1Zircon SHRIMP U-Pb geochronology of the Neoproterozoic Chengjiang Formation in central Yunnan Province(SW China) and its geological significance显示文摘The Chengjiang Formation is the earliest continental clastic deposit after the Jinning Orogeny in central Yunnan Province,and therefore its accurate depositional age is significant for understanding the formation and evolution of the Neoproterozoic rift basins in southern China.However,hampered by accuracy of the dating technique,the existing age data for the Chengjiang Formation are not very reliable.A large number of magmatic zircons were obtained from the tuff interbed in the lower part of the Chengjiang Formation in Jinyang area and the bottom part of the Chengjiang Formation in Dongchuan area,central Yunnan Province,and high-precision SHRIMP U-Pb dating was carried out on these zircons.The results show that the weighted mean 206 Pb/238 U ages are 797.8±8.2 and 803.1±8.7 Ma respectively.In combination with related geological evidence,it has been demonstrated that the bottom boundary age of the Chengjiang Formation should be 800±5 Ma,whereas the top boundary age could be ca 725 Ma.By synthetically analyzing the latest age data for the relevant strata,it has been confirmed that the Chengjiang Formation should be correlated with the Suxiong Formation and the Kaijianqiao Formation in western Sichuan Province,the Liantuo Formation in the middle and lower Yangtze,the Hongchicun Formation and the Shangshu Formation in northern Zhejiang Province,the Puling Formation in southern Anhui Province,the Luokedong Formation and the Majianqiao Formation in northwestern Jiangxi Province,the Wuqiangxi Formation in northwestern Hunan Province,the Fanzhao Formation and the Qingshuijiang Formation in southeastern Guizhou Province,and the Sanmenjie Formation and the Gongdong Formation in northern Guangxi Province.Sedimentary cycle analysis shows that the sedimentary filling sequence of the Neoproterozoic rift basins in southern China can be divided into four cycles.Among them,Cycle II began at ca.800 Ma,accompanied by intensive tectonic-thermal events.The zircon U-Pb ages from the bottom of the Chengjiang Formation reported in this paper indicate that the Neoproterozoic Kangdian rift subbasin probably started to develop at ca.800 Ma and therefore missed Cycle I of the Neoproterozoic sedimentary filling sequence in southern China.JIANG XinSheng WANG Jian CUI XiaoZhuang ZHUO JieWen XIONG GuoQing LU JunZe LIU JianHui 2012Science China Earth Sciences2012,55,11:25

3Radix Astragali-induced differentiation of rat bone marrow-derived mesenchymal stem cells显示文摘BACKGROUND:Chemical induction has been shown to be effective at promoting the differentiation of bone marrow-derived mesenchymal stem cells(MSCs).However,these inductors have cytotoxicity side effects that may damage cells over time.Traditional Chinese medicines avoid this disadvantage while still producing effective induction. OBJECTIVE:To investigate the influence of Radix Astragali(Huangqi) on the differentiation of MSCs. DESIGN,TIME AND SETTING:In vitro study of traditional Chinese medicine in neural stem cell differentiation.The experiment was performed at the Central Laboratory of Hebei North University between April and June 2007. MATERIALS:Radix Astragali solution(lot No.060105;license No.Z53021585) was purchased from Dali Pharmaceutical Co.,Ltd.,China;rabbit anti-rat nestin,rabbit anti-rat neuron-specific enolase (NSE),mouse anti-rat microtubule-associated protein 2,and rabbit anti-rat glial fibrillary acidic protein were purchased from Wuhan Boster,China. METHODS:Whole bone marrow was isolated from the femur and tibia of 6-week-old male Wistar rats and subcultured.The fourth passage of MSCs were harvested and induced by different concentrations(50,100,200,400 g/L) of Radix Astragali. MAIN OUTCOME MEASURES:Hematoxylin-eosin staining was used to observe MSC morphology after 24 hours of induction.Immunocytochemistry was employed to observe the expression of NSE (specific neuronal marker),nestin(marker of neural stem cell),glial fibrillary acidic protein and microtubule-associated protein 2(markers of astrocytes). RESULTS:Following Radix Astragalitreatment,changes occurred in cell morphology including:cell body pyknosis;thin and long processes formed in some cells,with growth corresponding to drug concentration and induction time;and the formation of network-like connections between some cells. With increasing drug concentration and induction time,nestin expression was upregulated,and the number of positive cells increased;cells produced NSE,glial fibrillary acidic protein and microtubule-associated protein 2;nestin was expressed earlier than glial fibrillary acidic protein and microtubule-associated protein 2 expression.In addition,the number of NSE-positive cells was increased significantly more than glial fibrillary acidic protein-positive cells. CONCLUSION:Radix Astragali promoted process formation in stem cells.It may induce the differentiation of MSCs into neural stem cells,and subsequently into neuronal-and glial-like cells. 4Polarity inverted intracloud discharges and electric charge structure of thunderstorm显示文摘The three-dimensional spatial and temporal development of impulsive VHF radiation events during lightning discharges has been analyzed based on the data measured by the lightning mapping array (LMA) system with high space and time resolution. The results indicate that intracloud discharges in the thunderstorm with a normal tripole charge structure occur not only between the upper positive and middle main negative charge region, but also with inverted polarity between the lower positive and middle main negative charge region. The polarity-inverted discharge originates from the middle negative charge and propagates downward to the lower positive region, where then the discharge develops horizontally. The characteristics of the discharge are similar to that of normal intracloud discharge except the polarity opposite. The results further confirmed the existence of the lower positive charge region involved in the lightning discharge. 5Acupuncture at Waiguan(SJ5) and sham points influences activation of functional brain areas of ischemic stroke patients: a functional magnetic resonance imaging study显示文摘Most studies addressing the specificity of meridians and acupuncture points have focused mainly on the different neural effects of acupuncture at different points in healthy individuals. This study examined the effects of acupuncture on brain function in a pathological context. Sixteen patients with ischemic stroke were randomly assigned to true point group(true acupuncture at right Waiguan(SJ5)) and sham point group(sham acupuncture). Results of functional magnetic resonance imaging revealed activation in right parietal lobe(Brodmann areas 7 and 19), the right temporal lobe(Brodmann area 39), the right limbic lobe(Brodmann area 23) and bilateral occipital lobes(Brodmann area 18). Furthermore, inhibition of bilateral frontal lobes(Brodmann area 4, 6, and 45), right parietal lobe(Brodmann areas 1 and 5) and left temporal lobe(Brodmann area 21) were observed in the true point group. Activation in the precuneus of right parietal lobe(Brodmann area 7) and inhibition of the left superior frontal gyrus(Brodmann area 10) was observed in the sham group. Compared with sham acupuncture, acupuncture at Waiguan in stroke patients inhibited Brodmann area 5 on the healthy side. 6Charge structures and cloud-to-ground lightning discharges characteristics in two supercell thunderstorms显示文摘The charge structures and temporal and spatial characteristics of cloud-to-ground (CG) light- ning discharges in two supercell thunderstorms have been analyzed based on the data of three-dimen- sional VHF radiation sources with high time and space resolution produced by lightning discharges. The results indicate that the charge structures in main part (convective region) of the thunderstorms were inverted tripole while a number of positive CG lightning discharges were occurring in the two thun- derstorms. The positive CG lightning discharges oc- curred in main part of the thunderstorms and origi- nated from the positive charge region located at the middle part of the thunderstorms. While a number of negative CG lightning discharges were occurring, the negative CG lightning discharges occurred in the anvil of the thunderstorm. The charge structure is inverted dipole in the region due to the slant of charge structure in main region toward anvil region. The negative charge region located at the upper part of anvil produced a lot of negative CG lightning dis- charges. No or less CG lighting was produced di- rectly by the charge region located at the lower part of the thunderstorm. 8Efficient generation of transgenic chickens using the spermatogonial stem cells in vivo and ex vivo transfection显示文摘The highly efficient novel methods to produce transgenic chickens were established by directly in-jecting the recombinant plasmid containing green fluorescent protein (GFP) gene into the cock's testis termed as testis-medianted gene transfer (TMGT), and transplanting transfected spermatogonial stem cells (TTSSCs). For the TMGT approach,four dosages of pEGFP-N1 DNA/cationic polymer complex were injected intratesticularly. The results showed: (1) 48 h after the injection,the percentages of testis cells expressing GFP were 4.0%, 8.7%, 10.2% and 13.6% in the 50, 100, 150 and 200 μg/mL group, re-spectively. The difference from the four dosage groups was significant (P<0.05). On day 25 after the injection, a dosage-dependent and time-dependent increase in the number of transgenic sperm was observed. The percentages of gene expression reached the summit and became stable from day 70 to 160, being 12.7%, 12.8%, 15.9% and 19.1%, respectively. The difference from the four dosage groups was also significant (P<0.05). (2) 70 d after the injection, strong green fluorescent could be observed in the seminiferous tubules by whole-mount in-situ hybridization. (3) 70 d after the injection, the semen was collected and used to artificially inseminate wild-type females. The blastoderms of F1 and F2 transgenic chicken expressed GFP were 56.2% (254/452) and 53.2% (275/517), respectively. The detec-tion of polymerase chain reaction (PCR) of F1 and F2 transgenic chicken blood genomic DNA showed that 56.5% (3/23) of F1 and 52.9% (9/17) of F2 were positive. Southern blot showed GFP DNA was in-serted in their genomic DNAs. (4) Frozen whole mount tissue sections of F1 and F2 transgenic chicken liver, heart, kidney and muscle showed that the rates of green fluorescent positive were between 50.0% and 66.7%. (5) With the TTSSCs method, SSCs ex vivo transfected with GFP were transplanted into recipient roosters whose endogenic SSCs had been resoluted. The donor SSCs settled and GFP ex-pression became readily detectable in the frozen whole mount tissue sections of recepient testes. Moreover, sperms carrying GFP could be produced normally. The results of artificially inseminating wild-type females with these sperms showed 12.5% (8/64) of offspring embryo expressed GFP and 11.1% (2/18) hatched chicks were tested transgenic. 9Late-Quaternary strong earthquakes on the seismogenic fault of the 1976 M_s7.8 Tangshan earthquake,Hebei,as revealed by drilling and trenching显示文摘Composite borehole profiling combined with trenching is an effective way to acquire evidence of past ruptures of buried active faults.In this study,three composite borehole profiles and a large-scale trench excavation were carried out across the surface rupture zone of the 1976 Ms 7.8 Tangshan earthquake.The following three major conclusions have been reached.(1) The surface rupture zone of the 1976 earthquake extends more than 47 km long to the south of Tangshan city,passing to the west of Sunjialou,to Daodi town in Fengnan County,to Xihe in Fengnan County.(2) The surface rupture zone is divided into south and north branches.The north branch has mainly right-lateral strike-slip motion,and the vertical displacement of the surface is up on the west and down on the east.On the other hand,the vertical displacement of the south branch is up on the east and down on the west,accompanied by some right-lateral slip.Such a faulting style cannot be explained by the movement of a single normal or reverse fault,but is consistent with the vertical displacement field induced by the right-lateral strike-slip of the fault belt.The drilling and trenching data from this study verify that such activity continued through the Late Quaternary on the Tangshan Fault.(3) The fault planes exhumed by trenching and the dislocations of strata revealed by the boreholes indicate that multiple faulting events occurred on the Tangshan Fault in the Late Quaternary.The timing of three ruptures prior to the 1976 earthquake was 7.61-8.13,>14.57,and 24.21-26.57 ka BP.Counting the earthquake of 1976,the recurrence interval of the four strong events is about 6.7 to 10.8 ka.On one of the three borehole profiles,the Niumaku profile,nine faulting events were detected since 75.18 ka BP with an average interval of 8.4 ka.In addition,this paper also discusses the difference between the Late Quaternary sedimentary environments to the north and south of Tangshan city based on stratum dating.GUO Hui JIANG WaLi XIE XinSheng 2011Science China Earth Sciences2011,54,11:13
10Opening time and filling pattern of the Neoproterozoic Kangdian Rift Basin,western Yangtze Continent,South China显示文摘The Neoproterozoic Kangdian Rift Basin is a continental rift basin in the western Yangtze Continent.Determining its time of opening and subsequent filling pattern is an important aspect of understanding the formation and evolution of the Yangtze Continent,South China.The Luliang Formation is the early filling in the eastern part of this rift basin,and its deposition age and filling sequence are significant for studies of the regional stratigraphic correlation,opening time,and filling pattern of this basin.Having been correlated to the upper part of the Mesoproterozoic Kunyang Group or to the Neoproterozoic Chengjiang Formation,the age and regional stratigraphic correlation of the Luliang Formation have long been uncertain.This is due to a lack of reliable absolute age constraints.To address this issue,the first zircon SHRIMP U-Pb geochronology has been established for this formation,yielding two high-precision isotopic ages of 818.6±9.2 and 805±14 Ma for the tuff layers at the bottoms of the lower and upper members of the Luliang Formation,respectively.Given the error factor,the bottom age of the lower member of the Luliang Formation can be interpreted as ca.820 Ma,corresponding to the bottom age of the lower part of the Banxi Group,which is the early filling of the Neoproterozoic Xianggui Rift Basin,a representative basin of the Neoproterozoic rift basins in the Yangtze Continent,South China.The bottom age of the upper member of the Luliang Formation can be interpreted as ca.800±5 Ma,corresponding to the bottom age of the Chengjiang Formation in the western part of the Neoproterozoic Kangdian Rift Basin and also corresponding to the bottom age of the upper part of the Banxi Group in the Neoproterozoic Xianggui Rift Basin.These ages indicate that the Neoproterozoic Kangdian Rift Basin shares the same opening time and filling sequences as those of the other Neoproterozoic rift basins in South China.Basin analysis shows that the Neoproterozoic Kangdian Rift Basin is a typical half-graben basin,with its main boundary fault in the west and basin center in the east.This basin consisted of mini unidirectional half-graben basins in its juvenile stage and simplified to become a large united half-graben basin in its mature stage,i.e.,a supradetachment basin.ZHUO JieWen JIANG XinSheng WANG Jian CUI XiaoZhuang XIONG GuoQing LU JunZe LIU JianHui MA MingZhu 2013Science China Earth Sciences2013,56,10:12

12Analysis of the CDR3 Length Repertoire and the Diversity of TCRα Chain in Human Peripheral Blood T Lymphocytes显示文摘Analysis of complementarity determining region 3 (CDR3) length of T lymphocyte receptors (TCRs) by immunoscope spectratyping technique has been used successfully to investigate the diversity of TCR in autoimmune diseases and infection diseases. In this study, we investigated the patterns of CDR3 length distribution for all 32 TCR AV gene families in human peripheral blood lymphocytes of four normal volunteers by the immunoscope spectratyping technique. It was found that PCR products exhibited an obscure band on 1.5% agarose gel electrophoresis. Each TCR AV family exhibited more than 8 bands on 6% sequencing gel electrophoresis. The CDR3 spectratyping of all TCR AV families showed a standard Gaussian distribution with different CDR3 length, and the expression frequency of CDR3 was similar among the gene families. Most of CDR3 in TCR AV family recombine in frame. However, some of the CDR3 showed out-of frame gene rearrangement. Additionally, we found that in some of TCR AV families there were 18 amino acid discrepancies between the longest CDR3 and shortest CDR3. 13Stem cell transplantation for treatment of cerebral ischemia in rats Effects of human umbilical cord blood stem cells and human neural stem cells显示文摘BACKGROUND: Exogenous neural stem cell transplantation promotes neural regeneration. However, various types of stem cells transplantation outcomes remain controversial. OBJECTIVE: To explore distribution, proliferation and differentiation of human neural stem cells (hNSCs) and human umbilical cord blood stem cells (hUCBSCs) following transplantation in ischemic brain tissue of rats, and to compare therapeutic outcomes between hNSCs and hUCBSCs. DESIGN, TIME AND SETTING: Randomized controlled animal studies were performed at the Experimental Animal Center of Nanjing Medical University and Central Laboratory of Second Affiliated Hospital of Nanjing Medical University of China from September 2008 to April 2009. MATERIALS: hNSCs were harvested from brain tissue of 10-13 week old fetuses following spontaneous abortion, and hUCBSCs were collected from umbilical cord blood of full-term newborns at the Second Affiliated Hospital of Nanjing Medical University of China. hNSCs and hUCBSCs were labeled by 5-bromodeoxyuridine (BrdU) prior to transplantation. METHODS: Rat models of cerebral ischemia were established by the suture method. A total of 60 healthy male Sprague Dawley rats aged 7-9 weeks were randomly assigned to hNSC transplantation, hUCBSC transplantation and control groups. The rat models in the hNSC transplantation, hUCBSC transplantation and control groups were infused with hNSC suspension, hUCBSC suspension and saline via the caudal vein, respectively. MAIN OUTCOME MEASURES: The distribution, proliferation and differentiation of hNSCs and hUCBSCs in ischemic brain tissue were observed using immunohistochemical methods. Neurological function in rats was assessed using the neurological severity score. RESULTS: The number of BrdU-positive cells was significantly greater in the hNSC transplantation group compared with hUCBSC transplantation group at 14 days following transplantation (P < 0.05). The number of BrdU-positive cells reached a peak at 28 days following transplantation. Nestin-positive, glial fibrillary acidic protein-positive, cyclic nucleotide 3' phosphohydrolase-positive and neuron specific enolase-positive cells were visible following transplantation. No significant difference was determined in the constituent ratio of various cells between hNSC and hUCBSC transplantation groups (P > 0.05). The neurological severity score was significantly decreased in rats at 21 days following transplantation (P < 0.05). No significant difference was detected in neurological severity score between hNSC and hUCBSC transplantation groups at various time points (P > 0.05). 15Inhibition of Rac1-dependent forgetting alleviates memory deficits in animal models of Alzheimer's disease显示文摘Accelerated forgetting has been identified as a feature of Alzheimer's disease(AD),but the therapeutic efficacy of the manipulation of biological mechanisms of forgetting has not been assessed in AD animal models.Ras-re-lated C3 botulinum toxin substrate 1(Rac1),a small GTPase,has been shown to regulate active forgetting in Drosophila and mice?Here,we showed that Rac1 activity is aberrantly elevated in the hippocampal tissues of AD patients and AD animal models.Moreover,amyloid-beta 42 could induce Rac1 activation in cultured cells.The elevation of Rac1 activity not only accelerated 6-hour spatial memory decay in 3-month-old APP/PS1 mice,but also significantly contributed to severe memory loss in aged APP/PS1 mice.A similar age-dependent Rac1 activity-based memory loss was also observed in an AD fly model.Moreover,inhibition of Rac1 activity could ameliorate cognitive defects and synaptic plasticity in AD animal models.Finally,two novel compounds,identified through behavioral screening of a randomly selected pool of brain permeable small molecules for their positive effect in rescuing memory loss in both fly and mouse models,were found to be capable of inhibiting Rac1 activity.Thus,multiple lines of evidence corroborate in supporting the idea that inhibition of Rac1 activity is effective for treating AD-related memory loss.Wenjuan Wu Shuwen Du Wei Shi Yunlong Liu Ying Hu Zuolei Xie Xinsheng Yao Zhenyu Liu Weiwei Ma Lin Xu Chao Ma Yi Zhong 2019Protein & Cell2019,10,10:10
16Broadband interferometer observations of a triggered lightning显示文摘The development of positive leader of an artificially triggered lightning has been analyzed based on the data of electric field change, location of radiation source and frequency spectrum obtained by using the broadband interferometer system. The results indicate that radiation from positive leader could be detected within close distance in spite of the relatively weak radiation, while the radiation from negative breakdown processes was relatively stronger. Positive leader developed with few branches, and the initial progression velocity was of the order of 105 m/s. 18Relationship between the number of tapping-induced secondary laticifer lines and rubber yield among Hevea germplasm显示文摘The lack of suitable early selection parameters means that traditional rubber breeding for yield is time consuming and inefficient. Laticifer is a tissue specific fo natural rubber biosynthesis and storage in rubber tree. The number of the secondary laticifers in the trunk bark tissues is positively correlated with rubber yield in the matured rubber trees that are regularly tapped. In the present study the rubber yield from 280 of 4–5 year-old virgin trees from7 cross combinations was compared with the number o newly differentiated secondary laticifers caused by tapping. 20Acupuncture at Waiguan (TE5) influences activation/deactivation of functional brain areas in ischemic stroke patients and healthy people A functional MRI study显示文摘In the present study, 10 patients with ischemic stroke in the left hemisphere and six healthy controls were subjected to acupuncture at right Waiguan (TE5). In ischemic stroke subjects, functional MRI showed enhanced activation in Broadmann areas 5, 6, 7, 18, 19, 24, 32, the hypothalamic inferior lobe, the mamillary body, and the ventral posterolateral nucleus of the left hemisphere, and Broadmann areas 4, 6, 7, 18, 19 and 32 of the right hemisphere, but attenuated activation of Broadmann area 13, the hypothalamic inferior lobe, the posterior lobe of the tonsil of cerebellum, and the culmen of the anterior lobe of hypophysis, in the left hemisphere and Broadmann area 13 in the right hemisphere. In ischemic stroke subjects, a number of deactivated brain areas were enhanced, including Broadmann areas 6, 11, 20, 22, 37, and 47, the culmen of the anterior lobe of hypophysis, alae lingulae cerebella, and the posterior lobe of the tonsil of cerebellum of the left hemisphere, and Broadmann areas 8, 37, 45 and 47, the culmen of the anterior lobe of hypophysis, pars tuberalis adenohypophyseos, inferior border of lentiform nucleus, lateral globus pallidus, inferior temporal gyrus, and the parahippocampal gyrus of the right hemisphere. These subjects also exhibited attenuation of a number of deactivated brain areas, including Broadmann area 7. These data suggest that acupuncture at Waiguan specifically alters brain function in regions associated with sensation, vision, and motion in ischemic stroke patients.
