| 1 | Effects of endostatin-vascular endothelial growth inhibitor chimeric recombinant adenoviruses on antiangiogenesis显示文摘AIM: To investigate the inhibitory effects of endostatinvascular endothelial growth inhibitor (VEGI151) recombinant adenoviruses on neovascularization.METHODS: We used recombinant adenoviruses to treat human vascular endothelial cell line ECV304, human hepatocellular carcinoma cell line HepG2, and murine fibroblast cell line L929, in order to study the chimeric gene expression in these cell lines. Chick choriallantic membrane(CAM) model, rabbit inflammatory corneal neovascularization(CNV) model, and liver cancer-bearing nude mice model were employed to investigate the negative biological effect of fusion molecules on neovascularization in vivo.RESULTS: Western blot showed that the molecular weight of fusion protein was about 41kD after infection of ECV304,HepG2 and L929 cells with supernatant of AdhENDO-VEGI151.The fusion protein showed a specific inhibitory effect on the proliferation of ECV304 cells, but no inhibitory effect on the growth of HepG2 and L929 cells (F=13112.13, P=0.0001). In the chick choriallantic membrane (CAM) assay, the expressed fusion protein significantly inhibited neovascularization. Rabbit inflammatory corneal neovascularization (CNV) induced by intrastromal sutures resulted in a uniform neovascular response. In this model, direct subconjunctival injection of AdhENDO-VEGI151 expressed the fusion protein in vivo and suppressed the development of CNV. Topical application of AdhENDO-VEGI151 led to a significant suppression of CNV (F=1413.11, P=0.0001),as compared with the control group of AdLacZ. Immunohistochemical staining showed the fusion protein dominantly expressed in corneal epithelium. Compared with the control group of AdLacZ (4075.9±1849.9 mm^3), the average tumor size of group AdhENDO-VEGI151 reduced in size (487.7±241.2 mm^3) (F=14.80, P=0.0085), with an inhibition rate of 88.03%. Immunohistochemical staining showed the adenoviruses carried the fusion gene expressed on liver cancer cell membrane. MVD decreased more significantly in treated mice (30.75±3.31%) than in AdLacZ control(50.25±8.65%) (F=17.72, P=0.0056) with an inhibition rate of 39%.CONCLUSION: Fusion protein expressed by recombinant adenoviruses has a significant inhibitory effect on neovascularization. | XinPan YongWang MinZhang WeiPan Zhong-TianQi Guang-WenCao | 2004 | World Journal of Gastroenterology2004,10,10: | 19 |
| 2 | Immune fluorescence test strips based on quantum dots for rapid and quantitative detection of carcino-embryonic antigen显示文摘At present,many researchers focused on the point-of-care testing(POCT),a method of disease markers detection without large-scale instruments and specialized persons.However,most POCT diagnostic methods were suffered from poor detection sensitivity or inefficiency in quantitative detection.Herein,we developed a newly QD-immune fluorescence test strips(QD-IFTS) based on quantum dots(QDs) as the fluorescence nanocarrier to prepare the immune fluorescence probes in the classical immunochromatography detection system for sensing carcino-embryonic antigen(CEA),a kind of glycoprotein produced by intestinal tissue and a broad spectrum of tumor marker for cancer diagnosis.And we designed a homemade strips fluorescence reader for detection of fluorescence intensity of QDs on the QD-IFTS.Under the optimized reaction conditions,chromatographic time of the newly QD-IFTS was only25 min,sample volume of the newly QD-IFTS was only 40 m L and the LOD of the newly QD-IFTS was 0.72 ng/m L.In addition,the efficiency and robustness of the newly QD-IFTS were confirmed by successfully application in 300 clinical serum samples,and the results revealed great potential in clinical POCT of other biomarkers. | Yudong Wu Weipan Peng Qian Zhao Jiafang Piao Bo Zhang Xiaoli Wu Hanjie Wang Zhihong Shi Xiaoqun Gong Jin Chang | 2017 | Chinese Chemical Letters2017,28,9: | 3 |

| 10 | Carbon-Coated-Nylon-Fiber-Reinforced Cement Composites as an Intrinsically Smart Concrete for Damage Assessment during Dynamic Loading显示文摘Concrete containing short carbon-coated-nylon fibers (0.4~2.0 vol. pct) exhibited quasi-ductile response by developing a large damage zone prior to fracture localization. In the damage zone, the material was microcracked but continued to local strain-harden. The carbon-coated-nylon-fiber-reinforced concrete composites (NFRC) were found to be an intrinsically smart concrete that could sense elastic and inelastic deformation, as well as fracture. The fibers served to bridge the cracks and the carbon coating gave the conduction path. The signal provided came from the change in electrical resistance, which was reversible for elastic deformation and irreversible for inelastic deformation and fracture. The resistance decrease was due to the reduction of surface touch resistance between fiber and matrix and the crack closure. | 11 | Identification of annual routines and critical stopover sites of a breeding shorebird in the Yellow Sea,China显示文摘Pied Avocets(Recurvirostra avosetta)are common migratory shorebirds in the East Asian–Australasian Flyway.From 2019 to 2021,GPS/GSM transmitters were used to track 40 Pied Avocets nesting in northern Bohai Bay to identify annual routines and key stopover sites.On average,southward migration of Pied Avocets started on 23 October and arrived at wintering sites(mainly in the middle and lower reaches of the Yangtze River and coastal wetlands)in southern China on 22 November;northward migration started on 22 March with arrival at breeding sites on 7 April.Most avocets used the same breeding sites and wintering sites between years,with an average migration distance of 1124 km.There was no significant difference between sexes on the migration timing or distance in both northward and southward migration,except for the departure time from the wintering sites and winter distribution.The coastal wetland of Lianyungang in Jiangsu Province is a critical stopover site.Most individuals rely on Lianyungang during both northward and southward migration,indicating that species with short migration distances also heavily rely on a few stopover sites.However,Lianyungang lacks adequate protection and is facing many threats,including tidal flat loss.We strongly recommend that the coastal wetland of Lianyungang be designated as a protected area to effectively conserve the critical stopover site. | Yang Wu Weipan Lei Bingrun Zhu Jiaqi Xue Yuanxiang Miao Zhengwang Zhang | 2022 | Avian Research2022,13,4: | 0 |
| 12 | High-contrast and real-time visualization of membrane proteins in live cells with malachite green-based fluorogenic probes显示文摘Imaging dynamics of membrane proteins of live cells in a wash-free and real-time manner has been a challenging task. Herein, we report unprecedented applications of malachite green(MG), an organic dye widely used in pigment industry, as a switchable fluorophore to monitor membrane enzymes or noncatalytic proteins in live cells. Conformationally flexible MG is non-fluorescent in aqueous solution, yet covalent binding with endogenous proteins of cells significantly enhances its fluorescence at 670 nm by restricting flexibility of dye. Integrating a phosphate-caged quinone methide precursor with MG yielded a covalent labeling fluorogenic probe, allowing real-time imaging of membrane alkaline phosphatase(ALP,a model catalytic protein) activity in live cells with over 100-fold enhancement of fluorescence intensity.Moreover, MG is also applicable to image non-catalytic protein by conjugation with protein-specific ligand. | 14 | Single-stranded RNA as primers of terminal deoxynucleotidyl transferase for template-independent DNA polymerization显示文摘Terminal deoxynucleotidyl transferase(Td T) has been characterized as template-independent polymerase using single-stranded DNA(ss DNA) as primers to generate random oligonucleotides. However, the extension performance of Td T to single-stranded RNA(ss RNA) is vague. By systematically comparing and contrasting the performance of Td T-catalyzed ss DNA and ss RNA extension, it is indicated that the catalytic efficiency of ss RNA as primers was about 3 times lower than ss DNA as primers.
