| 1 | Flavonoid profiling of a traditional Chinese medicine formula of Huangqin Tang using high performance liquid chromatography显示文摘The quality control processes for herbal medicines have been problematic. Flavonoids are the major active components of Huangqin Tang(HQT, a traditional Chinese medicine formula). In this study, we used a combinative method approach consisting of chromatographic fingerprinting(high performance liquid chromatography; HPLC), quantitative methods and a pharmacodynamic evaluation model to analyze the flavonoids of HQT obtained from different sources. Ten batches of HQT were analyzed by the HPLC fingerprinting method and 26 common peaks were detected, of which 23 peaks corresponded with the chemical profile of HQT. In addition, 11 major compounds were identified by LC–MS analysis(liquid chromatography–tandem mass spectrometer; LC–MS^n) and quantified by the HPLC quantitative method approach. The studied10 batches of HQT were found to be homogeneous in their composition with a similarity between 0.990 and1.000. The distribution of the 11 identified compounds was found to be very similar among the batches. Only slight pharmacodynamic differences were detected between the different batches, confirming the homogeneity of HQT. | 3 | The Electric Field Detector (EFD) onboard the ZH-1 satellite and firstobservational results显示文摘Previous studies have reported that, before or after occurrences of strong earthquakes, some low earth orbit satellites recorded ionospheric disturbances, including electromagnetic emissions and plasma fluctuations over the epicenter region or its conjugate point.Theoretically speaking, due to some electromagnetic coupling effect, electromagnetic emissions from the earthquake preparation zone could propagate from the lithosphere to the atmosphere, and could reach the ionosphere, even up to the inner magnetosphere. This paper introduces the electric field detector(EFD) onboard the ZhangHeng-1 satellite(ZH-1). The EFD is designed to measure electric field fluctuations within the broad frequency range of DC to 3.5 MHz, divided into 4 channels: ULF(DC–16 Hz), ELF(6 Hz–2.2 kHz), VLF(1.8 kHz–20 kHz) and HF(18 kHz–3.5 MHz). The sampling rates of the channels are 125 Hz, 5 kHz, 50 kHz and 10 MHz, respectively. The EFD includes4 spherical probes mounted on a over 4.5 m boom and an electronic box inside the satellite module. The resolution of the EFD is 1μV·m-1·Hz-1/2 at frequencies from DC to 16 Hz, and the sensitivity is 0.1 μV·m-1·Hz-1/2 at frequencies from 6 Hz to 2.2 kHz, 0.05 μV·m-1·Hz-1/2 in the band 1.8 kHz to 20 kHz, and 0.1μV·m-1·Hz-1/2 from 20 kHz to 3.5 MHz. The dynamic range from DC to 20 kHz is over 120 dB, and over96 dB from 20 kHz to 3.5 MHz. The EFD has two observation modes: survey mode and burst mode. The survey mode concentrates primarily on electric field power density values; the burst mode provides high sampling rate waveform data. The detailed configuration of the EFD onboard the ZH-1 is also introduced in this paper. During the six months' orbit test phase, the EFD recorded a number of natural electromagnetic emissions. | 5 | State-of-the-Art Review of High-Throughput Statistical Spatial-Mapping Characterization Technology and Its Applications显示文摘Macroscopic materials are heterogeneous,multi-elementary,and complex.No material is homogeneous or isotropic at a certain small scale.Parts of the material that differ from one another can be termed'natural chips.'At different spots on the material,the composition,structure,and properties vary slightly,and the combination of these slight differences establishes the overall material performance.This article presents a state-of-the-art review of research and applications of high-throughput statistical spatialmapping characterization technology based on the intrinsic heterogeneity within materials.Highthroughput statistical spatial-mapping uses a series of rapid characterization techniques for analysis from the macroscopic to the microscopic scale.Datasets of composition,structure,and properties at each location are obtained rapidly for practical sample sizes.Accurate positional coordinate information and references to a point-to-point correspondence are used to set up a database that contains spatialmapping lattices.Based on material research and development design requirements,dataset spatialmapping within required target intervals is selected from the database.Statistical analysis can be used to select a suitable design that better meets the targeted requirements.After repeated verification,genetic units that reflect the material properties are determined.By optimizing process parameters,the assembly of these genetic unit(s)is verified at the mesoscale,and quantitative correlations are established between the microscale,mesoscale,macroscale,practical sample,across-the-scale span composition,structure,and properties.The high-throughput statistical spatial-mapping characterization technology has been applied to numerous material systems,such as steels,superalloys,galvanization,and ferrosilicon alloys.This approach has guided the composition and the process optimization of various materials. | Haizhou Wang Lei Zhao Yunhai Jia Dongling Li Lixi Yang Yuhua Lu Guang Feng Weihao Wan | 2020 | Engineering2020,6,6: | 7 |
| 6 | The transcription factor SlHY5 regulates the ripening of tomato fruit at both the transcriptional and translational levels显示文摘Light plays a critical role in plant growth and development,but the mechanisms through which light regulates fruit ripening and nutritional quality in horticultural crops remain largely unknown.Here,we found that ELONGATED HYPOCOTYL 5(HY5),a master regulator in the light signaling pathway,is required for normal fruit ripening in tomato(Solanum lycopersicum).Loss of function of tomato HY5(SlHY5)impairs pigment accumulation and ethylene biosynthesis.Transcriptome profiling identified 2948 differentially expressed genes,which included 1424 downregulated and 1524 upregulated genes,in the Slhy5 mutants.In addition,genes involved in carotenoid and anthocyanin biosynthesis and ethylene signaling were revealed as direct targets of SlHY5 by chromatin immunoprecipitation.Surprisingly,the expression of a large proportion of genes encoding ribosomal proteins was downregulated in the Slhy5 mutants,and this downregulation pattern was accompanied by a decrease in the abundance of ribosomal proteins.Further analysis demonstrated that SlHY5 affected the translation efficiency of numerous ripening-related genes.These data indicate that SlHY5 regulates fruit ripening both at the transcriptional level by targeting specific molecular pathways and at the translational level by affecting the protein translation machinery.Our findings unravel the regulatory mechanisms of SlHY5 in controlling fruit ripening and nutritional quality and uncover the multifaceted regulation of gene expression by transcription factors. | Weihao Wang Peiwen Wang Xiaojing Li Yuying Wang Shiping Tian Guozheng Qin | 2021 | Horticulture Research2021,8,1: | 6 |
| 7 | Asymmetric distribution of cytokinins determines root hydrotropism in Arabidopsis thaliana显示文摘The phenomenon of plant root tips sensing moisture gradient in soil and growing towards higher water potential is designated as root hydrotropism,which is critical for plants to survive when water is a limited factor.Molecular mechanisms regulating such a fundamental process,however,are largely unknown.Here we report our identification that cytokinins are key signaling molecules directing root growth orientation in a hydrostimulation(moisture gradient)condition.Lower water potential side of the root tip shows more cytokinin response relative to the higher water potential side.Consequently,two cytokinin downstream type-A response regulators,ARR16 and ARR17,were found to be up-regulated at the lower water potential side,causing increased cell division in the meristem zone,which allows the root to bend towards higher water potential side.Genetic analyses indicated that various cytokinin biosynthesis and signaling mutants,including the arr16 arr17 double mutant,are significantly less responsive to hydrostimulation.Consistently,treatments with chemical inhibitors interfering with either cytokinin biosynthesis or cell division completely abolished root hydrotropic response.Asymmetrically induced expression of ARR16 or ARR17 effectively led to root bending in both wild-type and miz1,a previously known hydrotropism-defective mutant.These data demonstrate that asymmetric cytokinin distribution is a primary determinant governing root hydrotropism. | Jinke Chang Xiaopeng Li Weihao Fu Jiawen Wang Yueyuan Yong Hongyong Shi Zhaojun Ding Hong Kui Xiaoping Gou Kai He Jia Li | 2019 | Cell Research2019,29,12: | 4 |
| 8 | Air pollution and inhalation exposure to particulate matter of different sizes in rural households using improved stoves in central China显示文摘Household air pollution is considered to be among the top environmental risks in China.To examine the performance of improved stoves for reduction of indoor particulate matter(PM) emission and exposure in rural households, individual inhalation exposure to size-resolved PM was investigated using personal portable samplers carried by residents using wood gasifier stoves or improved coal stoves in a rural county in Central China.Concentrations of PM with different sizes in stationary indoor and outdoor air were also monitored at paired sites. The stationary concentrations of size-resolved PM in indoor air were greater than those in outdoor air, especially finer particles PM0.25. The daily averaged exposure concentrations of PM0.25, PM1.0, PM2.5 and total suspended particle for all the surveyed residents were 74.4 ± 41.1, 159.3 ± 74.3, 176.7 ± 78.1 and 217.9 ± 78.1 μg/m3,respectively. Even using the improved stoves, the individual exposure to indoor PM far exceeded the air quality guideline by WHO at 25 μg/m3. Submicron particles PM1.0 were the dominant PM fraction for personal exposure and indoor and outdoor air. Personal exposure exhibited a closer correlation with indoor PM concentrations than that for outdoor concentrations. Both inhalation exposure and indoor air PM concentrations in the rural households with gasifier firewood stoves were evidently lower than the reported results using traditional firewood stoves. | 11 | Microstructure and mechanical properties of hot isostatically pressed cermets with TiN coatings显示文摘To increase the adhesion strength between the coating and the substrate, sintered Ti(C,N)-based cermets were se- lected and deposited with monolayer TiN using a multiarc ion-plating technique; subsequently, hot isostatic pressing (HIP- ing) treatment was performed at 1000°C using nitrogen pressure up to 110 MPa. The mechanical properties of cermets after a coating process and subsequent HIPing treatment have been evaluated with respect to the hardness, the residual stress, and the coating adhesion. The results show that after the HIPing process, there was a higher increase in critical load in the TiN-coated cermets with lower surface roughness compared with those with higher surface roughness. In all cases, the re- sidual stress was found to be compressive. The effects of substrate surface roughness and posttreatment on the adhesion strength of the coatings were thus investigated. | 14 | ATP11B deficiency leads to impairment of hippocampal synaptic plasticity显示文摘Synaptic plasticity is known to regulate and support signal transduction between neurons, while synaptic dysfunction contributes to multiple neurological and other brain disorders;however, the specific mechanism underlying this process remains unclear. In the present study, abnormal neural and dendritic morphology was observed in the hippocampus following knockout of Atpllb both in vitro and in vivo. Moreover, ATP11B modified synaptic ultrastructure and promoted spine remodeling via the asymmetrical distribution of phosphatidylserine and enhancement of glutamate release, glutamate receptor expression, and intracellular Ca^2+ concentration. Fuithermoe experimental results also indicate that ATP11B regulated synaptic plasticity in hippocampal neurons through the MAPK14 signaling pathway. | 16 | Safety and efficacy of a modified XELOX adjuvant regimen for patients with operated stage III colon cancer:a Chinese single-center experience显示文摘Background:A fixed 8-cycle oxaliplatin and capecitabine(XELOX)regimen has been the standard adjuvant therapy for patients with stage III colon cancer.However,completing the full-cycle of oxaliplatin is often associated with severe neurotoxicity.To spare patients from the toxic effects,without comprising the required efficacy,we evaluated the safety and efficacy of a modified XELOX(mXELOX)adjuvant chemotherapy regimen with 6 cycles of oxaliplatin and a full cycle of capecitabine.Methods:We retrospectively analyzed 330 eligible patients with stage III colon cancer who underwent cura-tive tumor resection followed by mXELOX,standard XELOX or unfinished XELOX adjuvant chemotherapy between December 2007 and April 2015.Associated prognostic factors were investigated and their disease-free survival(DFS)and overall survival(OS)rates were also determined and compared among the different regimen groups.Results:Compared with the standard XELOX group,the mXELOX group had lower total incidence rates of neuro-toxicity(39.3%vs.76.2%,P<0.001),leucopenia(53.6%vs.69.8%,P=0.017)and thrombocytopenia(38.1%vs.56.3%,P=0.011).The standard XELOX and mXELOX adjuvant chemotherapy regimens presented with comparable 3-year DFS rates(86.3%vs.89.2%;P=0.838)and 3-year OS rates(92.7%vs.97.6%;P=0.227).Compared to unfinished XELOX chemotherapy,the oncologic benefits of the mXELOX regimen were greater for patients with T4 tumors(3-year DFS:Hazard ratio[HR],2.184;95%confidence interval[CI],1.051-4.540;P=0.036;3-year OS:HR,4.529;95%CI 1.245-16.479;P=0.022)and for high-risk patients(3-year DFS:HR,1.962;95%CI 0.964-3.993;P=0.044;3-year OS:HR,4.193;95%CI 1.182-14.874;P=0.026).Conclusions:The mXELOX adjuvant chemotherapy presented a comparable survival benefit and lower incidence of toxicity than standard XELOX chemotherapy.It could be an alternative treatment for high-risk patients with operated stage III colon cancer. | Jianhong Peng Weihao Li Rongxin Zhang Junzhong Lin Jinghua Tang Yongshan Wen Zhenhai Lu Xiaojun Wu Zhizhong Pan | 2019 | Cancer Communications2019,39,1: | 2 |
