| 1 | Changes in the power spectrum of electrical signals in maize leaf induced by osmotic stress显示文摘To quantify the characteristics of the power spectrum of plant electrical signals, we defined the following concepts:spectral edge frequency (SEF), spectral center frequency (SCF), power index (PI) and power spectral entropy (PSE). These parameters were used to examine and quantify changes in the power spectrum of electrical signals in maize leaves under osmotic stress. In the absence of osmotic stress, the SEF of the electrical signal in maize leaves was approx. 0.2 Hz and the SCF was approx. 0.1 Hz. The electrical signal in maize leaves was mainly a slow wave signal with a frequency of 0-0.1 Hz. After 2 h osmotic stress, the SEF and SCF of the electrical signal increased to higher frequencies. The proportion of the fast wave frequency also increased to 0.1-0.2 Hz, resulting in a dramatic increase in PSE. We also found that the changes in PSE and SCF were significantly correlated during osmotic stress. We propose that the changes in the PSE and SCF in maize leaves can be used as a sensitive signal indicating water deficit in leaf cells under osmotic stress. | 4 | Multifunctional MgF_2/Polydopamine Coating on Mg Alloy for Vascular Stent Application显示文摘Mg alloy is of great potential in the application of vascular stent due to its degradation in physical environment and proper mechanical property.However its mechanical integrity does not meet the clinical requirement due to relatively fast degradation.Besides,in order to accelerate the reendothelialization of Mg-based stents,it needs surface modification to improve the attachment,growth and adhesion of endothelial cells(ECs).To solve the main obstacles,an anti-corrosion and quick endothelialization coating was prepared on novel Mg-Zn-Y-Nd alloy via a simple two-step immersion method in the present study,first in hydrofluoric acid(HF) then in dopamine tris-Hydrochloric acid(tris-HC1) solution.The coating was uniform and thin,which consisted of two layers—the upper was polydopamine(PDA) layer and the lower was MgF_2 layer.The alloy with the coating demonstrated dramatic corrosion resistance enhancement in vitro by immersion test and electrochemical test.Moreover the HFPDA-treated Mg alloy exhibited great performance of cell adhesion and proliferation.The coating created a favorable environment for ECs to have a competitive advantage over vascular smooth muscle cells(VSMCs),which was preferable for re-endothelialization.The results suggest that HF-PDA-treated Mg-Zn-Y-Nd alloy has great potential in the application of vascular stent and the surface coating method is of great application value in biodegradable Mg alloy stent due to its simplicity and effectiveness. | Xiaoli Liu Zhen Zhen Jing Liu Tingfei Xi Yudong Zheng Shaokang Guan Yufeng Zheng Yan Cheng | 2015 | Journal of Materials Science & Technology2015,31,7: | 8 |

| 6 | A draft sequence of the rice (Oryza sativa ssp. indica) genome显示文摘The sequence of the rice genome holds fundamental information for its biology, including physiology, genetics, development, and evolution, as well as information on many beneficial phenotypes of economic significance. Using a 'whole genome shotgun' approach, we have pro-duced a draft rice genome sequence of Oryza sativa ssp. in-dica, the major crop rice subspecies in China and many other regions of Asia. The draft genome sequence is constructed from over 4.3 million successful sequencing traces with an accumulative total length of 2214.9 Mb. The initial assembly of the non-redundant sequences reached 409.76 Mb in length, based on 3.30 million successful sequencing traces with a total length of 1797.4 Mb from an indica variant cultivar 93-11, giving an estimated coverage of 95.29% of the rice genome with an average base accuracy of higher than 99%. | 7 | Development and Evaluation of a Loop-mediated Isothermal Amplification(LAMP) Assay for Rapid Detection of Chinese Giant Salamander Ranavirus显示文摘Loop-mediated isothermal amplification(LAMP) is a novel nucleic acid diagnostic method that can amplify rapidly a target template under isothermal conditions. In this study, a LAMP assay for rapid detection of Chinese giant salamander ranavirus(CGSRV) was developed from culture isolates and clinical samples. The LAMP assay was developed by designing one set of four specific primers, targeting the major capsid protein(MCP) gene of CGSRV. Reaction time and temperature were optimal for 40 min at 62°C. The developed LAMP assay is specific and highly sensitive for CGSRV detection, the detection limit could reach about 5 copies of cloned viral genomic fragments. The sensitivity of the LAMP assay was about 1000 and 10-fold higher than that of both conventional and nested PCR, respectively. The LAMP amplification produces a typical ladder-like pattern of products on an agarose gel that can be visually inspected after addition of ethidium bromide. | 8 | The equal-efficiency-proving of fluorescence quenching and enhancement equation显示文摘Through the fluorescence and ultraviolet- visible spectrophotometry we have studied the action of ce- foperazone sodium and worfloxacin, two kinds of medicine with albumin, and examined the equal efficiency quality of fluorescence enhancement and fluorescence quenching equa- tion. The result shows the equal efficiency of calculating dis- sociation constant. In this article, we obtain the dissociation constants of the action of the two kinds of medicine with bo- vine serum albumin, energy- transfer efficiency, the distance of donor-acceptor and quenching constants. The above shows that the action is not caused by dynamic collision but by the binding of medicine and albumin. This article specially points out that the [Mt]/[Pt], which is the ratio of the maxi- mum mol concentration of the quenching agent (medicine) to fluorescent (albumin) mol concentration when the quenching ends, is very important because it is the maximum binding number of quenching agent molecule to a fluorescent mole- cule. There is no more quenching after adding more quench- ing agent beyond this quantity to fluorescent solution. | 12 | Study on the binding mode of Mg(Sal_2trien) with DNA显示文摘In this study the complex Mg(Sal2trien) was synthesized for the first time, the binding mode of which with CT DNA was studied by the methods of UV spectra, fluorescence spectra, viscosity and CV (cyclic voltammetry). It was found that after the com- plex acted with CT DNA, the Abs of UV spectra rose obviously; the fluorescence intensity of EB-DNA was almost not changed; viscosity decreased. Determina- tion of cyclic voltammetry showed that DNA made the MgL’s formal potential negatively shift. Scatchard plot showed that the addition of the binding mode of the complex to EB was uncompetitive inhibition com- pared with EB to DNA. So the binding mode of the complex with CT DNA was stable-electricity binding. Then the interaction of the complex with pBR322 was studied by the method of gel electrophoresis. | 17 | Responses of the methanogenic pathway and fraction of CH4 oxidization in a flooded paddy soil to rice planting显示文摘Rice planting(RP)is significant to methane(CH4_(4))emissions from paddy fields,but its effect on the relative contribution of the acetoclastic methanogenesis to total CH4_(4) production(F_(ac))and the fraction of CH4_(4) oxidized(Fox)is poorly understood.To quantify the responses of the F_(ac) and Fox to RP,we investigated CH4_(4) fluxes,CH4_(4) production and oxidation potentials,dissolved CH4_(4) concentrations,and their stable carbon isotopes in a flooded paddy soil.The mcrA and pmoA gene copies were also determined by quantitative polymerase chain reaction(qPCR).Compared with the unplanted soil(control,CK),the seasonal CH4_(4) emissions from the planted soil were significantly enhanced,13.6 times,resulting in large decreases in the CH4_(4) concentrations in the soil solution.This indicated that much more CH4_(4) was released into the atmosphere by the RP than was stored in the soils.Acetoclastic methanogenesis became more important from the tillering stage(TS)to the ripening stage(RS)for the CK,with F_(ac) values increased from 17%-20% to 46%-55%.With RP,the F_(ac) values were enhanced by 10%-20%,and it significantly increased the copy numbers of the mcrA gene at the four rice stages(TS,booting stage(BS),grain-filling stage(GS),and RS).Furthermore,the effect of the RP on the abundance of the mcrA gene was highly concurrent with the effect on the F_(ac) values.At the TS,the Fox values at the soil-water interface were around 50%-75% for the CK,being 15%-20% lower than those of the RP in the rhizosphere.It increased to 65%-100% at the GS,but was reduced by 20%-30% after the RP.These differences might be because the copy numbers of the pmoA gene were significantly raised at the TS while lowered at the GS by the RP.This was further demonstrated by the strong correlations between the effect of the RP on the abundance of the pmoA gene and the effect on the Fox values.These findings suggest that RP markedly impacts on the abundances of the mcrA and pmoA genes,affecting the pathway of CH4_(4) production and the fraction of CH4_(4) oxidization,respectively. | Yuting YANG Guangbin ZHANG Jing MA Qiong HUANG Haiyang YU Kaifu SONG Xiaoli ZHU Xi MIAO Hua XU | 2021 | Pedosphere2021,31,6: | 1 |
| 18 | Glutamine mitigates murine burn sepsis by supporting macrophage M2 polarization through repressing the SIRT5-mediated desuccinylation of pyruvate dehydrogenase显示文摘Background:Alternative(M2)-activated macrophages drive the anti-inflammatory response against sepsis,a leading cause of death in patients suffering from burn injury.Macrophage M2 polarization is intrinsically linked with dominant oxidative phosphorylation(OXPHOS).Glutamine serves as a major anaplerotic source to fuel OXPHOS,but it remains unknown whether glutamine can modulate metabolic checkpoints in OXPHOS that favour M2 polarization.The study aims to explore whether glutamine essentially supports M2 polarization in IL-4-stimulated murine macrophages by sustaining the activity of PDH and whether glutamine augments macrophage M2 polarization and thus alleviates inflammation and organ injury in a murine burn sepsis model.Methods:To understand how glutamine promotes M2 activation in interleukin(IL-4)-treated murine macrophages,we detected glutamine-dependent M2 polarization and its relationship with the pyruvate dehydrogenase(PDH)complex by RT-PCR,flow cytometry and western blot.To explore how glutamine modulates PDH activity and thus supports M2 polarization,we compared the expression,phosphorylation and succinylation status of PDHA1 and then examined sirtuin SIRT5-dependent desuccinylation of PDHA1 and the effects of SIRT5 overexpression on M2 polarization by RT-PCR,flow cytometry and western blot.To determine whether glutamine or its metabolites affect M2 polarization,macrophages were cocultured with metabolic inhibitors,and then SIRT5 expression and M2 phenotype markers were examined by RT-PCR,flow cytometry and western blot.Finally,to confirm the in vivo effect of glutamine,we established a burn sepsis model by injecting Pseudomonas aeruginosa into burn wounds and observing whether glutamine alleviated proinflammatory injuries by RT-PCR,flow cytometry,western blot,immunofluorescent staining,hematoxylin-eosin staining and enzyme-linked immuno sorbent assay.Results:We showed that consumption of glutamine supported M2 activation in IL-4-treated murine macrophages by upregulating the activity of PDH.Mechanistically,glutamine did not affect the expression or alter the phosphorylation status of PDHA1 but instead downregulated the expression of SIRT5 and repressed SIRT5-dependent desuccinylation on PDHA1,which in turn recovered PDH activity and supported M2 polarization.This effect was implemented by its secondary metaboliteα-ketoglutarate(αKG)rather than glutamine itself.Finally,we demonstrated that glutamine pro-moted macrophage M2 polarization in a murine burn sepsis model,thereby repressing excessive inflammation and alleviating organ injury in model mice.Conclusions:Glutamine mitigates murine burn sepsis by essentially supporting macrophage M2 polarization,with a mechanism involving the repression of the SIRT5-mediated desuccinylation of pyruvate dehydrogenase that replenishes OXPHOS and sustains M2 macrophages. | Yuanfeng Zhu Xiaoli Chen Yongling Lu Lin Xia Shijun Fan Qianying Huang Xin Liu Xi Peng | 2022 | Burns & Trauma2022,10,1: | 1 |
