| 7 | Expression of reconstructive hF Ⅷ in the hrDNA by using hrDNA targeting vector显示文摘Our lab has constructed a new nonviral vec-tor—hrDNA targeting vector(pHrneo). pHrneo is a human derived vector that can target gene into human ribosomal DNA(hrDNA) locus. In this study, we inserted expression cassette of reconstructive hF Ⅷ (hFVIII-BDDAK39) to pHrneo to construct targeting vector: pHrneo-BDDAK39. Through electroporation of pHrneo-BDDAK39 into HT1080 cells, we identified the homologous recombinants by PCR and Southen blotting, and tested the expression of hFVIII- BDDAK39 in the hrDNA locus. The hFⅧ-BDDAK39 was successfully targeted into hrDNA locus of HT-1080 by pHrneo-BDDAK39, and the efficiency of site-specific inte-gration was 2.0×10?5. hFⅧ-BDDAK39 in hrDNA locus of HT-1080 is found to be able to express efficiently (32±5 ng·106 cells?1·24 h?1). | 9 | Study on Compressive Properties of Z-pinned Laminates in RTD and Hygrothermal Environment显示文摘Compressive tests of [0] 12 and [90] 12 unidirectional laminates and [45/0/-45/90] 2S quasi-isotropic laminates are accomplished in both room-temperature and dry (RTD) and hygrothermal environment. And simulation studies on the compressive strength of Z-pinned laminates of [0] 12 and [45/0/-45/90] 2S are conducted by using finite element analysis (FEA). A microstructural unit cell for FEA is created to simulate a representative laminates unit with one pin. Within the unit cell, the first directions of the elements’ material coordinate systems are changed to simulate the fibres’ deflecting around the pin. The hygrothermal effect is simulated by the material properties’ adjustments which are determined by the compressive tests of non-pined laminates. The experimental results indicate that the percentage of reduction in the compressive modulus of Z-pinned laminates caused by Z-pin becomes smaller with the percentage of 0° fibres decreasing in the laminates; the compressive strength of quasi-isotropic laminates reduces and the percentage of the reduction in the compressive strength declines with Z-pin volume content increasing, and the moisture absorption ratio of the Z-pinned specimens is greater than that of the non-pinned specimens, because the cracks around Z-pin increase the moisture absorption. In addition, the simulations show that the deflection of fibres around Z-pin is the main factor for the reduction in the compressive strength of Z-pinned unidirectional laminates, the dilution of fibre volume content caused by resin-rich pocket is the principal factor for the decline in the compressive strength of Z-pinned quasi-istropic laminates, and the compressive strength of Z-pinned specimens in hygrothermal environment reduces as the result of superimposition of some factors, including the changes in material properties caused by hygrothermal environment, the deflection of fibres and the resin-rich pocket caused by Z-pin. | LI Chenghu YAN Ying WANG Ping QI Desheng WEN Yonghai | 2012 | Chinese Journal of Aeronautics2012,25,1: | 4 |
| 10 | Single-cell RNA sequencing of meiocytes and microspores reveals the involvement of the Rf4 gene in redox homeostasis of CMS-C maize显示文摘Normal microsporogenesis is determined by both nuclear and mitochondrial genes. In maize C-type cytoplasmic male sterility, it is unclear how the development of meiocytes and microspores is affected by the mitochondrial sterility gene and the nuclear restorer gene. In this study, we sequenced the transcriptomes of single meiocytes(tetrad stage) and early mononucleate microspores from sterile and restorer lines. The numbers of expressed genes varied in individual cells and fewer than half of the expressed genes were common to the same cell types. Four comparisons revealed 3379 differentially expressed genes(DEGs), with 277 putatively associated with mitochondria, 226 encoding transcription factors,and 467 possibly targeted by RF4. KEGG analysis indicated that the DEGs in the two lines at the tetrad stage were involved predominantly in carbon metabolism and in amino acid biosynthesis and metabolism, whereas the DEGs during the transition from the tetrad stage to the early mononucleate stage were associated mostly with regulation of protein metabolism, fatty acid metabolism, and anatomical structure morphogenesis. | 14 | A lignified-layer bridge controlled by a single recessive gene is associated with high pod-shatter resistance in Brassica napus L.显示文摘Pod shattering causes severe yield loss in rapeseed(Brassica napus L.)under modern agricultural practice.Identification of highly shatter-resistant germplasm is desirable for the development of rapeseed cultivars for mechanical harvesting.In the present study,an elite line OR88 with strong shatter resistance and a lignified-layer bridge(LLB)structure was identified.The LLB structure was unique to OR88 and co-segregated with high pod-shatter resistance.The LLB structure is differentiated at stage 12 of gynoecium development without any gynoecium defects.Genetic analysis showed that LLB is controlled by a single recessive gene.By BSA-Seq and map-based cloning,the resistance gene location was delimited to a0.688 Mb region on chromosome C09.Transcriptome analysis suggested Bn TCP8.C09 as the gene responsible for LLB.The expression of Bn TCP.C09 was strongly downregulated in OR88,suppressing cell proliferation in the pod valve margin.KASP markers linked to the candidate gene were developed.This pod shatter-resistant line could be used in rapeseed breeding programs by direct transfer of the gene with the assistance of the DNA markers. | Wen Chu Jia Liu Hongtao Cheng Chao Li Li Fu Wenxiang Wang Hui Wang Mengyu Hao Desheng Mei Kede Liu Qiong Hu | 2022 | The Crop Journal2022,10,3: | 1 |

| 18 | Characterization of human IgM and IgG repertoires in individuals with chronic HIV-1 infection显示文摘Advancements in high-throughput sequencing(HTS)of antibody repertoires(Ig-Seq)have unprecedentedly improved our ability to characterize the antibody repertoires on a large scale.However,currently,only a few studies explored the influence of chronic HIV-1 infection on human antibody repertoires and many of them reached contradictory conclusions,possibly limited by inadequate sequencing depth and throughput.To better understand how HIV-1 infection would impact humoral immune system,in this study,we systematically analyzed the differences between the IgM(HIV-IgM)and IgG(HIV-IgG)heavy chain repertoires of HIV-1 infected patients,as well as between antibody repertoires of HIV-1 patients and healthy donors(HH).Notably,the public unique clones accounted for only a negligible proportion between the HIV-IgM and HIV-IgG repertoires libraries,and the diversity of unique clones in HIV-IgG remarkably reduced.In aspect of somatic mutation rates of CDR1 and CDR2,the HIV-IgG repertoire was higher than HIV-IgM.Besides,the average length of CDR3 region in HIV-IgM was significant longer than that in the HH repertoire,presumably caused by the great number of novel VDJ rearrangement patterns,especially a massive use of IGHJ6.Moreover,some of the B cell clonotypes had numerous clones,and somatic variants were detected within the clonotype lineage in HIV-IgG,indicating HIV-1 neutralizing activities.The in-depth characterization of HIV-IgG and HIV-IgM repertoires enriches our knowledge in the profound effect of HIV-1 infection on human antibody repertoires and may have practical value for the discovery of therapeutic antibodies. | Xiaolong Tian Binbin Hong Xiaoyi Zhu Desheng Kong Yumei Wen Yanling Wu Liying Ma Tianlei Ying | 2022 | Virologica Sinica2022,37,3: | 0 |
