1Mapping wetland changes in China between 1978 and 2008显示文摘Four wetland maps for all China have been produced,based on Landsat and CBERS-02B remote sensing data between 1978 and 2008 (1978,1990,2000 and 2008).These maps were mainly developed by manual interpretation and validated by substantial field investigation in 2009.Based on these maps,we analyzed the 2008 wetland distribution in China and discussed wetland changes and their drivers over the past 30 years.(i) There were about 324097 km 2 of wetlands in 2008,for which inland marshes or swamps were the most common wetland type (35%),with lakes (26%) second.Most of the wetlands were in Heilongjiang,Inner Mongolia,Qinghai and Tibet,occupying about 55% of the national wetland area.(ii) From 1978 to 2008,China's wetland area continually and significantly decreased,by about 33% based on changes in the wetland map.This was in sharp contrast to the increase in artificial wetlands,which increased by about 122%.Inland marshes accounted for the main loss of total wetlands from 1978 to 2000.From 2000 through 2008,riverine and lacustrine wetlands constituted the main wetland loss.Fortunately however,the rate of wetland loss decreased from 5523 to 831 km 2 /a.(iii) The change ratio of lost natural wetlands (including inland and coastal wetlands) to non-wetlands has decreased slightly over the past 30 years.From 1978 to 1990,nearly all natural wetlands (98%) lost were transformed into non-wetlands.However,the ratio declined to 86% from 1990 to 2000,and to 77% from 2000 to 2008.(iv) All Chinese provinces were divided into three groups according to patterns of wetland changes,which could relate to the driving forces of such changes.Tibet was completely different from other provinces,as it was one representative example in which there was a net wetland increase,because of global warming and decreased human activity since 1990.Increased economic development caused considerable wetland loss in most eastern provinces,and artificial wetlands increased.NIU ZhenGuo ZHANG HaiYing WANG XianWei YAO WenBo ZHOU DeMin ZHAO KuiYi ZHAO Hui LI NaNa HUANG HuaBing LI CongCong YANG Jun LIU CaiXia LIU Shuang WANG Lin LI Zhan YANG ZhenZhong QIAO Fei ZHENG YaoMin CHEN YanLei SHENG YongWei GAO XiaoHong ZHU WeiHong WANG WenQing WANG Hong WENG YongLing ZHUANG DaFang LIU JiYuan LUO ZhiCai CHENG Xiao GUO ZiQi GONG Peng 2012Chinese Science Bulletin2012,57,22:51
2AFLP analysis revealed differences in genetic diversity of four natural populations of Manila clam (Ruditapes philippinarum) in China显示文摘The amplified fragment length polymorphism (AFLP) technology was used to analyze the genetic diversities in four natural populations of Manila clam (Ruditapes philippinarum), distributed in four sea areas of China, i.e., the Bohai Sea, the Huanghai Sea, the East China Sea and the South China Sea. Two hundred and sixty-four AFLP loci were analysed in 195 individuals and revealed high levels of genetic diversity. The percentage of polymorphic loci ranged from 92.13% to 96.06% and the Shannon’s information index was from 0.256 8 to 0.275 6. By analyzing molecular variance (AMOVA), it was found that there were high levels of genetic differentiation between populations of Qingdao and the other three sea areas. Cluster analysis by Nei’s pairwise distances grouped specimens by geographical origin, except the population of Qingdao. A conclusion can be drawn that there are high genetic diversities in the four natural populations of Manila clam in China and some distinct differences existed among and between the four populations. 5Guided chemotherapy based on patient-derived mini-xenograft models improves survival of gallbladder carcinoma patients显示文摘Background:Gallbladder carcinoma is highly aggressive and resistant to chemotherapy,with no consistent strategy to guide first line chemotherapy.However,patient-derived xenograft(PDX)model has been increasingly used as an effective model for in preclinical study of chemosensitivity.Methods:Mini-PDX model was established using freshly resected primary lesions from 12 patients with gallblad-der to examine the sensitivity with five of the most commonly used chemotherapeutic agents,namely gemcitabine,oxaliplatin,5-fluorouracil,nanoparticle albumin-bound(nab)-paclitaxel,and irinotecan.The results were used to guide the selection of chemotherapeutic agents for adjunctive treatment after the surgery.Kaplan-Meier method was used to compare overall survival(OS)and disease free survival(DFS)with 45 patients who received conventional chemo-therapy with gemcitabine and oxaliplatin.Results:Cell viability assays based on mini-PDX model revealed significant heterogeneities in drug responsiveness.Kaplan-Meier analysis showed that patients in the PDX-guided chemotherapy group had significantly longer median OS(18.6 months;95%CI 15.9-21.3 months)than patients in the conventional chemotherapy group(13.9 months;95%CI 11.7-16.2 months)(P=0.030;HR 3.18;95%CI 1.47-6.91).Patients in the PDX-guided chemotherapy group also had significantly longer median DFS(17.6 months;95%CI 14.5-20.6 months)than patients in the conventional chemotherapy group(12.0 months;95%CI 9.7-14.4 months)(P=0.014;HR 3.37;95%CI 1.67-6.79).Conclusion:The use of mini-PDX model to guide selection of chemotherapeutic regimens could improve the out-come in patients with gallbladder carcinoma.Ming Zhan Rui-meng Yang Hui Wang Min He Wei Chen Sun-wang Xu Lin-hua Yang Qiang Liu Man-mei Long Jian Wang 2018Cancer Communications2018,38,1:11
6The Development of A Fluorescence Polarization Immunoassay for Aflatoxin Detection显示文摘A fluorescence polarization immunoassay(FPIA)was developed for the analysis ofaflatoxins(AFs)using an anti-aflatoxin B1(AFB1)monoclonal antibody and a novel fluorescein-labeled AFB1tracer.The FPIA showed an IC50 value of 23.33ng/mL with a limit of detection of 13.12 ng/mL for AFB1.The cross-reactivities of AFB1,AFB2,AFG1,AFG2,AFM1,and AFM2 with the antibody were100%,65.7%,143%,23.5%,111.4%,and 2%,respectively.The group-specificity of anti-AFB1mAb indicated that the FPIA could potentially be used in a screening method for the detection of total AFs,albeit not AFG2 and AFM2.The total time required for analyzing 96 samples in one microplate was less than 5 min.This study demonstrates the potential usefulness of the FPIA as a rapid and simple technique for monitoring AFs.SHENG Ya Jie EREMIN Sergei MI Tie Jun ZHANG Su Xia SHEN Jian Zhong WANG Zhan Hui 2014Biomedical and Environmental Sciences2014,27,2:10

8Hippocampal insulin resistance and the Sirtuin 1 signaling pathway in diabetes-induced cognitive dysfunction显示文摘In the peripheral nervous system,the activation of Sirtuin 1 can improve insulin resistance;however,the role played by Sirtuin 1 in the central nervous system remains unknown.In this study,rat models of diabetes mellitus were generated by a single injection of streptozotocin.At 8 weeks after streptozotocin injection,the Morris water maze test and western blot assays confirmed that the diabetic model rats had learning and memory deficits,insulin resistance,and Sirtuin 1 expression could be detected in the hippocampus.Insulin and the insulin receptor inhibitor S961 were intranasally administered to investigate the regulatory effects of insulin signaling on Sirtuin 1.The results showed that insulin administration improved the impaired cognitive function of diabetic model rats and increased the expression levels of phosphorylated insulin receptor,phosphorylated insulin receptor substrate 1,and Sirtuin 1 in the hippocampus.Conversely,S961 administration resulted in more severe cognitive dysfunction and reduced the expression levels of phosphorylated insulin receptor,phosphorylated insulin receptor substrate 1,and Sirtuin 1.The Sirtuin 1 activator SRT2104 and the inhibitor Sirtinol were injected into the lateral ventricle,which revealed that the activation of Sirtuin 1 increased the expression levels of target of rapamycin complex 1,phosphorylated cAMP-response elementbinding protein,and brain-derived neurotrophic factor.Hippocampal dendritic length and spine density also increased in response to Sirtuin 1 activation.In contrast,Sirtinol decreased the expression levels of target of rapamycin complex 1,phosphorylated cAMP-response elementbinding protein,and brain-derived neurotrophic factor and damaged the dendritic structure.These findings suggest that the Sirtuin 1 signaling pathway plays an important role in the development of insulin resistance-related cognitive deficits in diabetic rats.This study was approved by the Animal Ethics Welfare Committee of the First Affiliated Hospital of Hunan University of Chinese Medicine(approval No.ZYFY201811207)in November 2018.Hui Yang Lin Tang Zhan Qu Shi-Hui Lei Wei Li Yu-Hong Wang 2021Neural Regeneration Research2021,16,12:6
9AntarcticaLC2000:The new Antarctic land cover database for the year 2000显示文摘Antarctica plays a key role in global energy balance and sea level change.It has been conventionally viewed as a whole ice body with high albedo in General Circulation Models or Regional Climate Models and the differences of land cover has usually been overlooked.Land cover in Antarctica is one of the most important drivers of changes in the Earth system.Detailed land cover information over the Antarctic region is necessary as spatial resolution improves in land process models.However,there is a lack of complete Antarctic land cover dataset derived from a consistent data source.To fill this data gap,we have produced a database named Antarctic Land Cover Database for the Year 2000(AntarcticaLC2000) using Landsat Enhanced Thematic Mapper Plus(ETM+) data acquired around 2000 and Moderate Resolution Imaging Spectrometer(MODIS) images acquired in the austral summer of 2003/2004 according to the criteria for the 1:100000-scale.Three land cover types were included in this map,separately,ice-free rocks,blue ice,and snow/firn.This classification legend was determined based on a review of the land cover systems in Antarctica(LCCSA) and an analysis of different land surface types and the potential of satellite data.Image classification was conducted through a combined usage of computer-aided and manual interpretation methods.A total of 4067 validation sample units were collected through visual interpretation in a stratified random sampling manner.An overall accuracy of 92.3%and the Kappa coefficient of 0.836 were achieved.Results show that the areas and percentages of ice-free rocks,blue ice,and snow/firn are 73268.81 km2(0.537%),225937.26 km2(1.656%),and 13345460.41 km2(97.807%),respectively.The comparisons with other different data proved a higher accuracy of our product and a more advantageous data quality.These indicate that AntarcticaLC2000,the new land cover dataset for Antarctica entirely derived from satellite data,is a reliable product for a broad spectrum of applications.HUI FengMing KANG Jing LIU Yan CHENG Xiao GONG Peng WANG Fang LI Zhan YE YuFang GUO ZiQi 2017Science China Earth Sciences2017,60,4:5
10Primary structure of helix C to helix G of a new retinal protein in H.sp.xz515显示文摘The gene fragment encoding the retinal protein from helix C to helix G in a new strain of halobacteria, H.sp.xz515 has been amplified by PCR method. The nucleotide sequence of this fragment has been determined. The deduced amino acid sequence has been compared with halobium br and other two br-like proteins, ar-1 and ar-2. Results show that those amino acid residues in br, essential for proton pumping and binding to retinal, are conserved. 13An improved Landsat Image Mosaic of Antarctica显示文摘A revised Landsat Image Mosaic of Antarctica (LIMA) is presented, using the 1073 multi-band scenes of the original Landsat-7 ETM+ LIMA image collection available at the United States Geological Survey (USGS: http://gffzza64998f73d704ee0hu9vwfnfpv0k56nop.ffgz.tsg.suse.edu.cn/). Three improvements have been applied during the data processing: (1) DN saturation is adjusted by adopting a linear regression, which has a lower root mean square error than the ratio regression used by LIMA; (2) solar elevation angle is calculated using pixel-level latitude/longitude and the acquisition time and date of the central pixel of the scene, improving slightly upon the bilinear interpolation of the solar elevation angles of scene corners applied in LIMA; and (3) two additional image bands, Band 5 and Band 7, are sharpened using the panchromatic band (Band 8) and a Gram-Schmidt Spectral Sharpening algorithm to more easily distinguish snow, cloud and exposed rocks. The final planetary reflectance product is stored in 16-bit bands to preserve the full radiometric content of the scenes. 16Axial gradient excitation accelerates volumetric imaging of two-photon microscopy显示文摘Two-photon excitation fluorescence microscopy(TPM),owing to its capacity for subcellular resolution,intrinsic optical sectioning,and superior penetration depth in turbid samples,has revolutionized biomedical research.However,its layer-by-layer scanning to form a three-dimensional image inherently limits the volumetric imaging speed and increases phototoxicity significantly.In this study,we develop a gradient excitation technique to accelerate TPM volumetric imaging.The axial positions of the fluorophores can be decoded from the intensity ratio of the paired images obtained by sequentially exciting the specimen with two axially elongated two-photon beams of complementary gradient intensities.We achieved a 0.63μm axial localization precision and demonstrate the flexibility of the gradient TPM on various sparsely labeled samples,including bead phantoms,mouse brain tissues,and live macrophages.Compared with traditional TPM,our technique improves volumetric imaging speed(by at least sixfold),decreases photobleaching(i.e.,less than 2.07±2.89%in 25 min),and minimizes photodamages.Yufeng Gao Xianyuan Xia Lina Liu Ting Wu Tingai Chen Jia Yu Zhili Xu Liang Wang Fei Yan Zhuo Du Jun Chu Yang Zhan Bo Peng Hui Li Wei Zheng 2022Photonics Research2022,10,3:3
17Toll-like receptor 5-mediated signaling enhances liver regeneration in mice显示文摘Background:Toll-like receptor 5(TLR5)-mediated pathways play critical roles in regulating the hepatic immune response and show hepatoprotective effects in mouse models of hepatic diseases.However,the role of TLR5 in experimental models of liver regeneration has not been reported.This study aimed to investigate the role of TLR5 in partial hepatectomy(PHx)-induced liver regeneration.Methods:We performed 2/3 PHx in wild-type(WT)mice,TLR5 knockout mice,or TLR5 agonist CBLB502 treated mice,as a model of liver regeneration.Bacterial flagellin content was measured with ELISA,and hepatic TLR5 expression was determined with quantitative PCR analyses and flow cytometry.To study the effects of TLR5 on hepatocyte proliferation,we analyzed bromodeoxyuridine(BrdU)incorporation and proliferating cell nuclear antigen(PCNA)expression with immunohistochemistry(IHC)staining.The effects of TLR5 during the priming phase of liver regeneration were examined with quantitative PCR analyses of immediate early gene mRNA levels,and with Western blotting analysis of hepatic NF-κB and STAT3 activation.Cytokine and growth factor production after PHx were detected with real-time PCR and cytometric bead array(CBA)assays.Oil Red O staining and hepatic lipid concentrations were analyzed to examine the effect of TLR5 on hepatic lipid accumulation after PHx.Results:The bacterial flagellin content in the serum and liver increased,and the hepatic TLR5 expression was significantly up-regulated in WT mice after PHx.TLR5-deficient mice exhibited diminished numbers of BrdU-and PCNA-positive cells,suppressed immediate early gene expression,and decreased cytokine and growth factor production.Moreover,PHx-induced hepatic NF-κB and STAT3 activation was inhibited in Tlr5–/–mice,as compared with WT mice.Consistently,the administration of CBLB502 significantly promoted PHx-mediated hepatocyte proliferation,which was correlated with enhanced production of proinflammatory cytokines and the recruitment of macrophages and neutrophils in the liver.Furthermore,Tlr5–/–mice displayed significantly lower hepatic lipid concentrations and smaller Oil Red O positive areas than those in control mice after PHx.Conclusions:We reveal that TLR5 activation contributes to the initial events of liver regeneration after PHx.Our findings demonstrate that TLR5 signaling positively regulates liver regeneration and suggest the potential of TLR5 agonist to promote liver regeneration.Wen Zhang Lei Wang Xue-Hua Sun Xian Liu Yang Xiao Jie Zhang Ting Wang Hui Chen Yi-Qun Zhan Miao Yu Chang-Hui Ge Chang-Yan Li Guang-Ming Ren Rong-Hua Yin Xiao-Ming Yang 2021Military Medical Research2021,8,4:3

19Phosphate limitation increases coenzyme Q10 production in industrial Rhodobacter sphaeroides HY01显示文摘Coenzyme Q10(CoQ10)is an important component of the respiratory chain in humans and some bacteria.As a high-value-added nutraceutical antioxidant,CoQ10 has excellent capacity to prevent cardiovascular disease.The content of CoQ10 in the industrial Rhodobacter sphaeroides HY01 is hundreds of folds higher than normal physiological levels.In this study,we found that overexpression or optimization of the synthetic pathway failed CoQ10 overproduction in the HY01 strain.Moreover,under phosphate-limited conditions(decreased phosphate or in the absence of inorganic phosphate addition),CoQ10 production increased significantly by 12%to220 mg/L,biomass decreased by 12%,and the CoQ10 productivity of unit cells increased by 27%.In subsequent fed-batch fermentation,CoQ10 production reached 272 mg/L in the shake-flask fermentation and 1.95 g/L in a 100-L bioreactor under phosphate limitation.Furthermore,to understand the mechanism associated with CoQ10 overproduction under phosphate-limited conditions,the comparatve transcriptome analysis was performed.These results indicated that phosphate limitation combined with glucose fed-batch fermentation represented an effective strategy for CoQ10 production in the HY01.Phosphate limitation induced a pleiotropic effect on cell metabolism,and that improved CoQ10 biosynthesis efficiency was possibly related to the disturbance of energy metabolism and redox potential.Lu Zhang Leshi Liu Ke-Feng Wang Lan Xu Liming Zhou Weishan Wang Chuan Li Zheng Xu Tong Shi Haihong Chen Yuanhang Li Hui Xu XiuLiang Yang Zhichun Zhu Biqin Chen Dan Li Guanghuang Zhan Si-Liang Zhang Li-Xin Zhang Gao-Yi Tan 2019Synthetic and Systems Biotechnology2019,4,4:2
