| 1 | Response of sediment yield to vegetation restoration at a large spatial scale in the Loess Plateau显示文摘The impact of vegetation coverage on erosion and sediment yield in the Loess Plateau has been extensively studied,but the research has been primarily based on observations from slope runoff plots or secondary forest regions;the scaling method remains unresolved when it is applied at a large spatial scale,and it is difficult to apply to regions with severe soil and water loss given the predominance of herbs and shrubs.To date,there is little data on the quantitative impact of changes to vegetation on sediment concentration at a large spatial scale.This paper is based on vegetation information from remote sensing images,measured rainfall and sediment data over nearly 60 years,and results from previous runoff and sediment variation research on the Yellow River.We introduce the concepts of a sediment yield coefficient and the percentage of effective vegetation and erodible area,analyze the impact of different vegetation conditions on the flood sediment concentration and sediment yield,and evaluate the effect of rainfall intensity on sediment yield under different vegetation conditions at the watershed scale.We propose models to evaluate the impact of vegetation on sediment yield in the loess gully hilly region,which are based on remote sensing data and support an application at a large spatial scale.The models can be used to assess sediment reduction that results from the current significant improvement of vegetation in the Loess Plateau. | LIU XiaoYan YANG ShengTian DANG SuZhen LUO Ya LI XiaoYu ZHOU Xu | 2014 | Science China(Technological Sciences)2014,57,8: | 11 |

| 4 | The clinical and genetic characteristics in children with mitochondrial disease in China显示文摘Mitochondrial disease was a clinically and genetically heterogeneous group of diseases, thus the diagnosis was very difficult to clinicians. Our objective was to analyze clinical and genetic characteristics of children with mitochondrial disease in China. We tested 141 candidate patients who have been suspected of mitochondrial disorders by using targeted next-generation sequencing(NGS), and summarized the clinical and genetic data of gene confirmed cases from Neurology Department, Beijing Children's Hospital, Capital Medical University from October 2012 to January 2015. In our study, 40 cases of gene confirmed mitochondrial disease including eight kinds of mitochondrial disease, among which Leigh syndrome was identified to be the most common type, followed by mitochondrial encephalomyopathy, lactic acidosis, and stroke-like episodes(MELAS). The age-of-onset varies among mitochondrial disease, but early onset was common. All of 40 cases were gene confirmed, among which 25 cases(62.5%)with mitochondrial DNA(mtDNA) mutation, and 15 cases(37.5%) with nuclear DNA(nDNA) mutation. M.3243A>G(n=7)accounts for a large proportion of mtDNA mutation. | 5 | The chromosome-level Stevia genome provides insights into steviol glycoside biosynthesis显示文摘Stevia(Stevia rebaudiana Bertoni)is well known for its very sweet steviol glycosides(SGs)consisting of a common tetracyclic diterpenoid steviol backbone and a variable glycone.Steviol glycosides are 150–300 times sweeter than sucrose and are used as natural zero-calorie sweeteners.However,the most promising compounds are biosynthesized in small amounts.Based on Illumina,PacBio,and Hi-C sequencing,we constructed a chromosome-level assembly of Stevia covering 1416 Mb with a contig N50 value of 616.85 kb and a scaffold N50 value of 106.55 Mb.More than four-fifths of the Stevia genome consisted of repetitive elements.We annotated 44,143 high-confidence protein-coding genes in the high-quality genome.Genome evolution analysis suggested that Stevia and sunflower diverged~29.4 million years ago(Mya),shortly after the whole-genome duplication(WGD)event(WGD-2,~32.1 Mya)that occurred in their common ancestor.Comparative genomic analysis revealed that the expanded genes in Stevia were mainly enriched for biosynthesis of specialized metabolites,especially biosynthesis of terpenoid backbones,and for further oxidation and glycosylation of these compounds.We further identified all candidate genes involved in SG biosynthesis.Collectively,our current findings on the Stevia reference genome will be very helpful for dissecting the evolutionary history of Stevia and for discovering novel genes contributing to SG biosynthesis and other important agronomic traits in future breeding programs. | Xiaoyang Xu Haiyan Yuan Xiaqing Yu Suzhen Huang Yuming Sun Ting Zhang Qingquan Liu Haiying Tong Yongxia Zhang Yinjie Wang Chunxiao Liu Lei Wu Menglan Hou Yongheng Yang | 2021 | Horticulture Research2021,8,1: | 2 |
| 6 | Polyaniline-based electrocatalysts through emulsion polymerization:Electrochemical and electrocatalytic performances显示文摘One of the major challenges associated with fuel cells is the design of highly efficient electrocatalysts to reduce the high overpotential of the oxygen reduction reaction(ORR). Here we report Polyaniline(PANI)based micro/nanomaterials with or without transition metals, prepared by the emulsion polymerization and subsequent heat treatment. PANI microspheres with the diameter of about 0.7 μm have been prepared in basic(NH3solution) condition, using two different types of surfactant(CTAB, SDS) as the stabilizer, ammonium persulphate(APS) as oxidant with aniline/surfactants molar ratio at 1/1 under the hydrothermal treatment. PANI nanorods, Fe–PANI, and Fe–Co–PANI have been synthesized in acidic(HCl)medium with aniline/surfactants molar ratio at 1/2 and polymerization carried out without stirring for24 h. Products mainly Fe–Co–PANI have shown high current density with increasing sweep rate and excellent specific capacitance 1753 F/g at the scan rate of 1 m V/s. Additionally, it has shown high thermal stability by thermogravimetric analysis(TGA). Fe–PANI has been investigated for excellent performance toward ORR with four electron selectivity in the basic electrolyte. | 13 | Upregulation of RIN3 induces endosomal dysfunction in Alzheimer’s disease显示文摘Background In Alzheimer’s Disease(AD),about one-third of the risk genes identified by GWAS encode proteins that function predominantly in the endocytic pathways.Among them,the Ras and Rab Interactor 3(RIN3)is a guanine nucleotide exchange factor(GEF)for the Rab5 small GTPase family and has been implicated to be a risk factor for both late onset AD(LOAD)and sporadic early onset AD(sEOAD).However,how RIN3 is linked to AD pathogenesis is currently undefined.Methods Quantitative PCR and immunoblotting were used to measure the RIN3 expression level in mouse brain tissues and cultured basal forebrain cholinergic neuron(BFCNs).Immunostaining was used to define subcellular localization of RIN3 and to visualize endosomal changes in cultured primary BFCNs and PC12 cells.Recombinant flag-tagged RIN3 protein was purified from HEK293T cells and was used to define RIN3-interactomes by mass spectrometry.RIN3-interacting partners were validated by co-immunoprecipitation,immunofluorescence and yeast two hybrid assays.Live imaging of primary neurons was used to examine axonal transport of amyloid precursor protein(APP)andβ-secretase 1(BACE1).Immunoblotting was used to detect protein expression,processing of APP and phosphorylated forms of Tau.Results We have shown that RIN3 mRNA level was significantly increased in the hippocampus and cortex of APP/PS1 mouse brain.Basal forebrain cholinergic neurons(BFCNs)cultured from E18 APP/PS1 mouse embryos also showed increased RIN3 expression accompanied by early endosome enlargement.In addition,via its proline rich domain,RIN3 recruited BIN1(bridging integrator 1)and CD2AP(CD2 associated protein),two other AD risk factors,to early endosomes.Interestingly,overexpression of RIN3 or CD2AP promoted APP cleavage to increase its carboxyl terminal fragments(CTFs)in PC12 cells.Upregulation of RIN3 or the neuronal isoform of BIN1 increased phosphorylated Tau level.Therefore,upregulation of RIN3 expression promoted accumulation of APP CTFs and increased phosphorylated Tau.These effects by RIN3 was rescued by the expression of a dominant negative Rab5(Rab5S34N)construct.Our study has thus pointed to that RIN3 acts through Rab5 to impact endosomal trafficking and signaling.Conclusion RIN3 is significantly upregulated and correlated with endosomal dysfunction in APP/PS1 mouse.Through interacting with BIN1 and CD2AP,increased RIN3 expression alters axonal trafficking and procession of APP.Together with our previous studies,our current work has thus provided important insights into the role of RIN3 in regulating endosomal signaling and trafficking. | Ruinan Shen Xiaobei Zhao Lu He Yongbo Ding Wei Xu Suzhen Lin Savannah Fang Wanlin Yang Kijung Sung Brian Spencer Robert A.Rissman Ming Lei Jianqing Ding Chengbiao Wu | 2020 | Translational Neurodegeneration2020,9,2: | 1 |
