1Natural Polyphenol Inspired Polycatechols for Efficient siRNA Delivery显示文摘There is a continuing quest to rationally fabricate polymeric biomaterials with both high transfection efficiency and minimal toxicity for the emerging opportunities in small interfering RNA(siRNA)delivery.Recently,this goal was promoted highly by developing a robust and efficient strategy to facilitate polymer-mediated RNAi using natural polyphenols with multiple phenol groups that could condense siRNA effectively into negatively charged nanoparticles(NPs).Further coating of these NPs with cationic polymers of low molecular weight enabled their intracellular siRNA delivery.Inspired by the structural and functional features of natural polyphenols,we aimed to further the development of low molecular weight polycatechols as a new class of efficient and biocompatible polymers for siRNA delivery in our current study.The fabricated polycatechols have benefits of requiring only one-step fabrication toward efficient siRNA nanoformulations.Moreover,they could deliver siRNA into cells and silence target genes both in vitro and in vivo.The resulting polycatechol/siRNA formulations were also functionally competent,demonstrating a successful,profound downregulation of a proinflammatory enzyme to attenuate chronic intestinal inflammation in an intestinal injury model.This study provides a new approach in chemistry for the development of efficient synthetic polymers for therapeutic siRNA delivery.Wanwan Shen Ruojun Wang Qianqian Fan Xiao Gao Hui Wang Yang Shen Yiwen Li Yiyun Cheng 2020CCS Chemistry2020,2,3:1

3Integrating the second near-infrared fluorescence imaging with clinical techniques for multimodal cancer imaging by neodymiumdoped gadolinium tungstate nanoparticles显示文摘Second near-infrared(NIR-II)fluorescence imaging is a recently emerged technique and is highly useful for accurate diagnosis of cancer.Although a diverse array of fluorescent nanomaterials have been developed to enable NIR-II fluorescence in various situations,they normally fail to unify the clinical techniques,such as computed tomography(CT)and magnetic resonance imaging(MRI).Therefore,exploiting multimodal agents to integrate the newly emerged NIR-II fluorescence and traditional clinical techniques would be of key significance.Here,we report a rational fabrication of neodymium(Nd)-doped gadolinium tungstate nanoparticles(NPs)that are subsequentially decorated with a hydrophilic layer and demonstrate that they can achieve the harmonious integration of NIR-II fluorescence imaging,CT,and MRI.The NIR-II fluorescence emission was activated by an incident light with discrete wavelength ranging from 250 to 810 nm.NIR-II fluorescence-CT-MRI associated trimodal imaging was subsequently demonstrated for breast cancer by an 808 nm laser,along with the estimation of NIR-II fluorescence imaging for cervical cancer.The integration of newly emerged and traditional clinical imaging techniques highlights the huge potential of rare-earth-doped NPs for multimodal imaging of different types of cancer.Xujiang Yu Aodenggerile Zhao Jiang Jianliang Shen Zhiqiang Yan Wanwan Li Huibin Qiu 2021Nano Research2021,14,7:0
