1Abrupt Holocene changes of the Asian monsoon at millen-nial-and centennial-scales: Evidence from lake sediment document in Minqin Basin, NW China显示文摘Considerable research is now focusing on abrupt Holocene changes at millennial- and centennial-scales all over the world. This study shows that the changes in theHolocene summer monsoon can be divided into two main periods, based on climatic proxy records of lake sediments from the Sanjiaocheng section located at the NW boundary of the summer monsoon, Gansu Province. The early and middle Holocene was humid with stronger summer monsoon, while the late Holocene is dry with weak summer monsoon. Detailed results also show a series of millennial- and centennial-scale changes in the intensity of the summer monsoon, with a periodicity of 1600 years during the whole Holocene, and with a periodicity of 800 years during the early Holocene. Ten dry events during the Holocene are associated with the weakening of the summer monsoon. 2Autologous hematopoietic stem cell transplantation in chemotherapy-sensitive lymphoblastic lymphoma: treatment outcome and prognostic factor analysis显示文摘Objective: The study evaluated the effectiveness of autologous hematopoietic stem cell transplantation(AHSCT) in the treatment of lymphoblastic lymphoma(LL).Methods: We retrospectively analyzed the data from 41 patients with chemotherapy-sensitive LL who underwent hematopoietic stem cell transplantation(HSCT) from December 1989 to December 2009 in a single institution.Results: HSCT was conducted as first-line consolidation therapy and salvage therapy in 36 and 5 patients, respectively. The median follow-up was 97.1 months(range, 24.6-173.1 months). The 5-year overall survival(OS) and event-free survival(EFS) rate were 64% and 47% for the initially treated patients, respectively, and were both 20% for the relapsed ones. 3Autologous peripheral blood stem cell mobilization following dose-adjusted cyclophosphamide, doxorubicin, vincristine and prednisolone chemotherapy alone or in combination with rituximab in treating high-risk non-Hodgkin's lymphoma显示文摘Background: The regimen of cyclophosphamide, doxorubicin, vincristine, and prednisolone(CHOP) is an eicient treatment of non-Hodgkin's lymphoma(NHL). This study aimed to assess the eicacy and toxicity of dose-adjusted CHOP alone or in combination with rituximab(R-CHOP) by examining the stem cell mobilization in NHL patients. Factors afecting the collection of CD34+ cells were also explored.Methods: Our retrospective study included 39 patients eligible for autologous stem cell transplantation: 14 patients who expressed CD20 and were inancially eligible received R-CHOP for autologous peripheral blood stem cell(APBSC) mobilization; the remaining 25 patients received CHOP.Results: The median CD34+ cell yield was 7.01 × 106 cells/kg body weight(range 1.49–28.39 × 106 cells/kg body weight), with only two patients failing to meet the target CD34+ cell harvest of ber of apheresis procedures per patient was 1(range 1–3). The≥2.0 APBS× 106 cells/kg body weight. The median numC mobilization yield of the CHOP group appeared to be higher than that of the R-CHOP group(P response(CR) rate in = 0.005), whereas the success rate was similar between groups. R-CHOP elevated the completeB cell lymphoma patients as compared with CHOP(P = 0.01). 11Pathobiology of ovarian carcinomas显示文摘Ovarian tumors comprise a heterogeneous group of lesions, displaying distinct tumor pathology and oncogenic potentiel. These tumors are subdivided into three main categories: epithelial, germ cell, and sex-cord stromal tumors. We report herein the newly described molecular abnormalities in epithelial ovarian cancers(carcinomas). Immunohistochemistry and molecular testing help pathologists to decipher the significant heterogeneity of this disease. 13Biallelic variants in RBM42 cause a multisystem disorder with neurological,facial,cardiac,and musculoskeletal involvement显示文摘Here,we report a previously unrecognized syndromic neurodevelopmental disorder associated with biallelic loss-of-function variants in the RBM42 gene.The patient is a 2-year-old female with severe central nervous system(CNs)abnormalities,hypotonia,hearing loss,congenital heart defects,and dysmorphic facial features.Familial whole-exome sequencing(WEs)reveals that the patient has two compound heterozygous variants,c.304C>T(p.R102*)and c.1312G>A(p.A438T),in the RBM42 gene which encodes an integral component of splicing complex in the RNA-binding motif protein family.The p.A438T variant is in the RRM domain which impairs RBM42 pro-tein stability in vivo.Additionally,p.A438T disrupts the interaction of RBM42 with hnRNP K,which is the causa-tive gene for Au-Kline syndrome with overlapping disease characteristics seen in the index patient.The human R102*or A438T mutant protein failed to fully rescue the growth defects of RBM42 ortholog knockout△FgRbp1 in Fusarium while it was rescued by the wild-type(WT)human RBM42.A mouse model carying Rbm42 compound heterozygous variants,c.280C>T(p.Q94*)and c.1306_1308delinsACA(p.A436T),demonstrated gross fetal develop-mental defects and most of the double mutant animals died by E13.5.RNA-seq data confirmed that Rbm42 was involved in neurological and myocardial functions with an essential role in alternative splicing(As).Overall,we present clinical,genetic,and functional data to demonstrate that defects in RBM42 constitute the underlying etiology of a new neurodevelopmental disease which links the dysregulation of global AS to abnormal embryonic development.Yiyao Chen Bingxin Yang Xiaoyu Merlin Zhang Songchang Chen Minhui Wang Liya Hu Nina Pan Shuyuan Li Weihui Shi Zhenhua Yang Li Wang Yajing Tan Jian Wang Yanlin Wang Qinghe Xing Zhonghua Ma Jinsong Li He-Feng Huang Jinglan Zhang Chenming Xu 2024Protein & Cell2024,15,1:0
14Nickel-Carnosine complex:A new carrier for enzymes immobilization by affinity adsorption显示文摘Immobilization is an effective method to promote the application of enzyme industry for improving the stability and realizing recovery of enzyme.To some extent,the performance of immobilized enzyme depends on the choice of carrier material.Therefore,the development of new carrier materials has been one of the key issues concerned by enzyme immobilization researchers.In this work,a novel organic–inorganic hybrid material,nickel-carnosine complex(NiCar),was synthesized for the first time by solvothermal method.The obtained NiCar exhibits spherical morphology,hierarchical porosity and abundant unsaturated coordination nickel ions,which provide excellent anchoring sites for the immobilization of proteins.His-tagged organophosphate-degrading enzyme(Opd A)and x-transaminase(ω-TA)were used as model enzymes to evaluate the performance of NiCar as a carrier.By a simple adsorption process,the enzyme molecules can be fixed on the particles of NiCar,and the stability and reusability are significantly improved.The analysis of protein adsorption on NiCar verified that the affinity adsorption between the imidazole functional group on the protein and the unsaturated coordination nickel ions on NiCar was the main force in the immobilization process,which provided an idea way for the development of new enzyme immobilization carriers.Junyang Xu Yanjun Jiang Liya Zhou Li Ma Zhihong Huang Jiafu Shi Jing Gao Ying He 2021Chinese Journal of Chemical Engineering2021,34,10:0
15Fine mapping and validation of a stable QTL for thousand-kernel weight in wheat(Triticum aestivum L.)显示文摘Thousand-kernel weight(TKW)is a measure of grain weight,a target of wheat breeding.The object of this study was to fine-map a stable quantitative trait loci(QTL)for TKW and identify its candidate gene in a recombinant inbred line(RIL)population derived from the cross of Kenong 9204(KN9204)and Jing411(J411).On a high-density genetic linkage map,24,26 and 25 QTL were associated with TKW,kernel length(KL),and kernel width(KW),respectively.A major and stable QTL,QTkw-2D,was mapped to an8.3 cM interval on chromosome arm 2DL.By saturation of polymorphic markers in its target region,QTkw-2D was confined to a 9.13 Mb physical interval using a secondary mapping population derived from a residually heterozygous line(F6:7).This interval was further narrowed to 2.52 Mb using QTkw-2D near-isogenic lines(NILs).NILs~(KN9204)had higher fresh and dry weights than NILsJ411at various grain-filling stages.The TKW and KW of NILs~(KN9204)were much higher than those of NILsJ411in field trials.By comparison of both DNA sequence and expression between KN9204 and J411,TraesCS2D02G460300.1(TraesKN2D01HG49350)was assigned as a candidate gene for QTkw-2D.This was confirmed by RNA sequencing(RNA-seq)of QTkw-2D NILs.These results provide the basis of map-based cloning of QTkw-2D,and DNA markers linked to the candidate gene may be used in marker-assisted selection.Deyuan Meng Aamana Batool Yazhou Xuan Ruiqing Pan Na Zhang Wei Zhang Liya Zhi Xiaoli Ren Wenqing Li Jijie Li Yanxiao Niu Shuzhi Zheng Jun Ji Xiaoli Shi Lei Wang Hongqing Ling Chunhua Zhao Fa Cui Xigang Liu Junming Li Liqiang Song 2023The Crop Journal2023,11,5:0
16Single-cell analysis reveals an Angpt4-initiated EPDC-EC-CM cellular coordination cascade during heart regeneration显示文摘Mammals exhibit limited heart regeneration ability,which can lead to heart failure after myocardial infarction.In contrast,zebrafish exhibit remarkable cardiac regeneration capacity.Several cell types and signaling pathways have been reported to participate in this process.However,a comprehensive analysis of how different cells and signals interact and coordinate to regulate cardiac regeneration is unavailable.We collected major cardiac cell types from zebrafish and performed high-precision single-cell transcriptome analyses during both development and post-injury regeneration.We revealed the cellular heterogeneity as well as the molecular progress of cardiomyocytes during these processes,and identified a subtype of atrial cardiomyocyte exhibiting a stem-like state which may transdifferentiate into ventricular cardiomyocytes during regeneration.Furthermore,we identified a regeneration-induced cell(RIC)population in the epicardium-derived cells(EPDC),and demonstrated Angiopoietin 4(Angpt4)as a specific regulator of heart regeneration.angpt4 expression is specifically and transiently activated in RIC,which initiates a signaling cascade from EPDC to endocardium through the Tie2-MAPK pathway,and further induces activation of cathepsin K in cardiomyocytes through RA signaling.Loss of angpt4 leads to defects in scar tissue resolution and cardiomyocyte proliferation,while overexpression of angpt4 accelerates regeneration.Furthermore,we found that ANGPT4 could enhance proliferation of neonatal rat cardiomyocytes,and promote cardiac repair in mice after myocardial infarction,indicating that the function of Angpt4 is conserved in mammals.Our study provides a mechanistic understanding of heart regeneration at single-cell precision,identifies Angpt4 as a key regulator of cardiomyocyte proliferation and regeneration,and offers a novel therapeutic target for improved recovery after human heart injuries.Zekai Wu Yuan Shi Yueli Cui Xin Xing Liya Zhang Da Liu Yutian Zhang Ji Dong Li Jin Meijun Pang Rui-Ping Xiao Zuoyan Zhu Jing-Wei Xiong Xiangjun Tong Yan Zhang Shiqiang Wang Fuchou Tang Bo Zhang 2023Protein & Cell2023,14,5:0
