| 2 | 2-Methyl-5H-benzo[d]pyrazolo[5,1-b][1,3]oxazin-5-imine, an edaravone analog, exerts neuroprotective effects against acute ischemic injury via inhibiting oxidative stress显示文摘Oxidative stress plays an indispensable role in the pathogenesis of cerebral ischemia.Inhibiting oxidative stress has been considered as an effective approach for stroke treatment.Edaravone,a free radical scavenger,has been shown to prevent cerebral ischemic injury.However,the clinical efficacy of edaravone is limited because it has a low scavenging activity for superoxide anions(O_2^(·-)).Here,we report that 2-methyl-5 H-benzo[d]pyrazolo[5,1-b][1,3]oxazin-5-imine,a novel small-molecule compound structurally related to edaravone,showed a stronger inhibitory effect on oxidative stress in vitro.In vivo,2-methyl-5 H-benzo[d]pyrazolo[5,1-b][1,3]oxazin-5-imine reversed transient middle cerebral artery occlusion-induced dysfunctions of superoxide dismutases and malondialdehyde,two proteins crucial for oxidative stress,suggesting a strengthened antioxidant system.Moreover,2-methyl-5 H-benzo[d]pyrazolo[5,1-b][1,3]oxazin-5-imine decreased blood brain barrier permeability.Then,we found that 2-methyl-5 Hbenzo[d]pyrazolo[5,1-b][1,3]oxazin-5-imine had a stronger neuroprotective effect than edaravone.More importantly,2-methyl-5 H-benzo[d]pyrazolo[5,1-b][1,3]oxazin-5-imine decreased not only infarct size and neurological deficits in the acute phase but also modified neurological severity score and escape latency in Morris water maze task in the delayed period,indicating enhanced neuroprotection,sensorimotor function and spatial memory.Together,these findings suggest that 2-methyl-5 H-benzo[d]pyrazolo[5,1-b][1,3]oxazin-5-imine could be a preferable option for stroke treatment. | Huanyu Ni Yixuan Song Haiyin Wu Lei Chang Chunxia Luo Dongya Zhu | 2018 | The Journal of Biomedical Research2018,32,4: | 6 |
| 3 | The use of fire at Zhoukoudian:evidence from magnetic susceptibility and color measurements显示文摘In order to provide direct evidence for the use of fire by humans at Locality 1,Zhoukoudian,we examine the burnt and unburnt sediments of newly excavated area in Layer 4 by detailed measurements of magnetic susceptibility,color,and diffuse reflectance spectrum.Results show that the magnetic susceptibility and redness of the burnt sediments are remarkably higher than those of other areas on the same level:up to*22 times for magnetic susceptibility and*3 times for redness of those of the adjacent unburnt sediments.Fine-grained(superparamagnetic/stable single-domain)magnetite and hematite grains make dominant contributions to the distinctly high values of magnetic susceptibility and redness in the burnt sediments.Diffuse reflectance spectroscopy results show that the burnt sediments contain more hematite than those of other areas and localities 2 and 3.High-temperature magnetic susceptibility measurements demonstrate that the burnt sediments have been heated above 700°C.Those changes in low-frequency magnetic susceptibility and redness are impossibly resulted from natural fires,thus most likely signaling the human activities of controlled use of fire.However,further work is needed to confirm whether or not these heat-affected sediments were produced in situ. | Yan Zhang Zhengtang Guo Chenglong Deng Shuangquan Zhang Haibin Wu Chunxia Zhang Junyi Ge Deai Zhao Qin Li Yang Song Rixiang Zhu | 2014 | Chinese Science Bulletin2014,59,10: | 6 |

| 5 | Single copper sites dispersed on hierarchically porous carbon for improving oxygen reduction reaction towards zinc-air battery显示文摘The demand for high-performance non-precious-metal electrocatalysts to replace the noble metal-based catalysts for oxygen reduction reaction(ORR)is intensively increasing.Herein,single-atomic copper sites supported on N-doped three-dimensional hierarchically porous carbon catalyst(Cu_(1)/NC)was prepared by coordination pyrolysis strategy.Remarkably,the Cu_(1)/NC-900 catalyst not only exhibits excellent ORR performance with a half-wave potential of 0.894 V(vs.RHE)in alkaline media,outperforming those of commercial Pt/C(0.851 V)and Cu nanoparticles anchored on N-doped porous carbon(CuNPs/NC-900),but also demonstrates high stability and methanol tolerance.Moreover,the Cu_(1)/NC-900 based Zn-air battery exhibits higher power density,rechargeability and cyclic stability than the one based on Pt/C.Both experimental and theoretical investigations demonstrated that the excellent performance of the as-obtained Cu_(1)/NC-900 could be attributed to the synergistic effect between copper coordinated by three N atoms active sites and the neighbouring carbon defect,resulting in elevated Cu d-band centers of Cu atoms and facilitating intermediate desorption for ORR process.This study may lead towards the development of highly efficient non-noble metal catalysts for applications in electrochemical energy conversion. | Wenjie Wu Yan Liu Dong Liu Wenxing Chen Zhaoyi Song Ximin Wang Yamin Zheng Ning Lu Chunxia Wang Junjie Mao Yadong Li | 2021 | Nano Research2021,14,4: | 4 |

| 9 | A chromosome-scale genome assembly of Dasypyrum villosum provides insights into its application as a broad-spectrum disease resistance resource for wheat improvement显示文摘Dasypyrum villosum is one of the most valuable gene resources in wheat improvement,especially for disease resistance.The mining of favorable genes from D.villosum is frustrated by the lack of a whole genome sequence.In this study,we generated a doubled-haploid line,91C43^(DH),using microspore culture and obtained a 4.05-GB high-quality,chromosome-scale genome assembly for D.villosum.The assembly contains39727 high-confidence genes,and 85.31% of the sequences are repetitive.Two reciprocal translocation events were detected,and 7VS-4VL is a unique translocation in D.villosum.The prolamin seed storage protein-coding genes were found to be duplicated;in particular,the genes encoding low-molecular-weight glutenin at the Glu-V3 locus were significantly expanded.RNA sequencing(RNA-seq)analysis indicated that,after Blumeria graminearum f.sp tritici(Bgt)inoculation,there were more upregulated genes involved in the pattern-triggered immunity and effector-triggered immunity defense pathways in D.villosum than in Triticum urartu.MNase hypersensitive sequencing(MH-seq)identified two Bgt-inducible MH sites(MHSs),one in the promoter and one in the 3'terminal region of the powdery mildew resistance(Pm)gene NLR1-V.Each site had two subpeaks and they were termed MHS1(MHS1.1/1.2)and MHS2(MHS2.1/2.2).Bgt-inducible MHS2.2 was uniquely present in D.villosum,and MHS1.1 was more inducible in D.villosum than in wheat,suggesting that MHSs may be critical for regulation of NLR1-V expression and plant defense.In summary,this study provides a valuable genome resource for functional genomics studies and wheat-D.villosum introgression breeding.The identified regulatory mechanisms may also be exploited to develop new strategies for enhancing Pm resistance by optimizing gene expression in wheat. | Xu Zhang Haiyan Wang Haojie Sun Yingbo Li Yilong Feng Chengzhi Jiao Mengli Li Xinying Song Tong Wang Zongkuan Wang Chunxia Yuan Li Sun Ruju Lu Wenli Zhang Jin Xiao Xiue Wang | 2023 | Molecular Plant2023,16,2: | 1 |

| 15 | Physicochemical properties of gelatin produced from Nile tilapia skin using chemical and fermentation pretreatments显示文摘In this study,we aimed to verify the effectiveness of fermentation as a pretreatment in Nile tilapia(Oreochromis niloticus)skin gelatin production and to assess its effects on the extracted gelatin.Chemical-gelatin(CG)and fermentation-gelatin(FG)were extracted using hot water from tilapia skin after immersion in chemical reagents(sodium hydroxide and butanol)and after fermentation with Bacillus velezensis FEL-BM21,respectively.FG presented a higher recovery(27.40%),yield(49.99%),and imino acid(proline and hydroxyproline)content(177.17±0.35 residues/1000 residues)than CG,but had a lower molecular weight.Fourier transform infrared and circular dichroism spectroscopy confirmed that the collagen was denatured,and scanning electron microscopy revealed that FG had a more continuous filamentous structure than CG.Furthermore,except for transmittance(66.19%at 450 nm and 86.36%at 620 nm),FG exhibited higher gel strength(492.81 g),gelling point(28.93℃),and melting point(18.33℃)than CG.These findings suggest that fermentation can be used to treat gelatin raw materials without negatively affecting the structure of gelatin.Moreover,fermentation has potential industrial applications as an alternative to chemical treatment. | Tingju Chen Zengliu Song Huanming Liu Chunxia Zhou Pengzhi Hong Chujin Deng | 2022 | Food Bioscience2022,47,3: | 1 |
| 16 | Brasenia-inspired hydrogel with sustained and sequential release of BMP and WNT activators for improved bone regeneration显示文摘Although bone morphogenetic protein(BMP) and WNT signaling play pivotal roles in bone development,homeostasis, and regeneration, the applications of proteins to stimulate corresponding signaling pathways showed limited outcomes in the repair and regeneration of bone defects that might be attributed to the reciprocal interventions of these pathways. In order to satisfy the combinational and sequential activation of BMP and WNT pathways, inspired by the heterogeneous hydrogel-liked structures of Brasenia, heterogeneous alginate/chitosan hydrogels were fabricated and spatially loaded with FK506 and BIO to achieve sustained and sequential release of the activators. Alkaline phosphatase staining, alizarin red staining and q RT-PCR results suggested that FK506 and BIO enhanced osteoblastic differentiation in vitro when used separately. Besides, by mixing and matching the activators and the hydrogel layers, a superior releasing mode that a combination of early FK506 release and following BIO release was identified via both in vitro and in vivo explorations for most efficient bone regeneration. | 17 | A simple and sensitive assay for apurinic/apyrimidinic endonuclease 1activity based on host-guest interaction of β-cyclodextrin polymer and pyrene显示文摘Apurinic/apyrimidinic endonuclease 1(APE1) plays an important role in protecting the fidelity of genetic material and is also a promising biomarker of cancers. Herein, a simple and sensitive fluorescent method for APE1 activity detection was designed based on host-guest interaction between β-cyclodextrin polymer(β-CDP) and pyrene. In this method, pyrene-labelled DNA probes(AP-S1 S2) and β-CDP functioned as fluorescent signal producer and enhancer, respectively. When APE1 was absent, pyrene on the AP-S1 S2 could not enter the cavity of β-CDP because of steric hindrance, leading to a weak fluorescence intensity/anisotropy. When APE1 was present, it would recognize and cleave AP site in APS1 S2, thus producing a dissociated pyrene-labelled oligonucleotide segment. Pyrene labelled at the 50 end of oligonucleotide segment could be easily trapped into the cavity of β-CDP via host-guest interaction, leading to a significant enhancement of fluorescence intensity/anisotropy. This assay offered a dynamic range of 0.05 U/m L–5 U/m L and an estimated detection limit of 0.05 U/m L. The results have shown that this active ingredient can improve the proliferation, colony forming efficiency and Scratch repair ability of human primary keratinocytes. Moreover, it can significantly increase the expression of Perlecan which is key protein in the basal layer of the skin. Meanwhile, 'space ginseng yeast' can help the cell morphology recovery and improve Perlecan, collagen I and elastin expression on the fibroblasts after heavy ion irradiation.
