1Mycobacterium tuberculosis latency-associated antigen Rv1733c SLP improves the accuracy of differential diagnosis of active tuberculosis and latent tuberculosis infection显示文摘Background: Differential diagnosis of active tuberculosis (ATB) and latent tuberculosis infection (LTBI) has been a challenge for clinicians in high TB burden countries. The purpose of this study was to improve the accuracy of differential diagnosis of ATB and LTBI by using fluorescent immunospot (FluoroSpot) assay to detect specific Th1 cell immune responses. The novelmycobacterium tuberculosis (MTB) latency-associated antigens Rv1733c and synthetic long peptides derived from Rv1733c (Rv1733c SLP) were used based on virulence factors early secreting antigen target-6 (ESAT-6) and culture filtrate protein-10 (CFP-10).Methods: Fifty-seven ATB cases, including 20 pathogen-confirmed ATB and 37 clinically diagnosed ATB, and 36 LTBI cases, were enrolled between January and December 2017. FluoroSpot assay was used to detect the interferon γ (IFN-γ) and interleukin 2 (IL-2) secreted by the specific T cells after being stimulated with MTB virulence factors ESAT-6 and CFP-10, MTB latency-associated antigens Rv1733c and Rv1733c SLP. The receiver operating characteristic (ROC) curve was used to define the best cutoff value of latency-associated antigens in the use of differentiating ATB and LTBI. The sensitivity, specificity, predictive value, and likelihood ratio of ESAT-6 and CFP-10-FluoroSpot combined with latency-associated antigen in the differential diagnosis of ATB and LTBI were also calculated.Results: Following the stimulation with Rv1733c and Rv1733c SLP, the frequency of single IL-2-secreting T cells stimulated by Rv1733c SLP had the largest area under the ROC curve, which was 0.766. With a cutoff value of 1 (spot-forming cells [SFCs]/2.5 × 105 peripheral blood mononuclear cells) for frequency, the sensitivity and specificity of distinguishing ATB from LTBI were 72.2% and 73.7%, respectively. ESAT-6 and CFP-10-FluoroSpot detected the frequency and proportion of single IFN-γ-secreting T cells;the sensitivity and specificity of distinguishing ATB from LTBI were 82.5% and 66.7%, respectively. Combined with the frequency of single IL-2-secreting T cells stimulated by Rv1733c SLP on the basis of ESAT-6 and CFP-10-FluoroSpot, the sensitivity and specificity increased to 84.2% and 83.3%, respectively.Conclusion: Rv1733c SLP, combined with ESAT-6 and CFP-10, might be used as a candidate antigen for T cell-based tuberculosis diagnostic tests to differentiate ATB from LTBI.Lifan Zhang Huimin Ma Shijun Wan Yueqiu Zhang Mengqiu Gao Xiaoqing Liu 2022Chinese Medical Journal2022,,1:2
2Paleoecology of Oligocarpia gothanii Halle from the Middle Permian of Southeastern Shanxi,North China显示文摘Fossil ferns are abundant and diverse in the Permian Cathaysian Flora of Shanxi, North China. Although plants assemblages of the Carboniferous and Permian have been extensively investigated, their palaeoecology and especially the paleoecology of ferns has not been studied in detail. Recent reinvestigation of Oligocarpia gothanii has revealed its growth habit and cloning reproduction strategy, but most previous studies lack integrated sedimentological and taphonomic data. In this study, new materials of O. gothanii have been collected from a floodplain setting in the middle Permian Lower Shihhotse Formation of southeastern Shanxi. These plant fossils were found in a lenticular claystone associated with rooting structures. They are characterized by aphlebia at the base of the penultimate rachis. Sedimentological, taphonomic and morphological analyses were conducted to understand the growth habit and ecology of the fern. 4A Novel Material for Selective Separation of Mono-galactosyldiacylglycerols from Microalgae显示文摘Monogalactosyldiacylglycerols(MGDGs)have potential applications in food products,cosmetics and pharmaceuticals.MGDGs from microalgae with high amounts of polyunsaturated fatty acids(PUFAs)have potential functions,which arise the interest of the researchers.MGDGs were prepared by silica gel column chromatography with the appropriate mobile phase,while due to the similarity to molecular structure of MGDGs,digalactosyldiacylglycerols(DGDGs)were the most difficult impurities to separate during the extraction process of MGDGs from Nannochloropsis oceanica IMET1 and Arthrospira platensis.In order to obtain MGDGs from microalgae using low toxic solvent system,a novel material Click thiol-ene cysteine(Click TE-Cys)was employed to achieve its selective separation by differentiation of the hydrophilic interaction.The mixture of MGDGs and DGDGs standards were separated from each other by Click TE-Cys solid phase extraction(SPE),which was further confirmed by the result of LC-MS.The molecular interaction of MGDGs and DGDGs with Click TE-Cys demonstrated that DGDGs had more hydrogen bonds with Click TE-Cys material,which might cause a higher hydrophilic interaction.In this study,the Click TE-Cys material exhibited higher hydrophilicity with DGDGs and effectively separated MGDGs from 4 species microalgae by‘flow-through'mode using ethanol as mobile phase.JIANG Junpeng YANG Miao CAO Xupeng WAN Huihui LIU Shuqin ZHONG Shijun YAN Jingyu XUE Song 2021Journal of Ocean University of China2021,20,1:0
5Cretaceous integrative stratigraphy,biotas,and paleogeographical evolution of the Qinghai-Tibetan Plateau and its surrounding areas显示文摘The Cretaceous Period is a vital time interval in deciphering the evolutionary history of the Neo-Tethys Ocean and the convergence of different plates and blocks across the Qinghai-Tibetan Plateau.A detailed stratigraphic framework and paleogeographic patterns are the basis for understanding the evolution of the Neo-Tethys Ocean and the formation of the QinghaiTibetan Plateau.Here,the Cretaceous stratigraphy,biota,paleogeography,and major geological events in the Qinghai-Tibetan Plateau are analyzed to establish an integrative stratigraphic framework,reconstruct the paleogeography during the Cretaceous Period,and decode the history of the major geological events.The Cretaceous rocks of the Qinghai-Tibetan Plateau and its surrounding area are predominantly marine deposits,with a small amount of interbedded marine-terrestrial and terrestrial conponents.The Indus-Yarlung Tsangpo Suture Zone was responsible for the deposition of deep marine sediments dominated by ophiolite suites and radiolarian silicalite.To the south,the Tethys Himalayas and Indus Basin received marine sediments of varying depths and lithology;to the north,the Xigaze and Ladakh forearc basins are also filled with marine sediments.The Lhasa Block,Karakorum Block,western Tarim Basin,and West Burma block consist of shallow marine,interbedded marine-terrestrial,and terrestrial sediments.The Qiangtang Basin and other areas are dominated by terrestrial sedimentation.The Cretaceous strata of the Qinghai-Tibetan Plateau and its surrounding areas are widely distributed and diversified,with abundant foraminifera,calcareous nannofossils,radiolarians,ammonites,bivalves,and palynomorphs.On the basis of integrated lithostratigraphic,biostratigraphic,geochronologic,and chemostratigraphic analyses,we proposed herein a comprehensive stratigraphic framework for the Cretaceous Period of the eastern Neo-Tethys.By analyzing the Cretaceous biota of different biogeographic zones from eastern Neo-Tethys and its surrounding areas,we reconstructed the paleobiogeography of different periods of eastern NeoTethys.The Cretaceous paleogeographic evolution of the Qinghai-Tibetan Plateau and its surrounding areas can be divided into three phases:(1)gradual breakup of the Indian Plate from the Australia-Antarctica continent and the early collision phase of the Lhasa-Qiangtang blocks(145-125 Ma);(2)northward drift of the Indian Plate and the collision phase of the Lhasa-Qiangtang blocks(125-100 Ma);(3)rapid northward drift of the Indian Plate,formation of the Tarim-Tajik-Karakorum Bay,and early uplift of the Gangdise Mountains(100-66 Ma).The Indus-Tethys Himalayan biota underwent a transition from the cold-water type in the high latitudes of the southern hemisphere to the warm-water type near the equator from the Early Cretaceous to the MidCretaceous.The biodiversity and abundance of the eastern Neo-Tethys Ocean increased gradually in the Early Cretaceous,peaking in the Mid-Cretaceous,and decreased sharply during the late Late Cretaceous(late Maastrichtian).Along with the northward drift of the Indian Plate and subduction of the Neo-Tethys,the eastern Neo-Tethys and its surrounding areas experienced a series of major geological events,including the formation of the large igneous province,oceanic anoxia events,and mass extinction,etc.Dangpeng XI Guobiao LI Shijun JIANG Xin RAO Tian JIANG Tianyang WANG Zuohuan QIN Yasu WANG Jianzhong JIA Muhammad KAMRAN Zhongye SHI Yuyang WU Panxi WANG Lixin SUN Qinggao ZENG Xiaoqiao WAN 2024Science China Earth Sciences2024,67,4:0
