| 1 | Rhizospheric microbial communities are driven by Panax ginseng at different growth stages and biocontrol bacteria alleviates replanting mortality显示文摘The cultivation of Panax plants is hindered by replanting problems, which may be caused by plantdriven changes in the soil microbial community. Inoculation with microbial antagonists may efficiently alleviate replanting issues. Through high-throughput sequencing, this study revealed that bacterial diversity decreased,whereas fungal diversity increased, in the rhizosphere soils of adult ginseng plants at the root growth stage under different ages. Few microbial community, such as Luteolibacter, Cytophagaceae, Luteibacter, Sphingomonas,Sphingomonadaceae, and Zygomycota, were observed; the relative abundance of microorganisms, namely,Brevundimonas, Enterobacteriaceae, Pandoraea, Cantharellales, Dendryphion, Fusarium, and Chytridiomycota,increased in the soils of adult ginseng plants compared with those in the soils of 2-year-old seedlings. Bacillus subtilis 50-1, a microbial antagonist against the pathogenic Fusarium oxysporum, was isolated through a dual culture technique. These bacteria acted with a biocontrol efficacy of 67.8%. The ginseng death rate and Fusarium abundance decreased by 63.3% and 46.1%, respectively, after inoculation with B. subtilis 50-1. Data revealed that microecological degradation could result from ginseng-driven changes in rhizospheric microbial communities;these changes are associated with the different ages and developmental stages of ginseng plants. | 3 | Integrated metabolomic and transcriptomic analyses revealed the distribution of saponins in Panax notoginseng显示文摘Panax notoginseng is famous for its important therapeutic effects. Saponins are bioactive compounds found in different parts and developmental stages of P. notoginseng plants. Thus, it is urgently to study saponins distribution in different parts and growth ages of P. notoginseng plants. In this study,potential biomarkers were found, and their chemical characteristic differences were revealed through metabolomic analysis. High-performance liquid chromatography data indicated the higher content of saponins(i.e., Rg1, Re, Rd, and Rb1) in the underground parts than that in the aerial parts. 20(S)-Protopanaxadiol saponins were mainly distributed in the aerial parts. Additionally, the total saponin content in the 3-year-old P. notoginseng plant(188.0 mg/g) was 1.4-fold higher than that in 2-year-old plant(130.5 mg/g). The transcriptomic analysis indicated the tissue-specific transcription expression of genes, namely, PnFPS, PnSS, PnSE1, PnSE2, and PnDS, which encoded critical synthases in saponin biosyntheses. These genes showed similar expression patterns among the parts of P. notoginseng plants.The expression levels of these genes in the flowers and leaves were 5.2 fold higher than that in the roots and fibrils. These results suggested that saponins might be actively synthesized in the aerial parts and transformed to the underground parts. | 6 | Development of nutritious rice with high zinc/selenium and low cadmium in grains through QTL pyramiding显示文摘Enriching zinc(Zn) and selenium(Se) levels,while reducing cadmium(Cd) concentration in rice grains is of great benefit for human diet and health.Large natural variations in grain Zn, Se, and Cd concentrations in different rice accessions enable Zn/Sebiofortification and Cd-minimization through molecular breeding. Here, we report the development of new elite varieties by pyramiding major quantitative trait loci(QTLs) that significantly contribute to high Zn/Se and low Cd accumulation in grains. A chromosome segment substitution line CSSLGCC7 with the PA64s-derived GCC7 allele in the 93-11 background, exhibited steadily higher Mn and lower Cd concentrations in grains than those of 93-11. This elite chromosome segment substitution line(CSSL) was used as the core breeding material to cross with CSSLs harboring other major QTLs for essential mineral elements, especially CSSLGZC6 for grain Zn concentration and CSSLGSC5 for grain Se concentration. The CSSLGCC7+GZC6 and CSSLGCC7+GSC5 exhibited lower Cd concentration with higher Zn and Se concentrations in grains, respectively. | 8 | A chromosome-scale genome assembly of Artemisia argyi reveals unbiased subgenome evolution and key contributions of gene duplication to volatile terpenoid diversity显示文摘Artemisia argyi Le´vl.et Vant.,a perennial Artemisia herb with an intense fragrance,is widely used in traditional medicine in China and many other Asian countries.Here,we present a chromosome-scale genome assembly of A.argyi comprising 3.89 Gb assembled into 17 pseudochromosomes.Phylogenetic and comparative genomic analyses revealed that A.argyi underwent a recent lineage-specificwhole-genomeduplication(WGD)event after divergence fromArtemisia annua,resulting in two subgenomes.Wedeciphered the diploid ancestral genome of A.argyi,and unbiased subgenome evolution was observed.The recent WGD led to a large number of duplicated genes in the A.argyi genome.Expansion of the terpene synthase(TPS)gene family through various types of gene duplication may have greatly contributed to the diversity of volatile terpenoids in A.argyi.In particular,we identified a typical germacrene D synthase gene cluster within the expanded TPS gene family.The entire biosynthetic pathways of germacrenes,(+)-borneol,and(+)-camphor were elucidated in A.argyi.In addition,partial deletion of the amorpha-4,11-diene synthase(ADS)gene and loss of function of ADS homologs may have resulted in the lack of artemisinin production in A.argyi.Our study provides newinsights into the genome evolution of Artemisia and lays a foundation for further improvement of the quality of this important medicinal plant. | Hongyu Chen Miaoxian Guo Shuting Dong Xinling Wu Guobin Zhang Liu He Yuannian Jiao Shilin Chen Li Li Hongmei Luo | 2023 | Plant Communications2023,4,3: | 2 |
| 9 | Effects of warming on soil respiration during the non-growing seasons in a semiarid temperate steppe显示文摘尽管许多研究认为增温对陆地生态系统非生长季土壤呼吸的影响非常重要,但是关于这一问题仍缺乏充分的理解。本论文研究了非生长季增温对温带半干旱草原土壤呼吸的影响,并探讨了其影响机制。本实验位于温带半干旱典型草原,包括10对样地,每对样地包括一个对照和模拟增温处理。模拟增温样地自2014年10月开始,采用开顶箱(OTC)进行。2014年10月到2015年4月,2015年10月到2016年4月期间每月测定几次土壤呼吸、土壤温度与土壤湿度;并测定0-20 cm微生物量碳(MBC)、微生物量氮(MBN)和土壤速效氮。采用重复测量方差和成对t检验方法分析增温、时间及其交互作用对土壤呼吸的影响。采用简单回归确定各指标之间的因果关系。结果表明,对照样地非生长季平均土壤呼吸速率为0.24μmol m^(-2) s^(-1),约占当地生长季碳通量的14.4%。模拟增温在两个非生长季使土壤温度和土壤呼吸分别比对照样地提高了1.48℃(P<0.001)和42.1%(P<0.01)。2015-2016年非生长季模拟增温降低了0.66%的土壤湿度,降低幅度不显著。模拟增温处理下2015-2016年非生长季MBC和MBN分别显著提高了19.72%和20.99%(P<0.05)。此外,研究结果还表明,土壤温度、MBC和MBN的变化调控了土壤呼吸对模拟增温的响应。本结果揭示非生长季土壤呼吸变化能影响碳循环的其它组分,并可以有效促进气候变化背景下陆地生态系统碳循环的模型预测。 | Yuan Miao Mengzhou Liu Juan Xuan Wei Xu Shilin Wang Renhui Miao Dong Wang Wei Wu Yinzhan Liu Shijie Han | 2020 | Journal of Plant Ecology2020,13,3: | 2 |
