3Quantitative analysis of the ribbon synapse number of cochlear inner hair cells in C57BL/6J mice using the three-dimensional modeling method显示文摘In mammals,the ribbon synapses of cochlear inner hair cells are a synaptic structure of the first sensory nerve in the pathway of acoustical signal transmission to the acoustic center,and it is directly involved in voice coding and neurotransmitter release. It is difficult to quantitatively analyze the ribbon synaptic number only using an electron microscope,because the ribbon synaptic number is relatively limited and their location is deep. In this study,the specific presynaptic structure-RIBEYE,and non-specific postsynaptic structure-GluR 2 & 3 in C57BL/6J mouse basilar membrane samples were treated by immunofluorescent labeling. Serial section was performed on the samples using a laser scanning confocal microscope,and then the serial sections were used to build three-dimensional models using 3DS MAX software. Each fluorescein color pair indicates one synapse,so the number of ribbon synapses of inner hair cells is obtained. The spatial distribution and the number of ribbon synapses of cochlear inner hair cells were clearly shown in this experiment,and the mean number of ribbon synapses per inner hair cell was 16.10±1.03. Our results have demonstrated the number of ribbon synapses is accurately calculated by double immunofluorescent labeling to presynaptic and postsynaptic structures,serial sections obtained using a laser scanning confocal microscope,and three-dimensional modeling obtained using 3DS MAX software. 5Extranodal involvement in young patients with diffuse large B-cell lymphoma: distribution, prognostic value and treatment options显示文摘Objective: Extranodal involvement represents a peculiar presentation of diffuse large B-cell lymphoma(DLBCL). Previous studies have suggested that older patients are more prone to extranodal involvement. This study retrospectively addressed the distribution, prognostic value and treatment options of extranodal involvement in young patients with DLBCL.Methods: A total of 329 patients were enrolled according to the inclusion requirements. The effects of gender,extranodal involvement, age-adjusted international prognostic index(aa IPI), rituximab infusion and radiotherapy on patient outcomes were evaluated.Results: Among these patients, 59% presented extranodal involvement in 16 anatomic sites. More than one instance was linked to many poorer clinical characteristics and poorer survival compared with either nodal disease or one instance. In patients with one extranodal lesion, multivariate analysis revealed that the site of extranodal involvement, but not the aa IPI or rituximab infusion, was independently related to the outcome, and radiotherapy had a negative influence on survival.Conclusions: Extranodal involvement is common in younger patients and exhibits a ubiquitous distribution.The site of extranodal involvement is of strong prognostic significance. 6Association between 15 known or potential breast cancer susceptibility genes and breast cancer risks in Chinese women显示文摘Objective:There are many hereditary breast cancer patients in China,and multigene panel testing has been a new paradigm of genetic testing for these patients and their relatives.However,the magnitude of breast cancer risks related to multiple breast cancer susceptibility genes are largely unknown in Chinese women.Methods:We screened pathogenic variants in 15 established or potential breast cancer susceptibility genes from 8,067 consecutive Chinese female breast cancer patients and 13,129 Chinese cancer-free female controls.These breast cancer patients were unselected for age at diagnosis or family history.Results:We found that pathogenic variants in TP53[odds ratio(OR):16.9,95%confidence interval(CI):5.2–55.2];BRCA2(OR:10.4,95%CI:7.6–14.2);BRCA1(OR:9.7,95%CI:6.3–14.8);and PALB2(OR:5.2,95%CI:3.0–8.8)were associated with a high risk of breast cancer.ATM,BARD1,CHEK2,and RAD51D were associated with a moderate risk of breast cancer with ORs ranging from 2-fold to 4-fold.In contrast,pathogenic variants of NBN,RAD50,BRIP1,and RAD51C were not associated with increased risk of breast cancer in Chinese women.The pathogenic variants of PTEN,CDH1,and STK11 were very rare,so they had a limited contribution to Chinese breast cancer.Patients with pathogenic variants of TP53,BRCA1,BRCA2,and PALB2 more often had earlyonset breast cancer,bilateral breast cancer,and a family history of breast cancer and/or any cancer.Conclusions:This study provided breast cancer risk assessment data for multiple genes in Chinese women,which is useful for genetic testing and clinical management of Chinese hereditary breast cancer.Fenfen Fu Dongjie Zhang Li Hu Senthil Sundaram Dingge Ying Ying Zhang Shuna Fu Juan Zhang Lu Yao Ye Xu Yuntao Xie 2022Cancer Biology & Medicine2022,19,2:3
7Defect-density control of platinum-based nanoframes with high-index facets for enhanced electrochemical properties显示文摘Structure-engineered platinum-based nanoframes(NFs)at the atomic level can effectively improve the catalytic performance for fuel cells and other heterogeneous catalytic fields.We report herein,a microwave-assisted wet-chemical method for the preparation of platinum-copper-cobalt NFs with tunable defect density and architecture,which exhibit enhanced activity and durability towards the electro-oxidation reactions of methanol(MOR)and formic acid(FAOR).By altering the reduction/capping agents and thus the nucleation/growth kinetics,trimetallic platinum-copper-cobalt hexapod NFs with different density high-index facets are achieved.Especially,the rough hexapod nanoframes(rh-NFs)exhibit excellent specific activities towards MOR and FAOR,7.25 and 5.20 times higher than those of benchmark Pt/C,respectively,along with prolonged durability.The excellent activities of the rh-NFs are assigned to a synergistic effect,including high density of defects and high-index facets,suitable d-band center,and open-framework structure.This synergistic working mechanism opens up a new way for enhancing their electrocatalytic performances by increasing defect density and high-index facets in open-framework platinum-based NFs.Shaohan Yang Shuna Li Lianghao Song Yipin Lv Zhongyao Duan Chunsheng Li Raphael Francesco Praeg Daowei Gao Guozhu Chen 2019Nano Research2019,12,11:3
8Modification of physicochemical properties and degradation of barley flour upon enzymatic extrusion显示文摘In order to evaluate the application potential of enzyme-extruded barley flour(EEBF)in the brewing of whole-barley beer,the physicochemical properties,mashing and fermentation effects of EEBF were studied,and conventional extruded barley flour(CEBF)and steam-cooked barley flour(SCBF)were taken as controls.The water solubility index(WSI)of EEBF(46.88%-52.63%)was much higher than that of CEBF(6.70%)and SCBF(4.59%).The results of degree of gelatinization(DG)and enthalpy(ΔH)suggested that EEBF was almost completely gelatinized,and the rapid visco analyzer(RVA)exhibited that the peak viscosity of EEBF was drastically reduced.The weight-average molecular weight of EEBF(<10^(6) g/mol)was significantly lower than that of CEBF and SCBF.Scanning electron microscopy(SEM)showed that the heat-treatments caused starch granules to break and aggregate,and there were many hydrolyzed pores on the surface of EEBF.Both the long-and short-range order of EEBF were destroyed.Although EEBF caused serious filtration blocking,it greatly improved the alcohol content of finished beer(5.41%vol).Overall,the thermal modification of barley flour by enzyme-extrusion meets the needs of fermentation,and could improve the problem of limited leaching of unmalted barley,which was benefit for the production of whole-barley beer.Shuna Zhao Aiquan Jiao Yueyue Yang Qing Liu Wenqi Wu Zhengyu Jin 2022Food Bioscience2022,45,1:2
9Effect of aqueous extract of Sanweitanxiang powder on calcium homeostasis protein expression in ischemic-reperfusion injury rat heart显示文摘OBJECTIVE: To investigate the underlying mechanism of reduced myocardial ischemia-reperfusion (I/R) injury in rats using the traditionalTibetan medicine Sanweitanxiang powder (SWTX). METHODS: Rats were randomly divided into six groups (n=10) as follows: (a) propranolol dinitrate control group, given propranolol dinitrate 0.02 g/kg for 10 days before I/R, (b) SWTX with a high dose group, given SWTX 1.5 g/kg for 10 days before I/R, (c) SWTX with a medium dose group, given SWTX 1.25 g/kg for 10 days before I/R, (d) sham group (Sham), in which the rat heart was exposed by pericardiotomy but without I/R, (e) SWTX with a low dose group, given SWTX 1.0 g/kg for 10 days before I/R, and (f) I/R injury group. Rats were intragastrically pretreated with propranolol dinitrate orSWTX. After that, the operation to cause ischemia and reperfusion was conducted.The histopathologic changes of rat hearts were observed by hematoxylin and eosin staining and transmission electron microscopy. Ca2+ homeostasis protein expression was determined by western blot. RESULTS: After SWTX pretreatment, the development of ultrastructural pathological changes from IR injury was attenuated. A decrease in the expression of B-cell lymphoma 2 associated X protein, and an increase in the expression of B-cell lymphoma 2 were observed. An increased activation of extracellular signal regulated kinases were found. Compared with the sham group, the expression of sarcoplasmic reticulum calcium-ATPase, phospholamban, and calsequestrin were all up-regulated after pretreatment with SWTX. 11Secure and Efficient Data Storage and Sharing Scheme Based on Double Blockchain显示文摘In the digital era,electronic medical record(EMR)has been a major way for hospitals to store patients’medical data.The traditional centralized medical system and semi-trusted cloud storage are difficult to achieve dynamic balance between privacy protection and data sharing.The storage capacity of blockchain is limited and single blockchain schemes have poor scalability and low throughput.To address these issues,we propose a secure and efficient medical data storage and sharing scheme based on double blockchain.In our scheme,we encrypt the original EMR and store it in the cloud.The storage blockchain stores the index of the complete EMR,and the shared blockchain stores the index of the shared part of the EMR.Users with different attributes can make requests to different blockchains to share different parts according to their own permissions.Through experiments,it was found that cloud storage combined with blockchain not only solved the problem of limited storage capacity of blockchain,but also greatly reduced the risk of leakage of the original EMR.Content Extraction Signature(CES)combined with the double blockchain technology realized the separation of the privacy part and the shared part of the original EMR.The symmetric encryption technology combined with Ciphertext-Policy Attribute-Based Encryption(CP–ABE)not only ensures the safe storage of data in the cloud,but also achieves the consistency and convenience of data update,avoiding redundant backup of data.Safety analysis and performance analysis verified the feasibility and effectiveness of our scheme.Lejun Zhang Minghui Peng Weizheng Wang Yansen Su Shuna Cui Seokhoon Kim 2021Computers, Materials & Continua2021,,1:1
