4Subcellular localization of the stripe disease-specific protein encoded by rice stripe virus (RSV) in its vector,the small brown planthopper,Laodel-phax striatellus显示文摘The stripe disease-specific protein (SP) encoded by the rice stripe virus (RSV) was successfully used as a localization signal of the virus in its vector, the small brown lanthopper, Laodelphax striatellus Fallen. Immunogold particles in large numbers were detected in various parts of the viruliferous females: the ovum, surface of chorion, the mid-gut lumen, and the columnar cells. Whereas there was noneof these particles in the non-viruliferous females and males, and testis of viruliferous males. Endosymbionts (mycetocytes) were abundant, harboring ovaries of both viruliferous and non-viruliferous females, but none in the testis of males. The results provided us with the direct proof that RSV is a ciruculative and propagative plant virus and it was transovarially transmitted alongside with endosymbionts of its vector. 5Changes of protein oxidation,lipid oxidation and lipolysis in Chinese dry sausage with different sodium chloride curing salt content显示文摘The effect of sodium chloride(NaCl)curing salt content on protein oxidation,lipid oxidation and lipolysis of Chinese dry sausage was investigated.Two groups Chinese dry sausages with 2%and 4%(m/m)salt content were studied.The degree of protein oxidation increased during the processes in two groups sausages,while the content of phospholipids decreased,neutral lipids and free fatty acids increased.The degree of protein oxidation,lipid oxidation and lipolysis in 4%NaCl content group was higher than those in 2%NaCl content group,while 4%NaCl content group has higher lipase activity.In conclusion,4%NaCl may facilitate the protein oxidation,lipid hydrolysis and oxidation in Chinese dry sausage,and the protein oxidation had strong correlation with lipid oxidation and lipolysis.The results could provide a basis for improving the technology of industrial production.Bing Zhao Huimin Zhou Shunliang Zhang Xiaoqian Pan Su Li Ning Zhu Qianrong Wu Shouwei Wang Xiaoling Qiao Wenhua Chen 2020Food Science and Human Wellness2020,9,4:22

8Clinical role of (18)~F-fluorodeoxyglucose positron emission tomography/computed tomography in post-operative follow up of gastric cancer: Initial results显示文摘AIM: To evaluate the clinical role of 18F-fluorodeo-xyglucose positron emission and computed tomography (18F-FDG PET/CT) in detection of gastric cancer recurrence after initial surgical resection. METHODS: In the period from January 2007 to May 2008, 23 patients who had previous surgical resection of histopathologically diagnosed gastric cancer underwent a total of 25 18F-FDG PET/CT scans as follow-up visits in our center. The standard of reference for tumor recurrence consisted of histopathologic confirmation or clinical follow-up information for at least 5 mo after PET/CT examinations. RESULTS: PET/CT was positive in 14 patients (61%) and negative in 9 (39%). When correlated with final diagnosis, which was confirmed by histopathologic evidence of tumor recurrence in 8 of the 23 patients (35%) and by clinical follow-up in 15 (65%), PET/CT was true positive in 12 patients, false positive in 2, true negative in 8 and false negative in 2. Overall, the accuracy of PET/CT was 82.6%, the negative predictive value (NPV) was 77.7%, and the positive predictive value (PPV) was 85.7%. The 2 false positive PET/CT findings were actually chronic inflammatory tissue lesions. For the two patients with false negativePET/CT, the f inal diagnosis was recurrence of mucinous adenocarcinoma in the anastomosis in one patient and abdominal wall metastasis in the other. Importantly, PET/CT revealed true-positive findings in 11 (47.8%) patients who had negative or no definite findings by CT. PET/CT revealed extra-abdominal metastases in 7 patients and additional esophageal carcinoma in one patient. Clinical treatment decisions were changed in 7 (30.4%) patients after introducing PET/CT into their conventional post-operative follow-up program. 10Xuebijing injection alleviates liver injury by inhibiting secretory function of Kupffer cells in heat stroke rats显示文摘OBJECTIVE:To evaluate the effects of Xuebijing(XBJ) injection in heat stroke(HS) rats and to investigate the mechanisms underlying these effects.METHODS:Sixty anesthetized rats were randomized into three groups and intravenously injected twice daily for 3 days with 4 mL XBJ(XBJ group) or phosphate buffered saline(HS and Sham groups) per kg body weight.HS was initiated in the HS and XBJ groups by placing rats in a simulated climate chamber(ambient temperature 40 ℃,humidity 60%).Rectal temperature,aterial pressure,and heart rate were monitored and recorded.Time to HS onset and survival were determined,and serum concentrations of tumor necrosis factor(TNF)-α,inRESULTS:Time to HS onset and survival were significantly longer in the XBJ than in the HS group.Moreover,the concentrations of TNF-α,IL-1β,IL-6,ALT and AST were lower and liver injury was milder in the XBJ than in the HS group.Heat-stress induced structural changes in KCs and hepatic cells were more severe in the HS than in the XBJ group and the concentrations of TNF-α,IL-β and IL-6 secreted by KCs were lower in the XBJ than in the HS group.CONCLUSION:XBJ can alleviate HS-induced systemic inflammatory response syndrome and liver injury in rats,and improve outcomes.These protective effects may be due to the ability of XBJ to inhibit cytokine secretion by KCs.Yi Chen Huasheng Tong Xingqin Zhang Liqun Tang Zhiguo Pan Zhifeng Liu Pengkai Duan Lei Su 2013Journal of Traditional Chinese Medicine2013,33,2:14
11Baicalein ameliorates ulcerative colitis by improving intestinal epithelial barrier via AhR/IL-22 pathway in ILC3s显示文摘Ulcerative colitis(UC)is a chronic inflammatory disease of the gastrointestinal tract,which is closely related to gut barrier dysfunction.Emerging evidence shows that interleukin-22(IL-22)derived from group 3 innate lymphoid cells(ILC3s)confers benefits on intestinal barrier,and IL-22 expression is controlled by aryl hydrocarbon receptor(AhR).Previous studies show that baicalein protects the colon from inflammatory damage.In this study we elucidated the molecular mechanisms underlying the protective effect of baicalein on intestinal barrier function in colitis mice.Mice were administered baicalein(10,20,40 mg·kg^(-1)·d^(-1),i.g.)for 10 days;the mice freely drank 3%dextran sulfate sodium(DSS)on D1-D7 to induce colitis.We showed that baicalein administration simultaneously ameliorated gut inflammation,decreased intestinal permeability,restored tight junctions of colons possibly via promoting AhR/IL-22 pathway.Co-administration of AhR antagonist CH223191(10 mg/kg,i.p.)partially blocked the therapeutic effects of baicalein in colitis mice,whereas AhR agonist FICZ(1μg,i.p.)ameliorated symptoms and gut barrier function in colitis mice.In a murine lymphocyte line MNK-3,baicalein(5-20μM)dose-dependently increased the expression of AhR downstream target protein CYP1A1,and enhanced IL-22 production through facilitating AhR nuclear translocation,these effects were greatly diminished in shAhR-MNK3 cells,suggesting that baicalein induced IL-22 production in AhR-dependent manner.To further clarify that,we constructed an in vitro system consisting of MNK-3 and Caco-2 cells,in which MNK-3 cell supernatant treated with baicalein could decrease FITC-dextran permeability and promoted the expression of tight junction proteins ZO-1 and occluding in Caco-2 cells.In conclusion,this study demonstrates that baicalein ameliorates colitis by improving intestinal epithelial barrier via AhR/IL-22 pathway in ILC3s,thus providing a potential therapy for UC.Yan-yang Li Xiao-jing Wang Yu-lin Su Qing Wang Shao-wei Huang Zeng-feng Pan Yan-ping Chen Jun-jie Liang Mei-ling Zhang Xue-qian Xie Zhi-yun Wu Jin-yan Chen Lian Zhou Xia Luo 2022Acta Pharmacologica Sinica2022,43,6:14
12Comparison of breast-conserving surgery and mastectomy in early breast cancer using observational data revisited:a propensity score-matched analysis显示文摘Recent observational studies showed that breast-conserving surgery(BCS) resulted in superior survival compared to mastectomy in breast cancer patients. This study compared the clinical outcomes of BCS and mastectomy using propensity score(PS)matching analysis, which had advantages over conventional methods in reducing bias. Nonmetastatic breast cancer patients who underwent BCS and mastectomy were matched 1:1 based on their PS. We used the Kaplan-Meier method and Cox-regression model to estimate the treatment effects. A total of 2,866 patients with a median follow-up time of 67 months were included in the original study population. Although the mastectomy cohort(N=1,219) had more advanced disease compared to the BCS cohort(N=1,647), LRFS was similar between the two groups(93.8% vs. 92.4%, P>0.05). BCS(vs. mastectomy) was associated with improved DFS(73.8% vs. 58.7%, P<0.01) and CSS(91% vs. 78.2%, P<0.01) in the original population. In the PS-matched population(N=1,668), clinicopathological features were equally distributed between the two cohorts. BCS(vs. mastectomy) was not associated with improved DFS(70.7% vs. 66.9%, P>0.05) or CSS(87.5% vs. 84.9%, P>0.05). We found that PS methods reduce bias when estimating treatment effects using observational data. 13Early initiation renal replacement therapy for fluid management to reduce central venous pressure is more conducive to renal function recovery in patients with acute kidney injury显示文摘Background:Acute kidney injury (AKI) is a serious complication in critically ill patients with septic shock treated in the intensive care unit. Renal replacement therapy (RRT) is a treatment for severe AKI;however, the time of initiation of RRT and factors that affect the recovery of kidney function remains unclear. This study was to explore whether early initiation of RRT treatment for fluid management to reduce central venous pressure (CVP) can help to improve patients5 kidney function recovery. Methods: A retrospective analysis of septic patients who had received RRT treatment was conducted. Patients received RRT either within 12 h after they met the diagnostic criteria of renal failure (early initiation) or after a delay of 48 h if renal recovery had not occurred (delayed initiation). Parameters such as patients5 renal function recovery at discharge, fluid balance, and levels of CVP were assessed. Results: A total of 141 patients were eligible for enrolment: 40.4% of the patients were in the early initiation group (57 of 141 patients), and 59.6% were in the delayed initiation group (84 of 141 patients). There were no significant differences in the characteristics at baseline between the two groups, and there were no differences in 28-day mortality between the two groups (χ^2 = 2.142, P = 0.143);however, there was a significant difference in the recovery rate of renal function between the two groups at discharge (χ^2 = 4.730, P < 0.001). More importantly, early initiation of RRT treatment and dehydration to reduce CVP are more conducive to the recovery of renal function in patients with AKI. 14Precipitation Behavior of V-N Microalloyed Steels during Normalizing显示文摘The precipitation behavior of V-N microalloyed steel during normalizing process was studied by physicochemical phase analysis and transmission electron microscopy(TEM). The effect of precipitation behavior on mechanical properties was investigated by theoretical calculations. The results showed that 32.9% of V(C,N) precipitates remained undissolved in the austenite during the soaking step of the normalizing process. These precipitates prevented the growth of the austenite grains. During the subsequent cooling process, the dissolved V(C,N) re-precipitated and played a role in precipitation strengthening. The undissolved V(C,N) induced intragranular ferrite nucleation and refined the ferrite grains. 16Effect of Tempering Temperature on Microstructure and Mechanical Properties of Steel Containing Ni of 9%显示文摘Mechanical properties of quenching,intercritical quenching and tempering (QLT) treated steel containing Ni of 9% were evaluated from specimens subject to various tempering temperatures. The detailed microstructures of steel containing Ni of 9% at different tempering temperatures were observed by optical microscope (OM) and transmission electron microscope (TEM). The volume fraction of austenite was estimated by XRD. The results show that high strength and cryogenic toughness of steel containing Ni of 9% are obtained when the tempering temperature are between 540 and 580 ℃. The microstructure keeps the dual phase lamellar structure after the intercritical quenching and there is cementite created in the Ni-rich constituents when tempering temperature is 540 ℃. When tempering temperatures are between 560 and 580 ℃,the reversed austenites (γ′) grow up and the dual phase lamellar structure is not clear. The γ′ becomes instable at 600 ℃. When tempered at temperature ranging from 500 to 520 ℃,the increase of dislocation density in the lamellar matrix makes both tensile strength and yield strength decrease. When tempered at 540 ℃ and higher temperature,the yield strength decreases continuously because the C and alloying elements in the matrix are absorbed by the cementite and the γ′,so the yield ratio is decreased by the γ′. There are two toughness mechanisms at different tempering temperatures. One is that the precipitation of cementite absorbs the carbon in the steel which plays a major role in improving cryogenic toughness at lower temperature. Another is that the γ′ and the purified matrix become major role at higher tempering temperature. When the tempering temperature is 600 ℃,the stability of γ′ is decreased quickly,even the transformation takes place at room temperature,which results in a sharp decrease of Charpy-V impact energy at 77 K. 17Spatial and temporal variations of two cyanobacteria in the mesotrophic Miyun reservoir, China显示文摘Spatial variations in phytoplankton community within a large mesotrophic reservoir(Miyun reservoir, North China) were investigated in relation to variations in physico-chemical properties, nutrient concentrations, temperature and light conditions over a 5 month period in 2009. The dynamics of phytoplankton community was represented by the dominance of cyanobacteria through summer and fall, following with a short term dominance of chlorophyta in late fall, and a relatively high abundance of diatom in October; on the other hand, maximum phytoplankton biomass was recorded in the north shallow region of Miyun reservoir with a higher nutrients level. Particular attention was paid to the impacts of environmental conditions on the growth of two cyanobacteria genera, the toxin-producing Microcystis and the taste & odor-producing Oscillatoria. Microcystis biomass was in general greatly affected by water temperature and mixing depth/local water depth ratio in this reservoir, while the Oscillatoria biomass in the surface and middle layers was greatly affected by total dissolved phosphorus, and that in the bottom layer was related with the Secchi depth/local water depth ratio. 18Prioritizing natural-selection signals from the deep-sequencing genomic data suggests multi-variant adaptation in Tibetan highlanders显示文摘Human genetic adaptation to high altitudes(>2500 m)has been extensively studied over the last few years,but few functional adaptive genetic variants have been identified,largely owing to the lack of deep-genome sequencing data available to previous studies.Here,w e build a list of putative adaptive variants,including 63 missense,7 loss-of-function;1,298 evolutionarily conserved variants and 509 expression quantitative traits loci.Notably,the top signal of selection is located in TMEM 247,a transmembrane protein-coding gene.The Tibetan version of TMEM 247 harbors one high-frequency(76.3%)missense variant,rsl 16983452(c.248C>T;p.Ala83Val);with the T allele derived from archaic ancestry and carried by>94%of Tibetans but absent or in low frequencies(<3%)in non-Tibetan populations.The rsl 16983452-T is strongly and positively correlated with altitude and significantly associated with reduced hemoglobin concentration(p=5.78×10^-5),red blood cell count(p=5.72×10^-7)and hematocrit(p=2.57×10^-6).In particular,TMEM247-rs 116983452 shows greater effect size and better predicts the phenotypic outcome than any E P A S1 variants in association with adaptive traits in Tibetans.Modeling the interaction between TM£M247-rsl 16983452 and EPAS1 variants indicates weak but statistically significant epistatic effects.Our results support that multiple variants may jointly deliver the fitness of the Tibetans on the plateau,where a complex model is needed to elucidate the adaptive evolution mechanism.Lian Deng Chao Zhang Kai Yuan Yang Gao Yuwen Pan Xueling Ge Yaoxi He Yuan Yuan Yan Lu Xiaoxi Zhang Hao Chen Haiyi Lou Xiaoji Wang Dongsheng Lu Jiaojiao Liu Lei Tian Qidi Feng Asifullah Khan Yajun Yang Zi-Bing Jin Jian Yang Fan Lu Jia Qu Longli Kang Bing Su Shuhua Xu 2019National Science Review2019,6,6:9
