| 1 | Formyl tetrahydrofolate deformylase affects hydrogen peroxide accumulation and leaf senescence by regulating the folate status and redox homeostasis in rice显示文摘It is well established that an abnormal tetrahydrofolate(THF)cycle causes the accumulation of hydrogen peroxide(H_(2)O_(2))and leaf senescence,however,the molecular mechanism underlying this relationship remains largely unknown.Here,we reported a novel rice tetrahydrofolate cycle mutant,which exhibited H_(2)O_(2)accumulation and early leaf senescence phenotypes.Map-based cloning revealed that HPA1 encodes a tetrahydrofolate deformylase,and its deficiency led to the accumulation of tetrahydrofolate,5-formyl tetrahydrofolate and 10-formyl tetrahydrofolate,in contrast,a decrease in 5,10-methenyl-tetrahydrofolate.The expression of tetrahydrofolate cycle-associated genes encoding serine hydroxymethyl transferase,glycine decarboxylase and 5-formyl tetrahydrofolate cycloligase was significantly down-regulated.In addition,the accumulation of H_(2)O_(2)in hpa1 was not caused by elevated glycolate oxidation.Proteomics and enzyme activity analyses further revealed that mitochondria oxidative phosphorylation complex I and complex V were differentially expressed in hpa1,which was consistent with the H_(2)O_(2)accumulation in hpa1.In a further feeding assay with exogenous glutathione(GSH),a non-enzymatic antioxidant that consumes H_(2)O_(2),the H_(2)O_(2)accumulation and leaf senescence phenotypes of hpa1 were obviously compensated.Taken together,our findings suggest that the accumulation of H_(2)O_(2)in hpa1 may be mediated by an altered folate status and redox homeostasis,subsequently triggering leaf senescence. | Erhui Xiong Guojun Dong Fei Chen Chen Zhang Shan Li Yanli Zhang Jahidul Islam Shohag Xiaoe Yang Yihua Zhou Qian Qian Limin Wu Yanchun Yu | 2021 | Science China(Life Sciences)2021,64,5: | 1 |
| 2 | Characterization of ^(68)Zn uptake,translocation,and accumulation into developing grains and young leaves of high Zn-density rice genotype显示文摘Zinc(Zn) is an essential micronutrient for humans,but Zn deficiency has become serious as equally as iron(Fe) and vitamin A deficiencies nowadays.Selection and breeding of high Zn-density crops is a suitable,cost-effective,and sustainable way to improve human health.However,the mechanism of high Zn density in rice grain is not fully understood,especially how Zn transports from soil to grains.Hydroponics experiments were carried out to compare Zn uptake and distribution in two different Zn-density rice genotypes using stable isotope technique.At seedling stage,IR68144 showed higher 68Zn uptake and transport rate to the shoot for the short-term,but no signifi-cant difference was observed in both genotypes for the long-term.Zn in xylem sap of IR68144 was consistently higher,and IR68144 exhibited higher Zn absorption ratio than IR64 at sufficient(2.0 μmol/L) or surplus(8.0 μmol/L) Zn supply level.IR64 and IR68144 showed similar patterns of 68Zn accumulation in new leaves at seedling stage and in de-veloping grains at ripening stage,whereas 68Zn in new leaves and grains of IR68144 was consistently higher.These results suggested that a rapid root-to-shoot translocation and enhanced xylem loading capacity may be the crucial processes for high Zn density in rice grains. | Md.Jahidul Islam SHOHAG | 2011 | Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2011,12,5: | 1 |
