Effects of Grazing Exclusion on Soil Carbon and Nitrogen Storage in Semi-arid Grassland in Inner Mongolia, China显示文摘The semi-arid grasslands in Inner Mongolia, China have been degraded by long-term grazing. A series of ecological restoration strategies have been implemented to improve grassland service. However, little is known about the effect of these ecological restoration practices on soil carbon and nitrogen storage. In this study, characteristics of vegetation and soil properties under continued grazing and exclusion of livestock for six years due to a nationwide conservation program—′Returning Grazing Lands to Grasslands(RGLG)′ were examined in semi-arid Hulun Buir grassland in Inner Mongolia, China. The results show that removal of grazing for six years resulted in a significant recovery in vegetation with higher above and below-ground biomass, but a lower soil bulk density and pH value. After six years of grazing exclusion, soil organic C and total N storage increased by 13.9% and 17.1%, respectively, which could be partly explained by decreased loss and increased input of C and N to soil. The effects of grazing exclusion on soil C and N concentration and storage primarily occurred in the upper soil depths. The results indicate that removal of grazing pressure within the RGLG program was an effective restoration approach to control grassland degradation in this region. However, more comprehensive studies are needed to evaluate the effectiveness of the RGLG program and to improve the management strategies for grassland restoration in this area. | WU Xing LI Zongshan FU Bojie LU Fei WANG Dongbo LIU Huifeng LIU Guohua | 2014 | Chinese Geographical Science2014,24,4: | 10
The protective effects of peptides from Chinese baijiu on AAPH-induced oxidative stress in HepG2 cells via Nrf2 signaling pathway显示文摘Antioxidant peptides have been widely reported. However, only a few reports have been published examining the antioxidant peptides derived from Chinese baijiu. In this study,6 novel peptides derived from Chinese baijiu were identified successfully using high-performance liquid chromatography-quadrupoletime-of-flight mass spectrometry(HPLC-QTOF-MS)with a concentration of 0.835–24.540μg/L.The underlying molecular mechanisms were investigated, and their cytoprotective effects were examined against 2,2’-azobis(2-methylpropanimidamidine)dihydrochloride(AAPH)-induced oxidative stress in Hep G2 cells. The results showed that these peptides exerted protective effects by suppressing reactive oxygen species(ROS)generation, preventing malondialdehyde(MDA)formation, and upregulating cellular antioxidant enzyme activities(SOD,CAT,and GSH-Px)in a dose-dependent manner.Further experiments proved that these peptides exerted antioxidant effects via Nrf2/ARE-mediated signaling pathway by promoting Nrf2 nuclear translocation, inhibiting ubiquitination, and enhancing transcription capacity of Nrf2 in Hep G2 cells. These findings provide the molecular basis for the effects of antioxidant peptides derived from Chinese baijiu, which is important for a deeper understanding of the relationship between human health and moderate drinking. | Jiaying Huo Yuezhang Ming Huifeng Li Anjun Li Jiwen Zhao Mingquan Huang Weizheng Sun Jihong Wu Jinglin Zhang | 2022 | Food Science and Human Wellness2022,11,6: | 3
NUPR1 promotes the proliferation and metastasis of oral squamous cell carcinoma cells by activating TFE3-dependent autophagy显示文摘Oral squamous cell carcinoma(OSCC)is the most common type of oral malignancy, and metastasis accounts for the poor prognosis of OSCC.Autophagy is considered to facilitate OSCC development by mitigating various cellular stresses;nevertheless, the mechanisms of autophagy in OSCC cell proliferation and metastasis remain unknown. In our study,high-sensitivity label-free quantitative proteomics analysis revealed nuclear protein 1(NUPR1)as the most significantly upregulated protein in formalin-fixed paraffin-embedded tumour samples derived from OSCC patients with or without lymphatic metastasis. | Tengfei Fan Xiaoning Wang Sheng Zhang Ping Deng Yi Jiang Yidan Liang Sheng Jie Qing Wang Chuwen Li Guocai Tian Zhen Zhang Zhenhu Ren Bo Li Yanrong Chen Zhijing He Yan Luo Mingliang Chen Hanjiang Wu Zhengping Yu Huifeng Pi Zhou Zhou Zhiyuan Zhang | 2022 | Signal Transduction and Targeted Therapy2022,7,5: | 2
Characteristics of soil nutrients and their relationship with soil microbial properties in Artemisia sacrorum communities in the loess hilly region显示文摘Artemisia sacrorum communities with different growth years were selected to analyse soil nutrient characteristics, the variation in soil microbial properties, and their relationships in the loess hilly region. The results showed that with an increase in the number of growth years, soil microbial biomass carbon and nitrogen contents as well as soil phosphatase and urease activities initially decreased and then increased in the A.sacrorum communities. The soil organic carbon, organic nitrogen, and total nitrogen contents as well as soil respiration rate showed an increasing trend and reached a maximum at age(a)37.The soil available phosphorus content first decreased and then increased, with the lowest level observed at 18 a.By contrast, soil available potassium initially increased and then decreased.Soil microbial biomass carbon had a significant positive correlation with soil organic carbon, total nitrogen and organic nitrogen, while soil respiration had a significant positive correlation with organic nitrogen, soil phosphatase and organic carbon.Soil respiration had a highly significant positive correlation with organic carbon and total nitrogen, while soil phosphatase had a highly significant positive correlation with total nitrogen and organic nitrogen. In the A.sacrorum communities, the soil organic carbon and total nitrogen contents were mainly affected by soil respiration, soil available potassium content was mainly affected by soil urease activity, and soil organic nitrogen content was mainly affected by soil phosphatase activity. These findings indicate that soil basal respiration, urease activity and phosphatase activity were the major microbial factors affecting the characteristics of the soil nutrients in the A.sacrorum communities. In conclusion, the natural restoration process of A.sacrorum communities can enhance soil microbial activity and improve soil quality. | Lijuan Song Weiyang Liu Huifeng Wu Tian Gao Wenfang Hao | 2018 | International Journal of Agricultural and Biological Engineering2018,11,4: | 2

Discovery of the mechanisms of acupuncture in the treatment of migraine based on functional magnetic resonance imaging and omics显示文摘Migraine is one of the most prevalent and disabling neurological disease, but the current pharmacotherapies show limited efficacy and often accompanied by adverse effects.Acupuncture is a promising complementary therapy, but further clinical evidence is needed. The influence of acupuncture on migraine is not an immediate effect, and its mechanism remains unclear. This study aims to provide further clinical evidence for the anti-migraine effects of acupuncture and explore the mechanism involved.A randomized controlled trial was performed among 10 normal controls and 38 migraineurs. The migraineurs were divided into blank control, sham acupuncture, and acupuncture groups.Patients were subjected to two courses of treatment, and each treatment lasted for 5 days, with an interval of 1 day between the two courses. The effectiveness of treatment was evaluated using pain questionnaire. The functional magnetic resonance imaging(fMRI)data were analyzed for investigating brain changes induced by treatments.Blood plasma was collected for metabolomics and proteomics studies.Correlation and mediation analyses were performed to investigate the interaction between clinical,fMRI and omics changes. Results showed that acupuncture effectively relieved migraine symptoms in a way different from sham acupuncture in terms of curative effect, affected brain regions, and signaling pathways. The anti-migraine mechanism involves a complex network related to the regulation of the response to hypoxic stress, reversal of brain energy imbalance, and regulation of inflammation. The brain regions of migraineurs affected by acupuncture include the lingual gyrus, default mode network, and cerebellum. The effect of acupuncture on patients’metabolites/proteins may precede that of the brain. | Chong Li Xinyi Li Ke He Yang Wu Xiaoming Xie Jiju Yang Fan Zhang Yang Yue Huifeng Hao Shaokun Zhao Xin Li Guihua Tian | 2023 | Frontiers of Medicine2023,17,5: | 1

Multi‑omics reveals that the host‑microbiome metabolism crosstalk of differential rumen bacterial enterotypes can regulate the milk protein synthesis of dairy cows显示文摘Background Dairy cows’lactation performance is the outcome of the crosstalk between ruminal microbial metabo-lism and host metabolism. However,it is still unclear to what extent the rumen microbiome and its metabolites,as well as the host metabolism, contribute to regulating the milk protein yield(MPY).Methods The rumen fluid, serum and milk of 12 Holstein cows with the same diet(45%coarseness ratio),parity(2–3 fetuses)and lactation days(120–150 d)were used for the microbiome and metabolome analysis.Rumen metabolism(rumen metabolome)and host metabolism(blood and milk metabolome)were connected using a weighted gene co-expression network(WGCNA)and the structural equation model(SEM)analyses. Results Two different ruminal enterotypes, with abundant Prevotella and Ruminococcus, were identified as type1 and type2.Of these,a higher MPY was found in cows with ruminal type2.Interestingly,[Ruminococcus]gauvreauii group and norank_f_Ruminococcaceae(the differential bacteria)were the hub genera of the network. In addition, differential ruminal, serum and milk metabolome between enterotypes were identified, where the cows with type2 had higher L-tyrosine of rumen, ornithine and L-tryptophan of serum, and tetrahydroneopterin,palmitoyl-L-carnitine,S-lactoylglutathione of milk, which could provide more energy and substrate for MPY.Further, based on the identi-fied modules of ruminal microbiome,as well as ruminal serum and milk metabolome using WGCNA,the SEM analysis indicated that the key ruminal microbial module1,which contains the hub genera of the network([Ruminococcus]gauvreauii group and norank_f_Ruminococcaceae)and high abundance of bacteria(Prevotella and Ruminococcus),could regulate the MPY by module7 of rumen,module2 of blood, and module7 of milk, which contained L-tyrosine and L-tryptophan.Therefore,in order to more clearly reveal the process of rumen bacterial regulation of MPY,we established the path of SEM based on the L-tyrosine,L-tryptophan and related components. The SEM based on the metabolites suggested that[Ruminococcus]gauvreauii group could inhibit the energy supply of serum tryptophan to MPY by milk S-lactoylglutathione,which could enhance pyruvate metabolism.Norank_f_Ruminococcaceae could increase the ruminal L-tyrosine,which could provide the substrate for MPY.Conclusion Our results indicated that the represented enterotype genera of Prevotella and Ruminococcus, and the hub genera of[Ruminococcus]gauvreauii group and norank_f_Ruminococcaceae could regulate milk protein synthesis by affecting the ruminal L-tyrosine and L-tryptophan.Moreover,the combined analysis of enterotype,WGCNA and SEM could be used to connect rumen microbial metabolism with host metabolism, which provides a fundamental understanding of the crosstalk between host and microorganisms in regulating the synthesis of milk composition. | Chenguang Zhang Mengya Wang Huifeng Liu Xingwei Jiang Xiaodong Chen Tao Liu Qingyan Yin Yue Wang Lu Deng Junhu Yao Shengru Wu | 2023 | Journal of Animal Science and Biotechnology2023,14,6: | 1
