1Kinetics of the accumulation of group 2 innate lymphoid cells in IL-33-induced and IL-25-induced murine models of asthma:a potential role for the chemokine CXCL16显示文摘ILC2s are implicated in asthma pathogenesis, but little is known about the mechanisms underlying their accumulation in airways.We investigated the time course of ILC2 accumulation in different tissues in murine models of asthma induced by a serial per-nasalchallenge with ovalbumin (OVA), house dust mice (HDM), IL-25 and IL-33 and explored the potential roles of ILC2-attractingchemokines in this phenomenon. Flow cytometry was used to enumerate ILC2s at various time points. The effects of cytokines andchemokines on ILC2 migration were measured in vitro using a chemotaxis assay and in vivo using small animal imaging. Comparedwith saline and OVA challenge, both IL-25 and IL-33 challenge alone induced significant accumulation of ILC2s in the mediastinallymph nodes, lung tissue and bronchoalveolar lavage fluid of challenged animals, but with a distinct potency and kinetics. In vitro,IL-33 and CXCL16, but not IL-25 or CCL25, directly induced ILC2 migration. Small animal in vivo imaging further confirmed that asingle intranasal provocation with IL-33 or CXCL16 was sufficient to induce the accumulation of ILC2s in the lungs followinginjection via the tail vein. Moreover, IL-33-induced ILC2 migration involved the activation of ERK1/2, p38, Akt, JNK and NF-κB, whileCXCL16-induced ILC2 migration involved the activation of ERK1/2, p38 and Akt. These data support the hypothesis that epitheliumderived IL-25 and IL-33 induce lung accumulation of ILC2s, while IL-33 exerts a direct chemotactic effect in this process. 2Direct generation of an ultrafast vortex beam in a CVD-graphene-based passively mode-locked Pr:LiYF4 visible laser显示文摘We report on the direct generation of passively mode-locked vortex lasers in the visible spectral region, for the first time to the best of our knowledge, using a Pr :Li YF4(Pr:YLF) crystal as the gain medium. A stable mode-locked TEM00 mode has been achieved with a maximum average output power of 75 m W using a graphene saturable absorber mirror. The mode-locked pulse width is measured to be as short as about 73.4 ps at a repetition rate of about 140 MHz, and the laser wavelength is at about 721 nm with spectral width of about 0.5 nm. By slightly misaligning the laser resonator, a first-order Laguerre-Gaussian mode(LG0,1) has also been obtained with output power reduced to about 22 m W. The achieved LG0,1 mode has been verified via a home made improved Fizeau interferometer. 3Increased mtDNA copy number promotes cancer progression by enhancing mitochondrial oxidative phosphorylation in microsatellite-stable colorectal cancer显示文摘Colorectal cancer is one of the leading causes of cancer death worldwide.According to global genomic status,colorectal cancer can be classified into two main types:microsatellite-stable and microsatellite-instable tumors.Moreover,the two subtypes also exhibit different responses to chemotherapeutic agents through distinctive molecular mechanisms.Recently,mitochondrial DNA depletion has been shown to induce apoptotic resistance in microsatellite-instable colorectal cancer.However,the effects of altered mitochondrial DNA copy number on the progression of microsatellite-stable colorectal cancer,which accounts for the majority of colorectal cancer,remain unclear.In this study,we systematically investigated the functional role of altered mitochondrial DNA copy number in the survival and metastasis of microsatellite-stable colorectal cancer cells.Moreover,the underlying molecular mechanisms were also explored.Our results demonstrated that increased mitochondrial DNA copy number by forced mitochondrial transcription factor A expression significantly facilitated cell proliferation and inhibited apoptosis of microsatellitestable colorectal cancer cells both in vitro and in vivo.Moreover,we demonstrated that increased mitochondrial DNA copy number enhanced the metastasis of microsatellite-stable colorectal cancer cells.Mechanistically,the survival advantage conferred by increased mitochondrial DNA copy number was caused in large part by elevated mitochondrial oxidative phosphorylation.Furthermore,treatment with oligomycin significantly suppressed the survival and metastasis of microsatellite-stable colorectal cancer cells with increased mitochondrial DNA copy number.Our study provides evidence supporting a possible tumor-promoting role for mitochondrial DNA and uncovers the underlying mechanism,which suggests a potential novel therapeutic target for microsatellite-stable colorectal cancer.Xiacheng Sun Lei Zhan Yibing Chen Gang Wang Linjie He Qian Wang Feng Zhou Fang Yang Jin Wu Yousheng Wu Jinliang Xing Xianli He Qichao Huang 2018Signal Transduction and Targeted Therapy2018,3,1:3
4Segregation of Phosphorus and Precipitation of MNP-Type Phosphide at the Grain Boundary of IN706 Superalloy显示文摘The effect of phosphorus content and heat treatment on the segregation of phosphorus and precipitation of phosphide at the grain boundary of IN706 alloy has been investigated. The phosphide had a stoichiometry of MNP(M = Nb, Ti and N = Ni, Fe, Cr) and an orthorhombic crystal structure. The solubility of phosphorus in the grain matrix of IN706 alloy was determined to be between 0.008% and 0.013%.When soaked at 980 °C for 5 min, the phosphide was noticeably precipitated at grain boundaries with the grain size unchanged. When soaked at 980 °C for 3 h, the grains grew significantly, but the phosphide stayed at original grain boundaries which outlined the grain shape before soaking. Soaking at 1060 °C for 2 h fully dissolved the pre-existing phosphide but phosphorus atoms were not distributed uniformly in the grain matrix. When the holding time at 1060 °C was extended to 10 h, the phosphorus atoms were distributed uniformly in the grain matrix by sufficient diffusion, and the phosphide could only be formed at grain boundaries during the 980 °C soaking. 5Broadband radio spectral observations of the solar eclipse on 2008-08-01 and its implications on the quiet Sun atmospheric model显示文摘Based on the joint-observations of the radio broadband spectral emissions of the solar eclipse on August 1, 2008 at Jiuquan (total eclipse) and Huairou (partial eclipse) at the frequencies of 2.00-5.60 GHz (Jiuquan), 2.60-3.80 GHz (Chinese solar broadband radiospectrometer, SBRS/Huairou), and 5.20-7.60 GHz (SBRS/Huairou), the authors assemble a successive series of broadband spectra with a frequency of 2.60-7.60 GHz to observe the solar eclipse synchronously. This is the first attempt to analyze the solar eclipse radio emission under the two telescopes located at different places with broadband frequencies in the periods of total and partial eclipses. 6RNA Methylome Reveals the m^(6)A-mediated Regulation of Flavor Metabolites in Tea Leaves under Solar-withering显示文摘The epitranscriptomic mark N6-methyladenosine(m^(6)A),which is the predominant internal modification in RNA,is important for plant responses to diverse stresses.Multiple environmental stresses caused by the tea-withering process can greatly influence the accumulation of specialized metabolites and the formation of tea flavor.However,the effects of the m^(6)A-mediated regulatory mechanism on flavor-related metabolic pathways in tea leaves remain relatively uncharacterized.We performed an integrated RNA methylome and transcriptome analysis to explore the m^(6)Amediated regulatory mechanism and its effects on flavonoid and terpenoid metabolism in tea(Camellia sinensis)leaves under solar-withering conditions.Dynamic changes in global m^(6)A level in tea leaves were mainly controlled by two m^(6)A erasers(CsALKBH4A and CsALKBH4B)during solar-withering treatments.Differentially methylated peak-associated genes following solarwithering treatments with different shading rates were assigned to terpenoid biosynthesis and spliceosome pathways.Further analyses indicated that CsALKBH4-driven RNA demethylation can directly affect the accumulation of volatile terpenoids by mediating the stability and abundance of terpenoid biosynthesis-related transcripts and also indirectly influence the flavonoid,catechin,and theaflavin contents by triggering alternative splicing-mediated regulation.Our findings revealed a novel layer of epitranscriptomic gene regulation in tea flavor-related metabolic pathways and established a link between the m^(6)A-mediated regulatory mechanism and the formation of tea flavor under solar-withering conditions.Chen Zhu Shuting Zhang Chengzhe Zhou Caiyun Tian Biying Shi Kai Xu Linjie Huang Yun Sun Yuling Lin Zhongxiong Lai Yuqiong Guo 2023Genomics, Proteomics & Bioinformatics2023,21,4:2

10Dingxin recipe alleviates atherosclerosis injury in Apo E-knockout mice via downregulation of visfatin expression and inhibition of the visfatin-induced inflammatory response显示文摘OBJECTIVE:To further elucidate the mechanism underlying the anti-atherosclerotic effect of Dingxin recipe(DXR).METHODS:Fifty 6-week-old male Apo E^-/-mice were randomly divided into the following groups:model,simvastatin(5 mg·kg^-1·d^-1),DXR low-dose(9.30 g·kg^-1·d^-1),DXR middle-dose(18.59 g·kg^-1·d^-1)and DXR high-dose(37.18 g·kg^-1·d^-1)(n=10).Ten male C57BL/6J mice were used as the control group.All Apo E^-/-mice were fed a high-fat diet(HFD)and the control mice received a common diet.After HFD for 12 weeks,the mice were treated with DXR or simvastatin for another 12 weeks.The expression of inflammatory cytokines and visfatin was determined in serum and atherosclerotic lesions by enzyme-linked immunosorbent assay.Visfatin expression was also assessed in aortic atherosclerotic plaques.Cultured vessel endothelial cells(VECs)were pretreated with DXR sera prior to visfatin.The effects of DXR were analyzed to elucidate its protective mechanism against visfatin-induced inflammation in VECs.RESULTS:DXR regulated blood lipids and reduced tumor necrosis factor-α(TNF-α),interleukin-6(IL-6),intercellular adhesion molecules-1(ICAM-1),vascular cell adhesion molecule-1(VCAM-1)and visfatin expression in Apo E^-/-mice,particularly at the higher doses.The areas of atherosclerotic lesions in the DXR groups were significantly smaller than those in the model group.DXR alleviated visfatin-induced VEC injury via downregulation of TNF-α,IL-6,ICAM-1 and VCAM-1 through mitogen-activated protein kinase pathways.CONCLUSION:DXR alleviated atherosclerosis injury via downregulation of visfatin expression and inhibition of the visfatin-induced inflammatory response in VECs.Cui Xiaobing Huang Zhiyong Lou Linjie Cheng Saibo Zhang Yu Zhang Yaxin Jia Yuhua Zhou Fenghua 2020Journal of Traditional Chinese Medicine2020,40,6:1
11Continuous-variable quantum key distribution with on-chip light sources显示文摘Integrated quantum key distribution(QKD)systems based on photonic chips have high scalability and stability,and are promising for further construction of global quantum communications networks.On-chip quantum light sources are a critical component of a fully integrated QKD system;especially a continuous-variable QKD(CVQKD)system based on coherent detection,which has extremely high requirements for the light sources.Here,for what we believe is the first time,we designed and fabricated two on-chip tunable lasers for CV-QKD,and demonstrated a high-performance system based on these sources.Because of the high output power,fine tunability,and narrow linewidth,the involved on-chip lasers guarantee the accurate shot-noise-limited detection of quantum signals,center wavelength alignment of nonhomologous lasers,and suppression of untrusted excess noise.The system’s secret key rate can reach 0.75 Mb/s at a 50 km fiber distance,and the secure transmission distance can exceed 100 km.Our results mark a breakthrough toward building a fully integrated CV-QKD,and pave the way for a reliable and efficient terrestrial quantum-secure metropolitan area network.LANG LI TAO WANG XINHANG LI PENG HUANG YUYAO GUO LIANGJUN LU LINJIE ZHOU GUIHUA ZENG 2023Photonics Research2023,11,4:0
12An AIEgen/graphene oxide nanocomposite(AIEgen@GO)-based two-stage“turn-on”nucleic acid biosensor for rapid detection of SARS-CoV-2 viral sequence显示文摘The ongoing outbreak of Severe Acute Respiratory Syndrome Coronavirus 2(SARS-CoV-2)pandemic has posed significant challenges in early viral diagnosis.Hence,it is urgently desirable to develop a rapid,inexpensive,and sensitive method to aid point-of-care SARS-CoV-2 detection.In this work,we report a highly sequence-specific biosensor based on nanocomposites with aggregationinduced emission luminogens(AIEgen)-labeled oligonucleotide probes on graphene oxide nanosheets(AIEgen@GO)for one step-detection of SARS-CoV-2-specific nucleic acid sequences(Orf1ab or N genes).A dual“turn-on”mechanism based on AIEgen@GO was established for viral nucleic acids detection.Here,the first-stage fluorescence recovery was due to dissociation of the AIEgen from GO surface in the presence of target viral nucleic acid,and the second-stage enhancement of AIEbased fluorescent signal was due to the formation of a nucleic acid duplex to restrict the intramolecular rotation of the AIEgen.Furthermore,the feasibility of our platform for diagnostic application was demonstrated by detecting SARS-CoV-2 virus plasmids containing both Orf1ab and N genes with rapid detection around 1 h and good sensitivity at pM level without amplification.Our platform shows great promise in assisting the initial rapid detection of the SARS-CoV-2 nucleic acid sequence before utilizing quantitative reverse transcription-polymerase chain reaction for second confirmation.Qin Zhang Bohan Yin Jianhua Hao Linjie Ma Yingying Huang Xueying Shao Chuanqi Li Zhiqin Chu Changqing Yi Siu Hong Dexter Wong Mo Yang 2023Aggregate2023,4,1:0

14The chromosome-scale genome assembly of Jasminum sambac var. unifoliatum provides insights into the formation of floral fragrance显示文摘Jasmine [Jasminum sambac(L.) Ait.], a tropical and subtropical plant emits a sweet, heady fragrance during flower opening. However, the molecular mechanisms underlying this phenomenon remain largely unknown. In the present study, integrated Illumina sequencing, Pacbio sequencing, and high-throughput chromatin conformation capture(Hi-C) scaffolding was used to generate a 495.60 Mb genome assembly of J.sambac var. unifoliatum cultivar ‘Fuzhou Single-petal’(JSU-FSP), with contig N50 of 16.88 Mb;96.23% of the assembly was assigned to 13 pseudochromosomes. The genome harbors 30 989 protein-coding genes, and 49.47% of the assembled sequences are repetitive sequences. The analysis of duplication modes showed that 51% of genes were duplicated through dispersed duplication, and expanded gene families are mainly involved in photosynthesis, which may be responsible for the light-loving characteristic specific to jasmine. Transcriptome analysis revealed that at least 35 structural genes involved in the biosynthesis of volatile terpenes(VTs), volatile phenylpropanoid/benzenoids(VPBs),fatty acid-derived volatiles(FADVs), and indole were highly expressed in the flower-opening stage, both preharvest and postharvest, and are proposed to be important in endowing flower aroma. Additionally, at least 28 heat shock protein(HSP) and 11 β-glucosidase(BGLU) genes may be involved in the formation of floral fragrance. 16An orientated mass transfer in Ni-Cu tandem nanofibers for highly selective reduction of CO_(2) to ethanol显示文摘Electrochemically reducing CO_(2)to ethanol is attractive but suffers from poor selectivity.Tandem catalysis that integrates the activation of CO_(2)to an intermediate using one active site and the subsequent formation of hydrocarbons on the other site offers a promising approach,where the control of the intermediate transfer between different catalytic sites is challenging.We propose an internally self-feeding mechanism that relies on the orientation of the mass transfer in a hierarchical structure and demonstrate it using a one-dimensional(1D)tandem core-shell catalyst.Specifically,the carbon-coated Ni-core(Ni/C)catalyzes the transformation of CO_(2)-to-CO,after which the CO intermediates are guided to diffuse to the carbon-coated Cu-shell(Cu/C)and experience the selective reduction to ethanol,realizing the orientated key intermediate transfer.Results show that the Faradaic efficiency for ethanol was 18.2%at-1 V vs.RHE(V_(RHE))for up to 100 h.The following mechanism study supports the hypothesis that the CO_(2)reduction on Ni/C generates CO,which is further reduced to ethanol on Cu/C sites.Density functional theory calculations suggest a combined effect of the availability of CO intermediate in Ni/C core and the dimerization of key∗CO intermediates,as well as the subsequent proton-electron transfer process on the Cu/C shell.Xiaoxiong Huang Debin Kong Yingjie Ma Bin Luo Bin Wang Linjie Zhi 2023Fundamental Research2023,3,5:0

18Influence of Halide Choice on Formation of Low-Dimensional Perovskite Interlayer in Efficient Perovskite Solar Cells显示文摘Recent advances in heterojunction and interfacial engineering of perovskite solar cells(PSCs)have enabled great progress in developing highly efficient and stable devices.Nevertheless,the effect of halide choice on the formation mechanism,crystallography,and photoelectric properties of the lowdimensional phase still requires further detailed study.In this work,we present key insights into the significance of halide choice when designing passivation strategies comprising large organic spacer salts,clarifying the effect of anions on the formation of quasi-2D/3D heterojunctions.To demonstrate the importance of halide influences,we employ novel neo-pentylammonium halide salts with different halide anions(neoPAX,X=I,Br,or Cl).We find that regardless of halide selection,iodide-based(neoPA)_(2)(FA)_((n-1))PbnI_((3n+1))phases are formed above the perovskite substrate,while the added halide anions diffuse and passivate the perovskite bulk.In addition,we also find the halide choice has an influence on the degree of dimensionality(n).Comparing the three halides,we find that chloride-based salts exhibit superior crystallographic,enhanced carrier transport,and extraction compared to the iodide and bromide analogs.As a result,we report high power conversion efficiency in quasi-2D/3D PSCs,which are optimal when using chloride salts,reaching up to 23.35%,and improving long-term stability.Xueping Liu Thomas Webb Linjie Dai Kangyu Ji Joel A.Smith Rachel C.Kilbride Mozhgan Yavari Jinxin Bi Aobo Ren Yuanyuan Huang Zhuo Wang Yonglong Shen Guosheng Shao Stephen J.Sweeney Steven Hinder Hui Li David G.Lidzey Samuel D.Stranks Neil C.Greenham S.Ravi P.Silva Wei Zhang 2022Energy & Environmental Materials2022,5,2:0
19Research on influencing factors of urban building carbon emissions based on STIRPAT model--taking Suzhou as an example显示文摘城市建筑碳排放的影响因素研究对于城市建筑领域减碳,乃至实现城市“双碳”目标具有重要意义。本文运用STIRPAT模型并结合岭回归方法建立了苏州市建筑碳排放影响因素的模型,对城市化率、常住人口数量、人均建筑业和第三产业增加值、人均可支配收入等因素进行分析。分析结果表明,城市化率是苏州建筑碳排放的主要驱动因素,其次是常住人口数量,再次是人均建筑业和第三产业增加值,最后是人均可支配收入。研究结论表明,过去一段时间,工业化和城市化的发展有力地促进了苏州建筑碳排放的增长。未来,在工业化和城市化不断发展和人口持续增长的情况下,苏州市可以通过合理规划城市发展边界,促进城乡建设领域的绿色低碳转型发展,提高居民的低碳意识,倡导居民绿色低碳行为等措施减少建筑领域碳排放。Linjie Hao Ning Huang Qing Tong Yuefeng Guo Jing Qian Wenying Chen 2023Low-carbon Materials and Green Construction2023,1,1:0
