| 2 | Human CMTM2/CKLFSF2 enhances the ligand-induced transactivation of the androgen receptor显示文摘CKLF (chemokine-like factor)-like MARVEL (MAL and related proteins for vesicle trafficking and mem-brane link domain) transmembrane domain containing (CMTM) is a novel gene family. One member of this family, CMTM2, also named chemokine-like factor superfamily 2 (CKLFSF2), is expressed highly in the testis and moderately in the prostate, marrow and peripheral blood cells. However, the function of human CMTM2 remains unknown. Here, we found that CMTM2 was upregulated in 5α-dihydrotestos- terone (DHT)-treated LNCaP cells. We investigated the relationship between CMTM2 and the androgen receptor. Our results showed that CMTM2 enhanced DHT-mediated androgen receptor (AR) transacti-vation and the expression of prostate specific antigen (PSA). We also observed that CMTM2 enhanced the AR protein level, which was reversed by silencing endogenous CMTM2 expression, which sug-gested that CMTM2 might play an important role in maintaining the AR protein level. We also found that CMTM2 suppressed Akt activation. A previous study showed that Akt could phosphorylate AR at Ser210 and Ser790 and lead to AR ubiquitylation and degradation as well as suppression of AR activity. | 4 | RNA Methylome Reveals the m^(6)A-mediated Regulation of Flavor Metabolites in Tea Leaves under Solar-withering显示文摘The epitranscriptomic mark N6-methyladenosine(m^(6)A),which is the predominant internal modification in RNA,is important for plant responses to diverse stresses.Multiple environmental stresses caused by the tea-withering process can greatly influence the accumulation of specialized metabolites and the formation of tea flavor.However,the effects of the m^(6)A-mediated regulatory mechanism on flavor-related metabolic pathways in tea leaves remain relatively uncharacterized.We performed an integrated RNA methylome and transcriptome analysis to explore the m^(6)Amediated regulatory mechanism and its effects on flavonoid and terpenoid metabolism in tea(Camellia sinensis)leaves under solar-withering conditions.Dynamic changes in global m^(6)A level in tea leaves were mainly controlled by two m^(6)A erasers(CsALKBH4A and CsALKBH4B)during solar-withering treatments.Differentially methylated peak-associated genes following solarwithering treatments with different shading rates were assigned to terpenoid biosynthesis and spliceosome pathways.Further analyses indicated that CsALKBH4-driven RNA demethylation can directly affect the accumulation of volatile terpenoids by mediating the stability and abundance of terpenoid biosynthesis-related transcripts and also indirectly influence the flavonoid,catechin,and theaflavin contents by triggering alternative splicing-mediated regulation.Our findings revealed a novel layer of epitranscriptomic gene regulation in tea flavor-related metabolic pathways and established a link between the m^(6)A-mediated regulatory mechanism and the formation of tea flavor under solar-withering conditions. | Chen Zhu Shuting Zhang Chengzhe Zhou Caiyun Tian Biying Shi Kai Xu Linjie Huang Yun Sun Yuling Lin Zhongxiong Lai Yuqiong Guo | 2023 | Genomics, Proteomics & Bioinformatics2023,21,4: | 2 |
| 5 | The chromosome-scale genome assembly of Jasminum sambac var. unifoliatum provides insights into the formation of floral fragrance显示文摘Jasmine [Jasminum sambac(L.) Ait.], a tropical and subtropical plant emits a sweet, heady fragrance during flower opening. However, the molecular mechanisms underlying this phenomenon remain largely unknown. In the present study, integrated Illumina sequencing, Pacbio sequencing, and high-throughput chromatin conformation capture(Hi-C) scaffolding was used to generate a 495.60 Mb genome assembly of J.sambac var. unifoliatum cultivar ‘Fuzhou Single-petal’(JSU-FSP), with contig N50 of 16.88 Mb;96.23% of the assembly was assigned to 13 pseudochromosomes. The genome harbors 30 989 protein-coding genes, and 49.47% of the assembled sequences are repetitive sequences. The analysis of duplication modes showed that 51% of genes were duplicated through dispersed duplication, and expanded gene families are mainly involved in photosynthesis, which may be responsible for the light-loving characteristic specific to jasmine. Transcriptome analysis revealed that at least 35 structural genes involved in the biosynthesis of volatile terpenes(VTs), volatile phenylpropanoid/benzenoids(VPBs),fatty acid-derived volatiles(FADVs), and indole were highly expressed in the flower-opening stage, both preharvest and postharvest, and are proposed to be important in endowing flower aroma. Additionally, at least 28 heat shock protein(HSP) and 11 β-glucosidase(BGLU) genes may be involved in the formation of floral fragrance. | 6 | Achieving reliable and anti-collusive outsourcing computation and verification based on blockchain in 5G-enabled IoT显示文摘Widespread applications of 5G technology have prompted the outsourcing of computation dominated by the Internet of Things(IoT)cloud to improve transmission efficiency,which has created a novel paradigm for improving the speed of common connected objects in IoT.However,although it makes it easier for ubiquitous resource-constrained equipment that outsources computing tasks to achieve high-speed transmission services,security concerns,such as a lack of reliability and collusion attacks,still exist in the outsourcing computation.In this paper,we propose a reliable,anti-collusion outsourcing computation and verification protocol,which uses distributed storage solutions in response to the issue of centralized storage,leverages homomorphic encryption to deal with outsourcing computation and ensures data privacy.Moreover,we embed outsourcing computation results and a novel polynomial factorization algorithm into the smart contract of Ethereum,which not only enables the verification of the outsourcing result without a trusted third party but also resists collusion attacks.The results of the theoretical analysis and experimental performance evaluation demonstrate that the proposed protocol is secure,reliable,and more effective compared with state-of-the-art approaches. | Linjie Wang Youliang Tian Jinbo Xiong | 2022 | Digital Communications and Networks2022,8,5: | 0 |
