1Characterization and normalization factors of abiotic resource depletion for life cycle impact assessment in China显示文摘The availability of resources for economic activities differs between regions, and the importance of the resources is consequently observed to be different within regions compared to a global scale. With the current situation in Chinese mining industry and its statistic characteristics, the characterization procedures of abiotic resource in life cycle impact assessment (LCIA) have demonstrated certain limita-tions in the Chinese materials industry. The aim of this paper is to propose new characterization and normalization factors for abiotic resource depletion categories such as metals and non-renewable en- ergy resources in a Chinese context. The actual production of abiotic resources calculated by a modi- fied model is compared to the reserve base in line with the new national standard to determine char- acterization factors in equivalence units, with antimony as the reference mineral. The normalization factors are based on the total base reserves of the most important minerals in China. A case study on primary magnesium production using the Pidgeon process is used to compare LCIA results for abiotic resource categories that are between current LCIA factors and the new Chinese factors. 3Greenhouse gas emissions and reduction potential of primary aluminum production in China显示文摘A cradle-to-gate life cycle assessment was conducted in this paper to calculate the greenhouse gas (GHG) emissions, such as CO2, CH4, CF4 and C2F6 emissions, based on statistic data of Chinese aluminum industry of the year 2003. The results showed that the GHG emissions for 1 t primary aluminum production was 21.6 t CO2 equivalent which is 70% higher than that of worldwide average level of the year 2000. The main contributors of emission were the alumina refining and aluminum smelting process accounting for 72% and 22% in accumulative emission, respectively. According to the development and application of new process technologies for primary aluminum production and the ‘target of energy-saving and emissions-reducing’ of Chinese government, the reduction potential of the GHG emissions for alumina and aluminum production were estimated. 4Research and development of Chinese LCA database and LCA software显示文摘This paper detailed introduces the related work for building Chinese National Database, Sinocenter, of materials life cycle assessment (MLCA) and developing the environmental burden dataset of materials. The MLCA database was built in 2004, and the basic framework mainly includes LCA methodology, materials environmental dataset about energy consumption, resource input and environmental emissions. Nowadays, the database contains about fifty-thousand records of the main materials industries, such as cement, iron and steel, nonferrous metal, etc., and also includes the primary LCI data of fossil fuels and electricity grid in China. At the same time, the LCA method localization work is going on, for instance, calculated the resource characterization factors of 42 kinds of metal and 58 sorts of nonmetal, and also obtained some heavy metal impact factors in water. Based on the database, the iron and steel dataset has been developed with the data quality analysis, and some environmental burden data could be queried in our website, www.cnmlca.com, in the future. 6Grain size effect on wear resistance of WC-Co cemented carbides under different tribological conditions显示文摘The grain-size dependence of wear resistance of WC-Co cemented carbides(with mean WC grain sizes of 2.2μm,1.6μm,0.8μm and 0.4μm,respectively)was investigated under different tribological conditions.The results showed that the grain size had opposite effects on wear resistance of the cemented carbides in dry sliding wear and microabrasion tests.In the former condition,with decrease of WC grain size hence the increase of hardness,plastic deformation,fracture,fragmentation and oxidation were all mitigated,leading to a drastic decrease in the wear rate.In the latter condition,pull-out of WC grains after Co removal dominated the wear,so that the hardness of cemented carbide was not a core factor.As a result,the wear resistance of the cemented carbide generally showed a decreasing trend with decrease of the grain size,except for a slight increase in the ultrafine-grained cemented carbide.Single-pass scratching of the cemented carbides under various loads indicated the same failure mechanism as that in the sliding wear tests.Furthermore,the reasons for severe surface oxidation of the coarse-grained cemented carbides were disclosed.Haibin Wang Mark Gee Qingfan Qiu Hannah Zhang Xuemei Liu Hongbo Nie Xiaoyan Song Zuoren Nie 2019Journal of Materials Science & Technology2019,35,11:6
7Microstructure and mechanical properties of Al-Mg-Mn alloy with erbium显示文摘The effect of trace element Er on the microstructure and properties of Al-6Mg-0.7Mn-0.1Cr-0.15Ti alloy was investigated. The results show that the ultimate room-temperature and high-temperature strength sof the alloy have not increased obviously with the addition of erbium, but the uniform elongation of alloys decreases from 9% and 27.5% to 5.5% and 22.5%, respectively. The reason is that the addition of Er will first react with Ti, Cr to form Al20Ti2Er, Al8Cr4Er phases. The precipitation strengthening makes up the weakening of grain refinement strengthening. 8Overexpressed BRH1, a RING finger gene, alters rosette leaf shape in Arabidopsis thaliana显示文摘Leaves are the most important plant parts for photosynthesis and respiration. Many genes are involved in determining leaf shape;however, little is known about the effects of brassinosteroid (BR) signaling-pathway genes on the development of leaf shape. Here, the brassinosteroid-responsive RING-H2 (BRH1) gene, which is suppressed by 24-epi-brassinolide treatment, was isolated from Arabidopsis thaliana. The amino acid sequence contained a highly conserved RING finger domain. In a phylogenetic analysis,BRH1 clustered closely with GLYMA11G02470.1. The leaves of brh1 mutant plants were not much different to those of the wild-type, while transgenic plants with high BRH1 expression levels had rounder rosette leaves. Mutants of the BR synthesis pathway also had a similar round leaf phenotype, and greater BRH1 expression levels. Moreover, the related marker genes KNAT1,AtHB13 and ROT4, which are known to control leaf shape, altered transcriptional levels in both transgenic BRH1 and BR-synthesis mutant lines. Thus, BRH1 may be involved in the BR signaling pathway and regulate the growth and development of rosette leaves. 9Genome-wide identification and expression analysis of Gossypium RING-H2 finger E3 ligase genes revealed their roles in fiber development,and phytohormone and abiotic stress responses显示文摘Background: RING-H2 finger E3 ligase(RH2FE3) genes encode cysteine-rich proteins that mediate E3 ubiquitin ligase activity and degrade target substrates. The roles of these genes in plant responses to phytohormones and abiotic stresses are well documented in various species, but their roles in cotton fiber development are poorly understood. To date, genome-wide identification and expression analyses of Gossypium hirsutum RH2FE3 genes have not been reported.Methods: We performed computational identification, structural and phylogenetic analyses, chromosomal distribution analysis and estimated K_a/K_s values of G. hirsutum RH2FE3 genes. Orthologous and paralogous gene pairs were identified by all-versus-all BLASTP searches. We predicted cis-regulatory elements and analyzed microarray data sets to generate heatmaps at different development stages. Tissue-specific expression in cotton fiber, and hormonal and abiotic stress responses were determined by quantitative real time polymerase chain reaction(qRT-PCR) analysis.Results: We investigated 140 G. hirsutum, 80 G. arboreum, and 89 G. raimondi putative RH2FE3 genes and their evolutionary mechanisms and compared them with orthologs in Arabidopsis and rice. A domain-based analysis of the G.hirsutum RH2FE3 genes predicted conserved signature motifs and gene structures. Chromosomal localization showed the genes were distributed across al G. hirsutum chromosomes, and 60 duplication events(4 tandem and 56 segmental duplications) and 98 orthologs were detected. cis-elements were detected in the promoter regions of G. hirsutum RH2FE3 genes. Microarray data and qR T-PCR analyses showed that G. hirsutum RH2FE3 genes were strongly correlated with cotton fiber development. Additional y, almost al the identified genes were up-regulated in response to phytohormones(brassinolide, gibberel ic acid(GA), indole-3-acetic acid(IAA), and salicylic acid(SA)) and abiotic stresses(cold, heat,drought, and salt).Conclusions: The genome-wide identification, comprehensive analysis, and characterization of conserved domains and gene structures, as wel as phylogenetic analysis, cis-element prediction, and expression profile analysis of G. hirsutum RH2FE3 genes and their roles in cotton fiber development and responses to plant hormones and abiotic stresses are reported here for the first time. 10Advances in Thermionic Cathode of Tungsten and Molybdenum显示文摘Several kinds of tungsten thermonic cathodes have been introduced. As a promising alternative for thoriated tungsten, rare earth doped molybdenum cathode has been studied. Compared with the traditional thoriated tungsten, La- Mo cathode has higher emission current density at lower temperature, but it has poor emission stability. In order to im- prove the emission stability, systematical Study on the emission mechanism of La-Mo cathode has been carried out. The life of La-Mo cathode has been improved and has achieved 1400 h, which exceeds the minimum life for practical uses (1000 h). As another alternative for thoriated tungsten cathode, Y-Mo cathode has shown better performance. 11Genome-wide identification of Gossypium INDETERMINATE DOMAIN genes and their expression profiles in ovule development and abiotic stress responses显示文摘Background:INDETERMINATE DOMAIN(IDD)transcription factors form one of the largest and most conserved gene families in plant kingdom and play important roles in various processes of plant growth and development,such as flower induction in term of flowering control.Till date,systematic and functional analysis of IDD genes remained infancy in cotton.Results:In this study,we identified total of 162 IDD genes from eight different plant species including 65 IDD genes in Gossypium hirsutum.Phylogenetic analysis divided IDDs genes into seven well distinct groups.The gene structures and conserved motifs of GhIDD genes depicted highly conserved exon-intron and protein motif distribution patterns.Gene duplication analysis revealed that among 142 orthologous gene pairs,54 pairs have been derived by segmental duplication events and four pairs by tandem duplication events.Further,Ka/Ks values of most of orthologous/paralogous gene pairs were less than one suggested the purifying selection pressure during evolution.Spatiotemporal expression pattern by qRT-PCR revealed that most of the investigated GhIDD genes showed higher transcript levels in ovule of seven days post anthesis,and upregulated response under the treatments of multiple abiotic stresses.Conclusions:Evolutionary analysis revealed that IDD gene family was highly conserved in plant during the rapid phase of evolution.Whole genome duplication,segmental as well as tandem duplication significantly contributed to the expansion of IDD gene family in upland cotton.Some distinct genes evolved into special subfamily and indicated potential role in the allotetraploidy Gossypium hisutum evolution and development High transcript levels of GhIDD genes in ovules illustrated their potential roles in seed and fiber development Further,upregulated responses of GhIDD genes under the treatments of various abiotic stresses suggested them as important genetic regulators to improve stress resistance in cotton breeding.ALI Faiza QANMBER Ghulam LI Yonghui MA Shuya LU Lili YANG Zuoren WANG Zhi LI Fuguang 2019Journal of Cotton Research2019,2,1:5
12siRNA of ADAM17 gene induces apoptosis,proliferation inhibition and enhances the effects of genistein on HepG2 cells显示文摘Objective:To investigate the effects of siRNA of ADAM17 gene and genistein on apoptosis and the inhibition of proliferation in HepG2 cells in an attempt to seek an effective therapy for hepatocellular carinoma. Methods:Cells were divided into control groups and experimental groups and siRNA was used to silence the ADAM17 gene, alone and in combination with genistein. Cells were harvested at several time periods and assessed for proliferation and apoptosis. Proliferation was assayed by MTT at 24, 48, 72 and 96 hours following treatment and apoptosis was assessed by flow cytometric analysis at 48 hours. Results:In siRNA groups, proliferation of cells was significantly inhibited compared to the control groups at 24, 48 and 72 hours(P < 0.05), and apoptosis was significantly increased at 48 hours(P < 0.01); In genistein groups, proliferation was inhibited at 24, 48, 72 and 96 hours, and the apoptosis ratio was significantly increased at 48 hours(P < 0.01); while in the groups that received the combination of siRNA transfection and genistein treatment, there was a further significant decrease of proliferation and increase in apoptosis compared with either treatment alone. 13Prediction of VIGS efficiency by the Sfold program and its reliability analysis in Gossypium hirsutum显示文摘Genetic transformation in some plant species,including cotton(Gossypium hirsutum), is hampered by laborious and time-consuming processes and often unachievable. Virus-induced gene silencing(VIGS) by double-stranded RNAs can serve as a reverse-genetics tool to determine gene function. However, knockdown levels vary greatly when using a tobacco rattle virus-based vector that carries different c DNA fragments of a gene. How to choose the optional target fragment for high interference efficiency is very challenging. Addressing this challenge requires increasing the efficacy of small interference RNA(si RNA) in target fragment. Here, we describe a method to assess VIGS efficiency by comparing the following parameters of si RNA in target sequence: the disruption energy of the target(DGdisruption), the differential stability of si RNA duplex ends(DSSE), and the internal stability at positions 9–14 of the si RNA antisense strand(AIS), which are calculated by Sfold program(http://gffzz01a2382fbe924301h0wuokc0qv06q6w0c.ffgz.tsg.suse.edu.cn). We find that the si RNAs with low DGdisruption, high DSSE and high AIS have high activity and easily result in high VIGS efficiency by experimentally testing the actual knockdown levels of the four target genes, Gh PDS,Gh CLA1, Gh AOS1, and Gh CXE1 via choosing different target sequences for each gene.
