| 4 | Characterization of a novel regulatory pathway for mannitol metabolism and its coordination with biofilm formation in Mycobacterium smegmatis显示文摘Biofilm formation has been implicated to be tightly regulated in bacteria. Mycobacterial species possess a unique cell-wall structure; however, the underlying regulation mechanism for their biofilm formation remains largely unclear. In this study, we characterized a hypothetical mannitol metabolism and transportation gene cluster(Ms5571-Ms5576), designated as mmt operon, whose expression significantly contributes to the biofilm formation in Mycobacterium smegmatis. We showed that in the operon the Ms5575 gene encodes a GntR-like transcriptional repressor and the Ms5576 gene encodes a mannitol2-dehydrogenase which can produce D-mannitol from D-mannose. Strikingly, the D-mannitol molecule can derepress the negative regulation of Ms5575 on the mmt operon to stimulate the operon's expression. Consistently, addition of D-mannitol into the medium can obviously induce mycobacterial biofilm formation. Furthermore, we found that Ms0179 positively regulates the mmt operon through its downstream regulator Ms0180. Ms0180 directly binds the mmt operon to positively regulate its expression. Both Ms0179 and Ms0180 significantly affect the mycobacterial biofilm formation. Taken together, we explored a regulatory pathway for the mannitol metabolism and its coordination with the biofilm formation in M. smegmatis. | 5 | Inhibition of cystathionine β-synthase promotes apoptosis and reduces cell proliferation in chronic myeloid leukemia显示文摘Increased endogenous hydrogen sulfide(H_(2)S)level by cystathionine β-synthase(CBS)has been shown to closely relate tumorigenesis.H_(2)S promotes angiogenesis,stimulates bioenergy metabolism and inhibits selective phosphatases.However,the role of CBS and H_(2)S in chronic myeloid leukemia(CML)remains elusive.In this study,we found that CBS and H_(2)S levels were increased in the bone marrow mononuclear cells of pediatric CML patients,as well as in the CML-derived K562 cells and CBS expression levels were correlated with different disease phases.Inhibition of CBS reduced the proliferation of the CML primary bone marrow mononuclear cells and induced growth inhibition,apoptosis,cell cycle arrest,and migration suppression in K562 cells and tumor xenografts.The knockdown of CBS expression by shRNA and inhibiting CBS activity by AOAA decreased the endogenous H_(2)S levels,promoted mitochondrial-related apoptosis and inhibited the NF-xB-mediated gene expression.Our study suggests that inhibition of CBS induces cell apoptosis,as well as limits cell proliferation and migration,a potential target for the treatment of chronic myeloid leukemia. | Dan Wang Huan Yang Yun Zhang Rong Hu Dongjie Hu Qunxian Wang Yannan Liu Mingjing Liu Zijun Meng Weihui Zhou Weihong Song | 2021 | Signal Transduction and Targeted Therapy2021,6,3: | 0 |

| 7 | COM33 suppresses carboplatin-induced epithelial-mesenchymal transition via inhibition of Twist1 in ovarian cancer显示文摘Despite favorable responses to platinum-based chemotherapy in ovarian cancer(OC),chemoresistance is still a major cause of treatment failure.Hence,we develop a novel synthetic agent,COM33,to relieve the chemoresistance caused by carboplatin.The anti-cancerous effects of the combination of COM33 and carboplatin on OC are evaluated by cell viability,wound healing,and transwell invasion assays.A mechanistic investigation is carried out by using RNA-Seq analysis and then verified by western blot analysis and immunofluorescence microscopy.The safety and efficacy in vivo are evaluated using SKOV3 tumor-bearing nude mice.Results show that the co-administration of COM33 enhances the inhibitory effects of carboplatin on cancer cell viability,migration,and invasion in vitro and tumor growth in vivo.Furthermore,COM33 suppresses the carboplatin-induced epithelialmesenchymal transition(EMT)by inhibiting the ERK signaling pathway.Additionally,we show that Twist1,the effector of the ERK signaling pathway,participates in carboplatin-induced EMT and is also inhibited by COM33.Our data show that the combination of carboplatin with COM33 is beneficial for chemotherapy against OC,which may be a potential novel anti-tumor strategy. | Zhiyang Zhou Li Jin Jian Shen Weihui Shi Yue Xu Longyun Ye Junxi Liu Jiexue Pan | 2023 | Acta Biochimica et Biophysica Sinica2023,55,1: | 0 |
