1Bile acid nuclear receptor FXR and digestive system diseases显示文摘Bile acids(BAs) are not only digestive surfactants but also important cell signaling molecules,which stimulate several signaling pathways to regulate some important biological processes. The bileacid-activated nuclear receptor, farnesoid X receptor(FXR), plays a pivotal role in regulating bile acid,lipid and glucose homeostasis as well as in regulating the inflammatory responses, barrier function and prevention of bacterial translocation in the intestinal tract. As expected, FXR is involved in the pathophysiology of a wide range of diseases of gastrointestinal tract, including inflammatory bowel disease, colorectal cancer and type 2 diabetes. In this review, we discuss current knowledge of the roles of FXR in physiology of the digestive system and the related diseases. 2Notoginsenoside Ft1 acts as a TGR5 agonist but FXR antagonist to alleviate high fat diet-induced obesity and insulin resistance in mice显示文摘Obesity and its associated complications are highly related to a current public health crisis around the world.A growing body of evidence has indicated that G-protein coupled bile acid(BA) receptor TGR5(also known as Gpbar-1) is a potential drug target to treat obesity and associated metabolic disorders.We have identified notoginsenoside Ftl(Ftl) from Panax notoginseng as an agonist of TGR5 in vitro.However,the pharmacological effects of Ftl on diet-induced obese(DIO) mice and the underlying mechanisms are still elusive.Here we show that Ftl(100 mg/100 diet) increased adipose lipolysis,promoted fat browning in inguinal adipose tissue and induced glucagon-like peptide-1(GLP-1) secretion in the ileum of wild type but not Tgr5^(-/-) obese mice.In addition,Ftl elevated serum free and taurineconjugated bile acids(BAs) by antagonizing Fxr transcriptional activities in the ileum to activate Tgr5 in the adipose tissues.The metabolic benefits of Ftl were abolished in Cyp27 al^(-/-) mice which have much lower BA levels.These results identify Ftl as a single compound with opposite activities on two key BA receptors to alleviate high fat diet-induced obesity and insulin resistance in mice.Lili Ding Qiaoling Yang Eryun Zhang Yangmeng Wang Siming Sun Yingbo Yang Tong Tian Zhengcai Ju Linshan Jiang Xunjiang Wang Zhengtao Wang Wendong Huang Li Yang 2021Acta Pharmaceutica Sinica B2021,11,6:10
3Single-trial decoding of imagined grip force parameters involving the right or left hand based on movement-related cortical potentials显示文摘Time–domain feature representation for imagined grip force movement-related cortical potentials(MRCP)of the right or left hand and the decoding of imagined grip force parameters based on electroencephalogram(EEG)activity recorded during a single trial were here investigated.EEG signals were acquired from eleven healthy subjects during four different imagined tasks performed with the right or left hand.Subjects were instructed to execute imagined grip movement at two different levels of force.Each task was executed 60 times in random order.The imagined grip force MRCP of the right or left hand was analyzed by superposition and averaging technology,a single-trial extraction method,analysis of variance(ANOVA),and multiple comparisons.Significantly different features were observed among different imagined grip force tasks.These differences were used to decode imagined grip force parameters using Fisher linear discrimination analysis based on kernel function(k-FLDA)and support vector machine(SVM).Under the proposed experimental paradigm,the study showed that MRCP may characterize the dynamic processing that takes place in the brain during the planning,execution,and precision of a given imagined grip force task.This means that features related to MRCP can be used to decode imagined grip force parameters based on EEG.ANOVA and multiple comparisons of time–domain features for MRCP showed that movement-monitoring potentials(MMP)and specific interval(0–150 ms)average potentials to be significantly different among 4 different imagined grip force tasks.The minimum peak negativity differed significantly between high and low amplitude grip force.Identification of the 4different imagined grip force tasks based on MMP was performed using k-FLDA and SVM,and the average misclassification rates of 27%±5%and 24%±4%across 11 subjects were achieved respectively.The minimum misclassification rate was 15%,and the average minimum misclassification rate across 11 subjects was24%±4.5%.This investigation indicates that imagined grip force MRCP may encode imagined grip force parameters.Single-trial decoding of imagined grip force parameters based on MRCP may be feasible.The study may provide some additional and fine control instructions for brain–computer interfaces.Yunfa Fu Baolei Xu Yongcheng Li Yuechao Wang Zhengtao Yu Hongyi Li 2014Chinese Science Bulletin2014,59,16:8
4Preliminary analysis of species sensitivity distribution based on gene expression effect显示文摘Species sensitivity analysis is one of the major techniques applied to derive water quality criteria. Presently, the toxicity data used for development of water quality criteria are mainly in the biological individual level. With the increase of ecotoxicogenomics toxicity data, it is worth studying whether the gene expression effect data can be used to derive water quality criteria. Taking cadmium, copper and zinc as examples, we analyzed the toxic effects of the three heavy metals by constructing the species sensitivity distribution curves on the basis of extensive toxicity data. The results showed that the rank of species sensitivity for the acute, chronic and gene expression effect toxicity data of cadmium is 'chronic>gene>acute'. Although the gene expression effect data of copper and zinc are insufficient, the trend of data sensitivity of zinc is similar to cadmium. However, the trend of species sensitivity of copper is different from that of cadmium and zinc with higher sensitivity of gene expression data. It suggested that though the existing data of gene expression effects are not sufficient enough, they have the potential to be used in the development of chronic water quality criteria. 6Regional gravity field modeling based on rectangular harmonic analysis显示文摘Regional gravity field modeling with high-precision and high-resolution is one of the most important scientific objectives in geodesy,and can provide fundamental information for geophysics,geodynamics,seismology,and mineral exploration.Rectangular harmonic analysis(RHA)is proposed for regional gravity field modeling in this paper.By solving the Laplace’s equation of gravitational potential in local Cartesian coordinate system,the rectangular harmonic expansions of disturbing potential,gravity anomaly,gravity disturbance,geoid undulation and deflection of the vertical are derived,and so are the formula for signal degree variance and error degree variance of the rectangular harmonic coefficients(RHC).We also present the mathematical model and detailed algorithm for the solution of RHC using RHA from gravity observations.In order to reduce the edge effects caused by periodic continuation in RHA,we propose the strategy of extending the size of computation domain.The RHA-based modeling method is validated by conducting numerical experiments based on simulated ground and airborne gravity data that are generated from geopotential model EGM2008 and contaminated by Gauss white noise with standard deviation of 2 mGal.The accuracy of the 2.5′×2.5′geoid undulations computed from ground and airborne gravity data is 1 and 1.4cm,respectively.The standard error of the gravity disturbances that downward continued from the flight height of 4 km to the geoid is only 3.1 mGal.Numerical results confirm that RHA is able to provide a reliable and accurate regional gravity field model,which may be a new option for the representation of the fine structure of regional gravity field.JIANG Tao LI JianCheng DANG YaMin ZHANG ChuanYin WANG ZhengTao KE BaoGui 2014Science China Earth Sciences2014,57,7:4
7Mutual dependency between lncRNA LETN and protein NPM1 in controlling the nucleolar structure and functions sustaining cell proliferation显示文摘Fundamental processes such as ribosomal RNA synthesis and chromatin remodeling take place in the nucleolus,which is hyperactive in fast-proliferating cells.The sophisticated regulatory mechanism underlying the dynamic nucleolar structure and functions is yet to be fully explored.The present study uncovers the mutual functional dependency between a previously uncharacterized human long non-coding RNA,which we renamed LETN,and a key nucleolar protein,NPM1.Specifically,being upregulated in multiple types of cancer,LETN resides in the nucleolus via direct binding with NPM1.LETN plays a critical role in facilitating the formation of NPM1 pentamers,which are essential building blocks of the nucleolar granular component and control the nucleolar functions.Repression of LETN or NPM1 led to similar and profound changes of the nucleolar morphology and arrest of the nucleolar functions,which led to proliferation inhibition of human cancer cells and neural progenitor cells.Interestingly,this inter-dependency between LETN and NPM1 is associated with the evolutionarily new variations of NPM1 and the coincidental emergence of LETN in higher primates.We propose that this human-specific protein–lncRNA axis renders an additional yet critical layer of regulation with high physiological relevance in both cancerous and normal developmental processes that require hyperactive nucleoli.Xianteng Wang Xiaolin Hu Wanlu Song Hui Xu Zhengtao Xiao Rongyao Huang Qingran Bai Fan Zhang Yongzhen Chen Yu Liu Jianhuo Fang Xin Li Qin Shen Haitao Zhao Xuerui Yang 2021Cell Research2021,31,6:3
8Astragaloside Ⅳ suppresses post-ischemic natural killer cell infiltration and activation in the brain:involvement of histone deacetylase inhibition显示文摘Natural killer(NK)cells,a type of cytotoxic lymphocytes,can infiltrate into ischemic brain and exacerbate neuronal cell death.Astragaloside Ⅳ(ASIV)is the major bioactive ingredient of Astragalus membranaceus,a Chinese herbal medicine,and possesses potent immunomodulatory and neuroprotective properties.This study investigated the effects of ASIV on post-ischemic brain infiltration and activation of NK cells.ASIV reduced brain infarction and alleviated functional deficits in MCAO rats,and these beneficial effects persisted for at least 7 days.Abundant NK cells infiltrated into the ischemic hemisphere on day 1 after brain ischemia,and this infiltration was suppressed by ASIV.Strikingly,ASIV reversed NK cell deficiency in the spleen and blood after brain ischemia.ASIV inhibited astrocyte-derived CCL2 upregulation and reduced CCR2+NK cell levels in the ischemic brain.Meanwhile,ASIV attenuated NK cell activating receptor NKG2D levels and reduced interferon-γproduction.ASIV restored acetylation of histone H3 and the p65 subunit of nuclear factor-κB in the ischemic brain,suggesting inhibition of histone deacetylase(HDAC).Simultaneously,ASIV prevented p65 nuclear translocation.The effects of ASIV on reducing CCL2 production,restoring acetylated p65 levels and preventing p65 nuclear translocation were mimicked by valproate,an HDAC inhibitor,in astrocytes subjected to oxygen-glucose deprivation.Our findings suggest that ASIV inhibits post-ischemic NK cell brain infiltration and activation and reverses NK cell deficiency in the periphery,which together contribute to the beneficial effects of ASIV against brain ischemia.Furthermore,ASIV’s effects on suppressing NK cell brain infiltration and activation may involve HDAC inhibition.Baokai Dou Shichun Li Luyao Wei Lixin Wang Shiguo Zhu Zhengtao Wang Zunji Ke Kaixian Chen Zhifei Wang 2021Frontiers of Medicine2021,15,1:3

Relating to the constituents of theseherbs, triterpenes, phytosterols, furaldehyde, sesquitesand some phenolic glycosides have been reported, butnone of these compounds seems to be responsible for thebiological activities of this drug. As regards the quality evaluation of Radix Codonopsis, a HPLC analysis of atractylenolide Ⅲ has been described, but this sesquiterpene lactone exists in a very low content and is difficultfor conventional analysis. 10A NEW PUBLIC-KEY ENCRYPTION SCHEME BASED ON LUCAS SEQUENCE显示文摘Investigated the properties of LUCas sequence(LUC), the paper proposed a new variant of (probabilistic) public-key encryption scheme. Security analysis of the proposed encryption schemes shows that its one-wayness is equivalent to partial LUC discrete logarithm problem in ZN, and for the proposed probabilistic encryption scheme, its semantic security is equivalent to decisional LUC Diffie-Hellman problem in ZN. 11Clear mortality gap caused by graft macrosteatosis in Chinese patients after cadaveric liver transplantation显示文摘Background:Liver transplantation(LT)is one of the most effective surgical treatment for patients with end-stage liver disease.Steatosis is a contributor for inferior graft quality.But its impact and safety on transplantation was less assessed in Chinese patients.Methods:Graft steatosis and related information involved in recipients,donors and surgical procedures were retrospectively collected from 239 patients.Results:Donor macrosteatosis(MaS)caused about 2.14 and 2.80 folds of increment on patient and graft mortality.Dose-response analysis revealed prominent risk of grafts on overall patient/organ mortality when MaS content exceeded 10%(P<0.05).Noteworthy,deaths were only observed in MaS group when concurrent with extremely higher post-transplant alanine aminotransferase(ALT,64%).However,microsteatosis(MiS)grafts didn’t affect outcomes after LT.In a cohort of Chinese patients,MaS had comprehensive effects on post-transplant outcomes with relatively lower safety threshold at 10%.Mortality gap caused by MaS grafts was observed in patients with severer ischemia reperfusion injury.Conclusions:Our study revealled the graft MaS affected the post-transplant outcomes in lower risk cutoff in Chinese patients.Further study is worthy to validate these results and investigate inner mechanism under the phenomenon.Zhengtao Liu Wenchao Wang Li Zhuang Jingfeng Liu Shuping Que Dan Zhu Linfang Dong Jian Yu Lin Zhou Shusen Zheng 2020Hepatobiliary Surgery and Nutrition2020,9,6:2

13Human health ambient water quality criteria for 13 heavy metals and health risk assessment in Taihu Lake显示文摘Heavy metals are widely concerning because of their toxicity,persistence,non-degradation and bioaccumulation ability.Iluman health ambient water quality criteria(AWQC)are specific levels of chemicals that can occur in water without harming human health.At present,most countries do not consider the effects of aquatic vegetables in deriving human health AWQC.Therefore,the intake of aquatic vegetables(Brasenia schreberi)was added to the derivation of human health AWQC and a health risk assessment for 13 heavy metals in Taihu Lake.The human health AWQC(consumption of water,fish and aquatic vegetables)values of 13 heavy metals ranged from 0.04(Cd)to 710.87μg/L(Sn),and the intake of B.schreberi had a very significant effect on the human health AWQC for Cu,with a more than 62-fold difference.The hazard quotients of As(2.8),Cd(1.6),Cr(1.4)and Cu(4.86)were higher than the safe level(HQ=1),indicating that As,Cd,Cr and Cu in Taihu Lake posed a significant health risk.Sensitivity analysis showed that the contribution rate of B.schreberi intake to the human health risk from Cu was 91.6%,and all results indicated that the risk of Cu in B.schreberi to human health should be of particular concern.This study adds the consideration of aquatic vegetable consumption to the traditional method of human health AWQC derivation and risk assessments for the first time,and this approach can promote the development of risk assessments and water quality criteria.Liang Cui Ji Li Xiangyun Gao Biao Tian Jiawen Zhang Xiaonan Wang Zhengtao Liu 2022Frontiers of Environmental Science & Engineering2022,16,4:2

15Formation control with disturbance rejection for a class of Lipschitz nonlinear systems显示文摘In this paper, we consider the leader-follower formation control problem for general multi-agent systems with Lipschitz nonlinearity and unknown disturbances. To deal with the disturbances, a disturbance observer-based control strategy is developed for each follower. Then, a time-varying formation protocol is proposed based on the relative state of the neighbouring agents and sufficient conditions for global stability of the formation control are identified using Lyapunov method in the time domain. The proposed strategy and analysis guarantee that all signals in the closed-loop dynamics are uniformly ultimately bounded and the formation tracking errors converge to an arbitrarily small residual set. 17Detecting micro-blog user interest communities through the integration of explicit user relationship and implicit topic relations显示文摘In order to effectively utilize the explicit user relationship and implicit topic relations for the detection of micro-blog user interest communities, a micro-blog user interest community(MUIC) detection approach is proposed. First, through the analysis of the follow relationship between users, we have defined three types of such relationships to construct the user follow-ship network. Second, taking the semantic correlation between user tags into account, we construct the user interest feature vectors based on the concept of feature mapping to build a user tag based interest relationship network. Third, user behaviors, such as reposting, commenting, replying, and receiving comments from others, are able to provide certain guidance for the extraction of micro-blog topics. Hence, we propose to integrate the four mentioned user behaviors that are considered to provide guidance information for the traditional latent Dirichlet allocation(LDA) model. Thereby, in addition to the construction of a topic-based interest relationship network, a guided topic model can be built to extract the topics in which the user is interested. Finally, with the integration of the afore-mentioned three types of relationship network, a micro-blog user interest relationship network can be created. Meanwhile, we propose a MUIC detection algorithm based on the contribution of the neighboring nodes. 19Isotoosendanin exerts inhibition on triple-negative breast cancer through abrogating TGF-β-induced epithelial—mesenchymal transition via directly targeting TGFβR1显示文摘As the most aggressive breast cancer,triple-negative breast cancer(TNBC) is still incurable and very prone to metastasis.The transform growth factor β(TGF-β)-induced epithelial—mesenchymal transition(EMT) is crucially involved in the growth and metastasis of TNBC.This study reported that a natural compound isotoosendanin(ITSN) reduced TNBC metastasis by inhibiting TGF-β-induced EMT and the formation of invadopodia.ITSN can directly interact with TGF-β receptor type-1(TGFβR1) and abrogated the kinase activity of TGFβR1,thereby blocking the TGF-β-initiated downstream signaling pathway.Moreover,the ITSN-provided inhibition on metastasis obviously disappeared in TGFβR1-overexpressed TNBC cells in vitro as well as in mice bearing TNBC cells overexpressed TGFβR1.Furthermore,Lys232 and Asp351 residues in the kinase domain of TGFβR1 were found to be crucial for the interaction of ITSN with TGFβR1.Additionally,ITSN also improved the inhibitory efficacy of programmed cell death 1 ligand 1(PD-L1) antibody for TNBC in vivo via inhibiting the TGF-β-mediated EMT in the tumor microenvironment.Our findings not only highlight the key role of TGFβR1 in TNBC metastasis,but also provide a leading compound targeting TGFβR1 for the treatment of TNBC metastasis.Moreover,this study also points out a potential strategy for TNBC treatment by using the combined application of anti-PD-L1 with a TGFβR1 inhibitor.Jingnan Zhang Ze Zhang Zhenlin Huang Manlin Li Fan Yang Zeqi Wu Qian Guo Xiyu Mei Bin Lu Changhong Wang Zhengtao Wang Lili Ji 2023Acta Pharmaceutica Sinica B2023,13,7:1
