1Inhibition of epidermal growth factor receptor expression by RNA interference in A549 cells显示文摘AIM: To investigate the biological features of A549 cells in which epidermal growth factor (EGF) receptors expression were suppressed by RNA interference (RNAi). METHODS: A549 cells were transfected using short small interfering RNAs (siRNAs) formulated with Lipofectamine 2000. The EGF receptor numbers were determined by Western blotting and flowcytometry. The antiproliferative effects of sequence specific double stranded RNA (dsRNA) were assessed using cell count, colony assay and scratch assay. The chemosensitivity of transfected cells to cisplatin was measured by MTT. RESULTS: Sequence specific dsRNA-EGFR down-regulated EGF receptor expression dramatically. Compared with the control group, dsRNA-EGFR reduced the cell number by 85.0 %, decreased the colonies by 63.3 %, inhibited the migration by 87.2 %, and increased the sensitivity of A549 to cisplatin by four-fold. CONCLUSION: Sequence specific dsRNA-EGFR were capable of suppressing EGF receptor expression, hence significantly inhibiting cellular proliferation and motility, and enhancing chemosensitivity of A549 cells to cisplatin. 2Identification of effective siRNA against K-ras in human pancreatic cancer cell line MiaPaCa-2 by siRNA expression cassette显示文摘AIM: We shall construct the small interfering RNA (siRNA)expression cassette (SEC) targeting activated K-ras gene sequence, identify more effective siRNA sequence against K-ras gene in human pancreatic cancer cell line MiaPaCa-2 by SEC and reveal the anti-cancer effects of RNA interference (RNAi) and its therapeutic possibilities.WETHODS: Three different sites of SECs were constructed by PCR. K1/siRNA, K2/siRNA and K3/siRNA are located at sites 194, 491 and 327, respectively. They were transfected into MiaPaCa-2 cells by liposome to inhibit the expression of activated K-ras. In the interfering groups of sites 194 and 491, we detected the apoptosis in cells by FACS after they were incubated for 48 h, then we tested the alternation of K-ras gene in MiaPaCa-2 cells by RT-PCR immunofluorescence, respectively.RESULTS: Introduction of the K1/siRNA and K2/siRNA against K-ras into MiaPaCa-2 cells leads to increased apoptosis, and the number of apoptotic cells is increased compared with control cells. The tests of RT-PCR immunofluorescence show the effects of inhibiting expression of activated K-ras gene by RNA interference in the K1/siRNA and K2/siRNA groups. We also find that the introduction of K3/siRNA has no effect on MiaPaCa-2 cells.CONCLUSION: K1/siRNA and K2/siRNA can inhibit the expression of activated K-fas but K3/siRNA has no effect,demonstrating that K1/siRNA and K2/siRNA are effective sequences against K-ras gene and K3/siRNA are not. 3Clinical and experimental study on therapeutic effect of umbilical cord blood transplantation on severe viral hepatitis显示文摘AIM: To investigate the therapeutic effect of umbilical cordblood transplantation (UCBT) on patients with severe viralhepatitis and on liver lesions in rats.METHODS: One hundred and fifty three patients with severeviral hepatitis were included in the study between April 1990and July 2002. The patients were treated with adult plasmatransfusion (control), UCBT, plasma exchange (PE) and UCBTcombined with PE (UCBT+PE) respectively. The therapeuticeffectiveness was evaluated by serial determinations of liverfunction, lipids and immune function in all patients beforeand after the treatment. The model of experimental hepaticfailure was constructed in SD rats by injecting carbontetrachloride. Then, the rats were given normal saline, adultplasma or neonate cord blood intraperitoneally. After detectionof liver function, the rats were killed and morphologicalchanges of the liver were microscopically observed.RESULTS: UCBT group and UCBT+PE group had muchbetter improvement in liver and immune functions thancontrol group and PE group. The patients in UCBT+PE grouphad the best clinical efficacy. UCBT was safe and had noside effects. The animal experiment showed significantimprovements in liver function and survival rate in neonatecord blood group as compared with adult plasma group.The histopathology of rat′s liver indicated that neonate cordblood application could decrease the liver injury and increasehepatocellular regeneration.CONCLUSION: UCBT demonstrated a good therapeuticeffect on severe viral hepatitis and no obvious side effects.Umbilical cord blood can attenuate the liver lesions andreproduce hepatocyte. 4Effects of endostatin-vascular endothelial growth inhibitor chimeric recombinant adenoviruses on antiangiogenesis显示文摘AIM: To investigate the inhibitory effects of endostatinvascular endothelial growth inhibitor (VEGI151) recombinant adenoviruses on neovascularization.METHODS: We used recombinant adenoviruses to treat human vascular endothelial cell line ECV304, human hepatocellular carcinoma cell line HepG2, and murine fibroblast cell line L929, in order to study the chimeric gene expression in these cell lines. Chick choriallantic membrane(CAM) model, rabbit inflammatory corneal neovascularization(CNV) model, and liver cancer-bearing nude mice model were employed to investigate the negative biological effect of fusion molecules on neovascularization in vivo.RESULTS: Western blot showed that the molecular weight of fusion protein was about 41kD after infection of ECV304,HepG2 and L929 cells with supernatant of AdhENDO-VEGI151.The fusion protein showed a specific inhibitory effect on the proliferation of ECV304 cells, but no inhibitory effect on the growth of HepG2 and L929 cells (F=13112.13, P=0.0001). In the chick choriallantic membrane (CAM) assay, the expressed fusion protein significantly inhibited neovascularization. Rabbit inflammatory corneal neovascularization (CNV) induced by intrastromal sutures resulted in a uniform neovascular response. In this model, direct subconjunctival injection of AdhENDO-VEGI151 expressed the fusion protein in vivo and suppressed the development of CNV. Topical application of AdhENDO-VEGI151 led to a significant suppression of CNV (F=1413.11, P=0.0001),as compared with the control group of AdLacZ. Immunohistochemical staining showed the fusion protein dominantly expressed in corneal epithelium. Compared with the control group of AdLacZ (4075.9±1849.9 mm^3), the average tumor size of group AdhENDO-VEGI151 reduced in size (487.7±241.2 mm^3) (F=14.80, P=0.0085), with an inhibition rate of 88.03%. Immunohistochemical staining showed the adenoviruses carried the fusion gene expressed on liver cancer cell membrane. MVD decreased more significantly in treated mice (30.75±3.31%) than in AdLacZ control(50.25±8.65%) (F=17.72, P=0.0056) with an inhibition rate of 39%.CONCLUSION: Fusion protein expressed by recombinant adenoviruses has a significant inhibitory effect on neovascularization.XinPan YongWang MinZhang WeiPan Zhong-TianQi Guang-WenCao 2004World Journal of Gastroenterology2004,10,10:19

6Yin Yang 1(YY1) synergizes with Smad7 to inhibit TGF-β signaling in the nucleus显示文摘As a prototype of the TGF-βsuperfamily cytokines,TGF-βis well known for its diverse roles in embryogenesis and adult tissue homeostasis.TGF-βevokes cellular responses by signaling mainly through cell membrane receptors and transcription factor R-Smads and Co-Smad(Smad4),while an inhibitory Smad,Smad7,acts as a critical negative regulator of TGF-βsignaling.Smad7 antagonizes TGF-βsignaling by regulating the stability or activity of the receptors or blocking the DNA binding of the functional R-Smad-Smad4 complex in the nucleus.However,the function of Smad7 in the nucleus is not fully understood.Yin Yang 1(YY1)is a ubiquitously expressed transcription factor with multiple functions.It has been reported that YY1 can inhibit Smad-dependent transcriptional responses and TGF-β/BMP-induced cell differentiation independently of its DNA binding ability.In this study,we found that Smad7 interacts with YY1 and the interaction is attenuated by TGF-βsignaling.Reporter assays and target gene expression analyses revealed that Smad7 and YY1 act in concert to inhibit TGF-β-induced transcription in the nucleus.Furthermore,Smad7 could enhance the interaction of YY1 with the histone deacetylase HDAC1.Consistently,YY1 and HDAC1 augmented the transcription repression activity of Smad7 in Gal4-luciferase reporter analysis.Therefore,our findings define a novel mechanism of Smad7 and YY1 to antagonize TGF-βsignaling.YAN XiaoHua PAN Jun XIONG WanWan CHENG MinZhang SUN YingYuan ZHANG SuPing CHEN YeGuang 2014Science China(Life Sciences)2014,57,1:10
7Bathymetry predicted from vertical gravity gradient anomalies and ship soundings显示文摘In this paper,the admittance function between seafloor undulations and vertical gravity gradient anomalies was derived. Based on this admittance function,the bathymetry model of 1 minute resolution was predicted from vertical gravity gradient anomalies and ship soundings in the experimental area from the northwest Pacific. The accuracy of the model is evaluated using ship soundings and existing models,including ETOPO1,GEBCO,DTU10 and V15. 1 from SIO. 8Inhibition of mouse hepatocyte apoptosis via anti-Fas ribozyme显示文摘AIM: TO investigate the effects of anti-Fas ribozyme on Fas expression and apoptosis in primary cultured mouse hepatocytes.METHODS: Mouse hepatocytes were isolated by using collagenase irrigation. A hammerhead ribozyme targeting the Fas mRNA was constructed, and transfected into mouse hepatocytes via Effectene. Then Fas expression in mouse hepatocytes was detected by RT-PCR and western blotting.After being treated with anti-Fas antibody (JO2), hepatocytes viability was measured with MTI- assay. Caspase-3 proteolytic activity was detected, and cell apoptosis was measured according to Annexin V-FITC apoptosis detection kit.RESULTS: Fas expressed in primary mouse hepatocytes.Fas expression in hepatocytes transfected with anti-Fas ribozyme was decreased remarkably and correlated with the resistance to Fas-mediated apoptosis as determined by flow cytometry and caspase-3 proteolytic activity.CONCLUSION: AnU-Fas ribozyme can remarkably decrease the Fas expression in mouse hepatocytes, thus inhibit Fasmediated apoptosis in hepatocytes, It is suggested that anti-Fas ribozyme could significantly increase the resistance of transplanted hepatocytes to apoptosis and improve the survival of transplanted hepatocytes,MinZhang WeiHe FangLiu PingZou JuanXiao Zhao-DongZhong Zhong-BoHu 2004World Journal of Gastroenterology2004,10,17:8

10Inhibition of Fas-mediated Apoptosis in Yac-1 Cell via Anti-Fas Ribozyme显示文摘To detect a new and more effective way against apoptosis mouse lymphomatic cell line- Yac-1 in which fas gene was expressed highly was used as a model for studying the effects of anti-Fas ribozyme on Fas-mediated apoptosis. A hammerhead ribozyme gene targeting the fas mRNA was synthesized and its in vitro transcription vector was constructed, which was transfected into Yac-1 cells using electroporation. Rz596 expression was detected using RT-PCR, and Fas expression in Yac-1 cells was detected using RT-PCR, Western blot and flow cytometry. After treated with anti-Fas antibody (JO2), Yac-1 cell viability was measured with MTT assay, caspase-3 proteolytic activity was detected, and cell apoptosis was measured according to annexin V apoptosis detecting kit. Anti-Fas ribozyme could cleave fas mRNA efficiently in vivo and in vitro. Fas expression in Yac-1 cells transfected with anti-Fas ribozyme was decreased remarkably and correlated with resistance to Fas-mediated apoptosis as determined by flow cytometry and caspase-3 proteolytic activity. Anti-Fas ribozyme was detected in cells transfected with pU6-RZ596 and pU6-dRZ596 and could remarkably decrease the Fas expression in Yac-1 cells, which made Yac-1 cells get rid of Fas-mediated apoptosis. 11Numerical Simulation of Current Density Distribution in Keyhole Double-Sided Arc Welding显示文摘In the double-sided arc welding system (DSAW) composing of PAW+TIG arcs, the PAW arc is guided by the TIG arc so that the current mostly flows through the direction of the workpiece thickness and the penetration is greatly improved. To analyze the current density distribution in DSAW is beneficial to understanding of this process.Considering all kinds of dynamic factors acting on the weldpool, this paper discusses firstly the surface deformation of the weldpool and the keyhole formation in PAW+TIG DSAW process on the basis of the magnetohydrodynamic theory and variation principles. Hence, a model of the current density distribution is developed. Through numerical simulation, the current density distribution in PAW+TIG DSAW process is quantitatively analyzed. 12Temporal variation of gravity field prior to the Ludian Ms6.5 and Kangding Ms6.3 earthquakes显示文摘Using mobile gravity data from the central area of Sichuan and Yunnan Provinces, the relationship between gravity variation and earthquakes was studied based on the Ludian Ms6.5 earthquake that occurred on August 3rd, 2014, and the Kangding Ms6.3 earthquake that occurred on November 22 nd, 2014; the mechanism of gravity variation was also explored. The results are as follows:(1) Prior to both earthquakes, gravity variation exhibited similar characteristics as those observed before both the Tangshan and Wenchuan earthquakes, in which typical precursor anomalies were positive gravity variation near the epicenter and the occurrence of a high-gravity-gradient zone across the epicenter prior to the earthquake.(2) A relatively accurate prediction of the occurrence locations of the two earthquakes was made by the Gravity Network Center of China(GNCC) based on these precursor anomalies. In the gravity study report on the 2014 earthquake trends submitted at the end of 2013, the Daofu-Shimian section at the junction of the Xianshuihe and Longmenshan fault zones was noted as an earthquake-risk region with a predicted magnitude of 6.5, which covered the epicenter of the Kangding Ms6.3 earthquake. In another report on earthquake trends in southwestern China submitted in mid-2014, the Lianfeng, Zhaotong fault zone was also classified as an earthquake-risk region with a magnitude of 6.0, and the central area of this region basically overlapped with the epicenter of the Ludian Ms6.5 earthquake.(3) The gravity variation characteristics are reasonably consistent with crustal movements, and deep material migration is likely the primary cause of gravity variation.Hao Hongtao Wei Jin Hu Minzhang Liu Ziwei Li Hui 2015Geodesy and Geodynamics2015,6,6:3

16Terrestrial water storage variation in Hebei plain area of China,based on ground surface gravimetry显示文摘Variation of terrestrial water storage in the Hebei plain area from March 2010 to June 2014 was studied using ground gravimetry combined with vertical displacement data from the Global Navigation Satellite System.Results show that observed gravity variation in this area increased continuously,basically reflecting a trend toward land subsidence.With the effect of this subsidence removed,a dominantnegative change in gravity variation was evident,reflecting an average rate of decrease in terrestrial water in this area of 0.10±0.053 m/y,and this is equivalent to a volume of 81.5±43.2×108 m^(3)and is consistent with the spatial distribution of groundwater change from measured hydrologic data.These results can be an essential reference and supplement for the study of terrestrial water variation in the Hebei plain area,and indicate that ground surface gravimetry can be used as an important mean for studying changes in terrestrial water.Hongtao Hao Hongliang Liu Xinlin Zhang Jin Wei Bin Zhao Minzhang Hu 2021Geodesy and Geodynamics2021,12,3:1

20Novel Function of TNF Cytokines in Regulating Bone Marrow B Cell Survival显示文摘Two newly identified tumor necrosis factor (TNF) family cytokines, B cell activation factor from the TNF family (BAFF) and a proliferation-inducing ligand (APRIL), have recently been shown to enhance the maturation and survival of peripheral B cells. However, whether BAFF and APRIL are expressed in the bone marrow (BM) microenvironment and if these two cytokines modulate early B cell development remain unclear.In the present study, we have detected the abundant expression of BAFF and APRIL transcripts in BM non-lymphoid cells. Low levels of BAFF and APRIL mRNA are also found in developing B cells. Furthermore,we have determined the expression patterns of BAFF receptors during B lymphopoiesis. In cultures, both recombinant BAFF and APRIL significantly promote the survival of precursor B cells whereas only BAFF can suppress apoptosis of immature B cells. These findings suggest that BAFF and APRIL, in addition to their well established role in regulating peripheral B cell growth, can modulate the survival of developing B cells in the BM.
