In vitro differentiation of adipose-derived stem cells and bone marrow-derived stromal stem cells into neuronal-like cells显示文摘Adipose-derived stem cells and bone marrow-derived stromal stem cells were co-cultured with untreated or Aβ1-40-treated PC12 cells, or grown in supernatant derived from untreated or Aβ1-40-treated PC12 cells. Analysis by western blot and quantitative real-time PCR showed that protein levels of Nanog, Oct4, and Sox2, and mRNA levels of miR/125a/3p were decreased, while expression of insulin-like growth factor-2 and neuron specific enolase was increased. In comparison, the generation of neuron specific enolase-positive cells was most successful when adipose-derived stem cells were co-cultured with Aβ1-40-treated PC12 cells. Our results demonstrate that adipose-derived stem cells and bone marrow-derived stromal stem cells exhibit trends of neuronal-like cell differentiation after co-culture with Aβ1-40-treated PC12 cells. This process may relate to a downregulation of miR-125a-3p mRNA expression and increased levels of insulin-like growth factor-2 expression. | Jin Zhou Guoping Tian Jing'e Wang Xuefeng Cong Xingkai Wu Siyang Zhang Li Li Bing Xu Feng Zhu Xuedan Luo Jian Han Fengjie Hu | 2011 | Neural Regeneration Research2011,6,19: | 21
Detection for disease tipping points by landscape dynamic network biomarkers显示文摘A new model-free method has been developed and termed the landscape dynamic network biomarker(l-DNB) methodology. The method is based on bifurcation theory, which can identify tipping points prior to serious disease deterioration using only single-sample omics data. Here, we show that l-DNB provides early-warning signals of disease deterioration on a single-sample basis and also detects critical genes or network biomarkers(i.e. DNB members) that promote the transition from normal to disease states. As a case study, l-DNB was used to predict severe influenza symptoms prior to the actual symptomatic appearance in influenza virus infections. The l-DNB approach was then also applied to three tumor disease datasets from the TCGA and was used to detect critical stages prior to tumor deterioration using an individual DNB for each patient. The individual DNBs were further used as individual biomarkers in the analysis of physiological data, which led to the identification of two biomarker types that were surprisingly effective in predicting the prognosis of tumors. The biomarkers can be considered as common biomarkers for cancer, wherein one indicates a poor prognosis and the other indicates a good prognosis. | Xiaoping Liu Xiao Chang Siyang Leng Hui Tang Kazuyuki Aihara Luonan Chen | 2019 | National Science Review2019,6,4: | 12
Potentiating CD8^(+) T cell antitumor activity by inhibiting PCSK9 to promote LDLR-mediated TCR recycling and signaling显示文摘Metabolic regulation has been proven to play a critical role in T cell antitumor immunity. However, cholesterol metabolism as a key component of this regulation remains largely unexplored.Herein,we found that the low-density lipoprotein receptor(LDLR),which has been previously identified as a transporter for cholesterol, plays a pivotal role in regulating CD8+T cell antitumor activity.Besides the involvement of cholesterol uptake which is mediated by LDLR in T cell priming and clonal expansion,we also found a non-canonical function of LDLR in CD8+T cells:LDLR interacts with the T-cell receptor(TCR)complex and regulates TCR recycling and signaling, thus facilitating the effector function of cytotoxic T-lymphocytes(CTLs).Furthermore,we found that the tumor microenvironment(TME)downregulates CD8+T cell LDLR level and TCR signaling via tumor cell-derived proprotein convertase subtilisin/kexin type 9(PCSK9)which binds to LDLR and prevents the recycling of LDLR and TCR to the plasma membrane thus inhibits the effector function of CTLs.Moreover, genetic deletion or pharmacological inhibition of PCSK9 in tumor cells can enhance the antitumor activity of CD8+T cells by alleviating the suppressive effect on CD8+T cells and consequently inhibit tumor progression. While previously established as a hypercholesterolemia target, this study highlights PCSK9/LDLR as a potential target for cancer immunotherapy as well. | Juanjuan Yuan Ting Cai Xiaojun Zheng Yangzi Ren Jingwen Qi Xiaofei Lu Huihui Chen Huizhen Lin Zijie Chen Mengnan Liu Shangwen He Qijun Chen Siyang Feng Yingjun Wu Zhenhai Zhang Yanqing Ding Wei Yang | 2021 | Protein & Cell2021,12,4: | 12
An efficient methodology for utilization of K-feldspar and phosphogypsum with reduced energy consumption and CO2 emissions显示文摘The issues of reducing CO_2 emissions, sustainably utilizing natural mineral resources, and dealing with industrial waste offer challenges for sustainable development in energy and the environment. We propose an efficient methodology via the co-reaction of K-feldspar and phosphogypsum for the extraction of soluble potassium salts and recovery of SO_2 with reduced CO_2 emission and energy consumption. The results of characterization and reactivity evaluation indicated that the partial melting of K-feldspar and phosphogypsum in the hightemperature co-reaction significantly facilitated the reduction of phosphogypsum to SO_2 and the exchange of K^+(K-feldspar) with Ca^(2+)(CaSO_4 in phosphogypsum). The reaction parameters were systematically investigated with the highest sulfur recovery ratio of ~ 60% and K extraction ratio of ~ 87.7%. This novel methodology possesses an energy consumption reduction of ~ 28% and CO_2 emission reduction of ~ 55% comparing with the present typical commercial technologies for utilization of K-feldspar and the treatment of phosphogypsum. | Zhixi Gan Zheng Cui Hairong Yue Siyang Tang Changjun Liu Chun Li Bin Liang Heping Xie | 2016 | Chinese Journal of Chemical Engineering2016,24,11: | 11
Physiological effects of weightlessness: countermeasure system development for a long-term Chinese manned spaceflight显示文摘The Chinese space station will be built around 2020. As a national space laboratory, it will offer unique opportunities for studying the physiological effects of weightlessness and the efficacy of the countermeasures against such effects. In this paper, we described the development of countermeasure systems in the Chinese space program. To emphasize the need of the Chinese space program to implement its own program for developing countermeasures, we reviewed the literature on the negative physiological effects of weightlessness, the challenges of completing missions, the development of countermeasure devices, the establishment of countermeasure programs, and the efficacy of the countermeasure techniques in American and Russian manned spaceflights. In addition, a brief overview was provided on the Chinese research and development on countermeasures to discuss the current status and goals of the development of countermeasures against physiological problems associated with weightlessness. | Linjie Wang Zhili Li Cheng Tan Shujuan Liu Jianfeng Zhang Siyang He Peng Zou Weibo Liu Yinghui Li | 2019 | Frontiers of Medicine2019,13,2: | 9
Biothiol-specific fluorescent probes with aggregation-induced emission characteristics显示文摘Biothiols are important species in physiological processes such as regulating protein structures, redox homeostasis and cell signalling. Alternation in the biothiol levels is associated with various pathological processes, therefore non-invasive fluorescent probes with high specificity to biothiols are highly desirable research utilities. Meanwhile, fluorescent probes with aggregationinduced emission properties(AIEgens) possess unique photophysical properties that allow modulation of the sensing process through controlling the aggregation-disaggregation or the intramolecular rotational motions of the fluorophores. Herein we review the recent progress in the development of biothiol-specific AIEgens. In particular, the molecular design principles to target different types of biothiols and the corresponding sensing mechanisms are discussed, along with the potential of the future design and development of multi-functional bioprobes. | Siyang Ding Mengjie Liu Yuning Hong | 2018 | Science China Chemistry2018,61,8: | 7
Heparanase Expression Correlates with Angiogenesis and Lymphangiogenesis in Human Lung Cancer显示文摘Background and objective Heparanase has been thought to be a good molecular marker of tumor, and the heparanase expression level was correlated closely with tumor metastasis. In this study,we investigate the effects of heparanase on angiogenesis and lymphangiogenesis of lung cancer and the relationship between heparanase expression and vascular endothelial growth factor (VEGF),vascular endothelial growth factor-C (VEGF-C). Methods Immunohistochemistry was used to detect the expression of heparanase,VEGF,VEGF-C protein and microvascular density (MVD),lymphatic vessel density (LVD) in 115 cases of non-small cell lung cancer (NSCLC) and 45 cases of adjacent normal tissue samples. Results Our results showed that heparanase expression was significantly increased in 91 (79.13%) of the 115 cases and correlated with lymph node metastasis (node positive rate 87.0%; node negative rate 36.8%; P=0.003). Heparanase positive expression cases have significantly higher concentration of microvascular density (MVD) and lymphatic vessel density (LVD) as compared with heparanase negative expression cases (P<0.01,P<0.01,respectively),heparanase expression was significantly correlated with VEGF,VEGF-C expression in NSCLC. Conclusion Heparanase overexpression was associated with angiogenesis and lymphangiogenesis of lung cancer, targeting of heparanase may represent a significant therapeutic potential for lung cancer. | Qingfu ZHANG Jian MING Yang LI Siyang ZHANG Bo LI Xueshan QIU Enhua WANG | 2009 | 中国肺癌杂志2009,12,8: | 7
Data-based prediction and causality inference of nonlinear dynamics显示文摘Natural systems are typically nonlinear and complex, and it is of great interest to be able to reconstruct a system in order to understand its mechanism, which cannot only recover nonlinear behaviors but also predict future dynamics. Due to the advances of modern technology, big data becomes increasingly accessible and consequently the problem of reconstructing systems from measured data or time series plays a central role in many scientific disciplines. In recent decades, nonlinear methods rooted in state space reconstruction have been developed, and they do not assume any model equations but can recover the dynamics purely from the measured time series data. In this review, the development of state space reconstruction techniques will be introduced and the recent advances in systems prediction and causality inference using state space reconstruction will be presented. Particularly, the cutting-edge method to deal with short-term time series data will be focused on.Finally, the advantages as well as the remaining problems in this field are discussed. | Huanfei Ma Siyang Leng Luonan Chen | 2018 | Science China Mathematics2018,61,3: | 5

Coupled relationships among anammox, denitrification, and dissimilatory nitrate reduction to ammonium along salinity gradients in a Chinese estuarine wetland显示文摘Salinization in estuarine wetlands significantly alters the balance between their nitrogen(N)removal and retention abilities but these processes have not yet been characterized effectively. In the present study, the potential rates of sediment denitrification, anaerobic ammonium oxidation(anammox),and dissimilatory nitrate reduction to ammonium(DNRA)were mapped using N isotope tracing methods along salinity gradients across the Yellow River Delta wetland(YRDW)in China. The contribution of anammox to total dissimilatory N transformations in YRDW was merely 6.8%,whereas denitrification and DNRA contributed 52.3%and 40.9%,respectively.The potential rate of denitrification(5.82μmol/kg/h)decreased significantly along salinity gradients and markedly exceeded DNRA potential rate(2.7μmol/kg/h)in fresh wetlands, but was lower than that of DNRA in oligohaline wetlands(3.06 and 3.18μmol/kg/h,respectively).Moreover,a significantly positive relationship between salinity and DNRA/denitrification was obeserved, indicating that increased salinity may favor DNRA over denitrification.Furthermore, total sulfur(TS)content and ratio of total organic carbon to total nitrogen(C/N)increased with the salinity gradient and showed evident positive relationships with the DNRA/denitrification ratio. In this study,we proved that increased salinization resulted in the dominance of DNRA over denitrification, possible through the addition of S and alteration of the C/N in estuarine wetlands, leading to increased N retention in estuarine wetlands during salinization, which would enhance the eutrophication potential within wetlands and in downstream ecosystems. | Zijun Zhou Lei Ge Yufang Huang Yuqian Liu Siyang Wang | 2021 | Journal of Environmental Sciences2021,33,8: | 4
The complete reference genome for grapevine (Vitis vinifera L.) genetics and breeding显示文摘Grapevine is one of the most economically important crops worldwide. However, the previous versions of the grapevine reference genome tipically consist of thousands of fragments with missing centromeres and telomeres, limiting the accessibility of the repetitive sequences, the centromeric and telomeric regions, and the study of inheritance of important agronomic traits in these regions. Here,we assembled a telomere-to-telomere(T2T)gap-free reference genome for the cultivar PN40024 using PacBio HiFi long reads. The T2T reference genome(PN_T2T)is 69 Mb longer with 9018 more genes identified than the 12X.v0 version. We annotated 67%repetitive sequences,19 centromeres and 36 telomeres, and incorporated gene annotations of previous versions into the PN_T2T assembly. We detected a total of 377 gene clusters, which showed associations with complex traits, such as aroma and disease resistance.Even though PN40024 derives from nine generations of selfing,we still found nine genomic hotspots of heterozygous sites associated with biological processes, such as the oxidation–reduction process and protein phosphorylation. The fully annotated complete reference genome therefore constitutes an important resource for grapevine genetic studies and breeding programs. | Xiaoya Shi Shuo Cao Xu Wang Siyang Huang Yue Wang Zhongjie Liu Wenwen Liu Xiangpeng Leng Yanling Peng Nan Wang Yiwen Wang Zhiyao Ma Xiaodong Xu Fan Zhang Hui Xue Haixia Zhong Yi Wang Kekun Zhang Amandine Velt Komlan Avia Daniela Holtgräwe Jérôme Grimplet JoséTomás Matus Doreen Ware Xinyu Wu Haibo Wang Chonghuai Liu Yuling Fang Camille Rustenholz Zongming Cheng Hua Xiao Yongfeng Zhou | 2023 | Horticulture Research2023,10,5: | 3

Complement C3a activates osteoclasts by regulating the PI3K/PDK1/SGK3 pathway in patients with multiple myeloma显示文摘Objective:Myeloma bone disease(MBD)is the most common complication of multiple myeloma(MM).Our previous study showed that the serum levels of C3/C4 in MM patients were significantly positively correlated with the severity of bone disease. However, the mechanism of C3 a/C4 a in osteoclasts MM patients remains unclear. Methods:The formation and function of osteoclasts were analyzed after adding C3 a/C4 a in vitro.RNA-seq analysis was used to screen the potential pathways affecting osteoclasts, and the results were verified by Western blot,q RT-PCR,and pathway inhibitors. Results:The osteoclast area per view induced by 1μg/m L(mean±SD:50.828±12.984%)and 10μg/m L(53.663±12.685%)of C3 a was significantly increased compared to the control group(0μg/m L)(34.635±8.916%)(P<0.001 and P<0.001,respectively).The relative m RNA expressions of genes,OSCAR/TRAP/RANKL/cathepsin K,induced by 1μg/m L(median:5.041,3.726,1.638,and 4.752,respectively)and 10μg/m L(median:5.140,3.702,2.250,and 5.172,respectively)of C3 a was significantly increased compared to the control group(median:3.137,2.004,0.573,and 2.257,respectively)(1μg/m L P=0.001,P=0.003,P<0.001,and P=0.008,respectively;10μg/m L:P<0.001,P=0.019,P<0.001,and P=0.002,respectively).The absorption areas of the osteoclast resorption pits per view induced by 1μg/m L(mean±SD:51.464±11.983%)and 10μg/m L(50.219±12.067%)of C3 a was also significantly increased(33.845±8.331%)(P<0.001 and P<0.001,respectively)compared to the control. There was no difference between the C4 a and control groups.RNA-seq analysis showed that C3 a promoted the proliferation of osteoclasts using the phosphoinositide 3-kinase(PI3 K)signaling pathway. The relative expressions of PIK3 CA/phosphoinositide dependent kinase-1(PDK1)/serum and glucocorticoid inducible protein kinases(SGK3)genes and PI3 K/PDK1/p-SGK3 protein in the C3 a group were significantly higher than in the control group. The activation role of C3 a in osteoclasts of MM patients was reduced by the SGK inhibitor(EMD638683).Conclusions:C3 a activated osteoclasts by regulating the PI3 K/PDK1/SGK3 pathways in MM patients, which was reduced using a SGK inhibitor.Overall, our results identified potential therapeutic targets and strategies for MBD patients。 | Fengjuan Jiang Hui Liu Fengping Peng Zhaoyun Liu Kai Ding Jia Song Lijuan Li Jin Chen Qing Shao Siyang Yan Kim De Veirman Karin Vanderkerken Rong Fu | 2021 | Cancer Biology & Medicine2021,18,3: | 3
Experimental Investigation on Heat Transfer Characteristics of Water in Inclined Downward Tube of a Supercritical Pressure CFB Boiler显示文摘In this study, experiments have been performed for an investigation on heat transfer of water in an inclined downward tube with an inner diameter of 20 mm and an inclined angle of 45° from the horizon, with the range of pressure from 11.5 to 28 MPa, mass flux from 450 to 1550 kg/(m2 s), and heat flux from 50 to 585 k W/m2. Based on the experimental data, the temperature distribution in the tube wall was derived. The heat transfer characteristics of inclined downward flow were compared with that of vertical downward flow. The effects of heat flux on wall temperature were analyzed and the corresponding empirical correlations were presented. The results show that heat transfer characteristics of water in the inclined downward tube are not uniform along the circumference from the top surface to the bottom surface. An increase in heat flux exacerbates the non-uniformity. At subcritical pressures, both dry-out and departure from nucleate boiling(DNB) occur at the top surface of the inclined downward tube; inversely, only dry-out takes place on the bottom surface of the inclined downward tube and in the vertical downward tube. At near-critical pressures, DNB and dry-out occur in the comparing tubes with greater possibility. At supercritical pressures, heat transfer gets enhanced in the pseudo-critical enthalpy region; in the high enthalpy region, the top surface temperature of the inclined downward tube decreases obviously. | WANG Siyang YANG Dong XIE Beibei WANG Long WANG Yongjie | 2015 | Journal of Thermal Science2015,24,5: | 3
Selective oxidation of cyclopentene with H2O2 by using H3PW12O40 and TBAB as a phase transfer catalyst显示文摘The selective oxidation of cyclopentene by aqueous H2O2 using H3PW12O40 and tetrabutyl ammonium bromide(TBAB) as a phase transfer catalyst has been investigated. The results show that the presence of TBAB significantly improved the oxidation selectivity of cyclopentene. The effects of the reaction conditions on the conversion of cyclopentene were investigated in detail. The optimal reaction conditions are as follows: the H3PW12O40 to TBAB molar ratio, 1:1–1:3;H3PW12O40 to cyclopentene molar ratio,0.54:100–0.64:100;and molar ratio of H2O2 to cyclopentene, 1.6:1. The conversion reached to 59.8% in 4h at 35.0 ℃, while the selectivity of glutaraldehyde was 38.0% and the selectivity of 1,2-cyclopentanediol was 55.6%. In addition, a route for oxidation of cyclopentene by aqueous H2O2 using a heteropoly acid and quaternary ammonium salt as a phase transfer catalyst was proposed. | Yaling Luo Changjun Liu Hairong Yue Siyang Tang Yingming Zhu Bin Liang | 2019 | Chinese Journal of Chemical Engineering2019,27,8: | 3
Small GTPases:Structure, biological function and its interaction with nanoparticles显示文摘Small GTPase is a kind of GTP-binding protein commonly found in eukaryotic cells. It plays an important role in cytoskeletal reorganization, cell polarity, cell cycle progression, gene expression and many other significant events in cells, such as the interaction with foreign particles. Therefore,it is of great scientific significance to understand the biological properties of small GTPases as well as the GTPase-nano interplay, since more and more nanomedicine are supposed to be used in biomedical field. However, there is no review in this aspect. This review summarizes the small GTPases in terms of the structure, biological function and its interaction with nanoparticles. We briefly introduced the various nanoparticles such as gold/silver nanoparticles,SWCNT,polymeric micelles and other nano delivery systems that interacted with different GTPases. These current nanoparticles exhibited different pharmacological effect modes and various target design concepts in the small GTPases study. This will help to elucidate the conclusion that the therapeutic strategy targeting small GTPases might be a new research direction. It is believed that the in-depth study on the functional mechanism of GTPases can provide insights for the design and study of nanomedicines. | Siyang Song Wenshu Cong Shurong Zhou Yujie Shi Wenbing Dai Hua Zhang Xueqing Wang Bing He Qiang Zhang | 2019 | Asian Journal of Pharmaceutical Sciences2019,14,1: | 3
Comparing overall survival between first generation EGFR-TKIs and chemotherapy in lung cancer patients with Del19/L858R显示文摘Objective: Combined overall survival(OS) analysis of Lux-Lung 3 and Lux-Lung 6 demonstrated that patients with epidermal growth factor receptor(EGFR) exon 19 deletions(Del19) would benefit from first-line second generation EGFR tyrosine kinase inhibitors(TKIs) afatinib but not for those with L858 R. This study was to investigate the survival difference between first-line first generation EGFR-TKIs and chemotherapy in patients with either Del19 or L858 R, and to directly compare OS in these two mutation groups. Methods: Eligibles were all prospective and retrospective studies comparing EGFR-TKIs with conventional chemotherapy or receiving single agent EGFR-TKIs and demonstrating survival analysis based on mutation types. The primary outcome was OS measured as pooled hazard ratios(HRs). All measures were pooled using randomeffects models and 95% confidential interval(95% CI) was calculated. Results: A total of 14 studies incorporating 1,706 patients with either Del19 or L858 R were included. Enrolling patients with Del19 or L858 R in randomized controlled trials(RCTs), first-line first generation EGFR-TKIs were associated with no OS benefit, compared with chemotherapy(pooled HR_(TKI/Chemo) for Del19: 0.82, 95% CI: 0.64-1.06, P=0.14; pooled HR_(TKI/Chemo) for L858R: 1.15, 95% CI: 0.85-1.56, P=0.38). Direct comparison of Del19 with L858 R receiving with first-line first generation EGFR-TKIs demonstrated no significant survival difference(pooled HR_(19/21): 0.88, 95% CI: 0.67-1.16, P=0.37).Conclusions: Among patients with advanced non-small cell lung cancer(NSCLC) harboring Del19 and L858 R, first-line first generation EGFR-TKIs demonstrated no survival benefit comparing with chemotherapy. | 18 | Energy-Efficient Resource Allocation for Heterogeneous Network with Grouping D2D显示文摘In this paper,a new communication model is built named grouping D2D(GD2D).Different from the traditional D2D coordination,we proposed GD2D communication in licensed and unlicensed spectrum simultaneously. We formulate a resource allocation problem, which aims at maximizing the energy efficiency(EE)of the system while guaranteeing the quality-of-service(Qos)of users. To efficiently solve this problem, the non-convex optimization problem is first transformed into a convex optimization problem.By transforming the fractional-form problem into an equivalent subtractive-form problem,an iterative power allocation algorithm is proposed to maximize the system EE.Moreover, the optimal closedform power allocation expressions are derived by the Lagrangian approach.Simulation results show that our algorithm achieves higher EE performance than the traditional D2D communication scheme. | Jing Cao Xin Song Siyang Xu Zhigang Xie Yanbo Xue | 2021 | China Communications2021,18,3: | 2

Load Shedding and Restoration for Intentional Island with Renewable Distributed Generation显示文摘Due to the high penetration of renewable distributed generation(RDG),many issues have become conspicuous during the intentional island operation such as the power mismatch of load shedding during the transition process and the power imbalance during the restoration process. In this paper,a phase measurement unit(PMU)based online load shedding strategy and a conservation voltage reduction(CVR)based multi-period restoration strategy are proposed for the intentional island with RDG.The proposed load shedding strategy, which is driven by the blackout event, consists of the load shedding optimization and correction table. Before the occurrence of the large-scale blackout, the load shedding optimization is solved periodically to obtain the optimal load shedding plan, which meets the dynamic and steady constraints. When the blackout occurs, the correction table updated in real time based on the PMU data is used to modify the load shedding plan to eliminate the power mismatch caused by the fluctuation of RDG.After the system transits to the intentional island seamlessly,multi-period restoration plans are generated to optimize the restoration performance while maintaining power balance until the main grid is repaired.Besides,CVR technology is implemented to restore more loads by regulating load demand. The proposed load shedding optimization and restoration optimization are linearized to mixed-integer quadratic constraint programming(MIQCP)models. The effectiveness of the proposed strategies is verified with the modified IEEE 33-node system on the real-time digital simulation(RTDS)platform. | Jian Xu Boyu Xie Siyang Liao Zhiyong Yuan Deping Ke Yuanzhang Sun Xiong Li Xiaotao Peng | 2021 | Journal of Modern Power Systems and Clean Energy2021,9,3: | 2
