1Mucus production stimulated by IFN-AhR signaling triggers hypoxia of COVID-19显示文摘Silent hypoxia has emerged as a unique feature of coronavirus disease 2019(COVID-19).In this study,we show that mucins are accumulated in the bronchoalveolar lavage fluid(BALF)of COVID-19 patients and are upregulated in the lungs of severe respiratory syndrome coronavirus 2(SARS-CoV-2)-infected mice and macaques.We find that induction of either interferon(IFN)-P or IFN-y upon SARS-CoV-2 infection results in activation of aryl hydrocarbon receptor(AhR)signaling through an IDO-Kyn-dependent pathway,leading to transcriptional upregulation of the expression of mucins,both the secreted and membrane-bound,in alveolar epithelial cells.Consequently,accumulated alveolar mucus affects the blood-gas barrier,thus inducing hypoxia and diminishing lung capacity,which can be reversed by blocking AhR activity.These findings potentially explain the silent hypoxia formation in COVID-19 patients,and suggest a possible intervention strategy by targeting the AhR pathway.Yuying Liu Jiadi Lv Jiangning Liu Man Li Jing Xie Qi Lv Wei Deng Nannan Zhou Yabo Zhou Jiangping Song Peng Wang Chuan Qin Wei-Min Tong Bo Huang 2020Cell Research2020,30,12:5

5SARS-CoV-2 treatment effects induced by ACE2-expressing microparticles are explained by the oxidized cholesterolincreased endosomal pH of alveolar macrophages显示文摘Exploring the cross-talk between the immune system and advanced biomaterials to treat SARS-CoV-2 infection is a promising strategy.Here,we show that ACE2-overexpressing A549 cell-derived microparticles(AO-MPs)are a potential therapeutic agent against SARS-CoV-2 infection.Intranasally administered AO-MPs dexterously navigate the anatomical and biological features of the lungs to enter the alveoli and are taken up by alveolar macrophages(AMs).Then,AO-MPs increase the endosomal pH but decrease the lysosomal pH in AMs,thus escorting bound SARS-CoV-2 from phago-endosomes to lysosomes for degradation.This pH regulation is attributable to oxidized cholesterol,which is enriched in AO-MPs and translocated to endosomal membranes,thus interfering with proton pumps and impairing endosomal acidification.In addition to promoting viral degradation,AO-MPs also inhibit the proinflammatory phenotype of AMs,leading to increased treatment efficacy in a SARS-CoV-2-infected mouse model without side effects.These findings highlight the potential use of AO-MPs to treat SARS-CoV-2-infected patients and showcase the feasibility of MP therapies for combatting emerging respiratory viruses in the future.Zhenfeng Wang Jiadi Lv Pin Yu Yajin Qu Yabo Zhou Li Zhou Qiangqiang Zhu Shunshun Li Jiangping Song Wei Deng Ran Gao Yuying Liu Jiangning Liu Wei-Min Tong Chuan Qin Bo Huang 2022Cellular & Molecular Immunology2022,19,2:1

9An efficient approach for the treatment of radioactive waste perfluoropolyether lubricants via a synergistic effect of thermal catalysis and immobilization显示文摘Perfluoropolyether(PFPE)lubricants are a kind of high-molecular polymer with many excellent properties.However,the use of PFPEs in the nuclear industry can lead to partial decomposition and carrying radionuclides,resulting in a large amount of radioactive waste PFPE lubricants annually.Moreover,radioactive waste PFPE lubricants are difficult to be effectively treated due to their high stability,the risk of possible leakage of radionuclides,and hypertoxic fluorine-containing by-products.In this study,without any precedent,a strategy of Mn O_(2)-catalyzed decomposition and Na_(2)CO_(3)-immobilized conversion was proposed for PFPE lubricant decomposition and fluorine immobilization simultaneously based on the Lewis acid-base and oxygen vacancies concept.A high fluorine conversion efficiency of 95.4%was achieved.Meanwhile,the mechanism of decomposition suggested that Mn O_(2)mainly provided Lewis acid sites and attacked the(basic)fluorine or oxygen atoms in PFPE molecules.The decomposition of PFPE chains was proceed down and volatile fluorine-containing gas was released by partial electron transfer,intramolecular disproportionation reaction,and unzipping fashion.Subsequently,gas by-products could be further oxidized and then immobilized into fluoride salts by carbonate solid absorbents.Overall,this study provides a simple,safe,and potentially practical strategy for the harmless conversion of refractory fluorinated organic wastes,especially perfluoropolymers.Xinhang Du Wenhua Tong Xinyu Zhou Jia Luo Yuqiu Liu Yabo Wang Panyu Li Yongkui Zhang 2024Journal of Environmental Sciences2024,,2:0
10Thermodynamic behavior for generalized f(R) gravity with arbitrary coupling between matter and geometry显示文摘The thermodynamic behavior of field equations for the generalized f(R) gravity with arbitrary coupling between matter and geometry is studied in the two kinds of spacetime,i.e.,the both spatially homogenous,isotropic FRW universe and static,spherically symmetric black hole spacetime.The field equations of the generalized f(R) gravity with arbitrary coupling between matter and geometry can be cast to the form of the first law of thermodynamics with the reputed entropy production terms dS,which are quite general and can degenerate to the cases of Einstein's general relativity and pure f(R) gravity with non-coupling and nonminimal coupling as special cases.The appearance of the entropy production term dS illustrates that the horizon thermodynamics is non-equilibrium one for the generalized f(R) gravity with arbitrary coupling between matter and geometry.WU YaBo ZHAO YueYue LU JianBo ZHANG Xin TONG HaiDan YANG Hao 2012Science China(Physics,Mechanics & Astronomy)2012,55,12:0
