Clinical significance of serum procalcitonin in patients with ulcerative colitis显示文摘AIM:To investigate the association of procalcitonin(PCT)with ulcerative colitis(UC)activity.METHODS:Serum PCT levels,C-reactive protein(CRP)levels, the erythrocyte sedimentation rate, and the white blood cell count were analyzed in 18 patients with UC and 11 healthy volunteers.Serum PCT levels were analyzed by an electrochemiluminescence immunoassay.Severity assessments were based on Truelove and Witts’severity index.Correlation of serum PCT and CRP levels with UC activity was examined. Moreover,we assessed serum PCT and CRP levels in patients with a Mayo endoscopic subscore.RESULTS:Serum PCT levels in severe UC patients(n=7)(0.096±0.034 ng/mL)were significantly higher than in mild-to-moderate UC patients(n=11)(0.033±0.012 ng/mL)and healthy volunteers(n=11)(0.035±0.005 ng/mL)(P=0.0005 and P<0.0001,respectively).In addition, there was no difference in serum PCT levels between mild-to-moderate UC patients and healthy volunteers.Interestingly, patients with a Mayo endoscopic subscore of 3 points displayed significantly increased levels of serum PCT(0.075±0.043 ng/mL)compared with patients with a subscore of 2 points(0.03±0.011 ng/mL)(P=0.0302).Moreover,CRP levels in patients with severe UC or a Mayo endoscopic subscore of 3 points were not significantly higher than in patients with mild-to-moderate UC or a Mayo endoscopic subscore of 3 points.CONCLUSION:Serum PCT levels were significantly correlated with UC activity.Shigeo Koido Toshifumi Ohkusa Kazuki Takakura Shunichi Odahara Shintaro Tsukinaga Toyokazu Yukawa Jimi Mitobe Mikio Kajihara Kan Uchiyama Hiroshi Arakawa Hisao Tajiri 2013World Journal of Gastroenterology2013,19,45:15
Immunotherapy for colorectal cancer显示文摘The incidence of colorectal cancer(CRC)is on the rise, and the prognosis for patients with recurrent or metastatic disease is extremely poor.Although chemotherapy and radiation therapy can improve survival rates,it is imperative to integrate alternative strategies such as immunotherapy to improve outcomes for patients with advanced CRC.In this review,we will discuss the effect of immunotherapy for inducing cytotoxic T lymphocytes and the major immunotherapeutic approaches for CRC that are currently in clinical trials, including peptide vaccines, dendritic cell-based cancer vaccines, whole tumor cell vaccines, viral vector-based cancer vaccines, adoptive cell transfer therapy,antibody-based cancer immunotherapy, and cytokine therapy. The possibility of combination therapies will also be discussed along with the challenges presented by tumor escape mechanisms.Shigeo Koido Toshifumi Ohkusa Sadamu Homma Yoshihisa Namiki Kazuki Takakura Keisuke Saito Zensho Ito Hiroko Kobayashi Mikio Kajihara Kan Uchiyama Seiji Arihiro Hiroshi Arakawa Masato Okamoto Jianlin Gong Hisao Tajiri 2013World Journal of Gastroenterology2013,19,46:11
Research exploring greenhouse environment control over the last 50 years显示文摘Environments do not exist in isolation.Their main components in greenhouse systems are plants.Without consideration of plants, analysis of greenhouse environments and environmental control of greenhouses can be accomplished, although it is not simple to achieve.Initial attempts were undertaken to analyze greenhouse environments and then reproduce them.Ventilation rate effects on plant photosynthesis in a growth chamber were reported in 1966.Computer simulations then became a main subject of research. The first dynamic computer simulation of a greenhouse environment including plants was published in 1971.According to innovations of computer technology, the use of minicomputers and microcomputers spread in many areas.By measuring the net photosynthesis of lettuce plants grown under artificial lighting, air temperature was optimized using a minicomputer with the hill-climbing method. The method was designated as the Speaking Plant Approach to environment control(SPA).After the author developed the first reported environmental control system in Japan, systems using microcomputers spread widely for greenhouse environmental control.Knowledge-based expert systems were tested for plant management. Also,a machine vision system was developed to detect critical moments for watering of muskmelon plants. The first feed-forward control method for greenhouses with a large heat mass was reported. Then space farming was tested in 1996 to assess gravity effects on plants.Energy-saving aspects such as solar sterilization, ground heat storage system, and storage using phase change material(PCM)have been reported.Defects of ordinary solarimeters were reported in 2008 along with an approach to estimate evapotranspiration in a greenhouse without the effect of so-called cosine law.Later, this technique was expanded to estimate photosynthesis of the plant canopy in a greenhouse using newly developed sensor units.Tadashi Takakura 2019International Journal of Agricultural and Biological Engineering2019,12,5:5
Advances in inducing adaptive immunity using cell-based cancer vaccines: clinical applications in pancreatic cancer显示文摘The incidence of pancreatic ductal adenocarcinoma(PDA) is on the rise, and the prognosis is extremely poor because PDA is highly aggressive and notoriously difficult to treat. Although gemcitabine- or 5-fluorouracil-based chemotherapy is typically offered as a standard of care, most patients do not survive longer than 1 year. Therefore, the development of alternative therapeutic approaches for patients with PDA is imperative. As PDA cells express numerous tumor-associated antigens that are suitable vaccine targets, one promising treatment approach is cancer vaccines. During the last few decades, cell-based cancer vaccines have offered encouraging results in preclinical studies. Cell-based cancer vaccines are mainly generated by presenting whole tumor cells or dendritic cells to cells of the immune system. In particular, several clinical trials have explored cell-based cancer vaccines as a promising therapeutic approach for patients with PDA. Moreover, chemotherapy and cancer vaccines can synergize to result in increased efficacies in patients with PDA. In this review, we will discuss both the effect of cell-based cancer vaccines and advances in terms of future strategies of cancer vaccines for the treatment of PDA patients.Mikio Kajihara Kazuki Takakura Tomoya Kanai Zensho Ito Yoshihiro Matsumoto Shigetaka Shimodaira Masato Okamoto Toshifumi Ohkusa Shigeo Koido 2016World Journal of Gastroenterology2016,22,18:4
Direct therapeutic intervention for advanced pancreatic cancer显示文摘Currently, chemotherapy is an accredited, standard treatment for unresectable, advanced pancreatic cancer(PC). However,it has been still showed treatmentresistance and followed dismal prognosis in many cases. Therefore, some sort of new, additional treatments are needed for the better therapeutic results for advanced PC. According to the previous reports,it is obvious that interventional endoscopic ultrasonography(EUS) is a well-established,helpful and low-risky procedure in general. As the additional treatments of the conventional therapy for advanced PC,many therapeutic strategies, such as immunotherapies, molecular biological therapies, physiochemical therapies, radioactive therapies, using si RNA,using autophagy have been developing in recent years. Moreover, the efficacy of the other potential therapeutic targets for PC using EUS-fine needle injection, for example,intra-tumoral chemotherapeutic agents(paclitaxel, irinotecan),several ablative energies(radiofrequency ablation and cryothermal treatment, neodymiumdoped yttrium aluminum garnet laser,high-intensity focused ultrasound),etc.,has already been showed in animal models. Delivering these promising treatments reliably inside tumor, interventional EUS may probably be indispensable existence for the treatment of locally advanced PC in near future.Kazuki Takakura Shigeo Koido 2015World Journal of Clinical Oncology2015,6,6:4
Influence of texture feature size on spherical silicon solar cells显示文摘The effects of surface texturing on spherical silicon solar cells were investigated. Surface texturing for spherical Si solar cells was prepared by immersing p-type spherical Si crystals in KOH solution with stirring. Two kinds of texture feature sizes (1 and 5 μm pyramids) were prepared by changing stirring speed. After fabrication through our baseline processes, these cells were evaluated by solar cell performance and external quantum efficiency. The cell with 1 and 5 μm pyramids shows the short circuit current density (Jsc) value of 31.9 and 33.2 mA·cm-2, which is 9% and 13% relative increase compared to the cell without texturing. Furthermore, the cell with 5 μm pyramids has a higher open-circuit voltage (0.589 V) than the cell with 1 μm pyramids (0.577 V). As a result, the conversion efficiency was improved from 11.4% for the cell without texturing to 12.1% for the cell with 5 μm pyramids.HAYASHI Shota MINEMOTO Takashi TAKAKURA Hideyuki HAMAKAWA Yoshihiro 2006Rare Metals2006,25,z1:3

Dendritic cell-based cancer immunotherapy for colorectal cancer显示文摘Colorectal cancer(CRC) is one of the most common cancers and a leading cause of cancer-related mortality worldwide. Although systemic therapy is the standard care for patients with recurrent or metastatic CRC, the prognosis is extremely poor. The optimal sequence of therapy remains unknown. Therefore, alternative strategies, such as immunotherapy, are needed for patients with advanced CRC. This review summarizes evidence from dendritic cell-based cancer immunotherapy strategies that are currently in clinical trials. In addition, we discuss the possibility of antitumor immune responses through immunoinhibitory PD-1/PD-L1 pathway blockade in CRC patients.Mikio Kajihara Kazuki Takakura Tomoya Kanai Zensho Ito Keisuke Saito Shinichiro Takami Shigetaka Shimodaira Masato Okamoto Toshifumi Ohkusa Shigeo Koido 2016World Journal of Gastroenterology2016,22,17:3
Production of corticotropin-releasing factor and urocortin from human monocyte-derived dendritic cells is stimulated by commensal bacteria in intestine显示文摘AIM:To examine whether commensal bacteria are a contributing cause of stress-related mucosal inflammation.METHODS:Human peripheral blood monocyte-deriveddendritic cells(MoDCs)were stimulated by commensal bacterial strains, including Escherichia coli,Clostridium clostridioforme,Bacteroides vulgatus(B.vulgatus),Fusobacterium varium(F.varium),and Lactobacillus delbrueckii subsp.bulgaricus.After incubation,corticotropin-releasing factor(CRF)and urocortin 1(UCN1)mRNA in the cells was examined by real-time reverse transcription polymerase chain reaction.Supernatants from the cells were tested for CRF and UCN1 using an enzyme-linked immunosorbent assay.RESULTS:Both CRF and UCN1 were significantly augmented by B.vulgatus and F.varium at both the mRNA and protein levels. In particular,B.vulgatus stimulated human MoDCs,resulting in extremely high levels of CRF and UCN1.CONCLUSION:Stimulation of MoDCs by B.vulgatus and F.varium may be associated with CRF/UCN1-related intestinal disorders, such as irritable bowel syndrome and inflammatory bowel disease.Shigeo Koido Toshifumi Ohkusa Shin Kan Kazuki Takakura Keisuke Saito Hideo Komita Zensho Ito Hiroko Kobayashi Shinichiro Takami Kan Uchiyama Hiroshi Arakawa Masaki Ito Masato Okamoto Mikio Kajihara Sadamu Homma Hisao Tajiri 2014World Journal of Gastroenterology2014,20,39:2

Mouse models for investigating the underlying mechanisms of nonalcoholic steatohepatitis-derived hepatocellular carcinoma显示文摘As the incidence of hepatocellular carcinoma(HCC) caused by infection with the hepatotropic viruses hepatitis B and hepatitis C decreases, greater attention has become focused on HCC caused by nonalcoholic steatohepatitis(NASH), an advanced form of nonalcoholic fatty liver disease which has shown increasing prevalence in correspondence with the overall increase in metabolic syndrome over the recent decades. Several clinical population studies have shown a positive relationship between NASH and HCC, while also providing initial insights into the underlying mechanisms of HCC development from NASH. Research into the pathological progression of NASH to HCC has advanced by use of several beneficial rodent models. In this review, we summarize the established mouse models for preclinical research of NASH-associated HCC and discuss the underlying hepatic mechanisms of NASH-related tumorigenesis identified to date that could lead to new targets for treatment and prevention.Kazuki Takakura Tsunekazu Oikawa Yoichi Tomita Yusuke Mizuno Masanori Nakano Chisato Saeki Yuichi Torisu Masayuki Saruta 2018World Journal of Gastroenterology2018,24,18:2
Small hepatocellular carcinoma with ring calcification:A case report and literature review显示文摘Ring calcification in untreated hepatocellular carcinoma(HCC) is extremely rare, with only 3 previously reported cases in the English-language literature.A case of HCC with ring calcification was reported in this paper.Additionally,3 previously reported cases of HCC with ring calcification were reviewed. In 3 of these 4 cases(including our case),surgery was performed.Although the size of the ring-calcified lesion ranged from 3.0-3.7 cm in previously reported cases, the size was only 1 cm in ours. The differentiation of the tumor was moderate in the 2 previously reported cases in the histological findings and poor in ours.In spite of their poor differentiation for their sizes, these tumors showed no early enhancement in dynamic computed tomography. All calcified tumors showed a thick fibrous capsule and extensive necrosis histologically.Ring calcification was considered to result from a circulatory disturbance caused by the imbalance between the less abundant arterial blood flow and high inner pressure induced by either the thick fibrous capsule or vigorous proliferation due to the poor differentiation.Ring calcification in untreated HCC may suggest a lower differentiation of the tumor.Even if its size is small, hepatic resection should be performed for any tumor with ring calcification because poor differentiation is considered to be one of the risk factors for recurrence after local ablation therapy, including radio frequency ablation.Takashi Murakami Daisuke Morioka Hideki Takakura Yasuhiko Miura Shinji Togo 2013World Journal of Gastroenterology2013,19,1:2
