| 1 | Phase 1 clinical trial demonstrated that MUC1 positive metastatic seminal vesicle cancer can be effectively eradicated by modified Anti-MUC1 chimeric antigen receptor transduced T cells显示文摘Recent progress in chimeric antigen receptor-modified T-cell(CAR-T cell) technology in cancer therapy is extremely promising, especially in the treatment of patients with B-cell acute lymphoblastic leukemia. In contrast, due to the hostile immunosuppressive microenvironment of a solid tumor, CAR T-cell accessibility and survival continue to pose a considerable challenge, which leads to their limited therapeutic efficacy. In this study, we constructed two anti-MUC1 CAR-T cell lines. One set of CAR-T cells contained SM3 single chain variable fragment(sc Fv) sequence specifically targeting the MUC1 antigen and co-expressing interleukin(IL) 12(named SM3-CAR). The other CAR-T cell line carried the SM3 sc Fv sequence modified to improve its binding to MUC1 antigen(named p SM3-CAR) but did not co-express IL-12. When those two types of CAR-T cells were injected intratumorally into two independent metastatic lesions of the same MUC1+ seminal vesicle cancer patient as part of an interventional treatment strategy, the initial results indicated no side-effects of the MUC1 targeting CAR-T cell approach, and patient serum cytokines responses were positive. | 2 | Optical detection of acetylcholine esterase based on CdTe quantum dots显示文摘In this study, we have constructed a simple, sensitive and rapid biosensor for detection of acetylcholine esterase (AChE) based on CdTe quantum dots (QDs). The detection limit of AChE by one-step enzyme reaction based on the thiolglycolic acid (TGA) stabilized QDs (TGA-QDs) was 10 U/L and the linear range was 10-100 and 100-1200 U/L, respectively. The detection limit of AChE by two-step enzyme reaction based on the 3-mercaptopropionic acid (MGA) stabilized QDs (MGA-QDs) was found to be 20 U/L and the linear range was 100-2500 U/L. The experimental conditions of biosensors were optimized, and the detection mechanism was studied. We also detected AChE in serum samples based on TGA-QDs or MGA-QDs. The linear range was 10-140 and 50-1000 U/L, respectively. | 3 | Sensitive detection of dopamine and quinone drugs based on the quenching of the fluorescence of carbon dots显示文摘In this work,we demonstrated that the quinone structure can quench the fluorescence of the carbon dots(CDs).The sensitive determination of dopamine(DA) was studied primarily based on this principle.DA would be transformed into DA quinone under alkaline conditions,which resulted in fluorescence quenching of the CDs.A good linear range from 5 nmol/L to 0.4 mmol/L was obtained and the detection limit was 1 nmol/L.Moreover,the quenching effect of quinone structure on the fluorescence of CDs was confirmed by Fourier transform infrared spectra,time-correlated single-photon counting and X-ray photoelectron spectroscopy.Remarkably,CDs were firstly applied to detect the quinone drugs quantitatively which contained typical quinone structure based on the quenching mechanism.More than this,the sensing platform was demonstrated to provide credible selectivity and satisfactory stability in human serum solution with good liner range.Hence,our practical application and mechanism have showed great potential for diagnostic purposes. | Xiangling Ren Jiejie Ge Xianwei Meng Xiaozhong Qiu Jun Ren Fangqiong Tang | 2016 | Science Bulletin2016,61,20: | 3 |

| 8 | Safety Performance of a Precast Concrete Barrier: Numerical Study显示文摘The numerical simulation for a new type of precast concrete barrier for viaducts is carried out systematically.To obtain an accurate representation of the damage state of the concrete barrier under the impact of a vehicle,a stochastic damage-plasticity model of the concrete is adopted in the finite element model.Meanwhile,a simplified mathematical model of the impact between vehicles and the concrete barrier was established and the input energy was converted to the impact load to facilitate the investigation of the safety performance of the concrete barriers.On this basis,a refined finite element(FE)model of a precast concrete barrier was developed.The impact locations,impact load,boundary constraints,and reinforcement types were used as variables and the dynamic response of the precast concrete barrier was systematically analyzed under 36 working conditions.The simulated results indicated that the damage state of the concrete barrier and the stress of the reinforcing bars were accurately and quantitatively reflected.According to the computed results,some suggestions were put forward for selecting the appropriate reinforcement type of the precast concrete barrier and the connection between the superstructure and foundation. | Zishen Li Xiangling Gao Zicheng Tang | 2020 | Computer Modeling in Engineering & Sciences2020,,6: | 1 |
