hamsters showed a consider-able leftward shift of lower-critical temperature from 30°C to 20°C after the ambient temperature of acclimation from 30°C down to-15°C,whereas the upper-critical temperature of TNZ remainedfixed at 32.5°C.The rest-ing metabolic rate in thermoneutral zone(RMRt),nonshivering thermogenesis(NST),and COX activity of brown adipose tissue,liver,skeletal muscle,brain,and kidneys,increased significantly in hamsters acclimated at lower ambient temperatures.Following acute exposure to 5°C and-15°C,hamsters acclimated to 32.5°C had signifi-cantly lower maximal NST and lower serum thyroid tri-iodothyronine(T3)levels compared to those kept at 23°C.Thesefindings suggest that acclimation to the upper-critical temperature of TNZ impairs the hamsters’thermo-genic capacity to cope with extreme cold temperature.Reduced ambient temperature was mainly responsible for the leftward shift of TNZ in striped hamsters,which reflects the adaptation to cold environments.Shasha LIAO Song TAN Meizhi JIANG Jing WEN Jinsong LIU Jing CAO Ming LI Zhijun ZHAO 2023Integrative Zoology2023,18,2:0
10Recent advances in the application of MXenes for neural tissue engineering and regeneration显示文摘Transition metal carbides and nitrides(MXenes)are crystal nanomaterials with a number of surface functional groups such as fluorine,hydroxyl,and oxygen,which can be used as carriers for proteins and drugs.MXenes have excellent biocompatibility,electrical conductivity,surface hydrophilicity,mechanical properties and easy surface modification.However,at present,the stability of most MXenes needs to be improved,and more synthesis methods need to be explored.MXenes are good substrates for nerve cell regeneration and nerve reconstruction,which have broad application prospects in the repair of nervous system injury.Regarding the application of MXenes in neuroscience,mainly at the cellular level,the long-term in vivo biosafety and effects also need to be further explored.This review focuses on the progress of using MXenes in nerve regeneration over the last few years;discussing preparation of MXenes and their biocompatibility with different cells as well as the regulation by MXenes of nerve cell regeneration in two-dimensional and three-dimensional environments in vitro.MXenes have great potential in regulating the proliferation,differentiation,and maturation of nerve cells and in promoting regeneration and recovery after nerve injury.In addition,this review also presents the main challenges during optimization processes,such as the preparation of stable MXenes and long-term in vivo biosafety,and further discusses future directions in neural tissue engineering.Menghui Liao Qingyue Cui Yangnan Hu Jiayue Xing Danqi Wu Shasha Zheng Yu Zhao Yafeng Yu Jingwu Sun Renjie Chai 2024Neural Regeneration Research2024,19,2:0
