| 2 | Suppression of murine breast cancer metastasis by selective inhibition of CXCR4 by synthetic polypeptide derived from viral macrophage inflammatory protein Ⅱ显示文摘Stromal cell-derived factor-1 and its receptor CXC chemokine receptor-4(CXCR4)have been implicated in breast cancer metastasis.A significant association between HER2 and CXCR4 expression has been observed in human breast tumor tissues,and overexpression of CXCR4 is essential for HER2-mediated tumor metastasis.Moreover,CXCR4 expression is low in normal breast tissues and high in malignant tumors,suggesting that a blockade of CXCR4 may limit tumor metastasis.The present study investigated the action of a synthetic antagonist 21-mer peptide derived from viral macrophage inflammatory protein Ⅱ against CXCR4(NT21MP)in inhibiting metastasis in vitro and in vivo.The results showed that chemotaxis of SKBR3 cells toward SDF-1αwas reduced by NT21MP in a dose-dependent manner(P<0.05).NT21MP inhibited tumor growth at 500μg/kg and in combination with Herceptin,the anti-HER2 antibody.The in vivo metastatic assay showed that NT21MP significantly inhibited pulmonary metastasis,and the number of metastatic tumor nodes on the surface of the lung was greatly decreased.Compared with the saline-treated control group,PCNA expression was dose-dependently decreased by NT21MP,the percentage of apoptotic cells was increased,and CXCR4 mRNA and protein expression were downregulated.In conclusion,NT21MP inhibits cellular proliferation,promotes apoptosis by downregulating CXCR4 expression,and suppresses the progression of primary and metastatic tumors.CXCR4 may be a useful therapeutic target for breast cancer,and NT21MP may serve as a potential target drug for the treatment of breast cancer metastasis. | YANG QingLin DING YongXing CHEN ChangJie TANG Jie ZHANG Ju YANG ZhiFeng | 2010 | Chinese Science Bulletin2010,55,20: | 6 |
| 3 | Number of Species and Geographical Distribution of Diospyros L.(Ebenaceae) in China显示文摘Diospyros L. is the largest, most widely distributed, and economically important genus of the family Ebenaceae. However, various researchers disagree on the number of species and geographical distribution of Diospyros in China. For the purpose of providing a scientific basis for the effective protection and rational use of Diospyros, the present study involved a comprehensive review of the literature, electronic databases, and herbaria as well as field investigations. The results showed as the following.(1) While 174 scientific names have been published for various taxa of Diospyros in China, by screening and excluding synonyms, 65 species names were considered acceptable; these account for about 12%of the total species of Diospyros globally.(2) A review of herbarium specimens documented that Diospyros occurs in 28 administrative regions of China, more than the literature records. Species richness decreases from south to north in China while Guangxi, Yunnan, Guangdong, and Hainan have the greatest number of species.(3) Among the 65 relatively reliable Diospyros species, 45 species are endemic to China and 18 species are stenochoric. Some species may have become extinct as a result of environmental degradation and the status of some germplasm remain unresolved. In brief, Chinese Diospyros includes abundant, China-specific, and widely distributed features. | 10 | Expression of second mitochondria-derived activator of caspases, X-linked inhibitor of apoptosis protein, and caspase-3 in pituitary adenomas显示文摘Studies concerning correlations between pituitary adenomas and cell apoptosis have mainly focused on upstream apoptosis signaling, but seldom on downstream mediators. In the present study, second mitochondria-derived activator of caspases (Smac), X-linked inhibitor of apoptosis protein (XIAP), and caspase-3 protein were qualitatively analyzed using immunohistochemistry, and quantified by western blot. Smac, XIAP, and caspase-3 mRNA expressions were detected by reverse transcription-PCR. Results showed that XIAP protein and mRNA expressions were greater in the invasive pituitary adenoma group compared with the noninvasive pituitary adenoma group. However, Smac and caspase-3 protein and mRNA expressions were lower in the invasive pituitary adenoma group compared with the noninvasive pituitary adenoma group. In the invasive pituitary adenomas, Smac expression was positively correlated with caspase-3 protein and mRNA expression (Protein: r = 0.55, P < 0.01; mRNA: r = 0.50, P < 0.01). Smac and caspase-3 expressions were negatively correlated with XIAP protein and mRNA expression (Protein: r = -0.56, -0.64, P < 0.01; mRNA: r = -0.69, -0.67, P < 0.01). However, no significant differences in correlation among Smac, XIAP, and caspase-3 were detectable in noninvasive pituitary adenomas. These data indicated that high expression of XIAP and low expression of Smac and caspase-3 suppressed cell apoptosis and led to enhanced invasiveness of pituitary adenomas. | 11 | The K Domain Mediates Homologous and Heterologous Interactions Between FLC and SVP Proteins of Brassica juncea显示文摘The transcription factors FLOWERING LOCUS C(FLC) and SHORT VEGETATIVE PHASE(SVP) can interact to form homologous and heterologous protein complexes that regulate flowering time in Brassica juncea Coss.(Mustard). Previous studies showed that protein interactions were mediated by the K domain,which contains the subdomains K1,K2 and K3. However,it remains unknown how the subdomains mediate the interactions between FLC and SVP. In the present study,we constructed several mutants of subdomains K1–K3 and investigated the mechanisms involved in the heterologous interaction of BjFLC/BjSVP and in the homologous interaction of BjFLC/Bj FLC or BjSVP/BjSVP. Yeast two-hybrid and β-galactosidase activity assays showed that the 19 amino acids of the K1 subdomain in BjSVP and the 17 amino acids of the K1 subdomain in BjFLC were functional subdomains that interact with each other to mediate hetero-dimerization. The heterologous interaction was enhanced by the K2 subdomain of BjSVP protein,but weakened by its interhelical domain L2. The heterologous interaction was also enhanced by the K2 subdomain of BjFLC protein,but weakened by its K3 subdomain. The homologous interaction of BjSVP was mediated by the full K-domain. However,the homologous interaction of BjFLC was regulated only by its K1 and weakened by its K2 and K3 subdomains. | 13 | Symbology of Feynman integrals from twistor geometries显示文摘We study the symbology of planar Feynman integrals in dimensional regularization by considering geometric configurations in momentum twistor space corresponding to their leading singularities(LS). Cutting propagators in momentum twistor space amounts to intersecting lines associated with loop and external dual momenta, including the special line associated with the point at infinity, which breaks dual conformal symmetry. We show that cross-ratios of intersection points on these lines, especially those on the infinity line, naturally produce symbol letters for Feynman integrals in D = 4-2∈, which include and generalize their LS. At one loop, we obtain all symbol letters using intersection points from quadruple cuts for integrals up to pentagon kinematics with two massive corners, which agree perfectly with canonical differential equation(CDE) results. We then obtain all two-loop letters, for up to four-mass box and one-mass pentagon kinematics, by considering more intersections arising from two-loop cuts.
