1Complete chloroplast genome sequence of Magnolia grandiflora and comparative analysis with related species显示文摘Magnolia grandiflora is an important medicinal,ornamental and horticultural plant species.The chloroplast(cp) genome of M.grandiflora was sequenced using a 454 sequencing platform and the genome structure was compared with other related species.The complete cp genome of M.grandiflora was 159623 bp in length and contained a pair of inverted repeats(IR) of 26563 bp separated by large and small single copy(LSC,SSC) regions of 87757 and 18740 bp,respectively.A total of 129 genes were successfully annotated,18 of which included introns.The identity,number and GC content of M.grandiflora cp genes were similar to those of other Magnoliaceae species genomes.Analysis revealed 218 simple sequence repeat(SSR) loci,most composed of A or T,contributing to a bias in base composition.The types and abundances of repeat units in Magnoliaceae species were relatively conserved and these loci will be useful for developing M.grandiflora cp genome vectors.In addition,results indicated that the cp genome size in Magnoliaceae species and the position of the IR border were closely related to the length of the ycf1 gene.Phylogenetic analyses based on 66 shared genes from 30 species using maximum parsimony(MP) and maximum likelihood(ML) methods provided strong support for the phylogenetic position of Magnolia.The availability of the complete cp genome sequence of M.grandiflora provides valuable information for breeding of desirable varieties,cp genetic engineering,developing useful molecular markers and phylogenetic analyses in Magnoliaceae.LI XiWen GAO HuanHuan WANG YiTao SONG JingYuan HENRY Robert WU HeZhen HU ZhiGang YAO Hui LUO HongMei LUO Kun PAN HongLin CHEN ShiLin 2013Science China(Life Sciences)2013,56,2:17
2Preparation and Applications of Soybean Residue CNF Films显示文摘In this study, soybean residues were treated with HCl and soybean residue cellulose was extracted, which was used to prepare cellulose nanofiber (CNF) using the high-pressure homogenization method. The maximum yield of CNF, the reaction temperature, reaction time, and HCl concentration were optimized. The optimum HCl concentration for acid treatment was 6%, the reaction time was 60 min, the reaction temperature was 80℃, and the maximum yield of soybean residue cellulose was 78.8%. The different CNF films were then prepared;the color, mechanical property, and light transmittance of the CNF films were studied. Compared to the properties of the CNF film prepared with the soybean residue cellulose by high-pressure homogenization 15 times (HGT-15 film), the mechanical properties of the CNF film with soybean residue cellulose by decolorizing treatment decreased, but the light transmittance increased. The film prepared by adding HGT- 15 CNF to whey protein was investigated for its mechanical property, light transmittance, and solubility. Unlike the pure whey protein film, addition of 2.0% CNF to the whey protein enhanced the mechanical property and water vapor transmission rate (WVT) of the film. 3Multi-objective optimization for deepwater dynamic umbilical installation analysis显示文摘We suggest a method of multi-objective optimization based on approximation model for dynamic umbilical installation. The optimization aims to find out the most cost effective size, quantity and location of buoyancy modules for umbilical installation while maintaining structural safety. The approximation model is constructed by the design of experiment (DOE) sampling and is utilized to solve the problem of time-consuming analyses. The non-linear dynamic analyses considering environmental loadings are executed on these sample points from DOE. Non-dominated Sorting Genetic Algorithm (NSGA-II) is employed to obtain the Pareto solution set through an evolutionary optimization process. Intuitionist fuzzy set theory is applied for selecting the best compromise solution from Pareto set. 4Effect of High Pressure Homogenization Treatment on Structure and Properties of Soybean Residue Cellulose Nanofibers显示文摘To reduce the adverse effects of non-cellulose materials on subsequent homogenization,the effects of a high-pressure homogenization treatment on the structure and properties of cellulose nanofibers(CNF)prepared by acid treatment of soybean residue were studied.The effects of the number of homogenization step on the microfibrillation degree,crystalline structure and mechanical properties of the soybean residue were analyzed by SEM,FT-IR,XRD,TG and DTG.The results showed that an increase in the number of homogenization steps led to an increase in the degree of microfibrillation,a more uniform distribution of the CNF diameter,and an increase in the crystallinity of CNF.However,but when the number of homogenization steps exceeded 15,the rate of change decreased,and the crystallinity of CNF decreased.As the number of homogenization steps increased,the average degree of polymerization and average molecular weight of CNF decreased continuously,and after 15 homogenization steps,their rate of change also decreased.Therefore,15 steps of high-pressure homogenization represented a suitable number of steps to prepare the soybean residue CNF with an average diameter of 15 nm.Peiyi Li Yumeng Wang Binyao Zhou Qingqing Hou Hezhen Liu Haozhe Lei Boxing Jian Xinping Li 2019Paper And Biomaterials2019,4,4:4
5Compound Gaoziban tablet(复方高滋斑片) alleviates depression via toll-like receptor 4/myeloid differentiation factor 88/nuclear factor-kappa B pathway显示文摘OBJECTIVE: To evaluate the effect of compound Gaoziban tablet(复方高滋斑片, CGZBT) on depression, and to investigate the underlying mechanism. METHODS: The components of CGZBT were analysed by high-performance liquid chromatography. Then, we assessed the effects of varying doses of CGZBT on an established chronic unpredictable mild stress(CUMS) model in rats. Whether animals were depressed was evaluated by sucrose preference test, open field test and forced swimming test. Neurotransmitters of hippocampus were detected by liquid chromatography-mass spectrometry. Serum levels of tumor necrosis factor-alpha(TNF-α), interleukin(IL)-1β, IL-6, IL-4, and IL-10 were measured by enzyme-linked immunosorbent assay. Expressions of toll-like receptor 4(TLR4), myeloid differentiation factor 88(My D88), phospho-nuclear factorkappa B(p-NF-κB), cyclooxygenase-2(COX-2), ionized calcium binding adapter molecule-1(IBA-1) were assessed by immunohistochemical staining and western blotting. RESULTS: Eight compounds were identified from CGZBT, moreover, our results showed that CGZBT effectively reversed the CUMS-induced decrease in sucrose preference, shortened the movement distance and prolonged immobility time. CGZBT significantly increased levels of 5-hydroxytryptamine, dopamine, norepinephrine, 5-hydroxyindoleacetic acid levels, and reduced the expression of TNF-α, IL-1β, IL-6, yet increased IL-4 and IL-10. Furthermore, the expressions of TLR4, My D88, COX-2, p-NF-κB and IBA-1 in hippocampus were effectively reversed after treatment with CGZBT. 6Expression of cyclin-dependent protein kinase 5 in the hippocampus of vascular dementia mice after cerebral ischemia and reperfusion显示文摘BACKGROUND:The p25-activated cyclin-dependent protein kinase 5(Cdk5) may induce neuronal cell death and cause the development of dementia following cerebral ischemia and reperfusion. OBJECTIVE:To observe changes in the expression of Cdk5 and p25 in hippocampal tissue of vascular dementia mice at different time points following cerebral ischemia and reperfusion. DESIGN,TIME AND SETTING:A randomized,controlled animal experiment was performed in the clinical trial center of Hebei Provincial People's Hospital between September 2007 and October 2008. MATERIALS:Cdk5 rabbit anti-mouse polyclonal antibody,p35 rabbit anti-mouse polyclonal antibody,andβ-actin mouse monoclonal antibody were purchased from Santa Cruz Biotechnology, Inc.,USA;horseradish peroxidase-labeled goat anti-rabbit IgG and horseradish peroxidase-labeled goat anti-mice IgG were offered by Beijing Zhongshan Goldenbridye Biotechnology Co.,Ltd.,China; the protein quantitative kit was produced by Applygen Gene Technology Corp.,Beijing,China;cDNA reverse transcription and PCR amplification reagents were products of TianGen&Biotech(Beijing) Co.,Ltd.,China. METHODS:One hundred and sixty male Kunming mice were randomly divided into two groups:a sham-operated group(n=65) and a model group(n=95).Vascular dementia was induced with three periods of transient ischemia and reperfusion of the bilateral common carotid arteries.In the sham-operated group,the bilateral common carotid arteries were not blocked. MAIN OUTCOME MEASURES:Behavioral tests were done at four and six weeks post surgery. Pathological changes in the hippocampal CA1 region were observed with hematoxylin-eosin staining Cdk5 mRNA expression was examined by RT-PCR,and Western blots were used to evaluate Cdk5 and p25 expression.Learning and memory performance were assayed using the Morris water maze. RESULTS:Vascular dementia reduced learning and memory performance at 4 and 6 weeks post surgery.Vascular dementia also caused severe,time-dependent neuronal damage and death in the hippocampal CA1 region.Dementia induction also increased mRNA and protein expression of Cdk5 and p25 at both 4 and 6 weeks after surgery. 10The Impact of Urban Expansion on Plant Diversity Change in Karst Regions of Southwest China显示文摘Biodiversity is vital for the integrity and stability of ecosystems and sustainable development.Karst regions of Southwest China is featured for undulating and broken karst terrain as well as high plant diversity.Land use changes induced by the growing population and expanding human settlement have threatened biodiversity preservation in this region.However,the impact of urban expansion on plant diversity remains unclear here.This study focuses on how expanding countryside landscapes affect the recovery rate of plant diversity and demonstrate how urban expansion affects plant diversity conservation in karst regions of Southwest China.In situ biodiversity investigations and multisource remote sensing images were combined to analyze the role of human settlement evolution in the conservation of plant diversity using descriptive statistics and regression analysis.Unmanned vehicle images,historical aerial photographs,and long-term remote sensing images were used to observe the human settlement pattern changes over 40 yr and found that plant diversity is restored faster in countryside ecosystems than in island ecosystems restricted by water.Forests,however,contribute the most to plant diversity conservation in both ecosystems.While the forest area is stable during urban expansion,massive forest patches play an essential role in plant diversity conservation.Arable lands and grasslands shrank but with a fragmenting trend,which was conducive to preserving plant diversity,whereas increased and regularized large patches of built-up areas were not beneficial to plant diversity.Accordingly,forest protection should be prioritized to coordinate future socioeconomic development and plant diversity conservation in karst and broader regions.Furthermore,large built-up patches should be limited,and the irregularity should be improved during urban expansion.Irregular shaped cultivated land and grassland were suggested to promote biological information exchanges as landscape corridors.YANG Shengtian LI Chaojun LOU Hezhen LUO Ya WANG Pengfei ZHANG Jun LI Xi WU Xijin 2022Chinese Geographical Science2022,32,3:0
11Okara Cellulose Nanofibrils Produced by Pretreatment with Sustainable Deep Eutectic Solvent Coupled with Various Mechanical Treatments显示文摘In this study,a green,environmentally friendly method for rapid cellulose nanofribril(CNF)preparation with a significant cost advantage was developed.Pretreatment with a deep eutectic solvent(DES)synthesized from oxalic acid dihydrate and choline chloride(ChCl-O)was combined with various mechanical treatment methods to produce okara CNFs from agricultural waste,with different diameter distributions.The results showed that high-speed stirring produced CNFs with an average diameter of 27 nm.This method was advantageous because it consumed about 94%less energy than traditional high-pressure homogenization method.The DES recovery rate was more than 90%,and DES served as a highly effective treatment,indicating that DES pretreatment is an economical,convenient,and effective strategy for okara CNF preparation.Peiyi Li Haozhe Lei Boxing Jian Ruiyan Liu Miaomiao Zhou Yumeng Wang Hezhen Liu Yun Wang Bingyao Zhou 2022Paper And Biomaterials2022,7,2:0
