| 1 | Comparison of breast-conserving surgery and mastectomy in early breast cancer using observational data revisited:a propensity score-matched analysis显示文摘Recent observational studies showed that breast-conserving surgery(BCS) resulted in superior survival compared to mastectomy in breast cancer patients. This study compared the clinical outcomes of BCS and mastectomy using propensity score(PS)matching analysis, which had advantages over conventional methods in reducing bias. Nonmetastatic breast cancer patients who underwent BCS and mastectomy were matched 1:1 based on their PS. We used the Kaplan-Meier method and Cox-regression model to estimate the treatment effects. A total of 2,866 patients with a median follow-up time of 67 months were included in the original study population. Although the mastectomy cohort(N=1,219) had more advanced disease compared to the BCS cohort(N=1,647), LRFS was similar between the two groups(93.8% vs. 92.4%, P>0.05). BCS(vs. mastectomy) was associated with improved DFS(73.8% vs. 58.7%, P<0.01) and CSS(91% vs. 78.2%, P<0.01) in the original population. In the PS-matched population(N=1,668), clinicopathological features were equally distributed between the two cohorts. BCS(vs. mastectomy) was not associated with improved DFS(70.7% vs. 66.9%, P>0.05) or CSS(87.5% vs. 84.9%, P>0.05). We found that PS methods reduce bias when estimating treatment effects using observational data. | 6 | Dispersion of the Mechanical Parts Performance Indicators Based on the Concept of Random Vector显示文摘To solve the precision and reliability problem of various machinery equipments and military vehicles,some military organisations,the industrial sector and the academia at home and abroad begin to pay attention to the statistical distribution of machining dimensions,material properties and service loads,and the system reliability optimization design with constraints and reliability optimization design of various mechanical parts is studied in this way.However,the above researches focus on solving the strength and the life problem,and no studies have been done on the discrete degree and discrete pattern of other performance indicators.The concept of using a random vector to describe the mechanical parts performance indicators is presented;characteristics between the value of the vector variance matrix determinant and the sum of the diagonal covariance matrix in describing the performance indicators of vector dispersion are studied and compared.A clutch diaphragm spring is set as an example,the geometric dimension indicator is described with random vector,and the applicability of using variance matrix determinant and variance matrix trace of geometric dimension vector to describe discrete degree of random vector is studied by using Monte-Carlo simulation method and component discrete degree perturbation method.Also,the effects of different components of diaphragm spring geometric dimension vector on the value of covariance matrix determinant and the sum of covariance matrix diagonal of diaphragm spring performance indicators vector are analyzed.The present study shows that the impacts of the dispersion of diaphragm spring cone angle on every performance dispersion are all ranked first,and far exceed that of other dimension dispersion.So it must be strictly controlled in the production process.The result of the research work provides a reference for the design of diaphragm spring,and also it presents a proper method for researching the performance of other mechanical parts. | XIA Changgao ZHU Pei ZHANG Meng GAO Xiang LU Liling | 2012 | Chinese Journal of Mechanical Engineering2012,25,1: | 1 |

| 10 | Interleukin-13 promotes cellular senescence through inducing mitochondrial dysfunction in IgG4-related sialadenitis显示文摘Immunoglobulin G4-related sialadenitis(IgG4-RS)is an immune-mediated fibro-inflammatory disease and the pathogenesis is still not fully understood.The aim of this study was to explore the role and mechanism of interleukin-13(IL-13)in the cellular senescence during the progress of IgG4-RS.We found that the expression of IL-13 and IL-13 receptorα1(IL-13Rα1)as well as the number of senescent cells were significantly higher in the submandibular glands(SMGs)of IgG4-RS patients.IL-13 directly induced senescence as shown by the elevated activity of senescence-associatedβ-galactosidase(SA-β-gal),the decreased cell proliferation,and the upregulation of senescence markers(p53 and p16)and senescence-associated secretory phenotype(SASP)factors(IL-1βand IL-6)in SMG-C6 cells.Mechanistically,IL-13 increased the level of phosphorylated signal transducer and activator of transcription 6(p-STAT6)and mitochondrial-reactive oxygen species(mt ROS),while decreased the mitochondrial membrane potential,ATP level,and the expression and activity of superoxide dismutase 2(SOD2).Notably,the IL-13-induced cellular senescence and mitochondrial dysfunction could be inhibited by pretreatment with either STAT6 inhibitor AS1517499 or mitochondria-targeted ROS scavenger Mito TEMPO.Moreover,IL-13 increased the interaction between p-STAT6 and c AMP-response element binding protein(CREB)-binding protein(CBP)and decreased the transcriptional activity of CREB on SOD2.Taken together,our findings revealed a critical role of IL-13 in the induction of salivary gland epithelial cell senescence through the elevated mitochondrial oxidative stress in a STAT6–CREB–SOD2-dependent pathway in IgG4-RS. | Mengqi Zhu Sainan Min Xiangdi Mao Yuan Zhou Yan Zhang Wei Li Li Li Liling Wu Xin Cong Guangyan Yu | 2022 | International Journal of Oral Science2022,14,3: | 1 |
| 11 | Monolithic integration of nanorod arrays on microfluidic chips for fast and sensitive one-step immunoassays显示文摘Here,we present integrated nanorod arrays on microfluidic chips for fast and sensitive flow-through immunoassays of physiologically relevant macromolecules.Dense arrays of Au nanorods are easily fabricated through one-step oblique angle deposition,which eliminates the requirement of advanced lithography methods.We report the utility of this plasmonic structure to improve the detection limit of the cardiac troponin I(cTnI)assay by over 6x 105-fold,reaching down to 33.9fg mL^(-1)(-1.4fM) compared with an identical assay on glass substrates.Through monolithic integration with microfluidic elements,the device enables a flow-through assay for quantitative detection of cTnI in the serum with a detection sensitivity of 6.9 pg mL^(-1)(-0.3 pM)in<6 min,which was 4000 times lower than conventional glass devices.This ultrasensitive detection arises from the large surface area for antibody conjugation and metal-enhanced fluorescent signals through plasmonic nanostructures.Moreover,due to the parallel arrangement of flow paths,simultaneous detection of multiple cancer biomarkers,including prostate-specific antigen and carcinoembryonic antigen,has been fulfilled with increased signal-to-background ratios.Given the high performance of this assay,together with its simple fabrication process that is compatible with standard mass manufacturing techniques,we expect that the prepared integrated nanorod device can bring on-site point-of-care diagnosis closer to reality. | Ye Wang Jiongdong Zhao Yu Zhu Shurong Dong Yang Liu Yijun Sun Liling Qian Wenting Yang Zhen Cao | 2021 | Microsystems & Nanoengineering2021,7,4: | 0 |
