1Effects of narrow plant spacing on root distribution and physiological nitrogen use efficiency in summer maize显示文摘The objective of this study was to understand the effects of plant spacing on grain yield and root competition in summer maize(Zea mays L.). Maize cultivar Denghai 661 was planted in rectangular tanks(0.54 m × 0.27 m × 1.00 m) under 27 cm(normal) and 6 cm(narrow) plant spacing and treated with zero and 7.5 g nitrogen(N) per plant. Compared to normal plant spacing, narrow plant spacing generated less root biomass in the 0–20 cm zone under both N rates, slight reductions of dry root weight in the 20–40 cm and 40–70 cm zones at the mid-grain filling stage, and slight variation of dry root weights in the 70–100 cm zone during the whole growth period. Narrow plant spacing decreased root reductive activity in all root zones, especially at the grain-filling stage. Grain yield and above-ground biomass were 5.0% and 8.4% lower in the narrow plant spacing than with normal plant spacing, although narrow plant spacing significantly increased N harvest index and N use efficiency in both grain yield and biomass, and higher N translocation rates from vegetative organs. These results indicate that the reductive activity of maize roots in all soil layers and dry weights of shallow roots were significantly decreased under narrow plant spacing conditions, resulting in lower root biomass and yield reduction at maturity. 2Loss of stat3 function leads to spine malformation and immune disorder in zebrafish显示文摘STAT(Signal Transducers and Activators of Transcription) gene family members have been revealed to be involved in cell growth and differentiation in vertebrates. Despite their physiological importance,their functions are poorly studied at organ and systemic levels. In this study, we performed a genome-wide analysis using data from invertebrates to vertebrates to identify STAT genes and analyze their evolutionary history. Interestingly, the STAT gene family undergoes genome duplications during the evolutionary history with STAT3 homologues firstly appearing in the basal extant vertebrate, sea lamprey, suggesting its possible roles in spine formation. To investigate the functions of stat3 in fish species, TALEN technology was performed to generate mutant zebrafish lines. Stat3 mutant zebrafish showed no obvious defects at early developmental stage but displayed severe lateral and vertical curvature of the spine(scoliosis), spine fracture and the incomplete bone joints with narrower junction between vertebrae at early juvenile stage, as indicated by Alizarin red and Alcian blue staining,radiography and micro-computed tomography(Micro CT) analysis. Transcriptome analysis reveals dramatic alterations in a number of genes involved in immune and infection response, skeletal development and somatic growth, especially downregulated expression of collagen gene family, in the juvenile stat3 mutant zebrafish. Moreover, most of the collagen genes were detected to have abnormal expression pattern during the formation of spine deformities in stat3 mutants. 3A novel selective autophagy receptor,CCDC50,delivers K63 polyubiquitination-activated RIG-I/MDA5 for degradation during viral infection显示文摘Autophagy is a con served process that delivers cytosolic substa nces to the lysosome for degradatio n,but its direct role in the regulation of antiviral inn ate immu nity remains poorly un derstood.Here,through high-throughput screeni ng,we discovered that CCDC50 functions as a previously unknown autophagy receptor that negatively regulates the type I interferon(IFN)signaling pathway initiated by RIG-I-like receptors(RLRs),the sensors for RNA viruses.The expression of CCDC50 is enhanced by viral infection,and CCDC50 specifically recognizes K63-polyubiquitinated RLRs,thus delivering the activated RIG-I/MDA5 for autophagic degradation.The associatio n of CCDC50 with phagophore membrane protei n LC3 is con firmed by crystal structure analysis.In contrast to other known autophagic cargo receptors that associate with either the LIR-docking site(LDS)or the UIMdocking site(UDS)of LC3,CCDC50 can bind to both LDS and UDS,representing a new type of cargo receptor.In mouse models with RNA virus infection,CCDC50 deficiency reduces the autophagic degradation of RIG-I/MDA5 and promotes type I IFN responses,resulting in enhaneed viral resistance and improved survival rates.These results reveal a new link between autophagy and antiviral innate immune resp on ses and provide additional in sights into the regulatory mecha nisms of RLR-mediated antiviral signaling.Panpan Hou Kongxiang Yang Penghui Jia Lan Liu Yuxin Lin Zibo Li Jun Li Shuliang Chen Shuting Guo Ji'An Pan Junyu Wu Hong Peng Weijie Zeng Chunmei Li Yingfang Liu Deyin Guo 2021Cell Research2021,31,1:5
4Preparation, characterization, antimicrobial and cytotoxicity studies of copper/zincloaded montmorillonite显示文摘Background: A series of modified montmorillonites(Mt) including zinc-loaded Mt(Zn-Mt), copper-loaded Mt(Cu-Mt), copper/zinc-loaded Mt with different Cu/Zn ratio(Cu/Zn-Mt-1, Cu/Zn-Mt-2, Cu/Zn-Mt-3) were prepared by an ion-exchange reaction, and characterized using X-ray diffraction(XRD), fourier transformed infrared spectroscopy(FTIR) and transmission electron microscopy(TEM). The specific surface areas, antimicrobial activity and cytotoxicity of the modified Mt were investigated.Results: In the modified Mt, hydrated Cu ions and Zn ions were exchanged in the interlayer space of Mt and the particles were irregular shapes. The results showed that Cu/Zn-Mt enhanced antibacterial and antifungal activity compared with Zn-Mt and Cu-Mt possibly due to the synergistic effect between Cu and Zn. Among the Cu/Zn-Mt with different Cu/Zn raitos, Cu/Zn-Mt with a Cu/Zn ratio of 0.98 or 0.51 showed higher antimicrobial activity against gram-negative bacteria(Escherichia coli), gram-positive bacteria(Staphylococcus aureus), fungi(Candida albicans).Moreover, the antimicrobial activity of Cu/Zn-Mt was correlated with its specific surface area. 5A Virulent PEDV Strain FJzz1 with Genomic Mutations and Deletions at the High Passage Level Was Attenuated in Piglets via Serial Passage In Vitro显示文摘Highly virulent porcine epidemic diarrhea virus(PEDV)strains re-emerged and circulated in China at the end of 2010,causing significant economic losses in the pork industry worldwide.To understand the genetic dynamics of PEDV during its passage in vitro,the PEDV G2 strain FJzzl was serially propagated in Vero cells for up to 200 passages.The susceptibility and adaptability of the FJzzl strain increased gradually as it was serially passaged in vitro.Sequence analysis revealed that amino acid(aa)changes were mainly concentrated in the S glycoprotein,which accounted for 72.22%-85.71%of all aa changes.A continuous aa deletion(^(55)I^(56)G^(57)E→^(55)K^(56)Δ^(57)Δ)occurred in the N-terminal domain of S1(Sl-NTD).To examine how the aa changes affected its virulence,FJzzl-F20 and FJzzl-F200 were selected to simultaneously evaluate their pathogenicity in suckling piglets.All the piglets in the FJzzl-F20-infected group showed typical diarrhea at 24 h postinfection,and the piglets died successively by 48 h postinfection.However,the clinical signs of the piglets in the FJzzl-F200-infected group were significantly weaker,and no deaths occurred.The FJzzl-F200-infected group also showed a lower level of fecal viral shedding and lower viral loads in the intestinal tissues,and no obvious histopathological lesions.TypeⅠandⅢinterferon were induced in the FJzzl-F200 infection group,together with pro-inflammatory cytokines,such as TNF-α,IL-1βand IL-8.These results indicate that the identified genetic changes may contribute to the attenuation of FJzzl strain,and the attenuated FJzzl-F200 may have the potential for developing PEDV live-attenuated vaccines.Pengfei Chen Xiongwei Zhao Shuting Zhou Tianxing Zhou Xiangmei Tan Xia Wu Wu Tong Fei Gao Lingxue Yu Yifeng Jiang Hai Yu Zhibiao Yang Guangzhi Tong Yanjun Zhou 2021Virologica Sinica2021,36,5:4
6In-situ assembly of TiO2 with high exposure of(001)facets on three-dimensional porous graphene aerogel for lithium-sulfur battery显示文摘Resulting from the development of electric vehicles,high energy-density Li-S batteries have recently attracted ever-increasing attentions worldwide.However,continuous dissolution of cathodic sulfur and followed shuttle effect of polysulfides lead to very limited service lifetime for currently-applied Li-S batteries.Herein,a 3 D porous graphene aerogel(GA)decorated with high exposure of anatase TiO2(001)nanoplatelets is proposed as robust host to immobilize cathodic sulfur.Compared with commonly used TiO2(101)nanoparticles,the Ti O2(001)nanoplatelets have highly matched lattices with graphene(002)nanosheets,thus facilitating the electronic transfer.The in-site assembled TiO2@GA host exhibits superior sulfur-immobilized capability,which cannot only entrap sulfur by physical confinement,but also capture dissoluble sulfurous species by chemical bonding.The fabricated S@TiO2@GA cathode shows excellent electrochemical performance with high discharge capacity,superior rate capability,and durable cycling stability as well,supposed to be a promising cathode for high-performance Li-S battery applications.Ming Wang Shunyuan Tan Shuting Kan Yufeng Wu Shangbin Sang Kaiyu Liu Hongtao Liu 2020Journal of Energy Chemistry2020,29,10:3
7Construction of robust coupling interface between MoS2 and nitrogen doped graphene for high performance sodium ion batteries显示文摘As a layered inorganic material,MoS2 has recently attracted intensive attention as anode for sodium ion batteries(SIBs).However,this anode is plagued with low electronic conductivity,serious volume expansion and sluggish kinetics,resulting in capacity fading and poor rate performance.Herein,we develop an interface engineering strategy to substantially enhance the sodium storage performance of MoS2 by incorporating layered MoS2 into three dimensional N-doped graphene scaffold.The strong coupling-interface between MoS2 and N-doped graphene scaffold can not only stabilize the MoS2 structure during sodium insertion/extraction processes,but also provide plenty of anchor sites for additional surface sodium storage.The 3D MoS2@N-doped graphene composite as anode for SIBs performs an outstanding specific capacity of 667.3 mA h g^-1 at 0.2 A g^-1,a prolonged stability with a capacity retention of 94.4%after 140cycles and excellent rate capability of 445 mA h g^-1 even at a high rate of 10 A g^-1.We combined experiment and theoretical simulation to further disclose the interaction between MoS2 and N-doped graphene,adsorption and diffusion of sodium on the composite and the corresponding sodium storage mechanism.This study opens a new door to develop high performance SIBs by introducing the interface engineering technique.Yun Qiao Jiawei Wu Xiaoguang Cheng Yudong Pang Zhansheng Lu Xiangdong Lou Qingling Li Jin Zhao Shuting Yang Yang Liu 2020Journal of Energy Chemistry2020,29,9:3
8A chromosome-scale genome assembly of Artemisia argyi reveals unbiased subgenome evolution and key contributions of gene duplication to volatile terpenoid diversity显示文摘Artemisia argyi Le´vl.et Vant.,a perennial Artemisia herb with an intense fragrance,is widely used in traditional medicine in China and many other Asian countries.Here,we present a chromosome-scale genome assembly of A.argyi comprising 3.89 Gb assembled into 17 pseudochromosomes.Phylogenetic and comparative genomic analyses revealed that A.argyi underwent a recent lineage-specificwhole-genomeduplication(WGD)event after divergence fromArtemisia annua,resulting in two subgenomes.Wedeciphered the diploid ancestral genome of A.argyi,and unbiased subgenome evolution was observed.The recent WGD led to a large number of duplicated genes in the A.argyi genome.Expansion of the terpene synthase(TPS)gene family through various types of gene duplication may have greatly contributed to the diversity of volatile terpenoids in A.argyi.In particular,we identified a typical germacrene D synthase gene cluster within the expanded TPS gene family.The entire biosynthetic pathways of germacrenes,(+)-borneol,and(+)-camphor were elucidated in A.argyi.In addition,partial deletion of the amorpha-4,11-diene synthase(ADS)gene and loss of function of ADS homologs may have resulted in the lack of artemisinin production in A.argyi.Our study provides newinsights into the genome evolution of Artemisia and lays a foundation for further improvement of the quality of this important medicinal plant.Hongyu Chen Miaoxian Guo Shuting Dong Xinling Wu Guobin Zhang Liu He Yuannian Jiao Shilin Chen Li Li Hongmei Luo 2023Plant Communications2023,4,3:2
9Effects of sludge characteristics on electrical resistance and energy consumption during electro-dewatering process显示文摘The increase of electrical resistance(ER)and energy consumption(EC)during the later stage of dewatering is a major problem hindering the development of electro-dewatering(EDW)technology.As the variations of sludge characteristics are significant during the EDW process,the relationships between sludge characteristics and ER and EC during EDW remain unclear.In this study,the effects of moisture content(MC),thickness,pH,conductivity,zeta potential,temperature,and gas volume on the ER and EC during the EDW process were statistically investigated using correlation and multiple linear regression analyses.Herein,the results showed that the ER of the sludge near the anode was primarily affected by pH,whereas the sludge near the cathode was primarily affected by the MC and conductivity.Further,sludge temperature and conductivity were the most reliable indicators to predict the EC near the anode and cathode,respectively.The results of this study provide theoretical guidance useful for solving the increase of ER and EC during the later stage of the EDW process.Li Sha Xiaoyan Yu Zhangxiang Wu Xingxin Liu Shuting Zhang 2021Journal of Environmental Sciences2021,33,3:2
10Single-shot AAV-vectored vaccine against SARS-CoV-2 with fast and long-lasting immunity显示文摘Due to the insufficient long-term protection and significant efficacy reduction to new variants of current COVID-19 vaccines,the epidemic prevention and control are still challenging.Here,we employ a capsid and antigen structure engineering(CASE)strategy to manufacture an adenoassociated viral serotype 6-based vaccine(S663V-RBD),which expresses trimeric receptor binding domain(RBD)of spike protein fused with a biological adjuvant RS09.Impressively,the engineered S663V-RBD could rapidly induce a satisfactory RBD-specific IgG titer within 2 weeks and maintain the titer for more than 4 months.Compared to the licensed BBIBP-CorV(Sinopharm,China),a single-dose S663V-RBD induced more endurable and robust immune responses in mice and elicited superior neutralizing antibodies against three typical SARS-CoV-2 pseudoviruses including wild type,C.37(Lambda)and B.1.617.2(Delta).More interestingly,the intramuscular injection of S663V-RBD could overcome pre-existing immunity against the capsid.Given its effectiveness,the CASE-based S663VRBD may provide a new solution for the current and next pandemic.Fuhua Wu Shuang Luo Yongshun Zhang Yangsen Ou Hairui Wang Zhaofei Guo Chunting He Shuting Bai Penghui He Min Jiang Xiaoyan Chen Guangsheng Du Xun Sun 2023Acta Pharmaceutica Sinica B2023,13,5:2
11Electrodynamic Tethered Deorbit System Dynamics and Performance Analysis显示文摘Electrodynamic tethered deorbit technology is a novel way to remove abandoned spacecrafts like upper stages or unusable satellites. This paper investigates and analyses the deorbit performance and mission applicability of the electrodynamic tethered system. To do so, the electrodynamic tethered deorbit dynamics with multi-perturbation is firstly formulated, where the Earth magnetic field, the atmospheric drag, and the Earth oblateness effect are considered. Then, the key system parameters, including payload mass, tether length and tether type, are analyzed by numerical simulations to investigate their influences on the deorbit performance and to give the setting principles for choosing system parameters. 12Alterations in intestinal microbiota composition coincide with impaired intestinal morphology and dysfunctional ileal immune response in growing-finishing pigs under constant chronic heat stress显示文摘Background:Previous studies had shown that short-term acute heat stress(HS)affected the host’s metabolism and intestinal microbiota independent of feed intake(FI)reduction,and long-term calorie restriction caused intestinal morphological injuries and gut microbial alterations.However,research on the effects of constant chronic HS on intestinal microbial composition and the roles of FI reduction played in is limited.This study aimed to investigate the effects of 7-day constant chronic HS on the composition of intestinal microbes in growing-finishing pigs,and its relationship with pigs’performance,intestinal morphology,and ileal immune response.Twenty-four growingfinishing pigs(Duroc×Large White×Landrace,30±1 kg body weight)were randomly assigned to three treatments(n=8),1)thermal neutral(TN)conditions(25±1℃)with ad libitum FI,2)HS conditions(35±1℃)with ad libitum FI,3)pair-fed(PF)with HS under TN conditions to discriminate the confounding effects of dissimilar FI,and the FI was the previous day’s average FI of HS.The small intestinal segments(duodenum,jejunum,and ileum)and feces were collected on d 8.Results:Results indicated that HS drastically declined(P<0.05)average daily gain(ADG)and average daily feed intake(ADFI)(about 61%)in comparison with TN,and caused hyperpyrexia,meanwhile PF caused hypothermia.Morphological observation by light and electron microscopes showed that both HS and PF treatment decreased(P<0.05)the villus and microvillus height compared with TN.Additionally,HS increased(P<0.05)protein expression of heat shock protein 70 in the duodenum,jejunum,and ileum.Furthermore,the expression of tight junction protein zonula occluden-1(ZO-1)in the duodenum and ileum,and Occludin in the ileum were enhanced(P<0.05)compared with TN and PF.Moreover,HS significantly enhanced(P<0.05)the mRNA relative expression of inflammatory cytokines(TLR-2,TLR-4,and tumor necrosis factor-α(TNF-α),IL-6,IL-8,PG1–5,β-defensin 2(pBD-2)),mucins(mucin-1 and mucin-2)and P65 protein level in the ileal mucosa tissue.Intestinal microbiota analysis by 16S rRNA sequencing showed lower(P<0.10)αdiversity in both HS and PF,and a separated cluster ofβdiversity among groups.Compared with TN,HS but not PF mainly reduced(FDR<0.05)Bacteroidetes(phylum),Bacteroidia(class)and elevated the proportions of Proteobacteria(phylum,FDR<0.05),Bacillales(order,FDR<0.05),Planococcaceae(family,FDR<0.05),Kurthia(genus,FDR<0.05),Streptococcaceae(family,FDR<0.10)and Streptococcus(genus,FDR<0.10).Notably,Lactobacillales(order)was decreased(FDR<0.05)by PF alone.Furthermore,the Spearman correlation analysis indicated that the microbes prevalent in HS were positively(P<0.05)associated with intestinal morphological injuries indicators and ileal immune response parameters,and the microbes reduced in HS were negatively(P<0.05)with the performance data.Conclusions:Intestinal morphological injuries and ileal immune response caused by constant chronic HS independent of FI showed close connections with alterations in intestinal microbiota in growing-finishing pigs.Yunxia Xiong Shuting Cao Hao Xiao Qiwen Wu Hongbo Yi Zongyong Jiang Li Wang 2022Journal of Animal Science and Biotechnology2022,13,3:2
13Nlrc3-like is required for microglia maintenance in zebrafish显示文摘Microglia are tissue-resident macrophages residing in the central nervous system (CNS) and play critical roles in removing cellular debris and infectious agents as well as regulating neurogenesis and neuronal activities. Yet, the molecular basis underlying the establishment of microglia pool and the maintenance of their homeostasis in the CNS remain largely undefined. Here we report the identification and characterization of a mutant zebrafish, which harbors a point mutation in the nucleotide-binding oligomerization domain (NOD) like receptor gene nlrc3-like, resulting in the loss of microglia in a temperature sensitive manner. Temperature shift assay reveals that the late onset of nlrc3-like deficiency leads to excessive microglia cell death. Further analysis shows that the excessive microglia death in nlrc3-like deficient mutants is attributed, at least in part, to aberrant activation of canonical inflammasome pathway. 14Size effect of Pt nanoparticles in acid-assisted soot oxidation in the presence of NO显示文摘Pt/Al2O3 catalysts with mean Pt particle size ranged from 2.7 to 7.1 nm were synthesized by chemical reduction method,and the sulfated counterparts were prepared by impregnation of sulfuric acid.The turnover frequency of platinum for soot oxidation under loose contact conditions in a feed flow containing NO and O2 are positively correlated with the size of platinum.The sulfated Pt/Al2O3 exhibits higher catalytic activity for soot oxidation in the presence of NO despite their reduced ability for NO2 production.Such a contradiction is more significant for those catalysts with smaller platinum particles.Herein,the catalysts were characterized by X-ray diffraction(XRD),Brunauer-Emmett-Teller(BET),transmission electron microscopy(TEM),inductive coupled plasma(ICP)emission spectrometry,CO chemisorption,thermogravimetric analysis(TGA),NH3 temperature-programmed desorption(NH 3-TPD),NO temperature-programmed oxidation(TPO)and NO x temperatureprogrammed desorption(TPD).Possible effect of Pt particle size for the catalytic oxidation of soot in the presence of NO was presented based primarily on the promoted NO2 transfer efficiency onto the soot pushed by the acidic catalysts.Shuting Luo Xiaodong Wu Baofang Jin Shuang Liu Rui Ran Zhichun Si Duan Weng 2020Journal of Environmental Sciences2020,32,8:1

19Manipulations of micro/nanoparticles using gigahertz acoustic streaming tweezers显示文摘Contactless acoustic manipulation of micro/nanoscale particles has attracted considerable attention owing to its near independence of the physical and chemical properties of the targets,making it universally applicable to almost all biological systems.Thin-film bulk acoustic wave(BAW)resonators operating at gigahertz(GHz)frequencies have been demonstrated to generate localized high-speed microvortices through acoustic streaming effects.Benefitting from the strong drag forces of the high-speed vortices,BAW-enabled GHz acoustic streaming tweezers(AST)have been applied to the trapping and enrichment of particles ranging in size from micrometers to less than 100 nm.However,the behavior of particles in such 3D microvortex systems is still largely unknown.In this work,the particle behavior(trapping,enrichment,and separation)in GHz AST is studied by theoretical analyses,3D simulations,and microparticle tracking experiments.It is found that the particle motion in the vortices is determined mainly by the balance between the acoustic streaming drag force and the acoustic radiation force.This work can provide basic design principles for AST-based lab-on-a-chip systems for a variety of applications.Hang Wu Zifan Tang Rui You Shuting Pan Wenpeng Liu Hongxiang Zhang Tiechuan Li Yang Yang Chongling Sun Wei Pang Xuexin Duan 2022Nanotechnology and Precision Engineering2022,5,2:1
20Development of pretreatment protocol for DNA extraction from biofilm attached to biologic activated carbon(BAC)granules显示文摘The biologic activated carbon(BAC)process is widely used in drinking water treatments.A comprehensive molecular analysis of the microbial community structure provides very helpful data to improve the reactor performance.However,the bottleneck of deoxyribonucleic acid(DNA)extraction from BAC attached biofilm has to be solved since the conventional procedure was unsuccessful due to firm biomass attachment and adsorption capacity of the BAC granules.In this study,five pretreatments were compared,and adding skim milk followed by ultrasonic vibration was proven to be the optimal choice.This protocol was further tested using the vertical BAC samples from the full-scale biofilter of Pinghu Water Plant.The results showed the DNAyielded a range of 40μg·g^(-1) BAC(dry weight)to over 100μg·g^(-1) BAC(dry weight),which were consistent with the biomass distribution.All results suggested that the final protocol could produce qualified genomic DNA as a template from the BAC filter for downstream molecular biology researches.Shuting ZHANG Bo WEI Xin YU Bing LIU Zhuoying WU Li GU 2010Frontiers of Environmental Science & Engineering2010,4,4:1
