1A nanocleaner specifically penetrates the blood-brain barrier at lesions to clean toxic proteins and regulate inflammation in Alzheimer's disease显示文摘Insurmountable blood-brain barrier(BBB) and complex pathological features are the key factors affecting the treatment of Alzheimer's disease(AD).Poor accumulation of drugs in lesion sites and undesired effectiveness of simply reducing Aβ deposition or TAU protein need to be resolved urgently.Herein,a nanocleaner is designed with a rapamycin-loaded ROS-responsive PLGA core and surface modification with KLVFF peptide and acid-cleavable DAG peptide [R@(ox-PLGA)-KcD].DAG can enhance the targeting and internalization effect of nanocleaner towards neurovascular unit endothelial cells in AD lesions,and subsequently detach from nanocleaner in response to acidic microenvironment of endosomes to promote the transcytosis of nanocleaner from endothelial cells into brain parenchyma.Then exposed KLVFF can capture and carry Aβ to microglia,attenuating Aβ-induced neurotoxicity.Strikingly,rapamycin,an autophagy promoter,is rapidly liberated from nanocleaner in the high ROS level of lesions to improve Aβ degradation and normalize inflammatory condition.This design altogether accelerates Aβ degradation and alleviates oxidative stress and excessive inflammatory response.Collectively,our finding offers a strategy to target the AD lesions precisely and multi-pronged therapies for clearing the toxic proteins and modulating lesion microenvironment,to achieve efficient AD therapy.Ting Lei Zhihang Yang Xue Xia Yuxiu Chen Xiaotong Yang Rou Xie Fan Tong Xiaolin Wang Huile Gao 2021Acta Pharmaceutica Sinica B2021,11,12:4
2Co-delivery of photosensitizer and diclofenac through sequentially responsive bilirubin nanocarriers for combating hypoxic tumors显示文摘Considering that photodynamic therapy(PDT)-induced oxygen consumption and microvascular damage could exacerbate hypoxia to drive more glycolysis and angiogenesis, a novel approach to potentiate PDT and overcome the resistances of hypoxia is avidly needed. Herein, morpholine-modified PEGylated bilirubin was proposed to co-deliver chlorin e6, a photosensitizer, and diclofenac(Dc). In acidic milieu, the presence of morpholine could enable the nanocarriers to selectively accumulate in tumor cells, while PDT-generated reactive oxidative species(ROS) resulted in the collapse of bilirubin nanoparticles and rapid release of Dc. Combining with Dc showed a higher rate of apoptosis over PDT alone and simultaneously triggered a domino effect, including blocking the activity and expression of lactate dehydrogenase A(LDHA), interfering with lactate secretion, suppressing the activation of various angiogenic factors and thus obviating hypoxia-induced resistance-glycolysis and angiogenesis. In addition, inhibition of hypoxia-inducible factor-1a(HIF-1a) by Dc alleviated hypoxia-induced resistance. 3Temporal variability of visibility and its parameterizations in Ningbo,China显示文摘Simultaneous and continuous measurements of visibility, meteorological parameters and air pollutants were carried out at a suburban site in Ningbo from June 1, 2013 to May 31,2015. The characteristics of visibility and their relationships with air pollutants and meteorological factors were investigated using multiple statistical methods. Daily visibility ranged from 0.6 to 34.1 km, with a mean value of 11.8 km. During the 2-year experiment,43.4% of daily visibility was found to be less than 10.0 km and only 9.2% was greater than 20.0 km. Visibility was lower in winter with a frequency of 53.4% in the range of 0.0–5.0 km.Annual visibility had an obvious diurnal variation, with the lowest and highest visibility being 7.5 km at approximately 06:00 local time and 15.6 km at approximately 14:00 local time, respectively. Multiple correspondence analysis(MCA) indicated that the different ranges of visibility were significantly affected by different levels of pollutants and meteorological conditions. Based on the analyses, visibility was found to be an exponential function of PM_(2.5) concentrations within a certain range of relative humidity. Thus, nonlinear models combining multiple linear regressions with exponential regression were subsequently developed using the data collected from June 2014 to May 2015, and the data from June 2013 to May 2014 was used to evaluate the performance of the model. 14An Improved Elite Slime Mould Algorithm for Engineering Design显示文摘The Swarm intelligence algorithm is a very prevalent field in which some scholars have made outstanding achievements.As a representative,Slime mould algorithm(SMA)is widely used because of its superior initial performance.Therefore,this paper focuses on the improvement of the SMA and the mitigation of its stagnation problems.For this aim,the structure of SMA is adjusted to develop the efficiency of the original method.As a stochastic optimizer,SMA mainly stimulates the behavior of slime mold in nature.For the harmony of the exploration and exploitation of SMA,the paper proposed an enhanced algorithm of SMA called ECSMA,in which two mechanisms are embedded into the structure:elite strategy,and chaotic stochastic strategy.The details of the original SMA and the two introduced strategies are given in this paper.Then,the advantages of the improved SMA through mechanism comparison,balance-diversity analysis,and contrasts with other counterparts are validated.The experimental results demonstrate that both mechanisms have a significant enhancing effect on SMA.Also,SMA is applied to four structural design issues of the welded beam design problem,PV design problem,I-beam design problem,and cantilever beam design problem with excellent results.Li Yuan Jianping Ji Xuegong Liu Tong Liu Huiling Chen Deng Chen 2023Computer Modeling in Engineering & Sciences2023,,10:1
