1Transcriptome-Wide Identification and Validation of Reference Genes in Black Rockfish(Sebastes schlegelii)显示文摘The quantitative real-time reverse transcription PCR(qRT-PCR)is a widely used technique to analyze gene expression levels.Selecting a suitable reference gene is a crucial step to obtain an accurate result in qRT-PCR.However,most previous studies on fishes adopted reference genes that were commonly used in mammals without validation.In this study,we utilized 89 transcrip-tome datasets covering early developmental stages and different adult tissues,and carried out transcriptome-wide identification and validation of reference genes in Sebastes schlegelii.Finally,121 candidate reference genes were identified based on four criteria.Eight candidates(METAP2,BTF3L4,EIF5A1,TCTP,UBC,PAIRB,RAB10,and DLD)and four commonly used reference genes in mam-mals(TUBA,ACTB,GAPDH,RPL17)were selected for validation via qRT-PCR and four statistical analysis methods(delta-Ct,Best-Keeper,geNorm,and NormFinder).The results indicated that when the black rockfish are cultured in a general condition,the eight candidate reference genes are more stable than traditional reference genes in mammals,and RAB10,EIF5A1,PAIRB and BTF3L4 are the best reference genes in rockfish.This is the first study to conduct transcriptome-wide identification and validation of reference genes for quantitative RT-PCR in the black rockfish,and lay an important foundation for gene expression analysis in teleost.JIN Chaofan SONG Weihao WANG Mengya QI Jie ZHANG Quanqi HE Yan 2021Journal of Ocean University of China2021,20,3:1
2Large-Scale Screening of Growth-Related Variants in Chinese Tongue Sole(Cynoglossus semilaevis)显示文摘Growth is the most valuable economic trait for improving aquaculture fish species,since fast growth can reduce labor cost and make more economic benefits.However,the knowledge about how many and which genes are related to the growth of Chi-nese tongue sole is limited.High-throughput sequencing screening of variants is a fast,economical and accurate assay to identify genes related to growth in crops,livestock and other aquaculture products.In the present study,genome-wide resequencing of 30 female Chinese tongue sole individuals from large and small groups to screen growth trait-related variations(SNPs,InDels)was carried out.In total,6545735 SNPs and 1016745 InDels were detected,while 31 genes related to growth traits were identified.Their functions were mainly involved in muscle architecture,post-embryonic development,neurosensory development and hormone regulation.Fur-thermore,18 of the 31 genes related to growth trait are located on W chromosome,indicating that W chromosome plays an important role in regulating the body size of female tongue soles.The markers and genes identified in our study can be applied to selective breeding of tongue sole and thus will promote the aquaculture industry and increase economic efficiency.SONG Weihao ZHU He WANG Yujue ZHANG Kai ZHANG Quanqi HE Yan 2021Journal of Ocean University of China2021,20,3:1

5Inhibition of non-small-cell lung cancer cell proliferation by Pbx1显示文摘Lung cancer is one of the most deadly human cancers and continues to be a major unsolved health problem worldwide. Here, we evaluate the function of Pbx1 in the proliferation of non-smallcell lung cancer(NSCLC). In contrast with its known proliferative function, we found that Pbx1 inhibits the proliferation of lung cancer cells. In particular, Pbx1-specific RNA interference resulted in increased proliferation in lung cancer cells. In addition, histone H3 phosphorylation was also increased following inhibition of Pbx1 expression. In contrast, Pbx1 overexpression repressed the proliferation of lung cancer cells and inhibited DNA synthesis. 6New Neuraminidase Inhibitory Alkaloids from the Mangrove Soil-Derived Fungus Arthrinium sp.SCSIO 41305显示文摘New alkaloid,(E)-2-(hydroxyimino)-4-methylpentanamide(1)and a new cyclopentano[b]pyridine,4-hydroxy-7-methyl-6,7-dihydro-5H-cyclopenta[c]pyridin-5-one(2),together with ten known compounds(3–12)were isolated from the mangrove soil-derived fungus Arthrinium sp.SCSIO 41305.Extensive spectroscopic analysis and X-Ray crystallographic analysis were used to elucidate the structure of(E)-2-(hydroxyimino)-4-methylpentanamide(1),including its absolute configuration.All the isolated compounds(1–12)were evaluated for their antimicrobial and enzyme inhibitory activities against acetylcholinesterase(ACh E),neuraminidase(NAs),and phosphatidylinositol 3-kinase(PI3K).Among them,compounds 1 and 3 exhibited strong neuraminidase inhibitory activity with IC_(50)values of 12.04,1.92μmol L^(-1)(IC_(50)20μmol L^(-1)for oseltamivir acid),while compounds 5,6,8,and 10showed moderate neuraminidase inhibitory activity,and compounds 6–10 displayed weak enzyme inhibitory activities against PI3K.HU Yiwei ZHAO Kai ZHU Junheng QI Xin CHEN Weihao SONG Yingying PANG Xiaoyan LIU Yonghong WANG Junfeng 2023Journal of Ocean University of China2023,22,5:0
