1Disease profile and plasma neutralizing activity of postvaccination Omicron BA.1 infection in Tianjin,China:a retrospective study显示文摘Dear Editor,SARS-CoV-2 Omicron variant BA.1 first emerged on the Chinese mainland on January 8,2022,in Tianjin and caused a large wave of infections.1 Four rounds of mass testing of its~14 million residents were launched on January 9,12,15,and 20,respectively.As of February 7,2022,a total of 430 individuals were tested positive for Omicron BA.1,with no new infections detected for the following 16 days.In the Chinese mainland,the most delivered vaccines are inactivated vaccines.As of January 8,2022,up to 93.2%of Tianjin’s residents had been vaccinated with at least one dose.2 We investigated the protection conferred by vaccination against Omicron BA.1 by examining the breakthrough infections among vaccinees as compared with unvaccinated cases.Our study population comprised all 430 cases during the outbreak of Omicron BA.1 in Tianjin.Hong Zheng Yunlong Cao Xiaosu Chen Fengmei Wang Ye Hu Weiliang Song Yangyang Chai Qingqing Gu Yansong Shi Yingmei Feng Shuxun Liu Yan Xie Xiaoliang Sunney Xie Wentao Jiang Zhongyang Shen 2022Cell Research2022,32,8:12
2Interferon-inducible cytoplasmic lncLrrc55-AS promotes antiviral innate responses by strengthening IRF3 phosphorylation显示文摘Type I interferon (IFN-I) production is efficiently induced to ensure a potent innate immune response to viral infection. How this response can be enhanced, however, remains to be explored. Here, we identify a new cytoplasmic long non-coding RNA (lncRNA), lncLrrc55-AS, that drives a positive feedback loop to promote interferon regulatory factor 3 (IRF3) signaling and IFN-I production. We show that lncLrrc55-AS is virus-induced in multiple cell types via the IFN-JAK-STAT pathway. LncLrrc55-AS-deficient mice display a weakened antiviral immune response and are more susceptible to viral challenge. Mechanistically, lncLrrc55-AS binds phosphatase methylesterase 1 (PME-1), and promotes the interaction between PME-1 and the phosphatase PP2A, an inhibitor of IRF3 signaling. LncLrrc55-AS supports PME-1-mediated demethylation and inactivation of PP2A, thereby enhancing IRF3 phosphorylation and signaling. Loss of PME-1 phenocopies lncLrrc55-AS deficiency, leading to diminished IRF3 phosphorylation and IFN-I production. 6Micro-nanofiber composite biomimetic conduits promote long-gap peripheral nerve regeneration in canine models显示文摘Peripheral nerve injuries may result in severe long-gap interruptions that are challenging to repair.Autografting is the gold standard surgical approach for repairing long-gap nerve injuries but can result in prominent donor-site complications.Instead,imitating the native neural microarchitecture using synthetic conduits is expected to offer an alternative strategy for improving nerve regeneration.Here,we designed nerve conduits composed of high-resolution anisotropic microfiber grid-cordes with randomly organized nanofiber sheaths to interrogate the positive effects of these biomimetic structures on peripheral nerve regeneration.Anisotropic microfiber-grids demonstrated the capacity to directionally guide Schwann cells and neurites.Nanofiber sheaths conveyed adequate elasticity and permeability,whilst exhibiting a barrier function against the infiltration of fibroblasts.We then used the composite nerve conduits bridge 30-mm long sciatic nerve defects in canine models.At 12 months post-implant,the morphometric and histological recovery,gait recovery,electrophysiological function,and degree of muscle atrophy were assessed.The newly regenerated nerve tissue that formed within the composite nerve conduits showed restored neurological functions that were superior compared to sheaths-only scaffolds and Neurolac nerve conduit controls.Our findings demonstrate the feasibility of using synthetic biophysical cues to effectively bridge long-gap peripheral nerve injuries and indicates the promising clinical application prospects of biomimetic composite nerve conduits.Xianhao Dong Yueyue Yang Zheheng Bao Adam C.Midgley Feiyi Li Shuxin Dai Zhuangzhuang Yang Jin Wang Lihua Liu Wenlei Li Yayuan Zheng Siyang Liu Yang Liu Weijian Yu Jun Liu Meng Fan Meifeng Zhu Zhongyang Shen Gu Xiaosong Deling Kong 2023Bioactive Materials2023,,12:0
7Experimental study of NO_2 reduction in N_2/Ar and O_2/Ar mixtures by pulsed corona discharge显示文摘Non-thermal plasma technology has been regarded as a promising alternative technology for NOx removal. The understanding of NO2 reduction characteristics is extremely important since NO2 reduction could lower the total NO oxidation rate in the plasma atmosphere. In this study, NO2 reduction was experimentally investigated using a non-thermal plasma reactor driven by a pulsed power supply for different simulated gas compositions and operating parameters. The NO2 reduction was promoted by increasing the specific energy density(SED), and the highest conversion rates were 33.7%, 42.1% and25.7% for Ar, N2/Ar and O2/Ar, respectively. For a given SED, the NO2 conversion rate had the order N2/Ar > Ar > O2/Ar. The highest energy yield of 3.31 g/k Wh was obtained in N2/Ar plasma and decreased with increasing SED; the same trends were also found in the other two gas compositions. The conversion rate decreased with increasing initial NO2 concentration. 8Intracellular adenosine-tri-phosphate levels are lower in pediatric liver transplant recipients with infection显示文摘目的对儿童肝移植受者中CD4+T淋巴细胞内三磷酸腺苷(iATP)含量(ImmuKnow法)进行分析,并探讨其与受者临床状态的关系。方法本研究共纳入63例儿童肝移植受者,将其分为临床稳定组和感染组,分析CD4+T淋巴细胞内iATP含量与感染的关系。结果 63例患儿中,共有35例(55.6%)患儿进行了iATP的系统检测,共计120例次。其中,39例临床稳定组患儿的64例次iATP的平均值为(372±225)μg/L,且个体差异性较大。与临床稳定组患儿相比,24例感染组儿童受者的iATP更低,差异具有统计学意义(P<0.001)。系统监测显示,随着感染进展,iATP水平均值显著下降(P<0.001),且随着感染好转,其水平又有所升高(P=0.036)。另外,12例iATP水平偏低(iATP<225μg/L)的临床稳定组患儿中,并未发现活动性感染,而8例感染组患儿中,其iATP水平却偏高(iATP>225μg/L)。结论与临床稳定组的患儿相比,感染组患者的iATP水平更低,且随着感染好转,其水平逐渐升高。本研究发现,iATP水平的个体差异性较大,提示iATP水平检测可能只是反映患儿免疫状态的一个指标。iATP水平的系统监测将更有助于提示儿童肝移植受者免疫状态变化。Rao Wei Xie Man Sun Xiaoye Yang Tao Gao Wei Zhang Jianjun Liu Yihe Deng Yonglin Shen Zhongyang 2014实用器官移植电子杂志2014,2,4:0
9Innate immune imprints in SARS-CoV-2 Omicron variant infection convalescents显示文摘SARS-CoV-2 Omicron variant infection generally gives rise to asymptomatic to moderate COVID-19 in vaccinated people.The immune cells can be reprogrammed or“imprinted”by vaccination and infections to generate protective immunity against subsequent challenges.Considering the immune imprint in Omicron infection is unclear,here we delineate the innate immune landscape of human Omicron infection via single-cell RNA sequencing,surface proteome profiling,and plasma cytokine quantification.We found that monocyte responses predominated in immune imprints of Omicron convalescents,with IL-1β-associated and interferon(IFN)-responsive signatures with mild and moderate symptoms,respectively.Low-density neutrophils increased and exhibited IL-1β-associated and IFN-responsive signatures similarly.Mild convalescents had increased blood IL-1β,CCL4,IL-9 levels and PI3+neutrophils,indicating a bias to IL-1βresponsiveness,while moderate convalescents had increased blood CXCL10 and IFN-responsive monocytes,suggesting durative IFN responses.Therefore,IL-1β-or IFN-responsiveness of myeloid cells may indicate the disease severity of Omicron infection and mediate post-COVID conditions.Zhiqing Li Xiaosu Chen Junyan Dan Tianju Hu Ye Hu Shuxun Liu Yangyang Chai Yansong Shi Jian Wu Hailai Ni Jiaqi Zhu Yanfeng Wu Nan Li Yizhi Yu Zhongfang Wang Jincun Zhao Nanshan Zhong Xianwen Ren Zhongyang Shen Xuetao Cao 2022Signal Transduction and Targeted Therapy2022,7,12:0
