1Perfecting and extending the near-infrared imaging window显示文摘In vivo fluorescence imaging in the second near-infrared window(NIR-II)has been considered as a promising technique for visualizing mammals.However,the definition of the NIR-II region and the mechanism accounting for the excellent performance still need to be perfected.Herein,we simulate the photon propagation in the NIR region(to 2340 nm),confirm the positive contribution of moderate light absorption by water in intravital imaging and perfect the NIR-II window as 900–1880 nm,where 1400–1500 and 1700–1880 nm are defined as NIR-IIx and NIR-IIc regions,respectively.Moreover,2080–2340 nm is newly proposed as the third near-infrared(NIR-III)window,which is believed to provide the best imaging quality.The wide-field fluorescence microscopy in the brain is performed around the NIRIIx region,with excellent optical sectioning strength and the largest imaging depth of intravital NIR-II fluorescence microscopy to date.We also propose 1400 nm long-pass detection in off-peak NIR-II imaging whose performance exceeds that of NIR-IIb imaging,using bright fluorophores with short emission wavelength.Zhe Feng Tao Tang Tianxiang Wu Xiaoming Yu Yuhuang Zhang Meng Wang Junyan Zheng Yanyun Ying Siyi Chen Jing Zhou Xiaoxiao Fan Dan Zhang Shengliang Li Mingxi Zhang Jun Qian 2021Light(Science & Applications)2021,10,10:4
2Targeting β-secretase with RNAi in neural stem cells for Alzheimer's disease therapy显示文摘There are several major pathological changes in Alzheimer's disease,including apoptosis of cholinergic neurons,overactivity or overexpression of β-site amyloid precursor protein cleaving enzyme 1(BACE1) and inflammation.In this study,we synthesized a 19-nt oligonucleotide targeting BACE1,the key enzyme in amyloid beta protein(Aβ) production,and introduced it into the pSilenCircle vector to construct a short hairpin(shRNA) expression plasmid against the BACE1 gene.We transfected this vector into C17.2 neural stem cells and primary neural stem cells,resulting in downregulation of the BACE1 gene,which in turn induced a considerable reduction in reducing Aβ protein production.We anticipate that this technique combining cell transplantation and gene therapy will open up novel therapeutic avenues for Alzheimer's disease,particularly because it can be used to simultaneously target several pathogenetic changes in the disease.Zhonghua Liu Shengliang Li Zibin Liang Yan Zhao Yulin Zhang Yaqi Yang Minjuan Wang Feng Li 2013Neural Regeneration Research2013,8,33:4
3Confocal visible/NIR photoacoustic microscopy of tumors with structural, functional, and nanoprobe contrasts显示文摘Distinguishing early-stage tumors from normal tissues is of great importance in cancer diagnosis.We report fiberbased confocal visible/near-infrared(NIR)optical-resolution photoacoustic microscopy that can image tumor microvasculature,oxygen saturation,and nanoprobes in a single scanning.We develop a cost-efficient single laser source that provides 532,558,and 1064 nm pulsed light with sub-microseconds wavelength switching time.Via dual-fiber illumination,we can focus the three beams to the same point.The optical and acoustic foci are confocally aligned to optimize the sensitivity.The visible and NIR wavelengths enable simultaneous tumor imaging with three different contrast modes.Results show obvious angiogenesis,significantly elevated oxygen saturation,and accumulated nanoparticles in the tumor regions,which offer comprehensive information to detect the tumor.This approach also allows us to identify feeding and draining vessels of the tumor and thus to determine local oxygen extraction fraction.In the tumor region,the oxygen extraction fraction significantly decreases along with tumor growth,which can also assist in tumor detection and staging.Fiber-based confocal visible/NIR photoacoustic microscopy offers a new tool for early detection of cancer.JIANGBO CHEN YACHAO ZHANG XIAOZHEN LI JINGYI ZHU DENGFENG LI SHENGLIANG LI CHUN-SING LEE LIDAI WANG 2020Photonics Research2020,8,12:2
4Resolving and weighing the quantum orbits in strong-field tunneling ionization显示文摘Tunneling ionization of atoms and molecules induced by intense laser pulses contains the contributions of numerous quantum orbits.Identifying the contributions of these orbits is crucial for exploring the application of tunneling and for understanding various tunneling-triggered strong-field phenomena.We perform a combined experimental and theoretical study to identify the relative contributions of the quantum orbits corresponding to the electrons tunneling ionized during the adjacent rising and falling quarter cycles of the electric field of the laser pulse.In our scheme,a perturbative second-harmonic field is added to the fundamental driving field.By analyzing the relative phase dependence of the signal in the photoelectron momentum distribution,the relative contributions of these two orbits are unambiguously determined.Our results show that their relative contributions sensitively depend on the longitudinal momentum and modulate with the transverse momentum of the photoelectron,which is attributed to the interference of the electron wave packets of the long orbit.The relative contributions of these orbits resolved here are important for the application of strong-field tunneling ionization as a photoelectron spectroscopy for attosecond time-resolved measurements.Jia Tan Shengliang Xu Xu Han Yueming Zhou Min Li Wei Cao Qingbin Zhang Peixiang Lu 2021Advanced Photonics2021,3,3:2
5Design of antibacterial peptide-like conjugated molecule with broad spectrum antimicrobial ability显示文摘Increasing multidrug-resistant(MDR) superbugs emerge worldwide causing a public health crisis. Consequently, it is urgent to find new antibiotics with efficient broad-spectrum antimicrobial activity. By virtue of versatility in molecular design, a new peptide-like cell membrane-broken molecule, oligo-(7,7′-bifluoren-benzo[c][1,2,5]thiadiazole)(OFBT) possessing a conjugated backbone and eight pendant guanidyl moieties was designed and synthesized. OFBT exhibits favorable broad-spectrum of antimicrobial activity to pathogens including Gram-negative and Gram-positive bacteria, and fungi with the minimum inhibitory concentration(MIC)below 3.0 μM. 6Electrochemical detection of methyl-paraoxon based on bifunctional cerium oxide nanozyme with catalytic activity and signal amplification effect显示文摘A new electrochemical sensor for organophosphate pesticide(methyl-paraoxon)detection based on bifunctional cerium oxide(CeO_(2))nanozyme is here reported for the first time.Methyl-paraoxon was degraded into p-nitrophenol by using CeO_(2) with phosphatase mimicking activity.The CeO_(2) nanozymemodified electrode was then synthesized to detect p-nitrophenol.Cyclic voltammetry was applied to investigate the electrochemical behavior of the modified electrode,which indicates that the signal enhancement effect may attribute to the coating of CeO_(2) nanozyme.The current research also studied and discussed the main parameters affecting the analytical signal,including accumulation potential,accumulation time,and pH.Under the optimum conditions,the present method provided a wider linear range from 0.1 to 100 mmol/L for methyl-paraoxon with a detection limit of 0.06 mmol/L.To validate the proof of concept,the electrochemical sensor was then successfully applied for the determination of methyl-paraoxon in three herb samples,i.e.,Coix lacryma-jobi,Adenophora stricta and Semen nelumbinis.Our findings may provide new insights into the application of bifunctional nanozyme in electrochemical detection of organophosphorus pesticide.Yuzhou Sun Jinchao Wei Jian Zou Zehua Cheng Zhongming Huang Liqiang Gu Zhangfeng Zhong Shengliang Li Yitao Wang Peng Li 2021Journal of Pharmaceutical Analysis2021,11,5:1

13A Stable Open-Shell Conjugated Diradical Polymer with Ultra-High Photothermal Conversion Efficiency for NIR-Ⅱ Photo-Immunotherapy of Metastatic Tumor显示文摘Massive efforts have been concentrated on the advance of eminent near-infrared(NIR) photothermal materials(PTMs) in the NIR-Ⅱ window(1000–1700 nm), especially organic PTMs because of their intrinsic biological safety compared with inorganic PTMs. However, so far, only a few NIR-Ⅱresponsive organic PTMs was explored, and their photothermal conversion efficiencies(PCEs) still remain relatively low. Herein, donor–acceptor conjugated diradical polymers with open-shell characteristics are explored for synergistically photothermal immunotherapy of metastatic tumors in the NIR-Ⅱ window. By employing side-chain regulation, the conjugated diradical polymer TTB-2 with obvious NIR-Ⅱ absorption was developed, and its nanoparticles realize a record-breaking PCE of 87.7% upon NIR-Ⅱ light illustration. In vitro and in vivo experiments demonstrate that TTB-2 nanoparticles show good tumor photoablation with navigation of photoacoustic imaging in the NIR-Ⅱ window, without any side-effect. Moreover, by combining with PD-1 antibody,the pulmonary metastasis of breast cancer is high-effectively prevented by the efficient photo-immunity effect. 16Facile preparation of multifunctional Cu_(2−x)S/S/rGO composite for all-round residual water remediation during interfacial solar driven water evaporation process显示文摘Presently,interfacial solar water evaporation(ISWE)is now injecting new vitality into the field of water remediation.However,during the ISWE process,the nonvolatile pollutants might be concentrated in residual water,and further contaminate the environment.Preparing advanced photothermal materials is in need to get comprehensive purification of various pollutants in residual water.Herein,we report a facile laser thermal method to prepare Cu_(2−x)S/sulfur/reduced graphene oxide(Cu_(2−x)S/S/rGO)nanocomposites for realizing all-round residual water remediation during the ISWE process.The as-prepared Cu2−xS/S/rGO nanocomposites demonstrated excellent photothermal and photocatalytic properties.Through blending with GO nanosheets having excellent adsorption capacity,the synergetic effect of photothermal,photocatalytic,and adsorption properties resulted in highly efficient purification of rhodamine B,bacterial,and heavy metal ions in residual water during the ISWE process.The experimental results also showed that,increasing solar light intensity can promote the residual water remediation,but ultrafast water evaporation under high light intensity may deteriorate the purifying effect.This report may pave a new way to prepare multifunctional materials for water remediation through the ISWE technology.Chaorui Xue Yang Shen Qian Zhang Qing Chang Ning Li Ying Li Wenjing Zheng Shengliang Hu Jinlong Yang 2023Nano Research2023,16,4:0
17Neurons generated from carcinoma stem cells support cancerprogression显示文摘Recent evidences show that nervous system acts as a crucial part of cancer microenvironment.Infiltration of nerve fibers into cancer microenvironment has an important active role in cancer progression.The stimulations of both cancer growth and metastasis by members of nervous system such as neurons and glial cells have been demonstrated.However,how the nervous system is built in cancer is largely unknown.Here we show that a fraction of cancer stem cells(CSCs)derived from patients with gastric carcinoma and colorectal carcinoma are capable of producing neurons that are involved in tumor neurogenesis and tumor growth.Cancer stem cell monoclone derived from a single cancer stem cell was able to generate neurons including sympathetic and parasympathetic neurons to take part in the nervous system in cancer tissues.Knocking down the neural cell generating capability of the human CSCs inhibited the growth of xenograft tumors in mouse model.Our data demonstrate that human CSCs are able to produce one of most important components in the cancer microenvironment that are required for cancer development and progression.Ran Lu Chuanwen Fan Wenqi Shangguan Yuan Liu Yu Li Yanna Shang Dongqin Yin Shengliang Zhang Qiaorong Huang Xue Li Wentong Meng Hong Xu Zongguang Zhou Jiankun Hu Weimin Li Lunxu Liu Xianming Mo 2017Signal Transduction and Targeted Therapy2017,2,1:0
18Hydrogen bonding-induced oxygen clusters and long-lived room temperature phosphorescence from amorphous polyols显示文摘Developing non-conjugated luminescent polymers(NCLPs)with fluorescence and long-lived roomtemperature phosphorescence is of great significance for revealing the essence of NCLPs luminescence,which has gradually attracted the attention of researchers in recent years.Herein,polymethylol(PMO)and poly(3-butene-1,2-diol)(PBD)with polyhydroxy structures were prepared and their luminescence behaviors were investigated to reveal the clusteroluminescence(CL)mechanism.Compared with polyvinyl alcohol with non-luminescent behavior,PMO and PBD exhibit cyan-blue fluorescence with quantum yields of ca.12%and green room-temperature phosphorescence with lifetimes of ca.89 ms in the solid state.Both fluorescence and phosphorescence exhibit typical excitation-dependent CL behavior.Experimental and theoretical analyses show that the strong hydrogen-bonding interaction of PMO and PBD greatly promotes the formation of oxygen clusters and the through-space n-n interaction of oxygen atoms,enabling fluorescence and phosphorescence emission.Our results have enormous implications for understanding the CL mechanism of NCLPs and provide a new polymer design strategy for the rational design of novel NCLPs materials.Ya-Ling Wang Kang Chen Hai-Ru Li Bo Chu Zishan Yan Hao-Ke Zhang Bin Liu Shengliang Hu Yongzhen Yang 2023Chinese Chemical Letters2023,34,1:0
19De novo design of highly efficient type-Ⅰphotosensitizer based onπ-conjugated oligomer for photodynamic eradication of multidrug-resistant bacterial infections显示文摘Traditional photosensitizers show limited singlet oxygen generation in hypoxic infection lesions,which greatly suppress their performance in antibacterial therapy.Meanwhile,there still is lack of feasible design strategy for developing hypoxia-overcoming photosensitizers agents.Herein,radical generation ofπ-conjugated small molecules is efficiently manipulated by an individual selenium(Se)substituent.With this strategy,the first proof-of-concept study of a Se-anchored oligo(thienyl ethynylene)(OT-Se)with high-performance superoxide radical(O_(2)^(·-))and hydroxyl radical(·OH)generation capability is present,and achieves efficient antibacterial activities towards the clinically extracted multidrug-resistant bacteria methicillin-resistant S.aureus(MRSA)and carbapenem-resistant E.coli(CREC)at sub-micromolar concentration under a low white light irradiation(30 mW/cm^(2)).The water-dispersible OT-Se shows a good bacteria-anchoring capability,biocompatibility,and complete elimination of multidrug-resistant bacteria wound infection in vivo.This work offers a strategy to boost type-I photodynamic therapy(PDT)performance for efficient antibacterial treatments,advancing the development of antibacterial agents.Qi Zhao Guangchao Qing Jie Yu Ying Liu Jianliang Shen Yang Luo Xingjie Zan Shengliang Li 2024Chinese Chemical Letters2024,35,3:0
