| 8 | Association of Overlapped and Un-overlapped Comorbidities with COVID-19 Severity and Treatment Outcomes: A Retrospective Cohort Study from Nine Provinces in China显示文摘Objective Several COVID-19 patients have overlapping comorbidities. The independent role of each component contributing to the risk of COVID-19 is unknown, and how some non-cardiometabolic comorbidities affect the risk of COVID-19 remains unclear.Methods A retrospective follow-up design was adopted. A total of 1,160 laboratory-confirmed patients were enrolled from nine provinces in China. Data on comorbidities were obtained from the patients’ medical records. Multivariable logistic regression models were used to estimate the odds ratio(OR) and 95% confidence interval(95% CI) of the associations between comorbidities(cardiometabolic or non-cardiometabolic diseases), clinical severity, and treatment outcomes of COVID-19.Results Overall, 158(13.6%) patients were diagnosed with severe illness and 32(2.7%) had unfavorable outcomes. Hypertension(2.87, 1.30–6.32), type 2 diabetes(T2 DM)(3.57, 2.32–5.49),cardiovascular disease(CVD)(3.78, 1.81–7.89), fatty liver disease(7.53, 1.96–28.96), hyperlipidemia(2.15, 1.26–3.67), other lung diseases(6.00, 3.01–11.96), and electrolyte imbalance(10.40, 3.00–26.10)were independently linked to increased odds of being severely ill. T2 DM(6.07, 2.89–12.75), CVD(8.47,6.03–11.89), and electrolyte imbalance(19.44, 11.47–32.96) were also strong predictors of unfavorable outcomes. Women with comorbidities were more likely to have severe disease on admission(5.46,3.25–9.19), while men with comorbidities were more likely to have unfavorable treatment outcomes(6.58, 1.46–29.64) within two weeks.Conclusion Besides hypertension, diabetes, and CVD, fatty liver disease, hyperlipidemia, other lung diseases, and electrolyte imbalance were independent risk factors for COVID-19 severity and poor treatment outcome. | 9 | High performance and stability of double perovskite-type oxide NdBa0.5Ca0.5Co1.5Fe0.5O5+δas an oxygen electrode for reversible solid oxide electrochemical cell显示文摘In this study,we successfully synthesized double perovskite-type oxide NdBa0.5Ca0.5Co1.5Fe0.5O5+δ(NBCCF)using a conventional wet chemical method as the oxygen electrode for reversible solid oxide electrochemical cells(RSOCs).The polarization resistance(Rp)of the composite electrode NBCCFGd0.1Ce0.9O2(GDC)is only 0.079Ωcm^2 at 800℃under air.The single cell based on NBCCF-GDC electrode displays a peak power density of 0.941 W/cm^2 in fuel cell mode and a low Rp value of 0.134Ωcm^2.In electrolysis cell mode,the cell displays an outstanding oxygen evolution reaction(OER)activity and shows current density as high as 0.92 A/cm^2 with 50 vol%AH(Absolute Humidity)at 800℃and applied voltage of 1.3 V.Most importantly,the cell exhibits admirable durability of 60 h both in electrolysis mode and fuel cell mode with distinguished reversibility.All these results suggest that NBCCF is a promising candidate electrode for RSOC. | Yunfeng Tian Yun Liu Wenjie Wang Lichao Jia Jian Pu Bo Chi Jian Li | 2020 | Journal of Energy Chemistry2020,29,4: | 4 |
| 10 | Calcitriol attenuates liver fibrosis through hepatitis C virus nonstructural protein 3-transactivated protein 1-mediated TGF β1/Smad3 and NF-κB signaling pathways显示文摘BACKGROUND Hepatic fibrosis is a serious condition,and the development of hepatic fibrosis can lead to a series of complications.However,the pathogenesis of hepatic fibrosis remains unclear,and effective therapy options are still lacking.Our group identified hepatitis C virus nonstructural protein 3-transactivated protein 1(NS3TP1) by suppressive subtractive hybridization and bioinformatics analysis,but its role in diseases including hepatic fibrosis remains undefined.Therefore,additional studies on the function of NS3TP1 in hepatic fibrosis are urgently needed to provide new targets for treatment.AIM To elucidate the mechanism of NS3TP1 in hepatic fibrosis and the regulatory effects of calcitriol on NS3TP1.METHODS Twenty-four male C57BL/6 mice were randomized and separated into three groups,comprising the normal,fibrosis,and calcitriol treatment groups,and liver fibrosis was modeled by carbon tetrachloride(CCl4).To evaluate the level of hepatic fibrosis in every group,serological and pathological examinations of the liver were conducted.TGF-β1 was administered to boost the in vitro cultivation of LX-2 cells.NS3TP1,α-smooth muscle actin(α-SMA),collagen I,and collagen Ⅲ in every group were examined using a Western blot and real-time quantitative polymerase chain reaction.The activity of the transforming growth factor beta 1(TGFβ1)/Smad3 and NF-κB signaling pathways in each group of cells transfected with pcDNA-NS3TP1 or siRNA-NS3TP1 was detected.The statistical analysis of the data was performed using the Student’s t test.RESULTS NS3TP1 promoted the activation,proliferation,and differentiation of hepatic stellate cells(HSCs)and enhanced hepatic fibrosis via the TGFβ1/Smad3 and NF-κB signaling pathways,as evidenced by the presence of α-SMA,collagen I,collagen Ⅲ,p-smad3,and p-p65 in LX-2 cells,which were upregulated after NS3TP1 overexpression and downregulated after NS3TP1 interference.The proliferation of HSCs was lowered after NS3TP1 interference and elevated after NS3TP1 overexpression,as shown by the luciferase assay.NS3TP1 inhibited the apoptosis of HSCs.Moreover,both Smad3 and p65 could bind to NS3TP1,and p65 increased the promoter activity of NS3TP1,while NS3TP1 increased the promoter activity of TGFβ1 receptor I,as indicated by coimmunoprecipitation and luciferase assay results.Both in vivo and in vitro,treatment with calcitriol dramatically reduced the expression of NS3TP1.Calcitriol therapy-controlled HSCs activation,proliferation,and differentiation and substantially suppressed CCl4-induced hepatic fibrosis in mice.Furthermore,calcitriol modulated the activities of the above signaling pathways via downregulation of NS3TP1.CONCLUSION Our results suggest that calcitriol may be employed as an adjuvant therapy for hepatic fibrosis and that NS3TP1 is a unique,prospective therapeutic target in hepatic fibrosis. | Liu Shi Li Zhou Ming Han Yu Zhang Yang Zhang Xiao-Xue Yuan Hong-Ping Lu Yun Wang Xue-Liang Yang Chen Liu Jun Wang Pu Liang Shun-Ai Liu Xiao-Jing Liu Jun Cheng Shu-Mei Lin | 2023 | World Journal of Gastroenterology2023,29,18: | 1 |

| 13 | Species-level Microbiota of Biting Midges and Ticks from Poyang Lake显示文摘Objective The purpose of this study was to investigate the bacterial communities of biting midges and ticks collected from three sites in the Poyang Lake area,namely,Qunlu Practice Base,Peach Blossom Garden,and Huangtong Animal Husbandry,and whether vectors carry any bacterial pathogens that may cause diseases to humans,to provide scientific basis for prospective pathogen discovery and disease prevention and control.Methods Using a metataxonomics approach in concert with full-length 16S rRNA gene sequencing and operational phylogenetic unit(OPU)analysis,we characterized the species-level microbial community structure of two important vector species,biting midges and ticks,including 33 arthropod samples comprising 3,885 individuals,collected around Poyang Lake.Results A total of 662 OPUs were classified in biting midges,including 195 known species and 373 potentially new species,and 618 OPUs were classified in ticks,including 217 known species and 326 potentially new species.Surprisingly,OPUs with potentially pathogenicity were detected in both arthropod vectors,with 66 known species of biting midges reported to carry potential pathogens,including Asaia lannensis and Rickettsia bellii,compared to 50 in ticks,such as Acinetobacter lwoffii and Staphylococcus sciuri.We found that Proteobacteria was the most dominant group in both midges and ticks.Furthermore,the outcomes demonstrated that the microbiota of midges and ticks tend to be governed by a few highly abundant bacteria.Pantoea sp7 was predominant in biting midges,while Coxiella sp1 was enriched in ticks.Meanwhile,Coxiella spp.,which may be essential for the survival of Haemaphysalis longicornis Neumann,were detected in all tick samples.The identification of dominant species and pathogens of biting midges and ticks in this study serves to broaden our knowledge associated to microbes of arthropod vectors.Conclusion Biting midges and ticks carry large numbers of known and potentially novel bacteria,and carry a wide range of potentially pathogenic bacteria,which may pose a risk of infection to humans and animals.The microbial communities of midges and ticks tend to be dominated by a few highly abundant bacteria. | GONG Jian WANG Fei Fei LIU Yang Qing PU Ji DONG Ling Zhi ZHANG Si Hui HUANG Zhen Zhou HUANG Yu Yuan LI Ya Ben YANG Cai Xin TAO Yuan Meihui ZHAO Li Jun JIN Dong LIU Li Yun YANG Jing LU Shan | 2024 | Biomedical and Environmental Sciences2024,37,3: | 0 |

| 15 | A two-step lineage reprogramming strategy to generate functionally competent human hepatocytes from fibroblasts显示文摘Terminally differentiated cells can be generated by lineage reprogramming,which is,however,hindered by incomplete conversion with residual initial cell identity and partial functionality. Here, we demonstrate a new reprogramming strategy by mimicking the natural regeneration route, which permits generating expandable hepatic progenitor cells and functionally competent human hepatocytes. Fibroblasts were first induced into human hepatic progenitor-like cells (hHPLCs), which could robustly expand in vitro and efficiently engraft in vivo. Moreover, hHPLCs could be efficiently induced into mature human hepatocytes (hiHeps) in vitro, whose molecular identity highly resembles primary human hepatocytes (PHHs). Most importantly, hiHeps could be generated in large quantity and were functionally competent to replace PHHs for drug-metabolism estimation, toxicity prediction and hepatitis B virus infection modeling. Our results highlight the advantages of the progenitor stage for successful lineage reprogramming. | 16 | In vivo evaluation of early renal damage in type 2 diabetic patients on 3.0 T MR diffusion tensor imaging显示文摘AIM To investigate the utility of renal diffusion tensor imaging(DTI) to detect early renal damage in patients with type 2 diabetes.METHODS Twenty-six diabetic patients(12 with microalbuminuria(MAU), and 14 with normoalbuminuria) and fourteen healthy volunteers were prospectively included in this study. Renal DTI on 3.0 T MR was performed, and estimated glomerular filtration rate(e GFR) was recorded for each subject. Mean cortical and medullary fractional anisotropy(FA) values were calculated by placing multiple representative regions of interest. Mean FA values were statistically compared among groups. Correlations between FA values and e GFR were evaluated. RESULTS Both cortical and medullary FA were significantly reduced in diabetic patients compared to healthy controls(0.403 ± 0.064 vs 0.463 ± 0.047, P = 0.004, and 0.556 ± 0.084 vs 0.645 ± 0.076, P = 0.002, respectively). Cortical FA was significantly lower in diabetic patients with NAU than healthy controls(0.412 ± 0.068 vs 0.463 ± 0.047, P = 0.02). Medullary FA in diabetic patients with NAU and healthy controls were similar(0.582 ± 0.096 vs 0.645 ± 0.076, P = 0.06). | 17 | High-gravity-assisted green synthesis of rare-earth doped calcium molybdate colloidal nanophosphors显示文摘In this work,we report an innovative route for the synthesis of rare-earth doped calcium molybdate(CaMoO4)nanophosphors by using high gravity rotating packed bed(RPB)technology and paraffin liquid as the solvent.The significant intensified mass transfer and micromixing of reactants in the RPB reactor are benefiting for homogeneous doping of rare-earth ions in the host materials,leading to nanophosphors with high quantum efficiency.The use of liquid paraffin as the solvent eliminates the safety risks associated with volatile organic compounds,increasing the potential for clean production of nanophosphors.Under excitation of deep ultraviolet(DUV)light,the CaMoO4:Na+,Eu3+nanophosphors exhibit red emission at peak wavelength of 615 nm and quantum yield of up to 35.01%.The CaMoO4:Na+,Tb3+nanophosphors exhibit green emission at peak wavelength of543 nm with quantum yield of up to 30.66%.The morphologies of the nanophosphors are tunable from nanofibers through nanorods to nanodots and the possible mechanism of controlling the formation of different nanostructures is proposed on the basis of experimental results and theoretical analysis of mesoscience.These nanophosphors are highly dispersible in organic solvents and utilized for fabricating fabrication of flexible,freestanding luminescent films based on silicone resin.We also demonstrate the red LEDs consisting of the hybrid films of CaMoO4:Na+,Eu3+nanoparticles as color-converting phosphors and DUV LEDs as illuminators,offering strong potential for future nanophosphors-basedsolid-state lighting systems. | Yuan Pu Lifeng Lin Jun Liu Jiexin Wang Dan Wang | 2020 | Chinese Journal of Chemical Engineering2020,28,6: | 3 |
| 18 | Receptor-like kinase OsCR4 controls leaf morphogenesis and embryogenesis by fixing the distribution of auxin in rice显示文摘Cell differentiation is a key event in organ development;it involves auxin gradient formation, cell signaling, and transcriptional regulation. Yet, how these processes are orchestrated during leaf morphogenesis is poorly understood. Here, we demonstrate an essential role for the receptor-like kinase Os CR4 in leaf development. oscr4 loss-of-function mutants displayed short shoots and roots, with tiny,crinkly, or even dead leaves. The delayed outgrowth of the first three leaves and seminal root in oscr4 was due to defects in plumule and radicle formation during embryogenesis. The deformed epidermal,mesophyll, and vascular tissues observed in oscr4 leaves arose at the postembryo stage;the corresponding expression pattern of pro Os CR4:GUS in embryos and young leaves suggests that Os CR4 functions in these tissues. Signals from the auxin reporter DR5 rev:VENUS were found to be altered in oscr4 embryos and disorganized in oscr4 leaves, in which indole-3-acetic acid accumulation was further revealed by immunofluorescence. Os WOX3 A, which is auxin responsive and related to leaf development,was activated extensively and ectopically in oscr4 leaves, partially accounting for the observed lack of cell differentiation. | 19 | Arginine ADP-ribosyltransferase 1 Regulates Glycolysis in Colorectal Cancer via the PI3K/AKT/HIF1αPathway显示文摘Objective:Arginine ADP-ribosyltransferase 1(ART1)is involved in the regulation of a diverse array of pathophysiological processes,including proliferation,invasion,apoptosis,autophagy and angiogenesis of colorectal cancer(CRC)cells.However,how ART1 regulates glycolysis in CRC remains elusive.Methods:To elucidate the role of ART1 in glycolysis in CRC,we assessed the protein level of ART1,hypoxia-inducible factor 1α(HIF1α),and glucose transporter type 1(GLUT1)in 61 CRC tumor tissue specimens obtained from patients with different 2-[^(18)F]fluoro-2-deoxy-D-glucose(^(18)F-FDG)uptake as analyzed by PET/CT before surgery.Colon adenocarcinoma CT26 cells with ART1 knockdown and overexpression were established,respectively,and the molecular mechanism underlying the effect of ART1 on glycolysis in CRC was determined both in vivo and in vitro.Results:The expression of ART1 and GLUT1 was significantly associated with FDG uptake(P=0.037 and P=0.022,respectively)in CRC tissues.Furthermore,the expression of hexokinase 2(HK2)and lactate dehydrogenase(LDH)was upregulated in ART1-overexpressed CT26 cells,but was downregulated in ART1-knockdown CT26 cells.The volume and weight of subcutaneously transplanted tumors were markedly increased in the ART1-overexpressed BALB/c mice group and decreased in the ART1-knockdown group.In CT26 cells,the overexpression of ART1 promoted the expression levels of HK2 and LDH,and knockdown of ART1 suppressed them in the CT26 tumors.In both normal and hypoxic conditions,ART1 expression was associated with the protein level of phospho-serine/threonine kinase(p-AKT),HIF1α,and GLUT1 but not with that of AKT in CT26 cells and subcutaneous transplanted tumors.Conclusion:ART1 plays a crucial role in the elevation of glucose consumption in CT26 cells and may regulate GLUT1-dependent glycolysis in CRC via the PI3K/AKT/HIF1αpathway. | Wen-bo LONG Xia PU Yi TANG Ming LI Yun LIU Qin SHE Ya-lan WANG Qin-xi GUO | 2022 | Current Medical Science2022,42,4: | 0 |
